1.The study of the relationship between cerebral infarction in the young patients and prothrombin gene G20210A mutation

Objective To investigate the relationship between the G20210A mutation of prothrombin gene and cerebral infarclion(CI) in young patients. Methods The frequency of G20210A gene mutation of prothrombin in 40 young CI patients and 48 controls were studied by polymerase chain reaction (PCR) followed by Mnl- I and Hind-Ⅲ restriction enzyme analysis. Results No prothrombin gene G20210A mutation was found in both patients and controls. Conclusion The G20210A gene mutation of prothrombin was not the risk factor of this group of young CI patients.

3.Investigation and analysis on Knowledge, Attitude and Practice (KAP) of nosocomial infection in nursing undergraduate students

Objective To investigate the current status about nursing undergraduate students' knowledge,attitude and practice (KAP)regarding nosocomial infection.Methods The self-administered questionnaires were employed to survey 108 undergraduate nursing students on the basis of a simple random sampling method.Results In the knowledge dimension,the nursing students earned 78.3% of accuracy rate when responding to the questionnaires.The students also demonstrated positive attitudes towards nosocomial infection and occupational safety,particularly the female students.In terms of practice,the students performed relatively poor as 36.1% of the students were unclear about the classification of medical garbage and 22.2% of the students used their non-clean hands to touch their eye-glasses.Conclusions The undergraduate nursing students have demonstrated adequate knowledge and proper attitude towards nosocomial infection and occupational safety,however,some behaviors need to be changed.Nursing schools and hospitals should be aware of these findings and provide more training programs regarding nosocomial infection and occupational safety so that they can help students formulate good habits to prevent and control nosocomial infection.
4.The application and exploration of PBL teaching mode in laboratory medicine practice

ObjectiveTo investigate the effects and problems of problem-based learning ( PBL ) teaching methods that are adopted in the laboratory medicine practice teaching. MethodsOne hundred and four undergraduate students of 5-year system of laboratory medicine were selected to use the PBL teaching methods during the laboratory medicine practice. ResultsIt showed that the PBL teaching methods obtained good probation effect. But teachers and students are required to make greater efforts in PBL teaching mode than in traditional mode, and the teaching methods was also needed to be consummated.ConclusionsUsing the problem-based teaching methods the comprehensive ability of the student is enhanced. Therefore it deserves to be generalized during laboratory medicine practice teaching.
5.Immunosensor construction of label free detection of CD4+T lymphocytes

Objective To construct an immunosensor for detecting CD4+ T lymphocytes without labeling .Methods The staphy-lococcus protein A(SPA) method was adopted to conduct the oriented immobilization of CD4 monoclonal antibodies on the gold in-terdigitated microelectrode surface for capturing CD4+ T lymphocytes .Then cyclic voltammetry(CV) method was used to conduct the representation of modification situation on the gold interdigitated microelectrode surface .Finally the electrochemical impedance spectroscopy(EIS) was used to detect the impedance of CD4+ T lymphocytes captured by the immunosensor .The standard curve was drawn by the impedance values change obtained by the equivalent electric circuit fitting .Results The linear range of this im-munosensor for detecting CD4+ T lymphocytes was (5 × 103 -5 .0 × 106 )/mL ,with lower detection limit of 5 .0 × 102/mL .Conclu-sion The constructed immunosensor has accurate and reliable detection results uhidn is simple to operate accurate ,convenient and cheap ,which might be expected to be used in the real-time detection system ,and offers help for realizing rapid ,accurate and inex-pensive CD4+ T lymphocyte count .

8.Rapid detection of Trichomonas vaginalis antigens by quantum dots and magnetic beads

Objective To establish a new double-antibody sandwich ELISA to detect the antigen AP33(a 33 kDa adhesive protein) of Trichomonas vaginalis based on the quantum dots and magnetic beads.Methods After BALB/c mice were immunized by AP33,the multiclonal antibodies in the antiserum was conjugated with the quantum dots and magnetic beads by carbon diimine crosslinking method respectively.Then the antibodies combined with magnetic beads were coated to microwell of plate as capturing antibody,and the antibodies bound to quantum dots were regarded as the marked antibody which can be directly observed under fluorescence microscope and quatitated by spectrofluorometry.The specificity and sensitivity of our established system were investigated.Results AP33 was successfully detected at the concentration as low as 50 ng/ml by this with-filling method.No crossreaction was observed when this system was used to detect Trichomonas vaginalis and other common bacteria in the vagina.The accuracy was 88% and the specificity was 90%.Conclusion This new double-antibody sandwich ELISA to detect Trichomonas vaginalis is successfully prepared and of sound specificity and sensitivity.
9.Cardiac atrioventricular conduction improved by autologous transplantation of mesenchymal stem cells in canine atrioventricular block models

Objective Atrioventricular block (AVB) is a common and serious arrhythmia. At present, there is no perfect method of treatment for this kind of arrhythmia. The purpose of this study was to regenerate cardiac atrioventricular conduction by autologous transplantation of bone marrow mesenchymal stem cells (MSCs), and explore new methods for therapy of atrioventricular block. Methods Eleven Mongrel canines were randomized to MSCs transplantation (n=6) or control (n=5) group. The models of permanent and complete AVB in 11 canines were established by ablating His bundle with radiofrequency technique. At 4 weeks after AVB, bone marrow was aspirated from the iliac crest. MSCs were isolated and culture-expanded by means of gradient centrifugal and adherence to growth technique, and differentiated by 5-azacytidine in vitro. Differentiated MSCs (1ml, 1.5×107cells) labeled with BrdU were autotransplanted into His bundle area of canines by direct injection in the experimental group, and 1ml DMEM in the control group. At 1-12 weeks after operation,the effects of autologous MSCs transplantation on AVB models were evaluated by electrocardiogram, pathologic and immunohistochemical staining technique. Results Compared with the control group, there was a distinct improvement in atrioventricular conduction function in the experimental group. MSCs transplanted in His bundle were differentiated into analogous conduction system cells and endothelial cells in vivo, and established gap junction with host cardiomyocytes. Conclusions The committed-induced MSCs transplanted into His bundle area could differentiate into analogous conduction system cells and improve His conduction function in canine AVB models.
10. Effects of Geniposide on the Levels of Blood Sugar and Lipids in Streptozocin-Induced Diabetic Rats

OBJECTIVE: To investigate the regulation of geniposide on blood sugar and lipids in diabetic rats. METHODS: ELISA assay was used to determine the content of insulin in vitro and in vivo, and MTT was used to determine the cell viability.RESULTS Geniposide prevented pancreatic beta damage induced by high glucose, and attenuated glucose-stimulated insulin secretion in the presence of 25 mmol•L-1 glucose. Furthermore, geniposide ameliorated insulin resistance, and decreased the contents of total cholesterol (TC) and triglyceride (TC) in STZ-induced diabetic rats. CONCLUSION: Geniposide is beneficial to improve the dysfunction of pancreatic beta cells induced by high glucose, and might be a promising lead compound for the treatment of diabetes.
