1.Research in correlation of professional commitment and communication ability among nursing baccalaureate students in universities of Traditional Chinese Medicine

Objective To investigate the level of professional commitment of nursing baccalaureate students,and explore the relationship between the professional commitment and communication ability.Methods Questionnaires were used to investigate 430 nursing baccalaureate students who were studying in the school.Results The professional commitment of nursing students was in the middle level.Correlation analysis revealed that the communication ability was correlated with the professional commitment.Conclusions We should take measures to improve the competency of professional commitment of nursing undergraduate students,strengthen their communication ability,promote the construction of nursing staff.
2. Content changes of chemical constituents in Ginseng Radix and Atractylodis Macrocephalae Rhizoma before and after compatibility

Objective: To discuss the synergistic mechanism of compatibility of Ginseng Radix (GR) and Atractylodis Macrocephalae Rhizoma (AMR), and to determine the compounds in GR and AMR before and after compatibility using HPLC method. Methods: Hypersil ODS column (250 mm × 4.6 mm, 5 μm) was adopted. The mobile phase was acetonitrile (A) and water (B) for gradient elution (0-35 min, 5%-35% A; 35-40 min, 35% A; 40-80 min, 35%-95% A). The detection wavelength was set at 203 nm. The column temperature was 25℃. The flow rate was 1 mL/min. Results: The HPLC analysis suggested the extracts before and after compatibility were different on contents of GR and AMR. In GR and AMR co-decoction, the contents of ginsenosides Rg1, Re, Rf, Rd, and butenolide I increased obviously, and a new chromatographic peak appeared. There was no obvious change in other components. Conclusion: The synergistic mechanism of the compatibility of GR and AMR can be proved with the increased dissolution of ginsenoside and a new chromatographic peak.
3.Scientific Misconduct Encountered By APAME Journals: An Online Survey

In June 2015, invitations were sent by email to 151 APAME journals to participate in an online
survey with an objective of gaining insight into the common publication misconduct encountered by
APAME editors. The survey, conducted through SurveyMonkey over a 20-day-period, comprised 10
questions with expansions to allow anecdotes limited to 400 characters, estimated to take less than
10 minutes to complete. Only one invitation was issued per journal, targeting (in order of priority)
editors, editorial board members and editorial staff, and limited by email availability. 54 (36%) journals
responded. 98% of respondents held Editor or Editorial Board positions. All respondent journals
have editorial policies on publication ethics and 96% provide instructions related to ethics. 45% use
anti-plagiarism software to screen manuscripts, the most popular being iThenticate, CrossCheck and
Turnitin. Up to 50% of journals had encountered studies without IRB approval. Author misconduct
encountered were (in rank order): plagiarism (75%), duplicate publication (58%), unjustified
authorship (39%), authorship disputes (33%), data falsification (29%), data/image manipulation
(27%), conflict of interest (25%), copyright violation (17%) and breach of confidentiality (10%).
Reviewer misconduct encountered were: conflict of interest (19%), plagiarism (17%), obstructive
behavior (17%), abusive language (13%) and breach of confidentiality (13%). Notwithstanding the
limitations of the survey and the response rate, a few insights have been gained: (1) the need for
strengthening the ethical culture of researchers/authors and reviewers, (2) anti-plagiarism software
can improve plagiarism detection by about 15%, and (3) the need for technical support to detect
plagiarism, duplicate publication and image manipulation.
4.Changes in bone mineral density and bone turnover in postmenopausal osteoporotic patients treated with salmon calcitonin nasal spray

Objective To study the changes of bone mineral density(BMD)and bone turnover in postmenopausal osteoporotic patients treated with salmon calcitonin nasal spray. Methods Sixty-seven postmenopausal osteoporotic patients were enrolled in our trial. All of them received calcium and vitamin D; 37patients were treated with salmon calcitonin nasal spray for 12 months and the other 30 patients received calcium and vitamin D only. Dual-energy X-ray absorptiometry(DEXA)and measurements of a series of bone turnover indices were performed before and after medication for 6 and 12 months. Results After treatment with salmon calcitonin nasal spray for6 months, BMD in lumbar spine 2-4 increased but no change occurred in femoral neck. However, after treatment for 12 months, BMD in both lumbar spine 2-4 and femoral neck increased. In the control group, BMD in lumbar spine 2-4 decreased after treatment for 6 and 12 months, but BMD in femoral neck decreased only after 12months. Comparing with the control group, after treatment with salmon calcitonin nasal spray, BMD in lumbar spine 2-4 and femoral neck were increased obviously. The level of TRACP-5b and NTX/Cr decreased after treatment with salmon calcitonin nasal spray for6 months and 12 months, while BALP increased only after treatment for 12 months. In the control group, BALP decreased after treatment for 12 months. The level of 25-(OH)vitamin D increased after treatment for 6 months and 12 months in both groups. Conclusions Long-term treatment with salmon calcitonin nasal spray prevents bone loss and may increase bone mass.
5.De novo sequencing and analysis of root transcriptome to reveal regulation of gene expression by moderate drought stress in Glycyrrhiza uralensis.

Moderate drought stress has been found to promote the accumulation of active ingredients in Glycyrrhiza uralensis root and hence improve the medicinal quality. In this study, the transcriptomes of 6-month-old moderate drought stressed and control G. uralensis root (the relative water content in soil was 40%-45% and 70%-75%, respectively) were sequenced using Illumina HiSeq 2000. A total of 80,490 490 and 82 588 278 clean reads, 94,828 and 305,100 unigenes with N50 sequence of 1,007 and 1,125 nt were obtained in drought treated and control transcriptome, respectively. Differentially expressed genes analysis revealed that the genes of some cell wall enzymes such as β-xylosidase, legumain and GDP-L-fucose synthase were down-regulated indicating that moderate drought stress might inhibit the primary cell wall degradation and programmed cell death in root cells. The genes of some key enzymes involved in terpenoid and flavonoid biosynthesis were up-regulated by moderate drought stress might be the reason for the enhancement for the active ingredients accumulation in G. uralensis root. The promotion of the biosynthesis and signal transduction of auxin, ethylene and cytokinins by moderate drought stress might enhance the root formation and cell proliferation. The promotion of the biosynthesis and signal transduction of abscisic acid and jasmonic acid by moderate drought stress might enhance the drought stress tolerance in G. uralensis. The inhibition of the biosynthesis and signal transduction of gibberellin and brassinolide by moderate drought stress might retard the shoot growth in G. uralensis.

6.Prokaryotic expression of EV71 VP1 and initial evaluation of the biological activity

Objective To express EV71 VP1 in prokaryotic expression system,initially evaluate the ability of blocking EV71 infection and the neutralizing activity of its polyclonal antibody.Methods Construct the prokaryotic expression plasmid pET30a (+)-VP1.Induced expression in Transetta (DE3) with IPTG and identified by Western blot.After purified with HisBind Protein Purification Kit,its ability of blocking EV71 infection and the neutralizing activity of its polyclonal antibody were analyzed.Results Plasmid PET-30a(+)-VPI was constructed successfully and the objective protein was expressed effectively.The ELISA titer of the polyclonal antibody was 1:3200 while neutralizing titer was 1:16 and the recombination protein lost the ability of blocking EV71 infection.Conclusion The recombination protein can stimulate mice to produce antibodies and the polyclonal antibody shew neutralizing activity but the recombination protein lost the binding activity to receptors probably due to the wrong advanced structure.
7.Isolation,identification and sequence analysis of the strain JL/07/SW with deletion mutations of PRRSV

One strain of PRRSV was isolated from tissue of piglets died of obvious respiratory syndrome and high fever.It was identified as a strain of American type PRRSV by serological test and gene sequencing.According to American type PRRSV VR-2332,we designed 3 pairs of primers to aim directly at N gene,GP5 gene and variation sequence of NSP2.The results of sequence analysis indicated a discontinuous deletion of 30 amino acids in nonstructural protein 2 (NSP2).N and GP5 gene is conservative relatively.
8.Effect of Weifuchun on inhibiting inflammation of Helicobacter pylori-infected GES-1 cells and NF-kappaB signaling pathway.

OBJECTIVETo study the effect of Weifuchun on inflammation of Helicobacter pylori (Hp)-infected gastric epithelial cells (GES-1) and its correlation with NF-kappaB signaling pathway.
METHODSHp standard home-made strain (CagA +, VacA +) NCTCI 1637 infected GES-1 cells were used. Weifuchun was used as intervention. Weifuchun of different concentrations (5,10, and 20 microg/mL) were screened by MTT assay. A blank group and the model group were set up. Then the growth inhibition rate of drugs on gastric epithelial GES-1 cells was detected with MTT assay. Cell cycle was detected using flow cytometry. The supernatant liquid was separated to detect the contents of IL-8 and IL-4 by ELISA.The protein expression level of NF-kappaB was detected by Western blot analysis.
RESULTSMTT assay indicated significantly inhibitory effect of Weifuchun on GES-1 cells [5% inhibiting concentration (IC5)] was 10 microg/ml in the Weifuchun group. After GES-1 and Hp were cultured together,the contents of IL-8 in the supernatant were more obviously higher in the model group than in the blank group (P < 0.05), and then gradually decreased. After treatment with different concentrations of Weifuchun, the levels of IL-8 in the supernatant were less when compared with the model group at 12, 24, 48, and 72 h (P < 0.05). The decrement was the most significant in the high dose Weifuchun group. The IL-4 level in the supernatant was obviously lower in the model group than in the blank group. It obviously increased in the high concentration Weifuchun group (P < 0.05). There was no statistical difference in the IL-4 level between middle, low concentration Weifuchun group and the blank group (P > 0.05). The protein expression of intranuclear P65 increased and that of IkBalpha decreased 60 min after Hp infection. But the protein expression of intranuclear P65 decreased and the protein expression of IkBalpha increased after intervention of Weifuchun.
CONCLUSIONSWeifuchun adjusted H. pylori induced IL-8 and IL-4 production by gastric epithelial cells through blocking NF-kappaB pathways. Its mechanisms might possibly lie in inhibiting p65 from entry into nucleus and the degradation of IkBalpha. Weifuchun was an effective drug for treatment of Hp correlated chronic gastritis.

9.Immune response induced by human cytomegalovirus DNA vaccine.

Human cytomegalovirus is an important pathogen which widely infects human. In this study, an eukaryotic expression vector containing human cytomegalovirus gB680 and pp65m genes was injected into BALB/c mice to explore the immune response. Firstly, the recombinant plasmid pVAX1/gB680+pp65m and plasmid pVAX1 were transfected into COS-7 cells respectively and the transiently expressed product was detected by RT-PCR and Western blot. The maxiprepared plasmid pVAX1/gB680 + pp65m and pVAX1 by alkaline lysis with SDS and purified by Sepharose 4FF column chromatography were then used to immunize BALB/c mice and the humoral and cellular immune responses were determined. Recombinant plasmid pVAX1/gB680+pp65m could be expressed in COS-7 cells, and could induce antibodies and cellular immune responses in BALB/c mice.

The efficient and safe delivery of drugs to the therapeutic site through the biofilm has traditionally been a difficult and hot topic in the field of drug delivery. In recent years, alkyl polyglycoside (APG) have become ideal penetration enhancers for drug delivery systems because of their high permeability, good safety and biodegradability, which has attracted wide attention of domestic and foreign researchers. In this paper, the physical and chemical properties, characteristics, action mechanism and application of APG in drug delivery system are reviewed, and its application prospect in drug delivery system is prospected.
