1.Progress in research on the susceptibility of tuberculosis

Tuberculosis(TB),a chronic infectious disease caused by Mycobacterium tuberculosis(MTB), is the outcome of the interaction between polygenes and environmental factors,which has been shown in lots of genetic studies. In recent years,the research of TB susceptibility genes and their single nucleotide polymorphism(SNP)loci has become a hot topic,but genes and SNPs exactly associated with TB susceptibility are rarely reported. What's more,some susceptibility genes are different among races, populations and geographical distributions, resulting in inconsistency, or even contrary conclusions drawn from local studies of different populations or areas. In order to obtain susceptibility genes that are widely applicable and exactly associated with TB,it is necessary to screen the genes preliminarily in the laboratory animals with gene diversity, and thereafter, verify those genes in animals or humans. The Collabrative Cross(CC)mice have the advantages of diverse and clear genetic background,which can provide a new tool for the research of TB susceptibility. This review summarizes the advances in the studies of TB susceptibility genes and their SNPs, and looking forward to the application of CC mice in these studies and its significance.
3.F10 expressions in cervical cancer tissues.

OBJECTIVETo detect the expression of F10 at both mRNA and protein levels in cervical cancer tissues and explore its role in the occurrence and progression of cervical cancer.
METHODSF10 expressions at mRNA and protein levels were detected in 30 pairs of cervical cancer tissues and adjacent tissues using RT-PCR and immunohistochemistry.
RESULTSThe mRNA and protein expressions of F10 were significantly higher in cervical cancer tissues than in the adjacent normal tissues (P<0.05). F10 expression was significantly higher in poorly differentiated cervical cancer than in well differentiated cancer tissues, and was also lower in patients with preoperative chemotherapy than in those without chemotherapy.
CONCLUSIONF10 expression level is inversely correlated with the differentiation of cervical cancer and possible plays a role in the tumorigenesis and progression of cervical cancer.
4.The value of core fucosylated alpha 2 macroglobulin in the diagnosis of hepatocellular carcinoma

Objective To investigate the core fucosylated alpha 2-macroglobulin(LCA-α2M) level in patients with hepatocellular carcinoma (HCC), liver cirrhosis (LC) and chronic hepatitis B (CHB), and explore its diagnostic value in HCC. Methods A total of 193 HCC patients,104 LC patients and 71 HC patients in Shanghai Eastern Hepatobiliary Surgery Hospital and 45 CHB patients in Changzheng Hospital from January 2013 to December 2016 were included for retrospective study. The method for detecting LCA-α2M was set up, and then the levels of serum α2M and LCA-α2M in each group were detected. The diagnostic value of LCA-α2M for HCC was evaluated by receiver operating characteristic (ROC) analysis. Result The level of LCA-α2M/α2M × 100(LCA-α2M%) was significantly higher in HCC patients[31.25(26.61-35.42)] than that in LC patients [26.00(22.30-30.64)], CHB patients[26.23 (23.86-31.86)] and healthy controls[20.29(17.35-22.60)] (H values were 5.626, 3.388 and 10.942, respectively, P<0.05). The area under the receiver operating characteristic curve (AUC) of LCA-α2M%for identifying HCC was 0.768 (0.725-0.808). Combined α-fetoprotein(AFP) and LCA-α2M%, the area under the ROC curve was 0.890(0.856-0.919). For AFP negative HCC patients, the sensitivity of LCA-α2M%was 77.42%(24/31). Conclusion LCA-α2M% has some values in assistant diagnosis of HCC, and could improve the detection of AFP negative HCC patients.
5.Diagnosis and treatment of cervical Castleman disease presenting as a neck mass

Objective To investigate the clinical and pathological features, diagnosis and treatment of Castleman disease (CD).Methods Clinical features and related information on diagnosis and treatment of eight cases of CD were retrospectively analyzed.The size of involved lymph nodes ranged from (2 cm×2 cm×3 cm)-(4 cm×3 cm×2 cm).The lymph nodes were found in level Ⅰ (1 case), level Ⅱ (3cases) ,level Ⅲ (3 cases) and level Ⅳ (1 case).CT examination in eight patients showed the lesions manifested as ellipse soft masses.Dynamic contrast CT scan in four patients showed ring-enhanced area around the masses.Blood routine examination in eight patients were generally normal, with six patients had mild anemia.Results Based on the clinical classification, all lesions in this group were localized CD.Histopathology indicated that all lesions were of hyaline-vascular type. After surgery, there was no recurrence during the follow-up period. ConclusionsPatients with localized CD mainly have lymphadenectasis in a single location.The CT scan can give some evidence.Surgery should be given first priority.
6.Instructional design and exploration on the experiment teaching of forensic science and biological evidence based on post competency cultivation

The traditional experimental teaching mode of forensic science and biological evidence is mostly confined to experimental operation, which is not capable of cultivating students' comprehensive quality and post competency. Therefore, it is urgent to seek an innovative teaching and training mode. At present, the experimental teaching of forensic science and biological evidence is dominated by teachers. There are some problems, such as insufficient training of students' scientific thinking and innovation ability, single teaching and evaluation model, and disconnection from the practical application. This paper proposes an experimental teaching design scheme of forensic science and biological evidence based on post competency training. The course is implanted in the framework of simulated cases, and the virtual simulation experiment platform and group discussion learning method are used to achieve a training model oriented by social needs and centered on students. In the preliminary study on the students who were trained in this mode of selected sections, we found that, compared with traditional teaching, the time for students to complete the prescribed experimental operation in this teaching mode was shortened by 4 minutes on average, and the average score of theoretical course test case analysis questions was increased by 1.5 points. In conclusion, the instructional design of the experiment teaching forensic science and biological evidence can effectively improve students' post-competency, and it deserves further exploration and application.
7.Therapeutic Effects of Transthoracic Pulmonary Artery Denervation in a Rat Model of Pulmonary Arterial Hypertension

Objectives:The aim of this study was to observe whether transthoracic pulmonary artery denervation (TPADN) could reduce the pulmonary arterial pressure and attenuate pulmonary vascular remodeling in rats with pulmonary arterial hypertension induced by monocrotaline. Methods:Twenty-four healthy male SD rats were randomly divided into control group, sham operation group and operation group (n=8 per group). Rats in sham operation group and operation group received single subcutaneous injection of monocrotaline (MCT, 60 mg/kg). After four weeks, the mean pulmonary arterial pressure (mPAP) and other hemodynamic parameters were measured with the right heart catheter in rats of these two groups. Then, operation group received the surgery of TPADN, which included thoracotomy in left 2-3 rib, exposing pulmonary artery, and removing the near connective tissue of the pulmonary artery trunk. After two weeks of operation, the mPAP and other hemodynamic parameters were measured again by the right heart catheter. The microstructure changes of the heart and pulmonary vessels was observed by immunohistochemistry and immunofluorescence. Meanwhile, RV cardiomyocyte cross-sectional area (CSA) and the right hearthy pertrophy index (RVHI= RV/[LV+S]) were used to evaluate the degree of right ventricular hypertrophy. Results:After four weeks of injection of MCT, the mPAP was significantly higher in the operation group and the sham operation group than in control group (P<0.01). Two weeks after the surgery of TPADN, the mPAP was significantly reduced in the operation group than compared in the sham group(P<0.01). Meanwhile, the percentage of medial thickness to outer diameter of the small pulmonary arterioles, right ventricular myocardial cell cross-sectional area and RVHI were also significantly decreased in the operation group compared to sham operation group(all P<0.01). Conclusions:Our results show that TPADN could reduce the mean pulmonary arterial pressure and attenuate the hypertrophy of medial thickness of small pulmonary arterioles and of right ventricle in PAH rats induced by monocrotaline.
8.Analysis of measles immunity level in persistent populations in Beijing, 2012.

OBJECTIVETo analyze the measles immunity level of persistent population in Beijing.
METHODSA total of 2125 objects from 10 age groups, who had been living in Beijing for over 6 months, were selected from urban and rural areas in Beijing in 2012. Demographic characteristics, history of measles and vaccine immunization were investigated by questionnaire. 5 ml blood sample of each subject was collected, and the Measles IgG antibody was measured by ELISA assay.
RESULTSPositive rate of measles antibody was 84.71% (1800/2125) and standardized positive rate was 88.07% . Median of antibody was 960.46 IU/L. Positive rate and median of measles antibody were significantly different between population from different age groups (χ(2) = 341.60, P < 0.01; H = 216.27, P < 0.01). Antibody positive rate and median were lowest in the <1 year age group, which were separately 43.06% (90/209) and 185.80 IU/L; and highest in the 1-4 (97.31% (181/186) and 2448.81 IU/L) and 5-9 years age group (96.46% (218/226) and 1910.72 IU/L). The range of antibody positive rate and median in adults of ≥ 15 years were 81.98%-90.14% and 744.38-1474.84 IU/L. Antibody positive rate and median in persistent population, which were separately 82.45% (883/1071) and 899.82 IU/L, were lower than those in migrant population, which were 87.00% (917/1054) and 166.19 IU/L, respectively (χ(2) = 8.51, P < 0.01;U = 538 704.00, P < 0.01). Antibody positive rate and median in population with vaccination history, which were separately 91.95% (891/969) and 1443.11 IU/L, were higher than those population without vaccination history and people whose history unknown (32.95% (57/173) , 127.33 IU/L; 86.67% (852/983) , 923.73 IU/L). The difference showed statistical significance (χ(2) = 399.92, P < 0.01; H = 202.11, P < 0.01).
CONCLUSIONAmong the persistent population in China, measles antibody level among the children aging 1-9 years old was high enough to prevent outbreak and epidemic of measles. However, we should try our best to strengthen the measles antibody level among the babies younger than 1 year old and the migrant population aging between 15 and 40 years old.

Aim To explore the mechanism of Danshen decoction in the treatment of colon cancer using network pharmacology and molecular docking.Methods The active components and corresponding target proteins of Salvia miltiorrhiza, Santalum album and Amo-mum villosum in Danshen decoction were screened based on Traditional Chinese Medicine Systems Pharmacology database and analysis platform.The targets of colon cancer were searched by using Genecards database, and the common targets were selected.The network diagram of traditional Chinese medicine-active components-target-disease was constructed by using Cytoscape 3.7.0.The protein protein interaction network of common targets was constructed by using STRING database.The gene ontology(GO)and Kyoto Encyclopedia of Genes and Gnomes(KEGG)enrichment analysis were carried out based on R4.0.2.The important targets in the key pathways and the important active components in the network diagram of traditional Chinese medicine-active ingredients-target-disease network were selected for Surflex Dock.Results A total of 78 active components, 142 targets, 3 239 colon cancer targets, 105 overlapping targets and 69 corresponding active components were screened out.KEGG analysis showed that the key signaling pathway was PI3K/AKT.Luteolin and Tanshinone IIA with high correlation were selected to dock with protein kinase B(AKT1).Both active components had hydrogen bonding with AKT1.Conclusions Danshen decoction plays a positive role in colon cancer treatment.The mechanism may be related to the regulation of PI3K/AKT signaling pathway.
10.Two cases of neonatal Legionella pneumonia

Patient 1,a 12-day-old female infant,presented with fever,cough,dyspnea,and elevated infection markers,requiring respiratory support.Metagenomic next-generation sequencing(mNGS)of blood and bronchoalveolar lavage fluid revealed Legionella pneumophila(LP),leading to diagnoses of LP pneumonia and LP sepsis.The patient was treated with erythromycin for 15 days and azithromycin for 5 days,resulting in recovery and discharge.Patient 2,an 11-day-old female infant,presented with dyspnea,fever,elevated infection markers,and multiple organ dysfunction,requiring mechanical ventilation.mNGS of blood and cerebrospinal fluid indicated LP,leading to diagnoses of LP pneumonia,LP sepsis,and LP intracranial infection.The patient was treated with erythromycin for 19 days and was discharged after recovery.Neonatal LP pneumonia lacks specific clinical symptoms,and azithromycin is the preferred antimicrobial agent.The use of mNGS can provide early and definitive diagnosis for severe neonatal pneumonia of unknown origin.
