1.Detection and separation of side population cells in haematological tumor cell lines

BACKGROUND:Side population(SP)cells,with varied contents,are widely distributed in adult tissues,embryos,and certain tumor cells.Haematological tumor is one of the main pathological conditions,which endangers human life.Thus,it is important to recognize SP cells in haematological tumor cell lines.OBJECTIVE:To identify whether hematologic tumor cell lines contain SP cells,and to explore a stable detection and separation methods.METHODS:Cells with the characteristics of stem cells being capable of fluorescent dye Hoechst33342 were isolated by flow cytometry;whether there were SP cells in logarithmic growth period of NB4,Raji,K562/ADM and K562 or not were detected.After sorting K562 subpopulations,the purity of sorted cells was detected.RESULTS AND CONCLUSION:After Hoechst33342 staining,flow cytometry results showed that the SP cells appeared in the haematological tumor NB4,Raji,K562/ADM and K562 cell lines,which accounted for 0.8%,2.7%,1.3% and 2.7%,respectively.These cells could be blocked by Verapamil.The purity was greater after a second detection of SP and Non SP cells in K562 cells.
2.The effects of hesperidin in caries prevention in rats

Objective:To investigate the anticaries effects of hesperidin in rats.Methods:The MIC of hesperidin against Streptococ-cus sobrinus(S.sobrinus)was explored with disc diffusion method.The rats with artificial caries were administered with 1/2 MIC hes-peridin,0.12%Chlorhexidine and distilled water respectively.The results of S.sobrinus level,Keyes scoring and DIAGNOdent exam-ination were used to evaluate the effects of hesperidin on S.sobrinus and caries development.Results:The MIC of hesperidin agaist S. sobrinus was 8 mg/ml.The S.sobrinus level was not statistically different between hesperidin group and negative control group(P>0. 05).Keyes scores of grade E of smooth and pit and fissure caries in hesperidin group were lower than in negative control(P<0.05), those of grade Ds and Dm of pit and fissure caries in hesperidin group were lower than in negative control(P<0.05 ),while higher than in chlorhexidine group(P<0.05).Hesperidin showed significant anti-caries effect(P<0.05)examined by DIAGNOdent.Con-clusion:Hesperidin at 1/2 MIC has anti-caries effect in rats without influence of the oral microecological balance.
3.Determination of osthole and imperatorin in Compound Shajiziyou Suppository by RP-HPLC

AIM:To develop a HPLC method of determining osthole and imperatorin in Compound Shajiziyou Suppository(oil of Semen Hippophae,Fructus Cnidii,Radix Sophorae flavescentis,etc.). METHODS: Phenomnex(luna) C_(18) column was used at(25 ?C).The mobile phase consisted of methanol-water(65(∶)35).The flow rate was 1.0 mL/min.The detection wavelength was set at 310 nm. RESULTS: The linear ranges of osthole and imperatorin were 0.051-0.816 ?g(r=0.999 6) and 0.026-0.416 ?g(r=0.999 8),respectively,The average recoveries were 98,4% with RSD of 1.4% and 97.6% with RSD of 2.0% respectively. CONCLUSION: This method is simple,accurate,reproducible and can be used for the determination of osthole and imperatorin in Compound Shajiziyou Suppository.
4.Real-time RT-PCR for detection and quantification of AML1/ETO leukemia fusion gene

Objective To set up real-time quantitative RT-PCR technique and measure leukemia fusion gene transcripts in patients with AML of FAB-M_2 subtype,and also to investigate the positive rate in patients and the relationship between the AMLI/ETO mRNA levels and the response rate after chemical therapy.Methods The plasmid containing the AMLI/ETO fusion gene sequences were constructed from myeloid cell lines Kasumi-1 (expressing AML1/ETO)to establish the standard curves,A TaqMan based realtime quantitative RT-PCR was performed to measure aberrant fusion gene transcripts in 45 samples of peripheral blood(PB) or bone marrow (BM) from 25 newly diagnosed patients with AML-M_2.All these 25 patients were diagnosed by the FCM(flow cytometry)and bone marrow molecular cytogenetics,and received the induce remission therapy with MA (Mitoxantrone+Ara-C).Results As a result,the AML1/ETO fusion gene transcripts were detected in 7(28%)out of 25 AML-M_2 patients (the ratios of AML1/ETO/ABL vary from 0.01 to 19.2),in which 5 patients were found t(8;21)(q22;q22).The transcript level of AML1/ETO fusion gene varied from the clinical situation of patients.These 7 patients with AML1/ETO fusion gene got complete remission(CR) after the first MA therapy,and the fusion gene reduced by 3 log in AML1/ETO/ABL.Only 11 patients got CR in 18 patients without AML1/ETO fusion gene.By following up these 7 patients with AML1/ ETO fusion gene kept persistent CR for 6 months.Conclusion It was concluded that real-time quantitative PCR is a reliable,innovative and promising technology with high sensitivity and speciality.It has potential clinical value for diagnosis,tumor typing,treatment selection,measuring the tumor load,monitoring fusion gene expression level and evaluating therapeutic strategy.It is worthy to apply in the clinical practice.
5.Assciation between uric acid levels and paroxysmal atrial fibrillation

Objective To investigate the potential relationship between the paroxysmal Atrial Fibrillation(PAF) and serum uric acid level.Methods Consecutive patients with (patient group,n =65) and without(control group,n =41) PAF,who were hospitalized in the Second Hospital of Tianjin Medical University from September 2011 to June 2012,were included in this study.We excluded subjects with congestive heart failure,acute coronary syndrome,congenital heart disease,valvular heart disease,cardiomyopathy,thyroid dysfunction and acute infection or inflammatory conditions.Baseline clinical data,complications and laboratory examination results were collected.Left atrium diameter (LAD),left ventricular end-diastolic dimension (LVEDD) and left ventricular ejection fraction (LVEF) were determined by echocardiography.Univariate and logistic regression was conducted to detect risk factors for PAF.Results Serum uric acid level were significantly increased in patients with PAF compared with controls ((360.2 ± 103.9) μmol/L vs (296.0 ±68.1) μmol/L,P =0.001).Multivariate logistic regression analysis showed that higher level of serum uric acid (OR:1.007,95% CI:1.000-1.015) and LAD (OR:1.142,95% CI:1.031-1.265) were independent risk factors for the occurrence of PAF.Conclusion High serum uric acid level is an independent risk factor for the development of PAF.Future larger studies should further evaluate this potential association as well as the underlying mechanisms.
6.Study on the status quo and influencing factors of rehabilitation confidence in stroke patients

Objective? To investigate the status of rehabilitation confidence level of stroke patients and analyze the influencing factors. Methods? A convenient sampling method was used to select 200 stroke patients who were admitted to a tertiary hospital in Beijing from December 2018 to March 2019. The general data questionnaire and the Chinese version of the Confidence after Stroke Measure (CaSM) were used in this cross study and the infuencing factors were analyzed. Results? A total of 200 questionaires were distributed and 195 questionaires were collected. The score of CaSM confidence of stroke patients was (47.32±11.44). The result of univariate analysis showed that age, marital status, education, occupation, family income, living status, accompanying type, personality type, episode times were the influencing factors of the level of rehabilitation confidence of stroke patients (P<0.05). Using the scores of CaSM of stroke patients as dependent variables and the statistically significant variables in analysis of single factor variance as independent variables, a multiple stepwise regression analysis was performed. The result showed that living alone, occupation for farmers, staff, introverted personality and accompanying type were the influencing factors of the level of rehabilitation confidence of stroke patients, which accounted for 20.40% of the variance in rehabilitation confidence of stroke patients. Conclusions? Nursing staff should pay more attention to the individual characteristics of patients on the basis of overall nursing, and develop practical and feasible rehabilitation plans from person to person, to help patients recover their pre-disease roles or find new meaningful roles as early as possible, and comprehensively improve their confidence in rehabilitation from three aspects: social confidence, positive attitude and self-confidence.
7.Effects of quinolinic acid on autophagy and protein expressions of related signaling pathway in PC12 cells

OBJECTIVE To investigate whether quinolinic acid(QA)induces autophagy in PC12 cells and its relationship with glycogen synthase kinase-3β(GSK-3β)/β-catenin related signaling path?ways. METHODS PC12 cells were treated with QA 2.5,5.0 and 10.0 mmol·L-1 for 24 h. The cell viability was determined by MTT assay. Autophagy fluorescent spots labelled form of microtubule-associated protein 1 light chain 3(LC3)was examined by LC3 immunostaining. The expressions of GSK-3β,β-catenin,LC3 and Beclin 1 were determined by Western blotting. RESULTS QA inhibited PC12 cell survival in a concentration-dependent manner,and IC50 was 8.7 mmol · L- 1. Compared with normal control group,QA 2.5,5.0 and 10.0 mmol · L-1 increased autophagic intracellular LC3 fluorescence spots,elevated the expression ratio of LC3-Ⅱ/LC3-Ⅰ and expression of Beclin 1 in PC12 cells(P<0.05). In addition,QA enhanced GSK-3βexpression and decreasedβ-catenin expression(P<0.05,P<0.01). CONCLUSION QA induces autophagy in PC12 cells. This mechanism may be associated with the activation of GSK-3β/β-catenin related signaling pathways.
8.Retrospective analysis of fetoscopic photocoagulation of communicating placental vessels of twin-twin transfusion syndrome

Objective To investigate the effect of fetoscopic photocoagulation of communicating placental vessels in twin-twin transfusion syndrome(TTTS)(selective or non-selective) on the perinatal outcomes.Methods Six cases of TTTS admitted in our department from Dec.2006 to Jun.2008 underwent fetoscopic photocoagulation of communicating vessels.Under direct real-time sonographic guidance,a 3-mm-diameter fetoscope was percutaneously inserted through the maternal abdominal wall into the amniotic cavity of the recipient twin.A combination of ultrasonographic and fetoscopic vision was used to identify the crossing vessels which were systematically coagulated using Nd:YAG laser fiber or bipolar electrocoagulation.Results All the 6 mothers tolerated the procedure without major complications.Two fetal survival rate was 33.33%.Conclusion Fetoscopic photocoagulation of communicating placental vessels in TTTS can effectively improve perinatal outcomes.
9.Effects of E-Coaching self-management model on health behavior change in perimenopausal women

Objective:To analyze the influence of E-Coaching self-management model on health behavior change in perimenopausal women.Methods:In this randomized controlled trial, 230 perimenopausal women who participated in health management prograam in the Health Management Center of Hangzhou Wuyunshan Hospital from January 2020 to October 2021 were selected as study objects by convenience sampling method. The subjects were divided into the experimental group and the control group with random number table (115 cases in each group). The experimental group was managed by health coaches with E-Coaching self-management model, and the control group was routinely managed by health managers. The intervention lasted for 6 months. Finally, 29 cases were lost to follow-up due to the failure of the subjects to comply with protocol requirements or voluntary withdrawal. So, a total of 201 subjects were included in the analysis (107 cases in the experimental group and 94 cases in the control group). χ2 test and t test were used to analyze the differences in modified Kupperman symptom score, perimenopausal knowledge and belief, regular exercise and dietary healthy behavior stage between the two groups. And the influence of E-Coaching self-management model on health behavior change in perimenopausal women was analyzed too. Results:After the intervention, the total score of modified Kupperman scale and the scores of insomnia, anxiety and fatigue in the experimental group were all lower than those in the control group [(7.36±2.91) vs (10.01±2.78) points, (0.49±1.13) vs (1.27±1.20) points, (0.80±0.99) vs (1.68±1.39) points, (0.67±0.55) vs (0.93±0.64) points]( t=6.553, 4.785, 5.219, 3.013, all P<0.05); and the total score of knowledge and belief questionnaire and the score of knowledge or belief dimension in the experimental group were significantly higher than those in control group [(25.15±1.55) vs (21.05±1.64) points, (9.61±0.56) vs (9.03±0.68) points, (15.54±1.53) vs (12.02±1.28) points] ( t=-18.238, -6.570, -17.801, all P<0.05). After the intervention, the proportions of the experimental group in the precontemplation and contemplation stage of exercise and diet were both significantly lower than those before intervention ( χ2=116.616, 139.964, both P<0.001), and were lower than those in the control group (the proportion of precontemplation stage of exercise was 7.5% vs 38.3%, and the contemplation stage of exercise was 26.2% vs 34.0%, χ2=38.330; the proportion of precontemplation stage of diet was 3.7% vs 23.4%, and the contemplation stage of diet was 18.7% vs 29.8%, χ2=25.399; all P<0.001). After the intervention, the proportion of the subjects in the preparation stage and action stage the experimental group were significantly higher than those before intervention ( χ2=116.616, 139.964, both P<0.001), and were higher than those in the control group (the proportion in preparation stage of exercise 18.7% vs 8.5%, and the action stage of exercise 47.7% vs 19.1%, χ2=38.330; the proportion in preparation stage of diet 20.6% vs 14.9%, and the action stage of diet 57.0% vs 31.9%, χ2=25.399; all P<0.001). Conclusion:E-Coaching self-management model can improve women′s perimenopausal symptoms in certain degrees, it improves their understanding of perimenopausal knowledge, enhances self-management beliefs and promotes healthy behavior changes.
10.Pathological features of Aβ1-42 deposition and its correlation with ApoE in the brain of streptozotocin-induced type Ⅱ diabetic rats

Objective To explore the pathological features of Aβ1-42 deposition and its correlation with Apolipoprotein E in brain of streptozotocin (STZ)-induced type Ⅱ diabetic rats.Methods High fat diet combined with small dose of STZ induced diabetic rats were adopted as experiment rats,and randomly divided into 3 months old diabetes mellitus group and 6 months old diabetes mellitus group (n=15).Healthy Wistar rats were adopted as control rats,and divided 3 months old control group and 6 months old control group (n=15).The Aβ1-42 and ApoE location and expressions were detected by immtmohistochemistry.Real-time fluorogenic quantitative-PCR and Westem blotting were used to detect the mRNA and protein levels of ApoE in each group.Results Immunohistochemical results showed that Aβ1-42 in the brain of the diabetic rats gathered around the vessel wall and blood vessel.The number of Aβ1-42 and ApoE positively-stained cells and positively-stained vessels was significantly larger and the ApoE mRNA and protein expressions were significantly increased in the 6 months old diabetes mellitus group as compared with those in the 3 months old diabetes mellitus group (P<0.05).The number of Aβ1-42 and ApoE positively-stained cells and positively-stained vessels was significantly larger and the ApoE mRNA and protein expressions were significantly increased in the 6 months old control groupas compared with those in the 3 months old control group (P<0.05).As compared with that in the 6 months old control group,the number of Aβ11-42 and ApoE positively-stained cells and positively-stained vessels was significantly larger and the ApoE mRNA and protein expressions were significantly increased in the 6 months old diabetes mellitus group (P<0.05).As compared with that in the 3 months old control group,the number of Aβ1-42 and ApoE positively-stained cells and positively-stained vessels was significantly larger and the ApoE mRNA and protein expressions was significantly increased in the 3 months old diabetes mellitus group (P<0.05).Person correlation coefficient indicated that the number of positively-stained cells of Aβ-42 was positively correlated to ApoE protein expression level (r=0.9755,P=0.000).Conclusions Aβ1-42 in the brain of type Ⅱ diabetic rats expresses both in blood vessel wall and around blood vessel.Age and duration of diabetes can increase the deposition of A3 and ApoE in brain tissues,and there is a positive correlation between them.
