1.Study on Preventive Effects of Arhinhor-11 on Acute Alcoholism in Mice

OBJECTIVE:To study the preventive effects of arhinhor-11 on acute alcoholism in mice. METHODS:48 mice were randomly divided into model group (sodium carboxymethyl cellulose),arhinhor-11 low-dose,medium-dose and high-dose groups [1.23,2.46,4.92 g(crude drug)/kg],with 12 mice in each group. Each group was given relevant medicine intragastrically, and 30 min later given 56% liquor intragastrically to induce acute alcoholism model. The disappearance time and recovery time of righting reflex were determined. 160 mice were grouped,given medicine and modeled with the same method,with 40 mice in each group. The alcohol concentration and alcohol dehydrogenase(ADH)activity in liver of 10 mice were determined 0.5,1,2,3 h af-ter giving alcohol. Other 10 mice were included in normal control (ig administration of sodium carboxymethyl cellulose). RE-SULTS:Compared with model group,the disappearance time of righting reflex were prolonged while recovery time shortened in arhinhor-11 groups,and alcohol concentration and ADH activity increased 0.5,1,2,3 h after giving alcohol (P<0.05 or P<0.01),especially 1 h after giving alcohol. 3 h after giving alcohol,alcohol concentration in blood of arhinhor-11 high-dose group decreased while ADH activity increased (P<0.05);there was no statistical significance among other arhinhor-11 groups (P>0.05). CONCLUSIONS:Arhinhor-11 has preventive effect on acute alcoholism in mice. It has a positive control effects on the alco-hol concentration in blood and ADH activity in liver.

Objective To investigate the effects of azithromycin on hyperoxia induced lung injury in neonatal rats.Methods Ninety 2-day-old neonatal rats were randomly divided into 3 groups (30 cases in each group):air control group,hyperoxia model group,azithromycin treated group.The rats in air control group were exposed to indoor air,and the rats in hyperoxia model group and azithromycin group were exposed to 900 mL?L-1 O2 for 14 days.From 2-days-after birth,rats in treated group received intraperitoneal injection of azithromycin (200 mg?kg-1)per day,rats in air control group and hyperoxia model group received intraperitoneal injection of normal saline.Ten rats in each group were executed and their lung tissues were taken out and were observed under a optical microscope on the 3rd,7th,14th days from the start.The expression of interferon gamma(IFN-?),connective tissue growth factor(CTGF) and matrix metalloproteinase-9 (MMP-9) in the lung tissues were detected by immunohistochemistry.Results There was no expression of IFN-? positive cells in air control group,but the expressional intensity level was gradually enhanced as the time of inhaling hyperoxia prolonged in hyperoxia model group,it reached the peak on the 7th day,and then obviously declined on the 14th day,but still higher than that of air control group.The expressional intensity level of IFN-? in azithromycin treated group was lower than that of hyperoxia model group in each time segment,there were significant differences among them(Pa

7.Clinical study on long-snake moxibustion plus Western medicine in treating chronic heart failure due to heart-kidney yang deficiency

Objective: To observe the clinical efficacy of long-snake moxibustion plus Western medicine for chronic heart failure due to heart-kidney yang deficiency.Methods: A total of 80 patients were randomized into a control group and an observation group, with 40 cases in each group. The control group was treated with routine Western medicine, and the observation group was treated with long-snake moxibustion therapy on the basis of routine Western medicine. Left ventricular ejection fraction (LVEF), stroke volume (SV) and left ventricular end-diastolic diameter (LVEDd) were measured before and after treatment, and the heart function was assessed. The serum brain natriuretic peptide (BNP) level was measured, the 6 min walking distance was recorded, and the Minnesota living with heart failure questionnaire (MLHFQ) was assessed and scored. The serum galectin-3 (Gal-3) and soluble growth stimulation expressed gene 2 (sST2) protein levels were measured. Clinical efficacy was evaluated after treatment. Results: The total effective rate of the observation group was higher than that of the control group, and the difference between the two groups was statistically significant (P<0.05). After treatment, LVEF and LVEDd in both groups were significantly improved (all P<0.05), and LVEF, SV and LVEDd in the observation group were superior to those in the control group (all P<0.05). The heart function classification in both groups were improved after treatment (both P<0.05), and the heart function classification in the observation group was superior to that in the control group (P<0.05). The serum BNP level in both groups decreased (both P<0.05), and the serum BNP level in the observation group was lower than that in the control group (P<0.05). The 6 min walking distance and MLHFQ scores in both groups were improved (all P<0.05), and the 6 min walking distance and MLHFQ score in the observation group were superior to those in the control group (both P<0.05). The serum Gal-3 and sST2 levels in the observation group decreased significantly (both P<0.05), and all lower than those in the control group (both P<0.05). Conclusion: Long-snake moxibustion plus Western medicine has significant efficacy in treating chronic heart failure due to heart-kidney yang deficiency; it can improve heart function, reduce the serum BNP level, and improve the exercise endurance and quality of life in the patients. This may be related to the down-regulation of serum Gal-3 and sST2 levels.
8.Early outcome of peripheral cutting balloon in the treatment of peripheral vessel stenosis

Objective To investigate the early clinical outcome of peripheral cutting balloon(PCB) in the management of peripheral vessel stenosis.Methods Thirteen patients with peripheral limb vessel stenosis, in which 4 stenoses in hemodialysis access and 9 at lower limb arteries,underwent angioplasty by PCB.For multiple stenosis in the same vessel,the distal one should be expanded firstly.The balloon pressure was controlled in the range of 8 atm to 10 atm(1 atm=10.108 kPa).All the patient were given continuous anticoagulant therapy after the procedure.Results All the procedure were carried out successfully on the 13 patients,and no serious complications occured.The symptoms did not recur in all patients after the procedure.The 5 months' follow-up angiography proved that no restenosis occurred in one patient with previous stenosis at the hemodialysis access.Conclusion The angioplasty with PCB was a safe and reliable procedure in management of the peripheral limb vessel stenosis.The early outcome is satisfying.

Objective To evaluate the pattern of energy metabolism and nutrients intake in patients with chronic viral hepatitis and posthepatitic cirrhosis to effectively direct their nutrition therapy.Methods Resting energy expenditure (REE) was measured with open-circuit indirect Jorimetry in 60 patients with chronic viral hepatitis and 60 patients with posthepatitic cirrhosis.Their normal basal energy expenditure (BEE) was predicted by Harris-Benedict equation and energy intake (EI) was determined by diet recall. Tetradrine-tashionone II(A)-PLGA composite microspheres were prepared by the SPG membrane emulsification method, and the characterization of tetradrine-tashionone II(A) -PLGA composite microspheres were studied in this experiment. The results of IR, DSC and XRD showed that teradrine and tashionone II(A) in composite microspheres were highly dispersed in the PLGA with amorphous form. The results of tetradrine-tashionone II(A) -PLGA composite microspheres in vitro release experiment showed that the cumulative release amounts of tetradrine and tashionone II(A) were 6.44% and 3.60% in 24 h, and the cumulative release amounts of tetradrine and tashionone II(A) were 89.02% and 21.24% in 17 d. The process of drug in vitro release accorded with the model of Riger-Peppas. Tetradrine-tashionone II(A) -PLGA composite microspheres had slow-release effect, and it could significantly reduce the burst release, prolong the therapeutic time, decrease the dosage of drugs and provide a new idea and method to prepare traditional Chinese medicine compound.
