1.Determination of cholesteryl palmitate in Oviductus Ranae by HPLC.

OBJECTIVETo develop a new method for the determination of cholesteryl palmitate in Oviductus Ranae.
METHODA HPLC method was set up, using Zorbax Silica column and cyclohexane-diethyl ether (40:1) as mobile phase with a flow rate of 1.0 mL x min(-1), and the UV detection wavelength was 203 nm.
RESULTThe calibration curve was linear over the range of 0.60-8.92 microg (r = 0.9997), the average recovery of the method was 98.4%. RSD 1.8% (n = 6).
CONCLUSIONThe results showed that method was reliable and accurate.

Based on the full-length cDNA of BmalphaTX14 from Chinese scorpion Buthus martensii Karsch (BmK), gene of the mature peptide of BmalphaTX14 was cloned into the yeast expression vector pPIC9K. After transforming, screening and inducing, tricine-SDS-PAGE and Western blot proved that rBmalphaTX14 protein was expressed in the medium for up to 84 hours, getting nearly 120 mg/L. Recombinant BmalphaTX14 was purified rapidly and efficiently through Ni-NTA-agarose, polyethylene glycol precipitation and gel filtration chromatography. The purified rBmalphaTX14 proved to have the anti-insect activity by toxicity assay. Meanwhile, genomic gene of BmalphaTX14 was cloned and sequenced by PCR method, sequence analysis of this gene showed that BmalphaTX14 had an intron of 408 base pairs located at the signal peptide encoding region, which was similar with the characteristic of other alpha-type sodium ion-channel toxin. Considering both the genomic organization and the peptide function, BmaTX14 proved to be a membership belonging to alpha-type sodium ion-channel toxin.

6.The impact of community-based organizations in HIV testing mobilization among men who have sex with men.

OBJECTIVETo analyze the impact of efforts of community-based organizations (CBO) in HIV testing mobilization and case finding among men who have sex with men(MSM).
METHODSResults of HIV testing mobilization among MSM through CBOs in 15 program areas were collected and compared with corresponding HIV case reporting data to demonstrate the contribution of CBO-based HIV testing in HIV case finding among MSM from July 2008 to December 2011. Meanwhile,the proportion of screened HIV positives who received testing results notification,confirmatory test, following up and CD4 cell tests were analyzed and compared with those identified in medical institutions.
RESULTSA total of 196 075 HIV tests were performed for MSM, as a result of mobilization efforts of CBOs. Cumulatively 7704 new HIV cases were identified, accounting for 51.7% (7704/14 914) of all newly diagnosed HIV cases infected via homosexual sex in the program areas.Among the newly diagnosed MSM HIV infections in the program areas,the proportion of infections detected through the mobilization of CBOs increased from 35.4% (609/1722) in 2008 to 63.7% (2371/3722) in 2010, and 58.3% (3024/5189) in 2011. Compared with those identified through medical institutions, newly diagnosed MSM infections detected though CBOs testing mobilization have higher rates of receiving screening testing results notification (97.3% (4441/4563) vs 92.8% (13 140/14 153)) , (84.6% (2559/3024) vs 79.8% (5589/7002)) and CD4 cell tests (66.1% (1999/3024) vs 52.9% (3705/7002)), and a lower rate of receiving confirmatory test (78.6% (3588/4563) vs 85.6% (12 115/14 153)).
CONCLUSIONCBOs can take their advantages in mobilizing MSM to receive HIV test, and MSM HIV cases detected through CBOs have become the main source of MSM HIV case finding in program areas.

7.Clinical characteristics in patients with non-allergic rhinitis and allergic rhinitis:preliminary analysis

Objective To compare and analyze the clinical characteristics in patients with hyperreactive non-allergic rhinitis (HNA R) and allergic rhinitis (AR). Methods A questionnaire survey on AR and HNAR patients between January and August 2009 was conducted. The clinical data of 298 AR patients and 100 HNAR patients were analyzed, including gender, age distribution, seasonal, clinical symptom and induced factors. Results The number of male patients was more than female in AR, while in NAR, the number of female patients was more than male(χ2 =6. 415, P =0. 01). The highest morbidity age in AR was teenagers, aged between 10 - 19 (χ2 = 12. 772, P = 0. 00), while in HNAR, the highest morbidity age was middle-aged and youth, aged between 30-39(χ2 =51. 533, P =0. 00). The main onset seasons in AR was autumn, while there was no seasonal diversity in HNAR. The main allergen in AR was mugwort and ragweed, consistent with the vegetative cover characteristic in Jilin province. The main classification of AR was moderate-severe persistent(χ2 = 123.991, P =0. 00), while the main classification of HNAR was moderate-severe intermittent (χ2 = 97. 420, P = 0. 00) . The clinical symptoms were significantly different between AR and HNAR except rhinocnesmus(all P < 0. 05). There was consistency about non-specificity induced factors in AR and HNAR(all P > 0. 05). Conclusions There were significant differences between AR and HNAR in sex, age, classification and seasons. The severity of clinical symptoms in AR was higher than that in HNAR except sneezing and gasping. There was consistency about induced factors in AR and HNAR.
8.Coil selection and application during temporomandibular joint MRI imaging

Objective To evaluate the clinical application of 15-channel phased-array head coil and 6-channel dS Flex M surface coil in MRI imaging of temporomandibular joint.Methods Totally 50 patients were randomly equally assigned into two groups, of whom, one group used a 15-channel phase-array head coil for MRI scanning, and the other applied a 6-channel dS Flex M surface coil. The same regions of interest were set up to measure and analyze the signal to noise ratio (SNR)and contrast to noise ratio (CNR)of each region respectively.SPSS 19.0 software was used for quantitative analysis. Results The head coil had the SNR and the CNR higher than those of the surface coil.The head coil and Flex surface coil had the imaging diagnosis rates being 95.83% and 91.67% respectively,and there was no significant difference between them(P>0.05). Conclusion Both phase-array head coil and dS Flex M surface coil can be used for temporomandibular joint MRI imaging, and it's suggested that conventional imaging be executed with dS Flex M surface coil and functional imaging be implemented with head coil.
9.Overexpression of interleukin-l7 in tumor-associated macrophages is correlated with the differentiation and angiogenesis of laryngeal squamous cell carcinoma.

BACKGROUNDInterleukin-l7 (IL-17), which exerts strong pro-inflammatory effects, has emerged as an important mediator in inflammation-associated cancer. The aim of this study was to clarify the relationship between IL-17 and tumor associated macrophages (TAMs), and the correlation of the microvessel density in the development of laryngeal squamous cell carcinoma (LSCC).
METHODSHistopathological observations and immunohistochemistry staining for IL-17, CD68, and CD34 were performed on 72 specimens (32 cases of LSCC, 20 cases of adjacent tissues of carcinoma as controls, and 20 cases of chronic hypertrophic laryngitis). Double immunohistochemical staining was done to determine which cells expressed IL-17. Real-time quantitative PCR determined the mRNA expression of IL-17. ELISA was used to detect the expression of the serum level of IL-17 in the three groups.
RESULTSThe inflammation response had increased in LSCC. Overexpression of IL-17 and CD68 protein were seen in LSCC (P < 0.01). The expression of IL-17 was different between well and poorly differentiated LSCC (P < 0.01). The IL-17 expressing cells were mainly located in macrophages (CD68(+)/IL17(+)) as demonstrated by double immunohistochemical staining. IL-17 expression significantly correlated with high microvessel density (CD34(+)) in LSCC (P < 0.05). Relatively higher mRNA expression levels of IL-17 were seen in LSCC compared to the controls (P < 0.05). The serum expression of IL-17 was similar among the three groups (P > 0.05).
CONCLUSIONIL-17 was expressed by TAMs, and IL-17 may significantly correlate to the differentiation and angiogenesis in the development of LSCC.

10.Correlation analysis of two serum specific IgE test systems and skin prick test in allergic rhinitis patients

Objective To evaluate the correlation between two serum specific IgE and skin prick test (SPT) for the diagnosis of allergic rhinitis.Methods Two hundred and sixteen patients were referred to the allergist for a suspected allergic rhinitis between June and October in 2009. Patients were classified as positive for inhalant allergy if they had a positive clinical history and a related positive SPT for the suspected inhalant allergen. Statistical analysis was performed using SPSS13.0 software. Results One hundred and fifty-eight patients had a positive SPT, comparing with the SPT, the diagnostic indexes (accuracy,sensitivity, specificity) of the ImmunoCAP system and the AllergyScreen system were 0.810 and 0.819,0.872 and 0.780, 0.741 and 0.862 respectively. The accuracy was similar between the two systems (x2=0.112, P>0.05). The ImmunoCAP system had a higher sensitivity (x2=7.361, P<0.05). The AllergyScreen system had a higher specificity (x2=10.222,P<0.05). Conclusions This data supported the use of ImmunoCAP system and AllergyScreen system to identify potentially significant individual allergens in the diagnosis of allergic rhinitis. The ImmunoCAP system had a higher sensitivity. The AllergyScreen system had a higher specificity. The AllergyScreen system can be used as a complementary with the ImmunoCAP system.
