2.Influence of donepezil hydrochloride and hyperbaric oxygen on cognitive function in patients with vascular dementia

Objective To investigate the influence of donepezil hydrochloride and hyperbaric oxygen on cognitive function in patients with vascular dementia.Methods 61 cases of patients with vascular dementia were chosen to research,and were divided into research group(31 cases)and control group(30 cases),the control group was treated with piracetam,the research group was treated with donepezil hydrochloride and hyperbaric oxygen,after 12 weeks of treatment congnitive function,daily life quality and adverse reaction between two group were compared. Results The ADL score and MMSE score between the two group had no significant difference before treatment(all P <0.05).The MMSE score of the research group were higher than the control group[(18.71 ±3.29)points vs (15.83 ±3.45)points]after treatment,the difference was statistically significant (t =3.337,P <0.05).The ADL score of the research group were lower than control group[(37.12 ±7.13)points vs (42.53 ±6.74)points],the difference was statistically significant (t =3.026,P <0.05).4 cases of mild to moderate earache and 1 case of mild diarrhea occurred in the research group,and 1 case of upper abdominal pain occurred in the control group,there was no significant difference in the incidence of adverse reactions between the two groups (χ2 =0.803,P >0.05 ). Conclusion Donepezil hydrochloride combined with hyperbaric oxygen could effectively improve the cognitive function of patients with vascular dementia,and with good safety.
3.Repairment Effect of Qingqinye on Joint Immune Pathological Injury in Hyperuricemia Model Rats

TNF-? in Qingqinye group was lower than model group and positive medicine group, especially in Qingqinye middle dosage group. Pathological section with articular cavity and surrounding tissues were better in positive medicine group and Qingqinye group. Conclusion Qingqinye has a certain effect for repairing joint immune pathological injury in hyperuricemia model rats.
4.Variation in flow rates of anthraquinones in Anzhong Tongbian Capsule

AIM: To establish variation in flow rates of anthraquinones in Anzhong Tongbian Capsule(Radix et Rhizoma rhei,Radix paeoniae alba,Fructus cannabis and Semen trichosanthis). METHODS: The thin layer chromatographic method was adopted. The anthraquinones compounds from Radix et Rhizoma Rhei in Anzhong Tongbian Capsules were extracted by ultrasound and separated by outspread on silica gel G thin layer,and n-hexane-ethyl acetate-formic acid(30 ∶ 10 ∶ 0. 5)as the developing system,detection wavelength was set at 435 nm and reference wavelength was 610 nm by using CAMAG TLC ScannerⅢ. RESULTS: Anthraquinones was separated well and 5 peaks of emodin,chrysophanol,aloe-emodin,rhein and physcion were ascertained. CONCLUSION: The method is simple and accurate with a good reproducibility and can be used for the quality control for Radix et Rhizoma Rhei in Anzhong Tongbian Capsules.
5.Biological effects of recombinant pcDNA3.0-hugl-1 plasmid on colorectal cancer cell line LS174

Objective:To observe the biological behaviors of colorectal cancer LS174 cells before and after pcDNA3.0-hugl-1 transfection,so as to investigate the association of hugl-1 with colorectal cancer.Methods:The eukaryotic expression vector pcDNA3.0-hugl-1 was constructed and transfected into LS174 cells.RT-PCR and Western blotting methods were used to analyze the expression of hugl-1 mRNA and protein in LS174 cells before and after transfection.Soft agar colony formation assay,wound-healing experiment,adhesion assay and Matrigel invasion assays were used to study the effects of hugl-1 expression on the proliferation,adhesion,movement and invasion in LS174 cells.Results:The recombinant plasmid pcDNA3.0-hugl-1 was successfully constructed.RT-PCR and Western blotting showed that the hugl-1 expression was higher in cells transfected with pcDNA3.0-hugl-1 than in those un-transfected or empty vector-transfected cells (P
6.Development of a new type nerve tissue engineering scaffolds and its combination with bone marrow stromal cells

[Objective] To develop a new type nerve tissue engineering scaffolds to repair the defect of peripheral nerve and observe the growth of bone marrow stromal cells cultured in vitro on the scaffolds.[Method]The biomaterial was made of I-collagen and gelatin by freeze-drying method,the alignment regularities of microscopic channels and there course directions were observed under the scanning electronic microscope.The size of the micropores and the factor of porosity were also measured.The bone marrow stromal cells were seeded on the collagen scaffolds and cultured for 5 days,then the growth of bone marrow stromal cells cultured in the scaffolds was observed.[Result]All the scaffolds were circular cylinder,the microscopic channels were arranged in parallel manners,and the pore sizes of the channels were uniform.The bone marrow stromal cells were seeded in the scaffolds successfully.[Conclusion]The developed nerve tissue engineering scaffolds have good structure and biocompatibility,which can be used in the repair of the nerve injuries.

8.The clinical observation of stellate ganglion blockade combined with oxycodone and acetaminophen tablets on migraine

Objective To observe the therapeutic effects of stellate ganglion block (SGB) with oxycodone and acetaminophen tablets on the treatment of migraine. Methods 100 patients with migraine were divided into two groups randomly. They received SGB with oxycodone and acetaminophen tablets (experimental group) or only SGB (control group) respectively. The parameters included pain intensity before and 1 d,3 d,1 w,1 m and 3 m after the treatment, and pain relief rate of each patient. The adverse effects were also evaluated. Results The pain intensity described as numeric rating score (NRS) decreased in the two groups at each time point. In the experimental group, the value of NRS was significantly less than that in the control group at 3d,1m and 3m after the treatment. The effec-tive rate ha the experimental group and the control group were 88% and 68%. Conclusion SGB with oxycodone and acetaminophen tablets is more effective for migraine and deserves widely used in patients.
9.Effect of Sijunzi Decoction on Mitochondrial DNA Missing of the Aging Model Mice

Objective To observe the effect of Sijunzi Decoction (SD) on the mitochondrial DNA deletion of the aging model mice, and to explore its anti-aging biomolecular mechanism. Methods Kunming mice were divided into 6 groups according to the body weight: normal control group, .model control group, high-, middle-and low-dose SD groups (at the dose of 20, 10, 5 g·kg-1 respectively), and vitamin E group (250 mg·kg-1), 20 mice in each group.The mice models of aging were induced by D-galactose. Water maze test was used to evaluate the learning and memory ability of the aging model mice. The effect of SD on the mitochondrial DNA deletion of the aging model mice was ob-served. Results In the hippocampal mtDNA of model control group and low-dose SD group, there were about 3.87 kb fragment missing. But there was no missing pieces in high-and middle-dose SD groups. Conclusion The anti-aging biomolecular mechanism of SD is probably related to the preventive effect on the mutation and deletion of the mitochondrial DNA in the aging model mice.
10.Prospective Efficacy of Pulsed Radiofrequency Guided with Spiral CT on Trigeminal Neuralgia: Long-term Observation

Objective To investigate the prostecdtive therapeutic efficacy of pulsed radiofrenquency and radiofrequency thermocoagulationof the Gasserian ganglion guided with spiral CT on trigeminal neuralgia. Methods 100 patients with idiopathic trigeminal neuralgiawere treated with pulsed radiofrenquency or radiofrequency thermocoagulation of gasserian ganglion. The numeric rating scales (NRS) ofpain were recorded before and 0.5 year, 1 year, and 2 years after treatment. Results There was no difference in NRS within 1 year, but the recurrencerate was high in the paitents accepted pulsed radiofrenquency 2 years later (P<0.01). Conclusion Pulsed radiofrenquency of thegasserian ganglion is safe and effective on trigeminal neuralgia, but poor in long-term outcome.
