2.Correlation between ApoE gene polymorphism and cardio-cerebrovascular diseases

Objective To investigate the correlation between apolipoprotein E gene polymorphism and car-dio-cerebrovascular diseases. Methods Gene chip method was used to determine ApoE genotypes in 1427 pa-tients with cardiovascular disease and 450 health controls. Levels of serum lipid were compared. Results The highest genotype frequency of ApoE was ε3/3 in the patient group and control group. The allele frequency of ApoE from high to low wasε3,ε2 andε4. The genotype ofε3/3 in patient group was significantly lower than that in con-trol group(χ2 = 12.562,P < 0.01),whileε3/4 was significantly higher than that in control group(χ2 = 36.112, P < 0.01). No significant differences were found between patient groups in genotype or allele frequency of ApoE. The level of TCH,TG,LDL-c and sdLDL in the patient group were significantly higher than those in the control group,and HDL-c was significantly lower than those in the control group(P>0.05). The level of TCH and LDL-c significantly decreased in patients with E2 genotype,and increased in patients with genotype E4(P < 0.05). No significant differences were found in other indexes(P>0.05). Positive relationship was found between ApoE geno-type and phenotype of ApoE and LDL-c. Conclusions ApoE gene polymorphism plays an important role in the oc-currence and development of cardio-cerebrovascular diseases.
3.Effects of antenatal corticosteroids on the apoptosis of neurons and the expression of Bcl-2 gene in neonatal rat brain

Objective To investigate the effect and the possible mechanism of the apoptosis of neurons and the expression of bcl-2 gene in the neonatal rat brain after different courses of Dexamethasone (DXM). Methods Thirty-four Sprague-Dawley(SD) pregnant rats were randomized into four groups and received different courses of antenatal DXM at the dose of 0.8mg/(kg?d) intramuscularly: (1) Group 1 (n=8) received DXM on the 17th day of gestation only; (2) Group 2 (n=8) received DXM consecutively on the 17th,18th and 19th day of gestation; (3) Group 3 (n=10) received DXM consecutively for 4 days(on 17th,18th,19th and 20th day of gestation); (4) The control group (n=8) received equivalent volumes of isotonic saline intramuscularly consecutively for 4 days (gestational day 17, 18, 19, 20). On the postnatal day 1(P_1), day 7 (P_7) and day 14 (P_ 14 ), sixteen newborns in each group were randomly selected and sacrificed. The weights of the body and the whole brain were measured. Tunel was used to evaluate the apoptosis of neurons, and the content of bcl-2 protein was examined by improved Western Blot. Results (1) The brains weight of the newborn mice on P_1 were (0.301?0.030)g, (0.302?0.026)g, and (0.296?0.025)g in Group 1, 2 and 3, respectively and all significantly lower than that of the control [(0.363?0.041)g, P0.05). Conclusions Repeated doses of antenatal DXM therapy could significantly increase the apoptosis of neurons by down-regulate the bcl-2 expression and handicap the development of newborn mice′ brain.
4.Study on pharmacodynamics and toxicology of Gejie Dingchuan Capsule

Object To inquire into the effect on pharmacology of Gejie Dingchuan Capsule (GJDCC). Methods The observation of relieving cough and asthma, removing phlegm, immunity, antibiosis and antiinflammation, antianaphylaxis, and acute and long-term toxicity was carried out. Results GJDCC could resist the convulsion of the isolated trachea of guinea pigs, lengthen the asthma incubation period of guinea pigs, increase the phlegm liquid secreting capacity of isolated rat trachea, promote the pigeon trachea cilium motion, lengthen the mice cough incubation period, restrain the mice ear swelling and rat swelling hyperplasia of granulation, promote the mice producing of serum hemolysin and the lymphocyte conversion rate, and reduce the guinea pig allergic reaction index and shock death rate. It possessed the bacteriostasis, the biggest capacity of bearing consumption was 500 times the clinical daily use and in long-term toxicity test there were not the clear toxicity reactions. Conclusion GJDCC has the function of relieving asthma and cough, removing phlegm, antibiosis and antiinflammation, antianaphylaxis and immunity, without poisonous side effect.
5.Clinical value of soluble triggering receptor expressed on myeloid cells-1 in early diagnosis and prognosis of ventilator-associated pneumonia

Objective This study examined the value of soluble triggering receptor expressed on myeloid cells-1 (sTREM-1)in serum and exhaled ventilator condensate (EVC)in early diagnosis and prognosis of ventilator-associated pneumonia (VAP). Methods A total of 37 adult patients undergoing mechanical ventilation were evaluated after treatment,including 24 patients with infection,13 without infection.Of the 24 patients with infection,10 patients were assigned to ineffective subgroup and 14 to effective subgroup.Levels of sTREM-1 in serum and EVC were measured on days 1,3,5,and 7 for all patients.sTREM-1 in serum and EVC were determined in patients by double antibody sandwich enzyme-linked immunosorbant assay (DAS-ELISA).The early diagnostic and prognostic value was assessed by receiver operating characteristic (ROC)curve analysis.Re-sults On Day 1,the sTREM-1 levels in serum and EVC did not show significant difference between the three groups (P>0.05).On Day 3 and Day 5,the level of sTREM-1 in the infection group was higher than that in the non-infection group (P <0.01).On Day 7,the sTREM-1 levels of serum and EVC in ineffective subgroup were higher than those in the effective subgroup and non-infection group (P <0.01). There were no difference between the non-infection group and effective subgroup (P >0.05).For ROC analysis,area under the curve (AUC)of serum sTREM-1 was 0.897,and AUC of EVC sTREM-1 was 0.909 on Day 3.When the cut-off value of EVC sTREM-1 was set at 4.70 ng/mL on Day 3,the sensitivity was 85.8%,specificity was 92.3%.Conclusions The results suggest that the levels of sTREM-1 in serum and EVC are conducive to the early diagnosis of VAP.The levels of sTREM-1 in serum and EVC on Day 7 are helpful for estimating the prognosis.EVC sample is easier to collect than serum.
6.Relationship between the single nucleotide polymorphisms in has-mir-125a-5p rs12975333 and expression of has-mir-125a-5p in Han Chinese female breast cancer patients

Objective To investigate the relationship between the single nucleotide polymorphisms (SNPs) in has-mir-125a-5p rs12975333 and the expression of has-mir-125a-5p and clinicopathological cheracteristics of female breast cancer in Han Chinese women. Methods Genomic DNA was extracted from peripheral blood lymphocytes. taqman-MGB assay was used to type breast cancer of 338 cases and 338 controls. Expression levels of has-mir-125a-5p in 289 biopsies were examined using stem-loop real-time RTPCR and the clinicopathological cheracteristics of breast cancer were evaluated. Result The gene frequencies (GG,GT,TT) of rsl2975333 in the patients were GG 273 (94. 5% ), GT 16 (5.5%),TT 0(0%), while in breast fibroadenoma and controls there were GG 49 ( 100% ), 338 (100%). The expression level of has-mir-125a-5p in breast cancer(0. 19 ±0. 04) was lower than that in the matched nontumor adjacent tissue specimens (0. 37 ± 0. 05 ) ( P = 0. 04 ) .The expression level of minor T allele of mature miR-125a in breast cancer patients was lower than that in has-mir-125a-5p-GG carying(P =0.022). The expression of has-mir-125a-5p was down-regulated in primary breast cancer, especially in elder patients ( P = 0. 036) and lymph node metastasis groups (P = 0. 001) and with negative ERBB2 (P = 0. 007), ERBB3 (P =0. 04). Conclusions rs12975333 polymorphisms in has-mir-125a-5p gene may work as a risk factor of breast cancer in Han Chinese women. The altered expression of has-mir-125a-5p might play a role in the pathogenesis and progression of breast carcinoma.
7.Effect of RNA interference for OPN on MDA-MB-231 cells and the growth of transplanted tumor in nude mice

ObjectiveTo investigate the role of OPN in human breast cancer cell line ( MDA-MB-231) by using small interfering RNA to specifically knockdown OPN expression. MethodsOPN ShRNA expression vector was stably transfected to MDA-MB-231 cell line.The expression of OPN mRNA and protein were analyzed using reverse transcription polymerase chain reaction (RT-PCR)and Western blot,respectively.The growth of MDA-MB-231 cells were observed by MTT.The effect of OPN siRNA on the transplanted tumor growth and tumor hypoxia were assessed in nude mice. ResultsThe expression level of OPN in MDA-MB-231 cells were significantly lower under hypoxia or normoxia(P < 0.05 ).OPN silence with RNAi significantly inhibited the invasion ability and proliferation of MDA-MB-231 cell lines (P < 0.01 ).Inhibition of OPN with RNAi significantly inhibited the growth ability of MDA-MB-231 cells in vivo(P <0.05).The tumor hypoxia significantly decreased(P < 0.05). ConclusionsOPN silence with RNAi can effectively inhibit cell proliferation and tumor growth of MDA-MB-231 cells,and decrease the bypoxia level of MDA-MB-231 transplanted tumor in nude mice.

【Objective】The chemical constituents of Viscum liquidambaricolum Hayata.were studied to screen the constituents with antitumor activity.【Methods】Chromatography was used for the isolation and purification of chemical constituents of Viscum liquidambaricolum Hayata..And their structures were identified on the basis of spectroscopic evidences and physiochemical properties.Meanwhile,the in-vitro antitumor activities of two kinds of triterpenoids and one kind of sitosterol were investigated.【Results】Four compounds were isolated from Viscum liquidambaricolum Hayata.and their structures were identified as genkwanin(1),?-amyrin acetate(2),erythrodiol(3) and ?-sitosterol(4).Compounds 2 and 3 had an inhibitory effect on mice sarcoma 180(S180),Ehrlich ascites carcinoma(EAC) and ascties hepatoma(HeAP),and compound 4 had an inhibitory effect on mice S180 and EAC,their half-inhibitory concentrations being less than 400??g/mL.【Conclusion】Compounds 1~4 are isolated from Viscum liquidambaricolum Hayata.for the first time,and compound 1 is isolated from Loranthaceae for the first time.Triterpenoids and steroids from Viscum liquidambaricolum Hayata.exert certain antitumor activity.
9.Study on Hemolysis,Allergic Reaction and Stimulation of Polydatin Injection

Objective To observe the hemolysis,allergic reactions and stimulation of Polydatin Injection(PI),thus to provide evidence for clinical medication.Methods Hemolysis was observed by in-vitro rabbit blood hemolysis.Sys- temic allergy and passive skin allergy were tested on guinea pigs and rats.Vascular stimulation and muscle stimulation were tested on rabbits.Results There was no hemolysis or reaction in erythrocyte agglutination 30,60 and 180 rain after adding PI.No systemic anaphylaxis was found in guinea pigs.There was no skin allergy passive in rats.There was no vascular stimulation or muscle stimulation after injecting PI(0.39 mg/mL)5.6 mg/kg into rabbit ears.Con- clusion Under experimental conditions,PI(0.39 mg/mL)shows no hemolysis in vitro or agglutination reaction;it has no systemic anaphylaxis or passive skin allergy;it either has no stimulating effect in rabbit auricular vessels and muscles.
10.Anti-osteoporotic effect of raloxifene on mice with osteoprotegerin gene deficiency

Objective To observe the effect of raloxifene, a selective estrogen receptor modulator, on osteoporosis in the osteoprotegerin (OPG) gene knock-out female and male mice. Methods Two groups of OPG gene deficient (OPG-/-) female and male mice, 20 mice in each group, were assigned to raloxifene-treated (3 mg The effect of raloxifene was evaluated by comparing the values of bone mineral density (BMD) , bone strength,histomorphometric measurement and osteoclast number between the raloxifene treated group and placebo group.Results As compared with placebo group osteoporotic manifestations were improved in OPG-/- female mice treated with raloxifene orally. BMD was increased both in lumbar vertebrae (P<0.05) and femurs (P<0.01).Bone strength was measured in femurs by three-point bending test and vertebrae by stress test. Results showed that ultimate load, ultimate stress and Young's modulus were increased both at lumbar and femur bone, suggesting decreased risk of fracture. Tartrate-resistant acid phosphatase, a marker enzyme of osteoclasts, was detected, and the number of osteoclasts declined significantly after the treatment of raloxifene. At the same time, results of histomorphometric measurements indicated that bone trabecular volume was increased and bone formation rate decreased from(8.05±4.02)mm3·mm-2·year-1 to (5.48±1.89)mm3·mm-2· year-1(P<0.05).These findings were found in the group of OPG-/- female mice treated with reloxifene but not in male mice. Conclusions Raloxifene is effective in treating osteoporosis in female OPG-/- mice, indicating that its action is at least in part independent of OPG gene. But it is ineffective in male OPG-/- mice.
