Journal of Environment and Health 1992;0(04):-
Objective To study the mechanism of rat blood pressure lowered by SO2 and SO2 derivatives. Methods 6-Keto-PGF1? and TXB2 were determined by radioimmunoassay in the plasma and aorta tissue of rats. Results The 6-Keto increased significantly in the isolated aortic rings in all exposure groups except 8 mmol/L group. No change of TXB2 was observed in the rings in all exposure groups.Meanwhile,6-Keto/TXB2 ratios increased significantly at 2 mmol/L and 4 mmol/L.Compared with the control group,6-Keto level decreased significantly in the plasma of the rats exposed to SO2 at 14 mg/m3,28mg/m3 and 56 mg/m3,but the level of TXB2 increased significantly.6-Keto/TXB2 ratios decreased gradually with the increase of SO2 concentration. Conclusion PGI2 and TXA2 are possibly changed by SO2 and SO2 derivatives, which regulates partly rat blood pressure.

Objective To study the change of intestinal permeability in patients with acute pancreatitis (AP) to investigate the indication of early operation in AP patients. Methods Serum concentrations of glutamine, endotoxin and urine lactulose/mannito (L/M) ratio were measured and compared at different time in 18 AP patients received non-operative treatment and 7 underwent operation . Results Serum concentrations of glutamine, endotoxin and urine lactulose mannito (L/M) ratio were significantly higher in operation treatment AP patients than those in non-operation treatment AP patients. Conclusion In the early course of AP, continued increased intestinal permeability and serum concentration of endotoxin is the indications of operation.

Objective To explore the feasibility of establishing the model of chronic blliary(pancreatitis) by incompletely ligaturing common bile duct. Methods Fifty Wistar rats were randomly(divided) into three groups: control group, experimental control group and model group. The rats in model group were exposed the root of common bile duct to enter duodenum using the dissolubility bowel line to ligating a needle with common bile duct, then drew out the needle, closing the abdominal cavity. The rats in experiment control group were opened the abdominal cavity and dissociated common bile duct, then closing the abdominal cavity. Results Compared with control group, the serum bilirubin and(amylase) of blood in model group were increased in 3 days and 14 days respectively and fibrotic(proliferation) of pancreatic tissues were found in 30 days.In model group,23% of rats developed(pancreatic) pseudocyst. Conclusions Incompletely ligating the common bile duct of Wistar rats could(induce) chronic pancreatitis and pancreatic fibrosis in 30 days.Strictures of common biliary duct may play a key role in chronic biliary pancreatitis.

Objective To explore the pathogenesis-related imprinted genes in osteosarcoma and investigate the role of imprinted genes expression in osteosarcoma pathogenesis. Methods The model of malignant transformed immortalized human osteoblastic cells was established and the malignant transformed cells were collected on day 40,55,70,90.The untreated cells passaged from the normal cells on day 90 served as control. After the total RNA extraction, the finally synthesized biotinylated cDNAs were hybridized to CapitalBio Genechip CAP-F0017.2 of the differentially expressed genes. Two genes were chosen at random to further confirm the array results using the SYBR Green real-time PCR in samples of control, day 40 and day 90. Results By an entrance limit of ≥2.0 or ≤0.5,ten imprinted genes which biological functions are mainly related with cell growth/maintenance and signaling transduction were detectable for notablely differential expression, including down-regulated genes of CD81,GRB10,NDN,MEST and up-regulated genes of H19,MKRN3,SLC22A1L,TSSC3,CDKN1C in the malignant transformation of immortalized human osteoblastic cells. The array results were further confirmed by the real-time RT-PCR.Conclusion Ten imprinted genes were detectable for notablely differential expression. Our results will promote our understanding of the molecular mechanisms of imprinted genes in the course of osteosarcoma pathogenesis.

Objective To observe the efficacy of single Nd-YAG laser treatment for the sub-inner limiting membrane(sub-ILM) haemorrhages in the macular region. Methods This is an uncontrolled case series retrospective study.Nd-YAG laser was performed for 6 patients (6 eyes) with premacular sub-ILM haemorrhages. The best corrected visual acuity(BCVA) and optical coherence tomography(OCT) in macular were observed at 1 day,1 week,1 month and 3 months after operation. The conditation of hematopoietic drainage and complications of macular retina were recorded. Results Sub-ILM haemorrhages were clinically confirmed in 6 patients (aged from 23 to 60 years old),including 4 cases of paroxysm associated with cough,1 case of hypertension, and 1 case of diabetic retinopathy.Funduscopic examination showed sub-inner limiting membrane hemorrhage in the macular region,BCVA count finger (CF) was 0.05 (averagely 0.03).The onset time was from 5 hours to 12 days. All the patients were treated with Nd-YAG laser treatment once, and all of them resulted in excellent partial visual recovery immediately. The average hemorrhage absorption time was 2 weeks.OCT examination showed macular retinal inner limiting membrane calm and foveal shape recovery at 3 month after laser treatment. However, the two patients with hypertension and diabetes recieved vitrectomy at 2 weeks and 1 month after laser surgery respectively because of serious vitreous hemorrhage. Conclusion Single Nd-YAG laser is a safe and effective method in the treatment of simplicity sub-ILM haemorrhages.

Objective To improve the recognition of treatment of adult T lymphoblastic lymphoma (TLL) with Hyper-CVAD regime. Methods The turnovers of 7 cases treated with Hyper-CVAD regime were summarized. Results Among the 7 cases,1 case died after the first unfinished chemotherapy,1 case gained complete remission before autologous stem cell transplantation but relapsed after transplantation,3 cases gained complete remission until now,2 cases without transplantation relapsed and then gave up following treatments after treating with ICE regime or the virgin regime, but the therapeutic effects were poor. Conclusions Adult TLL should get early chemotherapy treatments, and some special characteristics should be noted, the treatment does not relate to the stage, the chemotherapy treatment should be carried out regularly and enough, radiation of mediastinum mass can be applied after 4 cycles of Hyper-CVAD regime, choose suitable transplantation method.

A liquid chromatography-tandem mass spectrometry (LC-MS/MS) method was developed and validated for the determination of butoconazole in human plasma. Human plasma samples of 0.2 μL were pretreated by a single step protein precipitation procedure and analyzed using a high performance liquid chromatography (HPLC) electrospray tandem mass spectrometer system. The compounds were eluted isocratically on an Inertsil ODS-SP column (100 mm×2.1 mm, 3 μm), ionized using a positive ion atmospheric pressure electrospray ionization source and analyzed using multiple reaction monitoring (MRM) mode. The ion transitions monitored were m/z 412.8→165.1 for butoconazole and m/z 453.4→230.3 for the internal standard. The chromatographic run time was 3.5 min per injection, with retention time of 2.47 min and 2.15 min for butoconazole and repaglinide, respectively. The method was validated to be linear over the range of 20 to 8000 pg/mL (r>0.999) by using a weighted (1/x(2)) quadratic regression. The mean recovery rate was more than 86.7%, and the intra- and inter-day precision of the quality control samples (QCs) was less than 8.3% and the accuracy ranged from 96.0% to 110.2%, which indicated that the quantitative method was reliable and accurate. The method is simple, rapid, and has been applied successfully to a pharmacokinetics study of butoconazole nitrate suppositories in healthy Chinese females.
