1.TCM-WM treatment of reflux esophagitis in 60 cases

Objective To observe treatment efficacy of chinese and western medicine treatment for reflux esophagitis. Methods 60 cases with reflux esophagitis were divided into treatment group and control group,treated for 1 month. Results Treatment group the total effective rate was 96. 67%, control group, the total effective rate was 86. 67%. Treatment group and control group treatment results were significantly different( P < 0. 05 ), the difference has statistical significance. The results showed that the treatment group was significantly better than the control group. Conclusion TCM-WM treatment of reflux esophagitis, symptoms could be markedly improved, worthy of further prmotion.

3.The prognosis effect of different Helicobacter pylori eradication methods in patients with chronic gastritis

Objective To compare the prognosis effect of different Helicobacter pylori (Hp)eradication methods in patients with chronic gastritis.Methods One hundred and twenty patients with chronic gastritis diagnosed by gastroscopy and pathology examination were divided into sequential therapy group (36 cases),triple-combined therapy group (34 cases),Hp positive without eradication group (30cases) and Hp negative group (20 cases) according to the Hp infection status.All patients were followed up by gastroscopy,biopsy and rapid urease test before and after therapy.Results The Hp eradication rate in sequential therapy group was 94.44% (34/36),in triple-combined therapy group was 73.53% (25/34),and there was statistical difference (x2 =5.775,P =0.016).The symptoms scores and gastroscopy scores after therapy in the four groups were significantly lower than those before therapy,and there were statistical differences (P < 0.05).The symptoms scores and gastroscopy scores after therapy in sequential therapy group were significantly lower than those in triple-combined therapy group,and there were statistical differences (P <0.05).The symptoms scores and gastroscopy scores after therapy in sequential therapy group,triple-combined therapy group and Hp negative group were significantly lower than those in Hp positive without eradication group [(0.84 ± 0.60),(1.34 ± 0.59),(1.49 ± 0.62) scores vs.(2.98 ± 0.54) scores,(0.47 ± 0.37),(0.83 ± 0.35),(0.96 ± 0.75) scores vs.(1.22 ± 0.40) scores],and there were statistical differences (P < 0.05).Conclusions The Hp eradication rate of sequential therapy is higher than that of triple-combined therapy.The two therapy methods can both improve the symptoms score and gastroscopy score,but the symptoms after sequential therapy are relieved faster compared with triple-combined therapy.
4.A Strategy for Bioproduction of Rare Sugars: Izumoring

Rare sugars were defined as monosaccharides and their derivatives that rarely exist in nature. They played an important role in food, health, medicine and etc A strategy for bioproduction of rare sugars, namely Izumoring, was described. By the Izumoring method, all monosaccharides and polyols could be linked, using enzymatic reaction with D-tagatose 3-epimerase, aldose isomerases and polyol dehydrogenases. Izumoring for hexoses, pentoses and tetroses were designed respectively. According to this strategy, the bioproduction routes of various rare sugars, using microbial and enzymatic reactions, could be obtained. In addition, the future research tendency of biotransformation of rare sugars was put forward.
5.Role of delta and kappa opioid receptors in sufentanil preconditioning-induced attenuation of myocardial ischemia-reperfusion injury in rats

Objective To evaluate the role of delta and kappa opioid receptors in sufentanil preconditioning (SPC)-induced attenuation of myocardial ischemia-reperfusion (I/R) injury in rats.Methods Forty adult male Sprague-Dawley rats,aged 2-3 months,weighing 250-330 g,were randomly divided into 5 groups (n =8 each) using a random number table:sham operation group (group S),group I/R,group SPC,δ receptor antagonist naltrindole + SPC group (NTD + SPC group),and κ receptor antagonist nor-binaltorphimine (nor-BNI) + SPC group (BNI + SPC group).Myocardial I/R was induced by 30 min occlusion of left anterior descending branch of coronary artery followed by 120 min reperfusion.In group SPC,5 min infusion of sufentanil 3μg/kg was repeated 3 times at 5 min interval before myocardial ischemia.In NTD + SPC group,naltrindole 5 mg/kg was intravenously injected,at 10 before SPC.In BNI + SPC group,nor-BNI 5 mg/kg was injected intravenously at 15 min before SPC.Arterial blood samples were collected at 120 min of reperfusion for measurement of the serum cardiac troponin I (cTnI) and creatine kinase isoenzyme-MB (CK-MB) concentrations.The rats were then sacrificed and hearts removed for determination of myocardial infarct size (IS) and area at risk (AAR).IS/AAR ratio was calculated.Results Compared with S group,the serum cTnI and CK-MB concentrations were significantly increased in I/R,NTD + SPC and BNI + SPC groups,and the serum CK-MB concentrations were increased in SPC group,and IS/AAR ratio was increased in I/R group (P < 0.05 or 0.01).The serum cTnI and CK-MB concentrations were significantly lower in SPC,NTD + SPC and BNI + SPC groups and IS/AAR ratio was lower in SPC and BNI + SPC groups than in I/R group,and higher in NTD + SPC and BNI + SPC groups than in SPC group (P < 0.05 or 0.01).Conclusion Both κ and δ opioid receptors mediate SPC-induced attenuation of myocardial I/R injury in rats.
6.GC/MS-based dynamic analysis of Eucalyptus camaldulensis extract on corre-lation of chromatography-activity against Oncomelania hupensis

Objective To determine the contributions of main chemical compositions of extracts of Eucalyptus camaldulensis represented by GC/MS elute peaks to the molluscicidal activities,and explore a shortcut of looking for the effective components from natural products. Methods E. camaldulensis leaves were collected consecutively in 12 months at the same place,extracted with dichloromethane,analyzed by GC/MS,and their LC50(s)of molluscicidal activities were tested according to the method rec-ommended by WHO. The correlation of the main components in 12 extracts and their molluscicidal activities were analyzed by the grey relative correlation analysis model with software GTMS 3.0. Result All the dichloromethane extracts of eucalyptus leaves showed excellent molluscicidal activities with the highest LC50 of 0.257mg/L and 0.242mg/L for the samples in June and July and the lowest LC50 of 6.802 mg/L and 5.406 mg/L in December and January respectively. The structures of 16 main chemical compo-nents were elucidated by GC/MS and NIST Mass Spectral Library,most of which were monoterpenes and sesquiterpenoids. The gray correlation coefficients with activity were all over 0.5,the first five over 0.9 were 4,4,8-Trimethyltricyclo[6.3.1.0(1,5)]do-decane-2,9-diol,(-)-Spathulenol,a structural isomer of(-)-Spathulenol,Eucalyptol and Ledol. Conclusion The most main in-gredients in the dichloromethane extracts of E. camaldulensis leaves show good correlations with the molluscicidal activity,which suggests that the molluscicidal role is synergistically played by the multiple components together.
7.Purification and Characterization of L-AI from Bacillus Stearothermophilis IAM 11001 Expressed in E.coli

Thermostable L-arabinose isomerase (L-AI) is the most potential enzyme for the biological production of D-tagatose from D-galactose, a novel functional factor. Gene araA encoding the L-arabinose isomerase from Bacillus stearothermophilis IAM 11001 was cloned and expressed in Escherichia coli. The araA gene of 1491 bp has 95% identity with L-AI from Thermus sp. IM6501. The GenBank accession number for the nucleotide sequence of this araA gene determined in this work is EU394214.The bacterium was induced by IPTG and analyzed by SDS-PAGE, approximately 59 kDa exogenous protein was observed on the SDS-PAGE. The recombinant L-AI was purified to electrophoretical homogeneity with affinity chromatography, and the activity of recombinant L-AI was also studied. The bioconversion rate of D-galactose to D-tagatose reached 39.4% after 24h whole cell reaction.
8.Research Progress on the Microbial L-Arabinose Isomerase

L-arabinose isomerase (L-AI) can isomerize L-arabinose and D-galactose into L-ribulose and D-tagatose, respectively, which is currently the most effective biological catalyst for D-tagatose production. The crystal structure of L-AI has been solved recently and its gene has been cloned, sequenced and overex- pressed. L-AI improved by protein engineering will be the dominant enzyme for industrial production of D-tagatose. This paper reviewed researches on protein structure and function, properties and application in D-tagatose production of L-AI, and the long-term potential development of L-AI was prospected.
9.Effects of gene silence on immune function of colonic dendritic cells in rats with irritable bowel syndrome visceral hypersensitivity

Objective To explore the role and mechanism of protein disulfide isomerase A3 (PDIA3) in abnormal colonic mucosa immune response of irritable bowel syndrome (IBS) rats with visceral hypersensitivity.Methods A total of 48 SD rats were divided into blank control group (n=12),empty virus group (IBS-1) (n=12),PDIA3 silencing group (IBS-2) (n=12) and model control group(IBS-3) (n=12).The expression of CD103 in ileocecus colonic tissues was detected by immunohistochemistry.Dendritic cells (D C) of mesenteric lymph node were isolated by flow cytometry.CD4+/CD8+ T cells of spleen were separated by immune-magnetic beads sorting technique.DC and CD4+/CD8+ T cells were co-cultured.The proliferation ability of lymphocytes promoted by DC was measured by methyl thiazolyl tetrazolium (MTT).The secretion levels of interlcukin (IL) 4 and IL-9 of CD4+/CD8+ T cells stimulated by DC were determined by enzyme linked immunosorbent assay (ELISA) method.Independent sample t-test was performed for statistical analysis.Results The cell number of CD103 positive DC of rats colon of blank control group,IBS-3 group was 6.25±1.14 and 10.83± 1.03(t=10.07,P<0.05);that of IBS-2 group was 7.42 ± 0.90,and compared with that of IBS-3 group,the difference was statistically significant (t=-9.25,P < 0.05).The MTT value of CD4+ T cells proliferation stimulated by DC of blank control group and IBm3 group was 0.54±0.01 and 0.60±0.01 (t=3.373,P<0.05);that of IBS-2 group was 0.53±0.01,and compared with that of IBS-3 group,the difference was statistically significant (t =-3.139,P < 0.05).The MTT value of CD8+ T cells proliferation stimulated by DC was 0.52±0.01 and 0.59±0.00(t=3.539,P<0.01);that of IBS-2 group was 0.54±0.01,and compared with that of IBS-3 group,the difference was statistically significant (t=-3.183,P<0.05).The level of IL-4 secreted by CD4+T cells promoted by DC of blank control group and IBS-3 group was 10.24±0.09 and 16.61±1.00 (t=3.222,P<0.05);that of IBS-2 group was 11.75±0.54,and compared with that of IBS-3 group,the difference was statistically significant (t=-3.539,P<0.01).The level of IL-9 secreted by CD4+T cells stimulated by DC of blank control group and IBS-3 group was 15.86±10.19 and 43.51±11.32 (t=4.529,P<0.05);that of IBS-2 group was 29.05±2.09,and compared with that of IBS-3 group,the difference was statistically significant (t=-6.841,P<0.01).The level of IL-4 secreted by CD8+T cells promoted by DC of blank control group and IBS-3 group was 7.35±0.12 and 13.91±0.57 (t=19.557,P<0.01);that of IBS-2 group was 8.63± 0.24,and compared with that of IBS-3 group,the difference was statistically significant (t =-14.782,P<0.01).The level of IL-9 secreted by CD8+T cells stimulated by DC of blank control group and IBS-3 group was 29.12±5.14 and 60.70±11.02 (t=4.122,P<0.05);that of IBS-2 group was 37.17±2.65,and compared with that of IBS-3 group,the difference was statistically significant (t=-3.255,P< 0.05).Conclusions Protein disulfide isomerase A3 may mediate the process of DC activating T lymphocyte levels of related cytokines,cause abnormal immune response of colonic mucosa and promote the genesis of IBS visceral hypersensitivity.
10.Effect and mechanism of acrylamide on learning and memory and long-term potential in female Wistar rats

OBJECTIVE To investigate the accumulated effect and the mechanism of repeated sc administration of acrylamide (AM) on learning and memory after 28 d,and the effect of AM on the amplitude of population spike(PS) potential after a single sc administration of AM. METHODS Female Wistar rats were sc adminstered with AM 10, 20 and 40 mg · kg-1 once a day for 28 d, and weighted every week. The ability of study and memory was evaluated, The morris water maze was used from 22nd to 28th day,followed by step down test on the 29th and 30th day. The escape latent period and the number of errors in those two days were recorded. Rats from normal control group and AM 40 mg·kg-1 group were taken to have their N-methyl-D-aspartate receptor (NR) and calmodulin-dependent protein kinaseⅡ (CaMKⅡ) protein levels detected by Western blotting. Additionally, some other female Wistar rats were sc administered with a single dose of AM 40 mg · kg-1 ,before the changes in PS potential amplitude induced by high frequency stimulation were recorded by long-term potential (LTP). RESULTS Compared with normal control group, the relative body mass gain was significantly decreased in AM exposure groups(P<0.01). Additionally, the escape latency period was significantly increased in AM 20 and 40 mg·kg-1 groups compared with normal control group, while the crossing frequency was not significantly different betmeen these four groups. Compared with the first day of step down test, the number of errors was significantly decreased(P<0.01) and the escape latency period was significantly extended(P<0.01) in normal control group on the 2nd day. However, the number of errors and the escape latency period did not significantly change in the AM groups between the two days. The results of Western blotting showed that the protein expressions and phosphorylation of NR2A and NR2B, as well as the phosphorylation of CaMKⅡ in AM 40 mg · kg-1 group were significantly increased compared with normal control group. LTP result showed that AM 40 mg·kg-1 significantly inhibited the amplitude of PS potential after a single percutaneous administration. CONCLUSION AM can inhibit the PS amplitude by inhibiting the release of glutamate, increasing the expressions and activities of NR,and inhibiting PS potential, thus affecting the hippocampal synaptic plasticity, and the function of learning and memory.
