3.Effect of benazepril on expression of RAGE in renal tissues of diabetic rats

Objective To investigate the effects of benazepril on the expression of receptors for advanced glycation end-products(RAGE)in diabetic rat kidneys,and to explore its mechanisms of renal protection in diabetic rats. Methods Thirty SD rats were randomly divided into three groups(n=10 in each group): normal control group,diabetic control group and diabetic with benazepril group.Blood glucose,blood lipid,HbA1c,kidney to body weight ratio and 24 h urinary protein excretion were detected after 12 weeks.RAGE mRNA level was analyzed by quantitative RT-PCR. Results Compared with the normal control group,the blood glucose,HbA1c,triglyceride,total cholesterol and low density lipoprotein cholesterol in the diabetic control group and diabetic with benazepril group were significantly increased(P0.05).The kidney to body weight ratio,24 h urinary protein excretion and RAGE mRNA level in the diabetic with benazepril group were significantly higher than those in the normal control group(P
4.Effect of valsartan on expression of receptor for advanced glycation end-products in renal tissues of diabetic rats

Objective To investigate the effects of valsartan on the expression of receptors for advanced glycation end-products(RAGE) in kidneys of diabetic rats,and to explore its renoprotection mechanisms. Methods Thirty rats were divided into normal control group,diabetes control group and diabetes with valsartan group(n=10).Blood glucose,blood lipid,HbA1c,kidney to body weight ratio and 24 h urinary protein excretion were measured after 12 weeks.RAGE mRNA level was detected by real-time quantitative PCR. Results Compared with normal control group,kidney to body weight ratio,24 h urinary protein excretion and RAGE mRNA were significantly increased in diabetes control group.Compared with diabetes control group,kidney to body weight ratio,24 h urinary protein excretion and RAGE mRNA were significantly decreased in diabetes with valsartan group. Conclusion Valsartan can inhibit renal hypertrophy and decrease urinary protein excretion in diabetic rats.The renoprotective effects may be related to its inhibition on RAGE expression.
5.Magnetic resonance imaging changes and analysis of related factors after treatment in childhood hepatolenticular degeneration

Objectives To observe the changes of magnetic resonance imaging(MRI) during the course of treatment in hepatolen-ticular degeneration (HLD) and to evaluate its related factors.Methods Sixty - three patients with HLD received routine SE sequence MRI of brain and liver.Twenty - two patients with abnormal MRI in 42 patients and each of them had 2-4 times of serial MRI ex-aminations at interval of 1.0- 1.5 year.Results Ten patients had lesion in both brain and liver in 22 patients. Nine patients only in brain, 3 patients in liver.The major lesions disappear or decrease after rational treatments. There were only slight or no changes of MRI abnormalities in patients receiving treatments or age of patients was over 15 year old. Abnormal MRI findings shown again after stopped treatments over 2 months. In such cases, the clinical and MRI improvements were slow.The rate of clinical and MRI improvement in patients with adequate treatment were markedly higher than patients with inadequate treatment.Conclusions Liver and brain lesion will show in MRI disappear after adequate treatment;There are only slight or no change or showing again in MRI abnormalities in pa-tients with inadequate treatments compared with poor clinical improvement. MRI is beneficial to understanding therapeutic effect and prognosis of HLD.
6.Therapeutic efficacy and mechanism of heat-sensitive moxibustion for adjuvant treatment of depression in Parkinson disease

Objective: To observe the clinical efficacy of heat-sensitive moxibustion for adjuvant treatment of depression in Parkinson disease and explore its mechanism. Methods: A total of 80 patients with Parkinson disease coupled with depression were randomized into an observation group and a control group, with 40 cases in each group. The control group was treated with levodopa and benserazide hydrochloride tablets and paroxetine tablets, while the observation group was treated with heat-sensitive moxibustion on the basis of the medications in the control group. The treatment course was 2 months. The Hamilton depression scale-17 (HAMD-17), unified Parkinson's disease rating scale (UPDRS) and Parkinson's disease quality of life questionnaire-39 (PDQ-39) were scored before and after the treatment, and the efficacy was evaluated after treatment. Levels of patients' serum dopamine (DA), 5-hydroxytryptamine (5-HT), tumor necrosis factor-α (TNF-α) and interleukin-6 (IL-6) were detected before and after the treatment. Results: After treatment, the total effective rate of the observation group was higher than that of the control group (P<0.05). The HAMD-17 scores in the two groups decreased significantly after treatment (both P<0.05), and the score in the observation group was obviously lower than that in the control group (P<0.05). The component scores and total scores of UPDRS in both groups decreased significantly (all P<0.05), and the scores in the observation group were lower than those in the control group (all P<0.05). The score of PDQ-39 in the observation group decreased significantly (P<0.05), and was lower than that in the control group (P<0.05). After treatment, the serum DA and 5-HT levels in the observation group increased significantly (both P<0.05) and the TNF-α and IL-6 levels decreased significantly (both P<0.05), which were statistically different from those in the control group (all P<0.05). Conclusion: Heat-sensitive moxibustion has certain auxiliary effect in treating depression in Parkinson disease, significantly improving clinical symptoms and the quality of life, which may be related to the up-regulation of DA and 5-HT levels and down-regulation of TNF-α and IL-6 levels.
7.Clinical observation of head and body acupuncture and moxibustion on stroke

ObjectiveTo observe effect of head and body acupuncture and moxibustion on stroke.Methods183 stroke patients were randomly divided into 3 groups, head acupuncture and moxibustion, body acupuncture and moxibustion, and head body acupuncture and moxibustion. After two months treatment, effects of 3 groups were evaluated.ResultsThere were no differences between head group and body group, body group and head body group, but there was significantly difference between head body group and head group (P<0.05).ConclusionThe head body acupuncture and moxibustion can gain the best clinical effect on stroke patients compared with simply head or body acupuncture and moxibustion.
9.Bioinformatic Analysis of Structural Proteins of Paramyxovirus Tianjin Strain

The amino acid sequences of the NP,P, M, F,HN and L proteins of the paramyxovirus Tianjin strain were analyzed by using the bioinformatics methods. Phylogenetic analysis based on 6 structural proteins among the Tianjin strain and 25 paramyxoviruses showed that the Tianjin strain belonged to the genus Respirovirus, in the subfamily Paramyxovirinae, and was most closely related to Sendal virus (SeV). Phylogenetic analysis with 14 known SeVs showed that Tianjin strain represented a new evolutionary lineage. Similarities comparisons indicated that Tianjin strain P protein was poorly conserved, sharing only 78.7%-91.9% amino acid identity with the known SeVs, while the L protein was the most conserved, having 96.0%-98.0% amino acid identity with the known SeVs. Alignments of amino acid sequences of 6 structural proteins clearly showed that Tianjin strain possessed many unique amino acid substitutions in their protein sequences, 15 in NP, 29 in P, 6 in M, 13 in F, 18 in HN, and 29 in L. These results revealed that Tianjin strain was most likely a new genotype of SeV. The presence of unique amino acid substitutions suggests that Tianjin strain maybe has a significant difference in biological, pathological, immunological, or epidemiological characteristics from the known SeVs.
