Objective To investigate the expressions of estrogen receptor (ER), progesterone receptor (PR), human epidermal growth factor receptor 2 (HER-2) and relationships of these expressions with the clinicopathology in breast cancer patients aged 70 years old and over. Methods The expressions of ER, PR and HER-2 were detected by immunohistochemical staining from 124 elderly breast cancer patients, and the relationships of these expressions with pathological classification, tumor size, lymph node metastasis and TNM stage were analyzed. Results Among 124 patients, the positive expression rate of ER and PR was 75.0% (93/124) and 71.0% (88/124),respectively. They were not associated with pathological classification, tumor size, lymph node metastasis or TNM stage (all P>0. 05). The positive expression rate of Her-2(+++) and Her-2 (++) was 11.3% (14/124) and 24.2% (30/124), respectively. And the expression of Her-2 was significantly associated with tumor size, lymph node metastasis and TNM stage (P<0. 05) but not with pathological classification (P>0. 05). Conclusions The positive expressions of ER and PR are not correlated with the clinicopathology in elderly breast cancer patients. Her-2 overexpression is positively associated with tumor size, lymph node metastasis and TNM stage.

The inhibitory effect of the crude phycoerythrin(PE) on the frequency of PCE micronucleus induced by CP and its antitumor effect on mice bearing S 180 sarcoma were studied. The result showed that the PE could decrease significantly the frequency of PCE micronucleus with a fair dose-effect relationship, and showed obvious antitumor effect on S 180 bearing mice with an inhibitory rate 44%.
3.The expression and function of miR-145 in papillary thyroid carcinoma

Objective To study the expression and function of miR-145 in papillary thyroid carcinoma (PTC).Methods PTC tissues and adjacent tissues were collected from 43 cases.Expression of miR-145 in PTC tissues and adjacent tissues was detected with RT-PCR.miR-145 analogue was used to transfect TPC-1cell to upregulate miR-145 expression.Brdu-ELISA method was used to detect the proliferation of TPC-1 cell.Flow cytometry instrument was used to detect the apoptosis and cell cycle of TPC-1 cell.Results RT-PCR test showed that relative expression of miR-145 in thyroid carcinoma tissue was 0.369±0.082,significantly lower than 1.029-±0.365 in tissue adjacent,and the difference was statistically significant (t=3.129,P=0.000).The expression of miR-145 in patients whose biggest tumor size ≥ 1 cm was lower than patients whose biggest tumor size <1 cm.Compared with patients with single tumor,the expression of miR-145 in patients with multiple tumor was lower,and the dif ferences were statistically significant (P<0.05).miR-145 expression was enhanced by miR-145 analogue.Compared with negative control,the proliferative ability of thyroid cancer cell TPC-1 was suppressed significantly,and the difference was statistically significant (P<0.05).In addition,up-regulation of miR-145 expression could block thyroid cancer TPC-1 in G2/M phase.The apoptosis rate of thyroid cancer cell TPC-1 increased significantly (P<0.05).Conclusions miR-145 expression is decreased significantly in PTC tissue,and is associated with clinical pathological features.Up-regulation of miR-145 expression can inhibit thyroid cancer cell proliferation,block the cell cycle,and promote apoptosis,miR-145 may play an important role in occurrence and development of thyroid cancer.
4.The significance of AgNORs of peripheral T lymphocytes in the diagnosis of elderly cancer

Objective To investigate the diagnostic significance of argyrophilic nuclear organizer regions(AgNORs) of peripheral T lymphocytes in elderly cancer patients. Methods AgNORs was examined using KL tumor image analysis system in 45 cases of health control,36 cases of elderly inflammatory patients,97 cases of elderly cancer patients and 30 cases of elderly tumor-cured patients. CD3,CD4,CD8,CD4/8 and NK were detected by flow cytometer in cancer patients. Results Compared with normal controls,inflammation and tumor-cured patients,AgNORs was decreased extremely in elderly patients with cancer. Moreover,AgNORs was decreased in tumor metastasis status. Conclusions In analyzed elderly cancer patient cases,the expression of AgNORs is significantly decreased. The AgNORs may serve as a marker for judgment of clinical treatment and supervisor.
5.Current status and influencing factors of health behaviors in stroke patients

Objective:To investigate the current status and influencing factors of stroke patients.Methods:Using stratified sampling method, 458 stroke patients was investigated for the health behavior from October to December, 2015. Based on PROCEDE-PROCEED model, logistic regression analysis was conducted with the health behavior level of the subjects as the dependent variable, and the characteristic factors in the chi-square test as the independent variables to analyze the factors which influence the health behavior level of stroke patients from predisposing factors, enabling factors and reinforcing factors.Results:The health behavior score of stroke patients was (14.5±3.9), which was at medium level. The score of each dimension was nutrition (0.80±0.25), disease management (0.60±0.26), interpersonal relationship (0.57±0.33), and medication compliance (0.49±0.41). The multiple logistic regression analysis showed that the predisposition factors including age ( OR=2.03, 95% CI=1.08-3.80) and history of hypertension( OR=2.35, 95%=1.16-4.76), the enabling factors including the allocation of social resources ( OR=1.73, 95% CI=0.89-3.36), and the reinforcing factors including family support ( OR=2.03, 95% CI=0.99-4.17) were important factors which affect the health behavior of stroke patients. Conclusion:The health behavior level of stroke patients need to be improved. The relevant community resource allocation system continuously, and focus on the impact of family environment and particularly the elderly and patients with a history of hypertension should be attentioned when improving the health behavior of patients.
6.The effect of combination treatment by paliperidone and psychological intervention on first-episode schizophrenic out-patient

Objective To evaluate the effect of combination treatment by psychological intervention and a new atypical antipsychotic Paliperidone extended-release tablets(paliperidone ER)on first-episode schizophrenic out-patients.Methods All 62 patients diagnosed as first-episode schizophrenia by DSM-Ⅳ were randomly allocated into,combination treatment group and control group,for a 12-month treatment study.The combination treatment group was treated by paliperidone ER once a day and psychological counseling once a month,while the control group was treated only by paliperidone ER.The efficacy and social function were assessed by Positive and Negative Syndrome Scale(PANSS)and Personal and Social Performance Scale(PSP)individually at baseline,and following 1,3,6,9,12 months.The remission rate was calculated at the end of the 3rd,6th,9th month.The safety and tolerability were assessed using Barnes Akathisia Scale(BARS),extrapyramidal side effects scale(SAS)and involuntary movement scale(AIMS).Results Twenty-seven patients of combination treatment group and seventeen patients of the control group completed the trial with the discontinuation rate 12.9% and 45.2%,respectively.The difference between the above two groups was statistically significant(P<0.05).The mean total score of PANSS and the PSP of the both groups were significantly improved(P<0.05)after treatment.PANSS total score reduced more than 30% compared with baseline,and PSP total score increased more than 7.The remission rate of combination treatment group was 74.2%,51.6% and 67.7% at the end of 3rd,6th,and 9th month,which was better than the control group(45.2%,38.7% and 38.7%)(P<0.05).Both incidence and types of adverse events were almost same between the two groups.The major adverse events were extrapyramidal symptoms,tachycardia,gastrointestinal discomfort and akathisia.Conclusion Paliperidone ER treatment combined with psychological intervention on first-episode schizophrenia out-patients can improve their treatment compliance by reducing the discontinuation rate.And this combination treatment can increase the remission rate and improve social functioning of the patients.
7.Laboratory investigation of reactive plasmacytosis in patients with severe fever with throbocytopenia syndrome

Objective To investigate the immunological characteristics and clinical significance of reactive plasmacytosis in patients with severe fever with throbocytopenia syndrome (SFTS).Methods Bunyavirus-infected patients who were diagnosed with SFTS were collected from March 2015 to October 2015 in Taizhou Hospital.Morphology analysis of bone marrow and peripheral blood (PB) smear, as well as flow cytometry analysis of plasma cell immune phenotype from peripheral blood were conducted.Serum immunoglobulin levels and helper T hymphocytes (Th)1/Th2 cytokine expressions were detected.Mann-Whitney U test was used.Results PB plasma cells from all of the SFTS patients increased in varying degrees, and the phenotype of the plasma cells was CD19+CD38++CD45+CD138+, which indicated normal mature plasma cells.The ratio of PB plasma cells was >0.030 in 10/16 patients, and >0.300 in 2/16 patients.The ratios of PB plasma cells in the patients with severe and critical groups were significantly higher than that in the mild group (0.052 vs 0.016, P<0.05).Monocytoid histiocytes and hemophagocytes were observed in the BM morphology of 9 patients.Three of them were diagnosed as hemophagocytic lymphohistiocytosis (HLH).The ratio of plasma cells was more than 0.030 in the bone marrow of 8 patients.The serum levels of interlewkin (IL)-6, IL-10 and interferon (IFN)-γ in acute phase were significantly elevated with the median level of 49.75 ng/L, 26.98 ng/L (reference value 2.6 to 4.9 ng/L) and 17.57 ng/L, respectively.While the levels of IL-2, IL-4 and twmor necrosis fautor(TNF)-α were not significantly changed.The serum IL-6 and IL-10 levels in the patients with severe and critical groups were both significantly higher than those in the mild group (IL-6: 132.36 vs 22.81 ng/L;IL-10: 75.28 vs 6.33 ng/L, both P<0.05), but the difference of IFN-γ level was not significant (P>0.05).The serum IgG, IgA and IgM levels did not increase in acute stage, with the median of 11.6 g/L, 2.56 g/L and 1.60 g/L (reference value 0.46 to 3.04 g/L), respectively.Conclusion The patients with SFTS show excessive humoral and cellular immunity, and the severity of disease is positively correlated with the ratio of peripheral plasma cells and the levels of cytokines IL-6 and IL-10.
8.Expression and significance of miR-21 and PDCD4mRNA in human colorectal cancer

Objective To investigate the expression of miR-21 and PDCD4mRNA in colorectal cancer and their correlation with clinicopathological characteristics,to elucidate relationship between PDCD4and miR-21 in vivo.Methods Expression of RNA (including miR-21and PDCD4mRNA) and PDCD4 protein were detected respectively by quantitative real-time reverse transcriptase-PCR and SP immunohistochemical staining in 43 cases of colorectal carcinoma and corresponding normal mucous membrane tissue.Results In colorectal carcinoma,expression of miR-21 was higher than that of the control ( P < 0.05 ),and expression of PDCD4mRNA lower than the control ( P < 0.05 ).Expression of miR21 was associated with lymph node metastasis and clinical stages ( Ⅰ + Ⅱ,Ⅲ + Ⅳ ) ( P < 0.05 ).On the other hand,no significant differences were observed regarding sex,tumor site,size,local invasion,distant metastases,clinical stages( Ⅰ,Ⅱ ) (P > 0.05 ).Expression of PDCD4mRNA was associated with local invasion,clinical stages ( Ⅰ,Ⅱ ) ( P < 0.05 ).While no significant differences were found concerning sex,site,size,lymph node metastasis,distant metastases,clinical stages( Ⅰ + Ⅱ,Ⅲ + Ⅳ ) ( P > 0.05 ).miR21 levels was negatively correlated with PDCD4 expression including nuclear and nuclear and cytoplasmic put together (P < 0.05 ),in contrast to PDCD4mRNA and PDCD4 expression in cytoplasmic ( P > 0.05 ).Conclusions ( 1 ) Abnormal expression of PDCD4 mRNA and miR-21 correlate with prognosis in colorectal cancer.(2) miR-21 suppress translation of PDCD4mRNA by binding to the 3'-untranslated region (3'-UTR)of target PDCD4mRNA.
9.Expression of RUNX3 and miR-130b in gastric carcinoma and clinical significance

ObjectiveTo explore the expression of miR-130b and RUNX3mRNA in human gastric carcinoma and the clinical significance. MethodsThe expression of miR-130b and RUNX3mRNA were detected by RT-PCR in 40 cases of gastric carcinoma and corresponding normal mucosa tissue. The expression of RUNX3protein was determined by immunohistochemistry SP method. ResultsThe expression of miR-130b was significantly up-regulated in gastric carcinoma than that in the adjacent normal gastric mucosa tissues (2.18 ± 3.75 ) vs.( 2.59 ± 3.45 ),P < 0.05 ; The expression of RUNX3mRNA in gastric carcinoma tissues was significantly lower than that in adjacent normal gastric mucosa tissues( 8.76 ±2.82) vs.( 7.58 ± 2.87 ),P < 0.05.The expression of miR-130b and RUNX3mRNA were positively correlated with lymph node metastasis and clinical stage ( P < 0.05 ) ; No significant association was found between the expression and age,gender,tumor size,distant metastasis and depth of tumor invasion ( P >0.05 ).The expression of miR-130b was negatively correlated with RUNX3 protein expression in nuclei and cytoplasm (P < 0.05 ).ConclusionsAbnormal expression of miR-130b and RUNX3mRNA correlates with prognosis of gastric carcinoma; Decreased RUNX3 protein expression may contribute to tumourigenesis.
10.Tissue culture and polyploidy induction of Morinda officinalis.

OBJECTIVETo establish an effective protocol for plant generation and induce polyploidy of Morinda offcinalis.
METHODCallus was induced from immature embryo of M. officinalis and polyploidy was inducted by using colchicine treatment method. Chromosome was detected by flow cytometry.
RESULT AND CONCLUSIONThe highest induction rate of polyploidy was 18.40%, which was obtained with 500 mg x L(-1) colchicine treatment for 5 days. Roots of polyploid were bigger than diploid. Advantages of using immature embryo as explants are easy for sterilization, higher rate of callus induction and low degree dedifferentiation. The induced polyploidy of M. officinalis may have a value for spread of cultivation.
