2.Effect of matrix metalloproteinase-3 on vitreous and vitreoretinal interface in rabbit's eye

Objective To investigate the effects of intravitreal injection of matrix metalloproteinase-3 (MMP-3) on the vitreoretinal adhesion and the vitreous gelatin. Methods Twenty-four pigmented rabbits were randomly divided into 3 experimental groups (group A, B, and C)and one control group with 6 rabbits (12 eyes) in each. Different concentrations of 0.1 ml MMP-3 (5,10, 20 ng in group A, B, and C, respectively) and equivalent dose of balanced salt solution were intravitreally injected to the rabbits, respectively. Clinical examinations (such as gross observation, slit-lamp biomicroscopy, indirect fundus ophthalmoscopy ), electroretinography (ERG) and fundus fluorecein angiography (FFA) were taken before and after injection. Results One week after injection, posterior vitreous detachment (PVD) and focal vitreous liquefaction were recognized clinically for the first time in 1 eye in group B. By the end of this study, clinically detected PVD developed in 1 eye in group A, 3 eyes in group B, but the synchisis developed slowly, and no liquefaction or PVD occurred in control group. As for the histological examination, partial PVD was observed in 1 eye in group A and 3 eyes in group B 60 minutes after injection. All of the eyes in group A and B showed partial PVD 1 week after injection, and the area of PVD enlarged in contrast with before. Complete PVD were recognized in 1 eye in group A and 3 eyes in group B 15 weeks after injection, and the cleavage was narrow and limited. In group C, inflammatory cell infiltration in the inner layer of retina, destruction of retinal structure, and fluorescein leakage at late phase was found in the early period after injection. Conclusions MMP-3 is effective in disrupting the adhesion of the posterior hyaloid to the inner limiting membrane leading to PVD, and helpful to some extent in producing vitreous liquefaction. The dose of 10 ng MMP-3 is safe and effective for the rabbits eyes, which may be used as an promising assistant of vitreous surgery.

Object To study the volatile constituents in the TCM preparation "Pao Chai Pill" made in Yangcheng (Guangzhou) (YPCP) in comparison with that made in Hongkong (HPCP) to gain some insight for the renewal of YPCP to a new dosage form Methods The volatile constituents were extracted with the aid of ultrasonic wave in organic solvent, and identified by GC-MS and elucidated on the standard MS data Their relative contents in percentage were calculated by area normalization Results 24 and 20 volatile constituents were identified in YPCP and HPCP respectively Conclusion YPCP showed more rich in sesquiterpene lactones but less sesquiterpenes and sesquiterpene alcohols than HPCP
4.Influence of Toufengyu Dropping Pill on vasoactive substances in migraine rat

AIM: To observe the influence of Toufengyu Dropping Pill(Rhizome chuanxiong,Radix angelicae dahuricae,etc) on neuropeptide and vasoactive substances in rat's migraine model. METHODS: Rat's migraine model caused by nitroglycerin were given Toufengyu Dropping Pill and rat's behavior and blood levels of NO,CGRP,ET,TXB_2 and 6-keto-PGF_(1a) were measured. RESULTS: The experimental results showed that Toufengyou Dropping Dill could obviously decrease rat's positive reaction number,the blood levels of NO,CGRP,TXB_2 and 6-keto-PGF_(1a),and increase the blood plasma level of ET,and rectify the imbalance of ET/NO ratio. CONCLUSION: Toufengyou Dropping Pill can affect the contents of neurotransmitter,neuropeptide in rat's migraine,then decrease the algoenic and inflammatory substances,relieve the neurogenic inflammation,and increase the pain threshold value,finally be quite effective for migraine.
5.Components of Pharmaceutical Chemicals in Newly Edited National Chinese Traditional Patent Medicines: Statistics and Reflection

OBJECTIVE:To ensure safe and efficient drug use.METHODS:Chinese Traditional patent Medicines containing components of pharmaceutical chemicals collected in the Newly Edited National Chinese Traditional Patent Medicines were listed and classified statistically.RESULTS:Of the total collected 5 017 Chinese Traditional patent Medicines,160 contained components of pharmaceutical chemicals(3.19%).In which,the coldrex,medicine for relieving cough and asthma,gastrointestinal medicines took the leads.However,some Chinese Traditional patent Medicines instructions failed to indicate the contained contents of pharmaceutical chemicals as specified.CONCLUSIONS:For pharmaceutical manufacturing enterprises,the related information concerning the components of pharmaceutical chemicals should be added in the notes of Chinese Traditional patent Medicines.Doctors and pharmacists should know well about the related information so as to ensure safe and efficient drug use.

7.Effect of Nalmefene Hydrochloride on Expressions of Pulmonary β-endorphin and Interleukin-17 in Rats with Lung Ischemia-reperfusion Injury

Objective To study the protective effect of nalmefene hydrochloride on lung ischemia-reperfusion injury and its mechanism.Methods 40 rats were randomly divided into model group,high dose of nalmefene group,low dose nalmefene group and sham operation group equally(n =10).The lung ischemia-reperfusion model was established by occlusion of the left pulmonary hilum.The intravenous injection of nalmefene (20,10 μg·kg-1) was applied at 10 minutes before occlusion of the left pulmonary hilum in the high dose of nalmefene group and the low dose of nalmefene group,respectively.The sham operation group without occlusion of the left pulmonary hilum was not given any treatment.At 2 h after reperfusion,all rats were detected arterial blood gas value and then sacrificed.The specimens from the upper lobe of the left lung tissue were preserved to observe pulmonary lesions,detect the ratio of wet / dry weight and the expressions of β-endorphin and interleukin(IL)-17.Results Compared with the model group,the value of PCO2,the degree of pulmonary lesions,the ratio of wet / dry weight and the expressions of β-endorphin and IL-17 in lung tissue were significantly decreased (P < 0.01),while the value of PO2 was significantly increased (P < 0.01) in the low dose of nalmefene group.Compared with the low dose of nalmefene group,thevalue of PCO2,the degree of pulmonary lesions,the ratio of wet/dry weight and the expressions of β-endorphin and IL-17 in lung tissue were significantly decreased (P < 0.01),while the value of PO2was significantly increased (P < 0.01) in the high dose of nalmefene group.Conclusion Nalmefene hydrochloride may prevent lung ischemia-reperfusion injury in a dose dependent manner by reducing the production of β-endorphin and inhibiting the expression of IL-17 in lung tissue.
8.The prognosis effect of different Helicobacter pylori eradication methods in patients with chronic gastritis

Objective To compare the prognosis effect of different Helicobacter pylori (Hp)eradication methods in patients with chronic gastritis.Methods One hundred and twenty patients with chronic gastritis diagnosed by gastroscopy and pathology examination were divided into sequential therapy group (36 cases),triple-combined therapy group (34 cases),Hp positive without eradication group (30cases) and Hp negative group (20 cases) according to the Hp infection status.All patients were followed up by gastroscopy,biopsy and rapid urease test before and after therapy.Results The Hp eradication rate in sequential therapy group was 94.44% (34/36),in triple-combined therapy group was 73.53% (25/34),and there was statistical difference (x2 =5.775,P =0.016).The symptoms scores and gastroscopy scores after therapy in the four groups were significantly lower than those before therapy,and there were statistical differences (P < 0.05).The symptoms scores and gastroscopy scores after therapy in sequential therapy group were significantly lower than those in triple-combined therapy group,and there were statistical differences (P <0.05).The symptoms scores and gastroscopy scores after therapy in sequential therapy group,triple-combined therapy group and Hp negative group were significantly lower than those in Hp positive without eradication group [(0.84 ± 0.60),(1.34 ± 0.59),(1.49 ± 0.62) scores vs.(2.98 ± 0.54) scores,(0.47 ± 0.37),(0.83 ± 0.35),(0.96 ± 0.75) scores vs.(1.22 ± 0.40) scores],and there were statistical differences (P < 0.05).Conclusions The Hp eradication rate of sequential therapy is higher than that of triple-combined therapy.The two therapy methods can both improve the symptoms score and gastroscopy score,but the symptoms after sequential therapy are relieved faster compared with triple-combined therapy.

Through reviewing the relative literatures at home and abroad in recent years, the research progress in the chemical compositions and pharmacological activities of Ficuspumila L.was summarized.The study situation of flavonoids, terpenoids and carbohydrates et al in Ficuspumila L.was reviewed, and such pharmacological activities as antibacterial, antioxidant, hypoglycemic and anti-tumor of Ficuspumila L.were also evaluated.The modern researches showed Ficuspumila L.could be used for the treatment of intestinal cancer, lymphoma, mammary cancer and esophagus cancer.The review provided scientific basis for the further research and clinical application of Ficuspumila L.
10.Alfentanil protects the isolated rat heart against ischemia and reperfusion injury via opioid receptors and NO linked mechanisms

METHODS: Langendorff rat hearts were perfused at constant pressure with Kreb-Henseleit(K-H) buffer for 20 min?and?then?were?perfused?with?test?solution:?K-H?buffer?or?K-H?buffer?containing?alfentanil? 50 ?g?L -1,?alfentanil? 100 ?g?L -1, naloxone 200 ?g?L -1, alfentanil 100 ?g?L -1+naloxone 200 ?g?L -1, L-NAME 100 ?mol?L -1 and alfentanil 100 ?g?L -1+L-NAME 100 ?mol?L -1. After 10 min of this, the hearts were subjected to 25 min normothermic( 37 ℃) global ischemia followed by 30 min reperfusion with the same test solution as before. To evaluate myocardial function, LVEDP, LVDP, ?dp/dt max, HR and CF were measured at the 20th, 25 and 30th minute of perfusion and the 1st, 3rd, 5th, 10th, 20th and 30th minute of reperfusion.
