1.Effect of Geraniin on Expression of Wnt3a in BMSC from Osteoporotic Rats

Objective To investigate the effect of geraniin on expression of Wnt3a protein and mRNA in bone marrow stromal cell (BMSC) from osteoporotic rats. Methods The model of osteoporosis (OP) was duplicated by ovariectomy in rats. BMSCs were isolated and cultured. BMSCs from shamed rats were routinly cultured and taken as normal control, and BMSCs from OP rats were divided into model group, 1μmol/L simvastatin positive group, and geraniin group (0.01, 0.1, 1, 10, 100 μmol/L), respectively. The methods of realtime-PCR and western blot were used to assay the protein and mRNA expression of wnt3a, respectively.Results As compared with normal control group, the protein and mRNA expression of wnt3a in model group were significantly suppressed;Compared with model group, 1 μmol/L simvastatin, and 0.1, 1, 10 and 100 μmol/L geraniin significantly increased the expression of wnt3a protein and mRNA. Conclusion It is suggested that geraniin activates wnt/β-catenin pathway though increasing the expression of signaling protein wnt 3a in BMSCs from OP rats. It would be beneficial to osteogenic differentiation of BMSCs and osteogenesis.

Chinese medicine talents are the foundation and guarantee for the development of TCM, and the first resource for the inheritance and innovation of traditional Chinese medicine. Reforming the curriculum system and training mode of Chinese medicine, training Chinese medicine professional who are suitable for research and development of Chinese medicine, such as cultivation of Chinese medicine materials, quality detection, species identification and resource census, is the direction of further expansion and reformation of Chinese medicine education. In this article, on account of the characteristic of Chinese medicine professional courses, we will combine practice education mode with professional knowledge competition, to probe innovative mode of comprehensive ability training for Chinese medicine based on the combination of"Multi-mode and Multi-Ways". The new mode of Chinese medicine education is to provide reference for trainers of Chinese medicine.
3.Exploration of creating a Chinese materia medica teaching system based on the Qinling plant gene database

Qinling is a treasury of medicine with abundant resources of herbs. Due to the goal of enriching teaching contents, quality of teaching and creating a Chinese materia medica teaching system. Based on the Qinling plant gene database, our education team presents a series of innovative thoughts, such as multidisciplinary contact, inheriting and carrying forward the folk medicine, attaching importance to the practical application of herbs in Qinling and theory with practice to enrich teaching contentsand creating Shanxi characteristic teaching system of Chinese materia madica.That makes a contribution to raise the level of teaching in modern traditional Chinese medicine and promote the development of medicine in Shannxi.
4.Study on the techniques of purification flavones from Gnaphalium japonicum Thunb. by D-101 macroporous adsorption resin

Objective To investigate the optimal isolation and purification conditions of flavonoids in Gnaphlium affine Thunb. and the content of flavonoids in different parts of Gnaphlium affine Thunb. was measured. Methods The separation and purification abilities of D-101 macroporoue adsorbing resins for flavonoids in Gnaphlium affine Thunb. were studied with adorption and desorption as index. The static adsorption and dynamic adsorption methods were used to analyze the effects of static saturated adsorption, static elution rate, sample concentration, sample pH value, eluent concentration and amount of eluent. The flavonoids concentrations were determined with rutin as standard. Results The D-101 macroporous adsorption resin had good effect on the separation and purification of flavonoids from Gnaphlium affine Thunb.; The optimal conditions for purification were: sample concentration 1.0 mg/ml, sample pH=4, 60% ethanol as desorption solvent, washing flow 2 BV/h, with these parameters. With such condition, the purity of flavonoids in Gnaphlium affine Thunb. was 60.92%; The content of flavonoids in Gnaphlium affine Thunb. showed the highest content was in the stem, the second in the flower, and the least in the root. Conclusions The purification of flavonoids from the D-101 macroporous adsorption resin increased in the best purified separation conditions and the content of flavonoids was the highest in the leaf.
5.Measurements of glomerular filtration rate with iohexol plasma clearance: method selection and evaluation

Objective To compare the consistency between single-(I-GFR-SS) and dual-(I-GFR-DS) sample methods with three-sample method (I-GFR-TS) of iohexol plasma clearance in chronic kidney disease (CKD) patients for choosing the optimizing project of glomerular filtration rate (GFR) measurement.Methods The multiple-sample methods were performed in 174 patients with CKD admitted to the Department of Nephrology,Shanghai Ruijin Hospital from August 2017 to July 2018.Plasma concentrations of iohexol were measured three times at different time points after receiving 5 ml iohexol (300 g/L) intravenous injection,according to estimated GFR (eGFR) grouping.The first blood sample was collected at 2 hours,and the time for the last sample was delayed from 4 hours to 6 hours with reduction of eGFR.The synchronized Gates (99mTc-Gates-GFR) method was detected as control.With I-GFR-TS as the golden standard,the accuracies of I-GFR-DS,I-GFR-SS and 99mTc-Gates-GFR were compared.Results The median differences of I-GFR-DS,I-GFR-SS and 99mTc-Gates-GFR in overall patients were-0.15,-1.00,6.76 ml· min-1· (1.73 m2)-1 comparing with I-GFR-TS;P10(percentage of the GFRmeasurements that was within 10% of the standard method) were 95.4%,74.1%,28.7%,and P30 were 100%,93.7%,72.4% separately.In the patients with eGFR < 30 ml· min-1· (1.73 m2)-1,I-GFR-SS was more accurate when last point collecting extended to 6 h from 4 h [P10:43.5% vs 17.4%,P=0.055;P30:73.9% vs 43.5%,P<0.05].Conclusions The dual-sample plasma clearance of iohexol is recommended in clinical practice,and the single-sample method can be a secondary option because of its slightly poor accuracy but more convenient.Sample-collection protocol should be adjusted according to eGFR especially in moderate-to-severe CKD patients.The Gates method is not recommended.
6.Achievements, main experiences, challenges and suggestions on control of endemic diseases in China in the last decade

Since the 18th National Congress of the Communist Party of China (CPC), the CPC Central Committee and the State Council have paid great attention to the prevention and control of endemic diseases. Through the implementation of the prevention and control measures of endemic diseases, especially the implementation of the "Six Major Actions" to tackle key problems in "three years' project", all kinds of diseases have met the relevant requirements. The prevention and control of endemic diseases in China has achieved remarkable results and made historic achievements. However, risk challenges remain. At present, there are still some problems in the management and treatment of patients with iodine deficiency disorders, water-borne iodine excess goiter, drinking water-borne endemic fluorosis and drinking tea-borne endemic fluorosis in China, which need to be solved by strengthening the prevention and control efforts.
7.HSDL2 overexpression promotes rectal cancer progression by regulating cancer cell cycle and promoting cell proliferation.

OBJECTIVE:
To analyze the expression of hydroxysteroid dehydrogenase like 2 (HSDL2) in rectal cancer tissues and the effect of changes in HSDL2 expression level on proliferation of rectal cancer cells.
METHODS:
Clinical data and tissue samples of 90 patients with rectal cancer admitted to our hospital from January 2020 to June 2022 were collected from the prospective clinical database and biological specimen database. The expression level of HSDL2 in rectal cancer and adjacent tissues was detected by immunohistochemistry, and based on the median level of HSDL2 expression, the patients were divided into high expression group (n=45) and low expression group (n=45) for analysis the correlation between HSDL2 expression level and the clinicopathological parameters. GO and KEGG enrichment analyses were performed to explore the role of HSDL2 in rectal cancer progression. The effects of changes in HSDL2 expression levels on rectal cancer cell proliferation, cell cycle and protein expressions were investigated in SW480 cells with lentivirus-mediated HSDL2 silencing or HSDL2 overexpression using CCK-8 assay, flow cytometry and Western blotting.
RESULTS:
The expressions of HSDL2 and Ki67 were significantly higher in rectal cancer tissues than in the adjacent tissues (P < 0.05). Spearman correlation analysis showed that the expression of HSDL2 protein was positively correlated with Ki67, CEA and CA19-9 expressions (P < 0.01). The rectal cancer patients with high HSDL2 expressions had significantly higher likelihood of having CEA ≥5 μg/L, CA19-9 ≥37 kU/L, T3-4 stage, and N2-3 stage than those with a low HSDL2 expression (P < 0.05). GO and KEGG analysis showed that HSDL2 was mainly enriched in DNA replication and cell cycle. In SW480 cells, HSDL2 overexpression significantly promoted cell proliferation, increased cell percentage in S phase, and enhanced the expression levels of CDK6 and cyclinD1 (P < 0.05), and HSDL2 silencing produced the opposite effects (P < 0.05).
CONCLUSION
The high expression of HSDL2 in rectal cancer participates in malignant progression of the tumor by promoting the proliferation and cell cycle progress of the cancer cells.
