Chronic inflammation of intestine mucosal has been considered as a critical component of colorectal cancer development.Recently,the studies based on high throughput sequencing of colorectal cancer patients released a potential molecular subtype of colorectal cancer,characterized by wound healing and innate immune genes.It suggests that a probably mechanism described by a subset of cells that may be dependent on inflammatory cells and cytokines of tumor development,extremely expanding our insight of cell malignant transformation,and highlight the anti-inflammation treatment.This article will review the normal mechanism of intestine mucosal renewal,wound healing and malignant transformation.

3.Effects of nimesulide combined with cetuximab on apoptosis and apoptosis-related gene expression of human colon carcinoma cell line HT-29

Objective:The study was to investigate the effects of epidermal growth factor receptor (EGFR) antibody (cetuximab,C225) combined with cyclooxygenase 2(COX-2)inhibitor (nimesulide) on the proliferation and apoptosis of colon cancer HT-29 cells, and explore the potential molecular mechanism. Methods:C225 and nimesulide alone or in combination were incubated with HT-29 cells. MTT assay was used to measure the cell proliferation. AO/EB staining and flow cytometry were used to measure the cell apoptosis. RT-PCR was used to detect the COX-2 and non steroidal anti-inflammatory drug-activated gene (NAG-1) mRNA expression levels. The expression of Akt and phosphor-Akt protein was examined by Western blotting. Results:The inhibitory effect of nimesulide combined with C225 was significantly stronger than that of nimesulide alone [(72.8±2.3)% vs (51.8±1.8)% , P<0.05]at 48 h. Typical changes of apoptosis in HT-29 cells were observed in nimesulide combined with C225 treatment group. The apoptosis rate of combined group was significantly higher than that of single nimesulide group [(57.67±0.86)% vs(33.27±1.47)%]and single C225 group [(6.27±0.55)%,P<0.05]. C225 down-regulated the expression of COX-2 mRNA. The expression of NAG-1 mRNA was up-regulated in all treatment groups and the expression of phosphor-Akt was significantly down-regulated in combined treatment group than single nimesulide or C225 group. Conclusion:Nimesulide combined with C225 has obvious synergistic effects in inducing apoptosis. The potential mechanisms may be that EGFR signaling pathway is involved in the regulation of cycloxygenase-2 expression in HT-29 cells and finally influence the proliferation and apoptosis of HT-29 cells through the up-regulation of NAG-1 and down-regulation of phosphor-Akt expression.
4.EXPERIMENTAL STUDY ON MOBILIZATION OF PERIPHERAL HEMATOPOIETIC STEM/PROGENITOR CELLS IN MICE BY A COMBINATION OF CYCLOPHOSPHAMIDE, RECOMBINANT HUMAN INTERLEUKIN-11 AND GRANULOCYTE-COLONY STIMULATING FACTOR

To explore the effect of recombinant human interleukin 11(rhIL-11), granulocyte-colony stimulating factor (rhG-CSF) combined with cyclophosphamide (CTX) on mobilizing peripheral blood stem/progenitor cells in C57BL/6 mice. A total of 48 C57BL/6 mice were randomized into four groups. CTX (200mg/kg) was injected intraperitonealy on the first day (d0) in the treatment group. rhIL-11 alone or in combination with rhG-CSF was administered subcutaneously, beginning from 24 hours after CTX for 15 consecutive days in two other groups. The changes in peripheral blood white cell counts, platelet counts and the percentage of hematopoietic stem/progenitor cells of the mice were observed. It was shown that rhIL-11 or/and rhG-CSF could increase the numbers of WBC, PLT, CFU-GM, CFU-E, CFU-MK progenitor cells, and the percentage of CD34 positive cells in peripheral blood of myelosupressed mice. The results demonstrated that rhIL-11 alone or in combination with G-CSF and CTX could mobilize bone marrow haematopoietic stem/progenitor cells into peripheral blood.
5.Evaluation of the undoubled-blind design in the clinical trial of Chinese herb

Bias in undoubled blind design in the clinical trial of Chinese herb affects the evaluation of efficacy and safety. With two examples, the paper recommended a "segregation" technique to minimize the bias in investigators during undoubled blind trial. The results showed that this method combined other strict means can achieve the conclusions the same as a double blind trial.
6.Detection of chromosomally abnormal fetuses by nuchal translucency

Objective To assess the ultrasonographic detection of nuchal translucency or nuchal edema in identifying chromosomally abnormal fetuses.Methods Fetal nuchal translucency thickness was measured between 10~14 weeks of gestation,or nuchal fold thickness after 14 weeks,with serial follow-up ultrasound examinations.Results In a 2-year period,12 aneuploid fetuses had been identified,together with 5 fetuses of normal karyotype but fatal prognosis.Compared with the fetuses of increased nuchal translucency but normal karyotype and normal follow-up,they had a much thicker nuchal translucency in the first trimester (average 6.1 mm vs 3.6 mm),and in most cases the nuchal translucency evolved into nuchal edema in the second trimester,with other abnormalities detected by ultrasound.Conclusions Detection of increased nuchal translucency by ultrasound is effective in detecting chromosomally abnormal fetuses or fetuses with other abnomalities,and the changes of nuchal translucency observed by follow-up ultrasound may play an important role in differential diagnosis and predicting the prognosis.
7.Color Doppler ultrasonographic study of crural arterial diseases

50%,0-patent lumen.Those legs with dominant crural arterial lesion (gradeⅠ,Ⅱor Ⅲ) on at least one segments and without femoropopliteal arterial obstructions were selected.Results Totally 75 legs were involved.There were 53 legs( 70.7%) with only 1 main artery damaged,14 legs( 18.7%) with 2 main arteries damaged,and 8 legs( 10.6%) with all 3 main arteries damaged.Among the 5 arterial segments,the anterior tibial arterial lesions were found most frequently ( 73.3%,55/75),in which the segmental occlusion (gradeⅡ) was the most common type.The peroneal arterial lesions appeared least ( 20.0%,15/75).In the damaged main arteries,1/3 anterior tibial-dorsal arteries and 1/5 posterior tibial arteries had greatly improved distal vascularity compared with their proximal and middle portion,owing to the collateral arteries around the ankle.Statistical analysis showed that there was close relationship between the blood flow damage of the distal portion of the posterior tibial arterial segments (medial trans-ankle flow) and the peroneal artery with the clinical ischemic manifestations.Conclusions The dominant occlusive crural arterial lesions and the collateral arteries can be well imaged and diagnosed by color Doppler ultrasonography,which is helpful in providing accurate information of the blood supply of the distal part of the lower extremity.

9.ITS sequence analysis on medicinal plants of Euphorbia L. in Anhui and Jiangsu Provinces

Objective To study the correlation and variation between plants of internal transcribed spacers (ITS) sequence of the six medicinal plants of Euphorbia L. in Anhui and Jiangsu Provinces, in (order) to provide their DNA molecular marker to identify and explore the phylogenetic relationship of the plants of [WTBX]Euphorbia L. Methods To determine rDNA ITS sequence for the plants of Euphorbia L. by PCR technology. Results The sequences of ITS1 in the six species ranged from 255 to 262 bp in length and those of ITS2 from 214 to 236 bp. The dendrogram was obtained with Mega2 analysis. The analysis result was consistent with those from morphology. Conclusion The method can be used to identify the plants of [WTBX](Euphorbia) L. among different species and to differentiate their fakes.
10.Progress of sphincter-preserving treatment for low rectal cancer

This paper reviews the research progress of sphincter-preserving treatment for low rectal cancer.
