Objective:To investigate the standardization construction of nursing information terminologies in Guizhou Province and provide reference for constructing a standard nursing terminology system in Guizhou Province. Methods:Using the convenient sampling method, a total of 153 nursing managers from hospitals at all levels in Guizhou Province were selected as the research objects in June, 2021. The research objects were surveyed using a self-developed survey questionnaire on the current status of standardized terminology construction for hospital nursing information. The questionnaire consisted of two parts. The first part was the basic information of the hospital and the general information of the person in charge of nursing information. The second part was the construction of standardized terminology for nursing information, including three dimensions, such as the basic dataset of nursing electronic medical records, the shared document of nursing electronic medical records and the construction of nursing information terminologies interconnection. Multiple linear regression analysis was used to analyze the influencing factors of standardized construction of nursing information terminologies. A total of 153 questionnaires were distributed in this study and 148 valid questionnaires were collected, with an effective response rate of 96.73% (148/153) .Results:The total score of the survey questionnaire on the standardization of nursing information terminologies construction in 148 hospitals in Guizhou Province was (102.22±24.42) . The basic dataset dimension score of nursing electronic medical records was (60.35±13.32) , the shared document dimension score of nursing electronic medical records was (27.78±19.08) and the dimension score of nursing terminology interconnection construction was (14.08±2.71) . The results of multiple linear regression analysis showed that hospital level and whether nursing managers had received terminology training were the influencing factors for the score of hospital nursing information standardization terminology construction ( P<0.05) . Conclusions:The standardization construction of nursing information terminologies in medical institutions in Guizhou Province is still at the primary level of information construction. The insufficient attention paid by secondary hospital managers to the standardization construction of nursing information terminologies and the lack of understanding of standardized nursing terminology by nursing information managers are important influencing factors.

Calcium-binding protein is an indispensable protein which performs extensive and important functions in the growth of Schistosoma japonicum. Based on our primary study on tegument surface proteins of S. japonicun, a cDNA encoding a 66 kDa calcium-binding protein of S. japonicum (Chinese strain) was cloned, sequence analysis revealed that it was identical with that of SjIrV1 of Philippines strains S. japonicum. The expression of SjIrV1 were detected by Real-time PCR, using cDNA templates isolated from 7, 14, 21, 28, 35 and 42 days worms and the results revealed that the gene was expressed in all investigated stages, and the mRNA level of SjIrV1 is much higher in 42 d female worms than that in 42 d male worms. The cDNA containing the open reading frame of IrV1 was subcloned into a pET28a (+) vector and transformed into competent Escherichia coli BL21 for expression. The recombinant protein was purified using a Ni-NTA purification system, and confirmed by high performance liquid chromatography (RP-HPLC) and tandem mass spectrometry (MS/MS). Western blotting analysis showed that recombinant SjIrV1 (rSjIrV1) could be recognized by the S. japonicum infected mouse serum and the mouse serum specific to rSjIrV1, respectively. Immunofluorescence observation exhibited that SjIrV1 was mainly distributed on the tegument of the 35-day adult worms. ELISA test revealed that IgG, IgG1 and IgG2a antibodies are significantly increased in the serum of rSjIrV1 vaccinated mice. The study suggested that rSjIrV1 might play an important role in the development of S. japonicum.

Objective To study the influence of SCNIA intronic mutations in mRNA splicing in febrile seizures related epilepsy, and investigate the association between splicing changes and genotype-phenotype-inheritance pattern. Methods Molecular cloning of 5 SCN1A intronic mutations was performed in patients with partial epilepsy with antecedent febrile seizures plus (PEFS+) and Dravet syndrome (DS) through constructing mutant and wild-type plasmids of pTragetE2-3-4-5 and E24-25-26 by using Minigene splicing assay, and the in vitro expressions in HENK293 cells were detected. The mRNA splicing changes were analyzed qualitatively and quantitatively by reverse transcription (RT)-PCR and real time quantitative (q)-PCR.Results (1) Using RT-PCR,DS mutants presented a whole exon skipping without significant remain of normal mRNA transcripts, while PEFS+ mutants showed partial exon skipping or intronic insertion with coexistence of normal and aberrant mRNA transcripts.(2) Statistical differences were found between relative quantity (RQ) of aberrant and normal mRNA in PEFS+ mutant (c.473+5G>A:4.92%±1.05% and 6.10%±0.21%;c.473+5G>C:7.97%±1.12% and 3.94% ±1.25%) and that in DS mutant (c.602+1G>A:60.51%±1.81% and 0.060%±0.022%,P<0.05);similarly,there were statistical differences between relative RQ of normal and aberrant mRNA in PEFS+ mutant c.4853-25T>A (71.22%±11.92% and 7.38%±1.61%) and that in DS mutant c.4853-1G>C (0.08%±0.01% and 22.11%±2.83%,P<0.05).Conclusion The position and difference of splicing patterns of SCNIA intronic mutations are potential molecular pathogenesis for phenotypic difference of febrile seizures related epilepsy.
