In order to provide scientific basics for exploitation and sufficient application of Dendrobium officinale leaves resources, the phenol-sulfuric acid method was applied to determine the polysaccharide content. The monosaccharides were derivated by PMP and the derivatives were identified by HPLC-DAD-ESI-MS(n) and the contents of mannose and glucose were determined simultaneously. Similarity evaluation system for chromatographic fingerprint of traditional Chinese medicine (2004A) was employed to generate the mean chromatogram and similarity analysis of the samples was carried out. The results demonstrated that polysaccharide content, monosaccharide compositions and composition ratio had an obvious difference between stems and leaves. The polysaccharide content of stems was higher than that of leaves. Monosaccharide composition in leaf was significantly different from that in stem. The polysaccharide from stems was composed of mannose and glucose, however the polysaccharide of leaves was acid heteropolysaccharide and was mainly composed of five monosaccharides, including mannose, galacturonic acid, glucose, galactose and arabinose. The similarity value of the 14 batches was above 0.9, indicating that similarity of fingerprints among different samples was high. The study can provide evidence for expanding the medicinal parts of D. officinale.

3.Dihydromyricetin induce apoptosis in AGZY-83-a tumor cells

Aim To investigate the effects of dihydromyricetin on the apoptosis in AGZY-83-a tumor cells and its relevant mechanisms.Methods The survival rate of AGZY-83-a cells was assayed by MTT dye reduction.The cellular apoptosis was analyzed by TUNEL method and caspase-3 activity.The intracellular free calcium i was detected to explore the apoptotic mechanism of dihydromyricetin.Results In the MTT assay,dihydromyricetin 50?mol?L-1 significantly inhibited survival ratios of AGZY-83-a cells at dose-and time-dependent manner.It was demonstrated in TUNEL assay that dihydromyricetin could induce the apoptosis of AGZY-83-a cells in a concentration-dependent manner.The examination of Caspase-3 activity indicated that the dihydromyricetin could induce the apoptosis of AGZY-83-a cells,which was dose-dependent activation of Caspase-3 in AGZY-83-a cells.The detection of the intracellular i showed that the average FI of the i could be markedly increased to 20-fold as the basic condition.Conclusions Dihydromyricetin can induce the apoptosis in AGZY-83-a cells,which is associated with the overload of the intracellular i.
4.Comparative study of the display in periampullary carcinoma by conventional ultrasound,hydrosonography and double-contrast enhanced ultrasound

Objective To compare the value of conventional ultrasound,hydrosonography and double-contrast enhanced ultrasound in the display capacity of periampullary carcinoma and its relationship with the surrounding tissue.Methods A total of 18 patients with the periampullary carcinoma were diagnosed pathologically or by endoscopic biopsy.Each patient had three imaging modalities by conventional ultrasound,hydrosonography and double-contrast enhanced ultrasound.The display capacity and the relationship between the mass and surrounding tissue were compared with three modalities.Results The display rates of limpid visibilities of carcinoma on conventional ultrasound,hydrosonography and double-contrast enhanced ultrasound were 16.7%(3/18),22.2%(4/18) and 94.4%(17/18),respectively;The display rates of duodenum protrusion on three modalities were 0(0/18),38.9% (7/18) and 72.2% (13/18),respectively,with significant differences among conventional ultrasound,hydrosonography and double-contrast enhanced ultrasound.The double-contrast enhanced ultrasound showed two cases of peripheral vascular invasion and one case of intrahepatic metastasis.Conclusion The double-contrast enhanced ultrasound can increase the display capacity of periampullary carcinoma as a new diagnostic modality for periampullary carcinoma.

Objective To estimate the cancer incidence and mortality in Chongqing in 2014.Methods On basis of the methods and criteria of data quality control made by NCCR,authors compiled and summarized the 2014 tumor registration data reported by 11 registries in Chongqing.The datas which were stratified by area (urban/rural),gender,age and cancer type,incidence and mortality were calculated combined with national population.Results All 11 cancer registries reported 24 506 new cancer cases (14 610 were male and 9 896 were female)in 2014.The crude incidence in Chongqing was 244.66/105 (male was 289.01/105,female was 199.47/105).Sex ratio was 1.45:1.00,male was significantly higher than female.The incidence of lung cancer,colorectum,liver,esophagus,breast,stomach were the top six tumor.The incidence rate of all age groups in 2014 increased with age,the incidence rate increased from 40-44 years old group,and reached the highest incidence rate in the 80-84 years old group.Conclusion The data quality and representativeness in Chongqing tumor registration are gradually improved.Cancer registration as the basis of cancer prevention and control work,play an irreplaceable role.
6.Change of microbial community in rhizoma sphere of cultivated Atractylodes lancea.

OBJECTIVETo find the development rules of microbial community in rhizoma sphere of the cultivated Atractylodes lancea.
METHODTotal bacteria, fungi and actinomyces were counted by CFU x g(-1) though dilution plate method. And genomic DNA of microbes were extracted and amplified by primers of E. coli's 27f and 1492r to get the 16S rDNA, then the restriction endonuclease Hinf was used to digest the 16S rDNA.
RESULTTotal bacteria, fungi and actinomyces in 2-year old soil were lower than in 1-year old soil, they decreased 46. 14%, 49. 25%, 31.88% respectively and made the ratio of themselves changed. At the same time, all the 8 soil samples got fine 16S rDNA bands, which were about 1500 bp. And the main bands of most of the samples were found at 1000 bp, but the weak bands of each were different although most bands in the same year samples were more similar than in different year ones.
CONCLUSIONIt is indicated that the change of soil microbial community may has some relation to the continous cropping barrier of A. lancea.

7.Study on medicinal plant allelopathy and soil sickness based on ecological niche.

8.Habitat characteristics for the growth of Atractylodes lancea based on GIS.

OBJECTIVETo study the habitat characteristics for the growth of Atractylodes lancea.
METHODThe leading factors for the growth of A. lancea are determined after stepwise regression, and the limiting factor are determined by field study, literature search,and weather records. The mean values of leading factors and the limiting factors of A. lancea for 30 years were applied to do spatial interpolation using IDIRIEIW softwares. An overlay was prerformed using data obtained in Mt. Maoshan and the general habitat of A. Iancea.
RESULTIt was found that precipitation is one of the dominant climate factors that affects the quality of A. lancea. High temperature was one of the limiting factor which influences the living of A. lancea. The best habitat was determined to be: annual mean temperature more than 15 degrees C, monthly mean minimum temperature between - 2- -1 degrees C, monthly mean maximum temperature about 32 degrees C, lowest tolerable temperature - 17- -15 degrees C, drought season 1 - 2 months, annual mean precipitation between 1000 - 1160 mm.
CONCLUSIONMt. Maoshan was characterized by having high temperature, short drought season and adequate precipitation. The growth of A. Lancea is a result of environmental stress.

9.Determination of naringenin in Dendrobium officinale by HPLC.

OBJECTIVETo explore a characteristic chemical marker of Dendrobium officinale, establish determination method of its content and determine the naringenin content in D. officinale from different sources and growth years.
METHODThe content of naringenin was determined by HPLC. HPLC analysis was made on a XB -C18 (4.6 mm x 250 mm, 5 microm) with methanol and water containing 0.2% phosphoric acid as mobile phase. The detection wavelength was 290 nm.
RESULTThe HPLC method showed good linearity within the range of 0.026-0.208 microg (r = 1). The average recovery of naringenin was 96.3% (RSD 1.8%). The naringenin content was the highest in 3 years D. officinale and had some differences from different sources.
CONCLUSIONThe method is accurate and reliable. It is appropriate for the quantitative determination of naringenin in D. officinale and it's production.

10.HPLC specific chromatogram of Dendrobium officinale.

OBJECTIVETo establish the method of specific chromatogram analysis of ether extract of Dendrobium officinale for identification of D. officinale.
METHODChromatographic separation was carried out at 30 degrees C on an Ultimate C18 column (4.6 mm x 250 mm, 5 microm) eluted with methanol and water containing 0.2% phosphoric acid in a gradient elution at a flow rate of 1.0 mL x min(-1). The detection wavelength was set at 280 nm. The similarity evaluation system for chromatographic fingerprint of NPC (National Pharmacopoeia Committee) was adopted to specific chromatogram construction.
RESULTThe HPLC specific chromatogram of D. officinale was constructed with 6 common specific chromatographic peaks including naringenin as a reference peak.
CONCLUSIONThe method shows good precision and repeatability of relative retention time. It can be used to identify D. officinale.
