Leptin,a proteohormone primarily made by adipocytes,directly interacts with the hypothalamus for energy balance regulation.It plays an important role in the morbility of obesity.The soluble leptin receptor(sOb-R) is one of the leptin receptor isoforms.The examination of the sOb-R concentration is valuable because of its function in the regulation of leptin concentration.Leptin shows a great potential as a childhood obesity biomarker,and its concentration could be used in assessing the responsiveness of obese children to intervention programs.
2.Treatment for locally advanced pancreatic cancer

Combined therapy consisting of radiotherapy,chemotherapy and palliative surgery is the primary management for locally advanced pancreatic cancer,with the purpose of prolonging survival and improving life quality.Gemcitabine is the standard chemotherapy at present.Gemcitabine-based combinations show a definite effect,and the combination with targeted drugs also has a certain degree of efficacy.The development of new chemotherapy drugs and targeted drugs provides more ways for the teatment of locally advanced pancreatic cancer.

EML4-ALK fusion oncogene represents a new molecular target which appears mainly in lung adenocarcinoma.EML4-ALK is detected more frequently in young non-small-cell lung cancer patients who never or light smoke.ALK inhibitors(crizotinib)for the treatment of the EML4-ALK-positive non-small cell lung cancer has a high response rate.This finding can improve the individual treatment of non-small cell lung cancer.

5.The effect of aminoguanidine intraperitoneal injection on apoptosis in chondrocyte with osteoarthritis

Objective To observe the effect of aminoguanidine by intraperitoneal injection on apoptosis of chondrocyte with osteoarthritis.Methods Totally 32 rabbits were divided into 3 groups at random.Both the experiment and the model groups were fixed by plaster cast in the right hind straighten limb,while the control group received no treatment.The experiment groups were injected with aminoguanidine in abdominal cavity,100mg/(kg?d),and the control groups were injected the same voluminal normal saline in abdominal cavity.At the 4th week,we killed 4 rabbits from the normal group,and 6 from the control group and the experiment group,respectively.At the 8th week,we put the rest to death.We drew the materials from each sample for pathological observation on the whole and under the light microscope by HE and immunohistochemical staining,and counted apoptosis cells by TUNEL for statistical analysis.Results Compared with the control group,the experiment group was more similar to the normal group in general observation and optical microscope observation by HE staining.Both iNOS positive expression cell by immunohistochemical staining and apoptosis cell calculation by TUNEL of the experiment group were fewer than those of the control group(P
6.Expression and biological significance of COX-2, PCNA, CD44v6 protein in different procancerous lesion and esophageal squamous cell carcinoma tissue

Objective To examine the expression of COX-2, PCNA, CD44v6 protein in different procancerous lesion and esophageal squamous cell carcinoma tissue, and to investigate the relationship between their expression and the biological level,lymph node metastasis in esophageal squamous cell carcinoma tissue. Methods Immunohistochemistry was demonstrated to show these tumor markers expression in different procancerous lesions and esophageal squamous cell carcinoma tissue. Results Stepwise over-expression of COX-2, CD44v6, PCNA protein were observed in the multi-step process of esophageal tissue. Significant difference in expression of these tumor markers(P <0.05) was observed between normal esophageal epithelia and dysplasia. The expression of COX-2 (P <0.05) had statistical different in biological level and lymph node metastasis.The expression of CD44v6 (P <0.05)had statistical different in lymph node metastasis. The relationship between COX-2 and CD44v6 was statistical significant in esophageal squamous cell carcinoma tissue(P <0.01). Conclusion Stepwise over-expression of COX-2,CD44v6 PCNA protein have relation with esophageal squamous cell carcinoma. The overexpression of COX-2 and CD44v6 have effect on prognosis of esophageal squamous cell carcinoma. The expression of COX-2 have relationship with that of CD44v6 in esophageal squamous cell carcinoma.
7.Determination of the Related Substances in Nortriptyline Hydrochloride by HPLC

Objective: To establish a determination method for the related substances in nortriptyline hydrochloride by HPLC. Methods:An Agilent C18 (250 mm × 4. 6 mm,5 μm) column was adopted with a mobile phase consisting of acetonitrile-0. 2% trieth-ylamine(pH 3. 0)(34 ∶66). The flow rate was 1. 0 ml·min-1, the detection wavelength was set at 210 nm, the column temperature was 40℃ and the injection volume was 20 μl. Results:The resolutions of the related substances were acceptable. The detection limit and the quantitation limit of the samples was 0. 40 ng and 1. 35 ng, respectively. Conclusion: The method is sensitive, accurate and specific, and can be used to determine the related substances in nortriptyline hydrochloride.
8.Suppression Effect of Di-n-butyl Phthalate in vitro on Superoxide Dismutase of Various Organs of Mice

Objective The present study was conducted to study the oxidation damage effects of DBP on organs of mice and the molecular mechanisms of the effects. Methods The cells of the organs (liver,kidney,testis,lung) were exposed to different concentration of DBP (0?mol/L,5?mol/L,20?mol/L,80?mol/L) for one hour in vitro,and then the superoxide dismutase (SOD) activities of those cells of different organs were measured. Results With statistical significance,the SOD activities in liver,kidney,testis,lung cells were increased gradually with the increase of DBP concentrations. Conclusion The results suggested that DBP could induce the oxidative damage in liver,lung,kidney and testis cells of mice.
9.Ultrasound radiofrequency-data technique assessment of carotid: a reproducibility study

Objective To evaluate the reproducibility of quality intima-media thickness(QIMT) and quality arterial stiffness (QAS) technique in assessment of carotid under different measuring methods.Methods Between December 2012 and January 2014,carotid QIMT and QAS examinations were carried out in 30 health volunteers.QIMT and QAS indicators included IMT in QIMT and distensibility coefficient (DC),compliance coefficient(CC),stiffness index α(α),stiffness index β(β),pulse wave velocity(PWV) in QAS.The measurement employed unilateral/once,bilateral/twice,and unilateral/twice methods.Using intra observer and inter-observer variability,the reproducibility was compared between different QIMT and QAS indicators and measuring methods.Results Extremely high level of intra-observer reproducibility was found for both QIMT and QAS technique (ICC>0.8).QIMT also showed an excellent inter observer reproducibility (ICC>0.8).In contrast,the reproducibility of QAS technique varied in different indicators (PWV > β ≈ α > CC > DC) and method ( unilateral/once > bilateral/twice > unilateral/twice).Conclusions QIMT measurement was highly reproducible.Whereas the reproducibility of QAS technique varied in different indicators and methods.Due to low reproducibility,the study result did not support the clinical application of DC indicator and unilateral/once method.
10.Preparation and performance of LID-MWCNT based sustained release targeted drug delivery system

Objective To prepare a targeted antitumor drug delivery system using large-inner-diameter multi-walled carbon nanotubes (LID-MWCNTs) for sustained release and to study its performance. Methods LID-MWCNTs were puri?fied and oxidized,then use nanocarriers and USTs as homologous blockers. Folic acid and fluorescent labels were conjugat?ed onto the external surfaces of nanocarriers. CDDP (cisplatin) was encapsulated and ultrashort tubes (USTs) were employed to block the drug entry/exit paths. The microstructure of resulted drug delivery system (DDS) was observed, while drug load?ing efficiency and drug release profile in vitro were determined. The tumor-targeting property and cytotoxicity of DDS were also assessed. Results LID-MWCNT based sustained release targeted drug delivery system was established. Drug loading efficiency of CDDP@UST-FA-LID-MWCNTs was as high as 70.97%. A typical biphasic sustained release pattern was dem?onstrated, and the accumulating release time was 18 h. DDS exhibited a certain kind of tumor-targeting property, and inhibit?ed proliferation of tumor cells in a dose-dependent manner. Conclusion CDDP@UST-FA-LID-MWCNT drug delivery system exhibited an improved drug loading efficiency and a sustained drug release profile. It could specifically target the tu?mor cells and had a significant antitumor effect.
