BACKGROUND: Previous studies demonstrated that freeze-drying had great influence on internal heat transfer, such as pore distribution and void connectivity, however, the mechanisms involved in heat and mass transfer during drying is poorly understood.OBJECTIVE: Based on the previous studies, micro CT was served as analytical tool, to observe the movement characteristics of sublimation interface in freeze-drying process.METHODS: With aorta of pig, micro-CT scanning, image reconstruction and the technique of grey value analysis were used to observe and analyze the sublimation process in freeze-drying. RESULTS AND CONCLUSION: The slope of freeze-drying temperature curve was large at first, causing quick sublimation rate. The grey value of freeze-drying and ice crystal area was very different in second-dimensional cross-sectional reconstructed image.Sublimation interface was clear and sublimation took place simultaneously both in the outer'surface and inner surface. The movement of sublimation interface was obvious at this time. Along with the time, the slope of freeze-drying temperature curve became small, leading to slow sublimation rate. The movement of sublimation interface turned unciearly. For the vertically placed artery, when heated at the bottom, the sublimation takes place both in the outer surface and inner surfaceat the same time. The sublimation interface slowly approaches to the middle of vessel wall. With time prolonged, resistance for heat and mass transfer in freeze-drying is increased, and the sublimation rate becomes slowly.

Objective To investigate the current status of nursing undergraduate students′ self-differentiation and career adaptability and analyze the correlation of the two, so as to provide a reference for nursing educators to offer employment guidance. Methods The Undergraduate Self-Differentiation Scale and The College Students′Career Adaptability Questionnaire were used to investigate 341 nursing undergraduates. Results The mean scores of the self-differentiation and the career adaptability were (3.70±0.60) points and (3.60±0.42) points, respectively. It showed that the total score of self-differentiation and its each dimension were positively correlated with career adaptability, the Pearson correlation coefficients were career adjustment 0.26, career interpersonal 0.38, career curiosity 0.32, career confidence 0.39, career concerns 0.27, career control 0.46, and the coefficients between two total scales were 0.46 (all P<0.05). Conclusions The self-differentiation level of nursing undergraduates could affect their career adaptation level, and clinical nursing educators can take corresponding measures to improve their career adaptability based on the characteristics of nursing students′self-differentiation.

Aim By establishing mouse acute myocardial infarction model,to observe the protection of isoflavones on ischemic myocardium and research the mechanism. Methods Mouse acute myocardial infarction model was established by ligating the left anterior descending(LAD)coronary artery.Danshen was used as the positive control. The effect of isoflavones on myocardial infarct area, serum myocardium creatase and serum levels of SOD and MDA was observed.By Real Time PCR,it was found that isoflavones could affect the expression of β-adrenergic receptor kinase(β-ARK_1).Results Isflovones could obviously reduce the myocardial infarct area and lower the levels of serum myocardial creatase and MDA.It could downregulate the expression of β-ARK_1 as the doses are increased. Conclusions Isoflavones can protect the myocardium of acute myocardial infarction mouse. The mechanism is related to the reduction of the oxidative damage, and the downregulation of the expression of β-ARK_1.

BACKGROUND: The freeze-drying blood vessel is an excellent material for vasotransplantation. However, the reports concerning freeze-drying are few, and the mechanical property changes of freeze-dried blood vessel remains poorly understood. OBJECTIVE: Using micro-CT to explore the morphological changes of blood vessel during procedure of freeze-drying. DESIGN, TIME AND SETTING: A single sample observation was performed at the Institute of Refrigeration and Cryogenics, University of Shanghai for Science and Technology. MATERIALS: Fresh pig aorta was obtained from Shanghai Slaughter House. The lyophilizer was produced by SP Industries (USA). TRAPEZIUM LITE texture analyzer was purchased from Shimadzu (Hong Kong) Co., Ltd.METHODS: The differences of mechanical property between fresh aorta and freeze-dried aorta were compared.MAIN OUTCOME MEASURES: ① Temperature curve of freeze-drying procedure. ②The reeze-drying procedure was scanned by micro-CT. ③ Changes of mechanical property were detected by TRAPEZIUM LITE texture analyzer and puncture experiments.RESULTS: During freeze-drying procedure, there were presented layer phenomenon in pig aorta. Compared to the fresh aorta, the freeze-dried aorta after recover water would reduce 40% axial tensile force, increase 45% circumferential tensile force, with 75% puncture stress.CONCLUSION: The freeze-dried blood vessel maintains mechanical property as fresh vessel, thus, freeze-drying can be used for vascular preservation.

Objective To study the drug dosage and time dependency characteristics of high-fructose-high-fat-feeding plus dexamethason for inducing the mouse acute fatty liver model and to optimize the condition of drug induced fatty liver model. Methods Male KM mice were divided into the normal control group and high-fructose-high-fat-feeding plus peritoneal injection of dexamethason group. The mice were killed at 3 different time points. The mouse body mass and liver mass were detected. The liver index was calculated. The serum and liver tissue homogenate TG and serum glucose(GLU) levels were detected. The liver tissue pathological change was observed by HE staining.Total RNA reverse expression related gene was extracted from the liver tissue. The total protein was extracted from the liver tissue and the related protein expression was detected by Western Blot. Results Compared with the control group, blood and liver homogenate TG after 7 d in the dexamethason model group was increased, the liver index was increased, the pathological section displayed that the fatty liver was formed.RT-PCR showed that lipid metabolism related gene expression had obvious change.Western Blot showed that SIRT1 was significantly decreased. But with the dexamethason dosage decrease and time extending, the fatty liver related indexes were decreased, lipid metabolic gene PPAR,FOXO3 and FXR were gradually increased, while LXR was gradually decreased and protein SIRT1 was gradually increased. Conclusion High-fructose-high-fatfeeding plus peritoneal injection of dexamethason could establish the mouse acute fatty liver model, moreover the model maintenance has dependency on dexamethason dosage and medication time, which has a guidance significance for the drug interventional experiment.
