1.The Professor ChenBaoyi's Experience to Test Infants viral Myocarditis Ayyhythmia

[Aim] Discuss Chen's treatment effect of infants viral myocarditis ayyhythmia. [Methods] He made suit medicine on two cases and analysed the disease'classification and main clinical manifestation and relevant therapy and his empirical formula, according to some factors ,such as the etiology and pathogens, course of disease and so on. [Results] (1)For viral myocarditis early disease caused by tachyarrhythmia, Mr. Chen advocated using the methods to treat which is clearing away heat and toxin and calming the pulse, the prescription used is Qingxinjiedu Decoction;however, those ones with il ness are offered a way of suppiementing qi and nourishing yin and warming the heart yang in order to remove pulse frequency, Chen used the modified YiQiShengMai Decoction for treatment.(2) The professor Chen paid attention to clearing hot and humid and supplementing qi and warming yang to cure slow arrhythmia due to acute viral myocarditis,the modified Huolian Decoction is used for this disease; For the people with a long il ness delayed healing, Chen worked a Decoction by himself which is Wenyangfumai Decoction,so that it could supplement qi and warm yang and invigorate the circulation of blood through pulse; For the slow arrhythmia appeared to sequela period, Chen thought that treatment should be insisted on promoting blood circulation to remove blood stasis and warming and activating the heart yang, the prescription used is Tongmai oyal liquid.[Conclusion]Professor Chen had gained a satisfied effect to treat infants viral myocarditis arrhythmia with the methods of clearing away heat and toxin, supplementing qi and nourishing yin and warming the heart yang and removing pulse frequency from the different stages and types.

3.Effect of ascending blood formula on immune function and bone marrow suppression in patients with colon cancer chemotherapy

Objective To explore the influence of the old Chinese national match the experience of haplostele liters of blood prescription of chemotherapy for colorectal cancer patients with immune function and bone marrow suppression .Methods 94 cases of metastatic nodes in patients with rectal cancer treated in our hospital from February 2011 to June 2015 were selected as the re‐search object ,and were randomly divided into observation group and control group according to different regimen in the treatment , with 47 cases in each group .Patients in control group was treated with conventional XELON chemotherapy ,and patients in the ob‐servation group recieved rise blood prescription treatment based on the chemotherapy in the control group .The immune function and bone marrow suppression situation in both groups were observed after two treatment cycles .Results The stable rate of the obser‐vation group was 42 .55% ,significantly higher than that of the control group (P<0 .05);the CD4+ /,CD8+ CD8+ and NK cells were significantly higher than those in the control group (P<0 .05) ,while those of the control group was significantly higher than that in control group (P>0 .05);the CD4+ /,CD8+ CD8+ and CD4+ /cells were significantly higher than those in control group (P<0 .05);The incidence rate of KPS in the observation group and control group was 70 .21% ,70 .21% ,17 .02% ,14 .89% , 87 .23% ,25 .53% and 23 .40% respectively .The KPS score of the observation group and the control group were significantly higher than that in control group (P<0 .05) ,and the G‐CSF in the observation group was significantly higher than that in control group (P<0 .05) .Conclusion The treatment of advanced metastatic colorectal cancer patients in the course of chemotherapy to give rise to blood formula can effectively improve the patient′s immune function ,reduce the bone marrow suppression effect ,improve the quality of life ,and it is worthy of clinical application .
4.(-)-Epigallocatechin-3-gallate suppresses migration and invasion of cancer cell by decreasing expression of small GTP-binding protein RhoA

AIM: It is tested that the suppressive effects of (-)-Epigallocatechin-3-gallate (EGCG ) on the migration, invasion and RhoA expression of human lung carcinoma 95-D cells and B16BL6 melanoma cells invasion in vivo,which will possibly help to understand the molecular mechanisms by which EGCG inhibits the invasion of tumor cells. METHODS: The inhibitory effect of EGCG on the migration of 95-D cells was tested by cell migration assay. Cell invasion was analyzed by the matrigel invasion assay. Assay of tumor metastasis in an animal model, RT-PCR and Western blot analysis of expression of RhoA was also performed. RESULTS:EGCG was effective in inhibiting the migration of 95-D cells in a dose-dependent manner. EGCG dose-dependently inhibited 95-D cells invasion in vitro and 40 μmol·L-1 EGCG exhibited 79.9% inhibition and EGCG 50 mg·kg-1·d-1 for 3 weeks inhibited B16BL6 melanoma cells invasion by 71.7% in vivo. EGCG could down-regulate the expression of RhoA. CONCLUSION: EGCG strongly inhibits metastasis of 95-D cells, and the mechanism of EGCG is possible associated with the inhibition of RhoA expression.
5.Study on Determination Method for Dissolution of Pentoxyverine Citrate and Guaifenesin Tablets

OBJECTIVE:To establish a method for the dissolution determination of Pentoxyverine citrate and guaifenesin tab-lets,and to compare the difference of preparations from different manufactures. METHODS:The paddle method was used to deter-mine the dissolution,using water as medium with medium volume of 1000 mL and rotation speed of 75 r/min,sampling at 45 min. HPLC method was used to determine the accumulative dissolution of pentoxyverine citrate and guaifenesin:CAPCELL PAK C18 MGⅡ S-5 column;mobile phase of acetonitrile-triethylamine phosphoric buffer solution(pH 2.6±0.05)(33:67,V/V),flow rate of 1.0 mL/min,detection wavelength of 215 nm(pentoxyverine citrate)and 275 nm(guaifenesin),column temperature of 35 ℃,sample size of 20 μL. RESULTS:The linear range of pentoxyverine citrate and guaifenesin were 5.0916-50.9155 μg/mL(r=0.9999)and 29.9995-299.9952 μg/mL(r=0.9999),respectively. RSDs of precision,stability and reproducibility tests were all lower than 2.0%. Average recoveries were 97.90%-100.68%(RSD=0.95%,n=9)and 97.09%-101.85%(RSD=1.32%,n=9). Average accu-mulative dissolution of pentoxyverine citrate in 6 manufactures of samples were 12%,98%,66%,97%,91%,32%(n=3);those of guaifenesin were 8%,95%,68%,90%,93%,18%(n=3),respectively. CONCLUSIONS:The method is simple,ac-curate,sensitive,specific and suitable for the dissolution determination of Pentoxyverine citrate and guaifenesin tablets. The dissolu-tion of samples between different manufactures and the same manufactures different batches are quite different.
6.Diagnosis and treatment of 64 children with viral encephalitis complicated with respiratory failure

Objective To summarize the clinical features of children with viral encephalitis accompa-nied with respiratory failure,and to improve the early diagnosis and treatment. Methods The clinical data of 64 cases with viral encephalitis combined with respiratory failure in our unit from May 2005 to May 2015 were analyzed retrospectively. Results All children were characterized by sudden onset. Among them, 60 cases (93. 7%) had fever,50 cases(78. 1%) had convulsion onset,46 cases(71. 8%) had consciousness disorders, 30 cases(46. 8%) had positive pathological signs. Most of them developed respiratory failure in acute stage. Total 56 cases occurred central respiratory failure,6 cases occurred central respiratory failure with peripheral respiratory failure,2 cases occurred respiratory and circulatory failure. Total 46 cases underwent cerebrospinal fluid examination. Routine biochemical test found 32 abnormal cases,of which 10 cases had intracranial hyper-tension and 19 cases had leukocytosis,and 17 cases had increased protein content. Ten cases were positive in cerebrospinal fluid etiology examination,including herpes simplex virus positive in 8 cases,EB virus positive in 1 case,and coxsackie virus positive in 1 case. There were 6 of 64 cases with abnormal CT scans and 29 of 34 cases with abnormal MRI. The results of EEG examination were abnormal in all patients for the first time. The EEG of 48 cases showed diffuse slow waves-δ activity. EEG examination showed generalized discharges or focal discharges during treatment in 22 cases. Five cases of electrophysiological examination showed cervical spinal cord anterior horn injury. Total 24 cases were complicated with stress ulcer,4 cases with liver damage,6 cases with heart damage,4 cases with renal damage,1 case with lung damage. All cases underwent mechanical ventilation for 2-50 days. Total 33 cases(51. 5%) improved and discharged,14 cases died during hospitaliza-tion,17 cases were given up treatment. Total 25 cases had variety of neurological dysfunctions left. Total 14 cases with epilepsy were followed up for 3 months to 6 years in our department,of which 8 cases were medical-ly intractable epilepsies. Conclusion The children with viral encephalitis complicated with respiratory failure have acute onset,rapid progress,high disability and mortality. Early diagnosis and evaluation,effective mechani-cal ventilation in time,and protecting organ function,help to improve the prognosis.
7.Pharmacodynamic Study of Chongcaodihuang Syrupus

OBJECTIVE:To study the effects of Chongcaodihuang(CCDH)syrupus on immune function and antistress ability.METHODS:The experiments were carried out to investigate the effects of CCDH on weight of immune organs,the phagocytization of the monocytes by using the method of carbon particles expurgation,the content of antibody of hemolysin in serum,and time of swimming and hypoxia tolerance in mice.RESULTS:CCDH could enhance weight of immune organs,the phagocytization of the monocytes and increase the content of antibody of hemolysin in serum.It can also delay the time of swimming and strengthen hypoxia tolerance in mice.CONCLUSION:CCDH significantly improve immune function and antistress ability of mice.
8.Association between fetal ventricular septal defects and chromosomal abnormalities

Objective To evaluate the association between fetal ventricular septal defects (VSD)and chromosomal abnormalities.Methods The 214 fetuses diagnosed VSD in the First Affiliated Hospital of Sun Yat-sen University from January 2008 to September 2011 were included.The VSD were categorized into 3 types:perimembranous,muscular and mixed (the defect could not be classified because the dimensions were larger than 5 mm) type.The perimembranous defect was subdivided into inlet and outlet subtypes.Complicated with other cardiac abnormalities/extracardiac abnornalities or not,the cases were divided into isolated VSD group,VSD complicating cardiac anomalies group (other cardiac and/or great vessels malformation),VSD complicating extracardiac anomalies group (include organ malformation and sonographic soft markers) and VSD with both cardiac and extracardiac anomalies group.G-banding chromosome analysis was advised for all cases.In cases that no karyotype was obtained,the phenotype of the newborns was examined by the pediatricians.And those appeared normal were defined as normal karyotype.Results (1) There were 134 (62.6%,134/214) perimembranous defects,including 91 (42.5%,91/214) inlet lesions and 43 (20.1%,43/214) outlet lesions.There were 35 (16.4%,35/214) muscular defects and 45 (21.0%,45/214) mixed type lesions.(2) Among the 214 VSD fetuses,46 (21.5%) were isolated VSD,34 (15.9%) were cases with other cardiac anomalies,87 (40.6%) were cases with extracardiac anomalies and 47 (22.0%) were cases with both cardiac and extracardiac anomalies.(3) The chromosomal karyotypes were obtained in 105 cases,and 21 cases were considered as normal according to the phenotype.Of all these 126 cases,46 (36.5%,46/126) had chromosomal abnormalities.(4) Inlet defects had the highest risk of chromosomal abnormalities (28/55,50.9%),while the muscular defects had the lowest risk (2/25,8.0%).The incidence of chromosomal abnormalities in outlet and mixed type was 33.3% (9/27) and 7/19,respectively.The types of VSD were significantly correlated with chromosomal defects (P < 0.01).(5) The incidence of chromosomal abnormalities in the 4 groups were 3.4% (1/29),2/14,53.6% (30/56) and 48.1% (13/27),respectively.The risk of chromosomal abnormalities in the cases complicating extracardiac or both extracardiac and cardiac anomalies was significantly higher than the isolated VSD group (P < 0.01).Conclusion Fetal VSD had a highest risk of chromosomal abnormalities,especially the inlet type and VSD with extracardiac abnormalities,and then the fetal karyotype should be recommended.
9.A cross-sectional study of urinary iodine and salt iodine content among schoolchildren and their families in Haimen City, Jiangsu Province

Objective To understand the level of urinary iodine of schoolchildren and salt iodine content of their families in a primary school and to analysis influence between salt iodine content and urinary iodine level in Haimen City,Jiangsu Province.Methods A cross-sectional study was conducted in a primary school of Haimen City in 2012.With the method of stratified cluster sampling,all students aged 9 ~ 12 were extracted in grades 3-5,urine samples and salt samples of their family were collected.Urinary iodine and salt iodine were determined according to Method for Determination of Iodine in Urine by As3+-Ce4+ Catalytic Spectrophotometry (WS/T 107-2006) and Direct Titration of Universal Test method in Salt Industry (GB/T 13025.7-1999).Results Totally,452 students from grades 3-5,including 233 boys and 219 girls were selected by cluster-stratified sampling.The median of children's urinary iodine was 231.90 μg/L,while the median of boys and girls was 235.40 and 222.60 μg/L,respectively.The median of urinary iodine of 9,10,11 and 12 year old students was 253.65,195.70,236.40 and 241.70 μg/L,respectively.The proportion of less than 100 μg/L and ≥300 μg/L was 7.7% (35/452) and 27.7% (125/452),respectively.There were no significant differences between different gender and age students.Among the 452 salt samples collected the median of salt iodine was 27.50 mg/kg.The coverage rate of iodized salt,the qualified rate of iodized salt and the intake rate of qualified iodized salt were 98.9% (447/452),95.1% (425/447) and 94.0% (425/452),respectively.After consumptionn of non-iodized salt (< 5 mg/kg),unqualified iodized salt(5 ~ < 20 mg/kg or > 50 mg/kg) or qualified iodized salt [(35 ± 15)mg/kg],the median of urinary iodine of children was 177.30,211.95 and 232.90 μg/L,respectively.Correlation analysis showed that there was no relationship between iodine content of salt and urinary iodine level of schoolchildren (r =0.085,P > 0.05).Conclusions Current nutritional level of schoolchildren in Haimen City is higher than the appropriate amount.Household salt iodine content of edible salt does not affect urinary iodine level of the children significantly.
10.Effect of adeno-hTGF-β1 transfection on in vitro chondrogenic differentiation of bone marrow stromal cells

Objective To investigate the action of chondrogenesis differentiation of bone marrow stromal cells (BMSCs) transfected with adeno-hTGF-β1. Methods In the experiment group, replication-deficient a denoviruses carrying human hTGF-β1 complementary DNA (adeno-hTGF-β1 was constructed and applied to transfect to the first generation BMSCs. As a control, each BMSCs was transduced with 200 pfu of adeno-LacZ gene. One day after transfer, BMSCs were trypsinized, counted, and 5×105 cells aliuots were spun down at 500 rpm per minute in 15 ml polypropylene conical tubes and then cultured in a defined medium in an incubator at 37℃ for 21 days. The aggregates were harvested at time points to 21 days and assessed by gross observation, histological analyses and immunohistochemical localization of type Ⅱ collagen. Results When harvested at 21 days, each pellet shrinked to spheroid tissue with apearly opalescence in gross morphology and found to be relatively firm. H.E staining showed elongate dlining cells appeared as perichon drium-like cells at the surface. Some nests of cartilage were observed at the substrate of the tissue. Mature chon drocytes were embeded in the lacuna in the experiment group. In addition, Safranin'O staining confirmed the presence of sulfated proteoglycans in the ECM of chondrogenesis region. Immunohistochemical staining revealed the presence of type Ⅱ collagen in chondrogenesis region. By contrast, HE staining showed no evidence of cartilage formation in the control group. They were fibrous tissue with no architectural feature. Safranin'O staining and Immunohistochemical staining showed no evidence of sulfated proteoglycans or typeⅡ collagen expression. Conclusion BMSCs transfected with adeno-hTGF-β1 could induce its chondro-genesis when aggregate cultured in a defined medium in vitro, laying a foundation for the application of hTGFβ1 gene-transfected BMSCs in cartilage tissue engineering.
