Due to the extensive pharmacological effects, pyrazolo[3, 4-d] pyrimidine derivatives have aroused great attention. There are a variety of ways in their synthesis, including using pyrazole ring, pyrimidine ring and other materials as parent ring. The compounds have strong bactericidal, anti-inflammatory, anti-cancer and anti-virus activities, etc. In this paper, the synthesis and pharmacological activities of pyrazolo[3, 4-d] pyrimidine derivatives taking the synthesis as the main line are summarized.

Objective To investigate the relationship between single nucleotide polymorphisms of surfactant protein C(SP-C)gene and respiratory distress syndrome(RDS)in the Han nationality newborns in Inner Mongolia and whether there is a mutation occurs on SP-C gene exon 4 and 5.Methods One hundred newborns with RDS(case group)and 100 newborns without RDS(control group)were selected.PCR gene analysis was used to establish the genotype and allele frequencies of exon 4 (T138N)and 5 (S186N)on SP-C.Results In the Han nationality newborns of Inner Mongolia region,there was no mutation on SP-C gene exon 4 and 5.Exon 4(T138N)on SP-C could be checked out three genotypes:namely AA,AC and CC.The genetic polymorphisms of exon 4 on SP-C were not statistically different between the case group and the control group(χ2 ﹦0.744,P ﹦0.689).Besides,exon 5(S186N)on SP-C could also be checked out three genotypes:namely AA,AG and GG.The genetic polymorphisms of exon 5 on SP-C were also not statistically different between the case group and the control group(χ2 ﹦0.770,P ﹦0.681 ).Conclusion There is no mutation on SP-C gene exon 4 and 5.The genetic polymorphism of exon 4 and 5 on SP-C displays no signifi-cant correlation with RDS of the Han nationality newborns in Inner Mongolia.

Objective A retrospective analysis of the situation of newborn disease screening in Longwan Wenzhou from 2002 to 2009 was conducted.Methods The restdts of newborn diaease screening of 46 202 neonatus in Longwan district of Wenzhou between 2002 and 2009 were analyzed.The neonatns were screened within 72 hours after birth for congenital hypothyroidism(CH),phenylketonuria(PKU)by measuring thyroid-simulating hormone (TSH)and phenylalanine concentration on dried blood spots on filter paper cards.Neonatus weYe recalled after screened positive and the diagnosis was made by testing serum concentration.Results A total of 46 202 newborns were screened from 2002 to 2009,and 23 cases was positive and the total positive rate was 0.05%,22 cases were confirmed positive for CH.with an incidenee of 1/2 100.1 case was confirmed positive for PKU,with an incidence of 1/46 202.All patients were followed-up by experts of newborn disease screening centre of Zhejiang Province.Conclusion The results of this study confirm that newborn discease sereaning was one of the effective methods of early diagnosis of congenital CH and PKU,which could effectively prevent development low of intelligence or physique,and other organs impairment.

AIM: To explore the effects of rheum tanguticum polysaccharides(RTP) on TNBS-induced colitis in mice and its probable mechanisms.METHODS: Mice colitis model was induced by 2,4,6-trinitrobenzene sulfonic acid(TNBS).The mice were randomly divided into 5 groups,normal group,model group,RTP-100,200 and 400 group.The macroscopical and histological changes of the colon were evaluated and the cytokines IL-8,IL-10,TNF-?and IL-4 produced by splenocyts were analyzed with ELISA.RESULTS: Compared with the model group,both symptoms and the lesions of colonic mucosa of RTP-200 and 400 group were slighter on ulcerative colitis induced by TNBS in mice. Objective: To observe the clinical efficacy of heat-sensitive moxibustion for adjuvant treatment of depression in Parkinson disease and explore its mechanism. Methods: A total of 80 patients with Parkinson disease coupled with depression were randomized into an observation group and a control group, with 40 cases in each group. The control group was treated with levodopa and benserazide hydrochloride tablets and paroxetine tablets, while the observation group was treated with heat-sensitive moxibustion on the basis of the medications in the control group. The treatment course was 2 months. The Hamilton depression scale-17 (HAMD-17), unified Parkinson's disease rating scale (UPDRS) and Parkinson's disease quality of life questionnaire-39 (PDQ-39) were scored before and after the treatment, and the efficacy was evaluated after treatment. Levels of patients' serum dopamine (DA), 5-hydroxytryptamine (5-HT), tumor necrosis factor-α (TNF-α) and interleukin-6 (IL-6) were detected before and after the treatment. Results: After treatment, the total effective rate of the observation group was higher than that of the control group (P<0.05). The HAMD-17 scores in the two groups decreased significantly after treatment (both P<0.05), and the score in the observation group was obviously lower than that in the control group (P<0.05). The component scores and total scores of UPDRS in both groups decreased significantly (all P<0.05), and the scores in the observation group were lower than those in the control group (all P<0.05). The score of PDQ-39 in the observation group decreased significantly (P<0.05), and was lower than that in the control group (P<0.05). After treatment, the serum DA and 5-HT levels in the observation group increased significantly (both P<0.05) and the TNF-α and IL-6 levels decreased significantly (both P<0.05), which were statistically different from those in the control group (all P<0.05). Conclusion: Heat-sensitive moxibustion has certain auxiliary effect in treating depression in Parkinson disease, significantly improving clinical symptoms and the quality of life, which may be related to the up-regulation of DA and 5-HT levels and down-regulation of TNF-α and IL-6 levels.

ObjectiveTo demonstrate the effect of moderate hypothermia and doule carotid ice applied to acute intracerebral hemorrhage (ICH) and the best opportunity of treatment.Methods174 cases with ICH were randomly divided into treated group and control group, 87 cases for each group. Each case of both groups was treated by routine therapy, moreover, all cases in treated group were treated by moderate hypothermia (MHT).The clinical effect and prognosis between treated group and control group were contrasted,so to do between the cases in the treated group who were treated no more than 3 hours and more than 3 hours after attack.ResultsCompared with the control group, the neurological function improved in the treated group(P<0.05), as well as survival rate and recovery(P<0.05).Cases who treated no more than 3 hours after attack was better than that of more than 3 hours(P<0.05).ConclusionModerate hypothermia can effectively ease the neurologic deficits after ICH, decrease mortality and disable rate.The best therapy time is within 3 hours after ICH.

ObjectiveTo study the rehabilitation effect of physical therapy on patients with above knee amputation. Methods50 patients with above knee amputation were evaluated with FIM scale before and after physical therapy. ResultsThere was a significant difference before and after physical therapy(P<0.05). Conclusions Physical therapy is effective on patients with above knee amputation.

AIM: To evaluate the protection of Mn~(2+) on injury of human kidney tubular epithelial cell line induced by gentamicin, and investigate its principals. METHODS: Gentamicin was used to injury human kidney tubular epithelial cell line (HK-2) to produce model of kidney injury. MTT method was used to measure effect of Mn~(2+) on proliferation of cells after they were injured by gentamicin. The spectrophotometry method was used to observe the change of LDH,NAG and SOD effected by Mn~(2+). Ultrastructure was examined by electron microscopy. RESULTS: Although HK-2 cells were injured, Mn~(2+) promoted cells proliferation, decreased LDH activity and NAG content, increased SOD activity, and alleviated dilation of the mitochondria and crushing of lysosomes. CONCLUSION: Mn~(2+) can protect human kidney tubular epithelial cell line from injury induced by gentamicin. It may be in relation to inhibiting the oxidative injury, lightening dilation of the mitochondria, protecting the integrity of lysosomes and decrease the leakage of enzyme.
