1. Trichophyton rubrum: Relationship between susceptibilities to antifungal agents and species specificities

Objective: To study the relationship between susceptibilities of Trichophyton rubrum strains to antifungal agents and their species specificities. Methods: The susceptibilities of Trichophyton rubrum strains to itraconazole, ketoconazole, fluconazole, terbinafine, naftifine, 5-flucytosine and amphotericin B were evaluated using a modified microdilution method. The relationship between susceptibilities and genotypes and phenotypes of Trichophyton rubrum strains with different origins was analyzed by random amplified polymorphic DNA (RAPD). Results: The Trichophyton rubrum strains showed narrow minimum inhibitory concentration (MIC) ranges to terbinafine (0.016-0.032 μg/ml), naftifine (0.032-0.063 μg/ml) and itraconazole (0.25-1 μg/ml), whereas they showed broader MIC ranges to ketoconzole (0.25-2 μg/ml) and fluconazole (1-32 μg/ml). MICs of Trichophyton rubrum strains to terbinafine (M0 = 0.032 μg/ml) and naftifine (M0 = 0.032 μg/ml) were the lowest among 7 antifungal agents. Wilcoxon test (Kruskal-Wallis test) suggested that there was no significant relationship of MICs to terbinafine, naftifine, itraconazole and amphotericin B with the genotypes, phenotypes and origins of the Trichophyton rubrum strains. Conclusion: The antifungal susceptibility of Trichophyton rubrum strains may not be related to their genotypes, phenotypes or from which part of the body they are isolated.
2.Analysis on corneal spherical aberration in patients with cataract in Kaifeng area

AIM:To find out the mean corneal spherical aberration and its changes with age in Kaifeng population.
METHODS: A total of 466 eyes of 427 Kaifeng subjects with cataract, whose age ranged from 52 to 75 years old, were divided into three groups according to the age. All the subjects were included in measuring K-readings of cornea, the corneal anterior and posterior surfaces of the spherical aberration and the total corneal spherical aberration using Pentacam(Oculus, Germany) at the 6-mm optical zone.
RESULTS:The mean corneal spherical aberration( CSA) of the whole groups was 0. 339±0. 153μm. Patients from 50 to 60 years old had CSA of 0. 322±0. 137μm, 0. 326±0.157μm was the CSA of the patients from 60 to 70 years old, while those from 70 to 80 years old have a CSA of 0.401 ± 0. 182μm. The corneal K - readings had no correlation with the total corneal spherical aberration ( r=0. 034, P>0. 05), anterior surfaces CSA, posterior surfaces and total CSA were positively correlated with age ( r =0.253, 0. 327, 0. 233, all P<0. 01).
CONCLUSION:CSA is varied among the population and CSA of elderly in Kaifeng area are slightly higher than white people, and positively correlated with age.
3.Relationship between Tumor Necrosis Factor-? and Interferon-? Polymorphism with Infant Cytomegalovirus Hepatitis

Objective To explore the relationship between tumor necrosis factor(TNF)-? promoter G238A,G308A and interferon(IFN)-?+874A/T polymorphism and susceptibility to cytomegalovirus(CMV) hepatitis.Methods A TaqMan fluorescence polymerase chain reaction(PCR) in the IFN-?+874A/T and TNF-? gene promoter region single nucleotide polymorphism was tested in the subjects,including 87 infants with CMV hepatitis and 89 children without CMV hepatitis.Results No evident difference of TNF-? +238G/A and+308G/A allele frequency was found between infants with CMV hepatitis and controls.The cases of IFN-?+874 AA,AT and TT genotype were 64,20,3 cases in the infant CMV hepatitis group,and 45,26 and 18 cases in the control group,respectively.There was significant difference in IFN-?+874 allele genotype and frequency between infants with CMV hepatitis and the control group(P=0.001,P
4.Clinical Study on Treatment of Hepatocyte Growth - Promoting Factor for Infant Hepatitis Syndrome

Objective To explore the clinical efficacy and side effect of hepatocyte growth - promoting factor in treatment of infant hepatitis syndrome. Methods Sixty one cases of infant hepatitis syndrome were chased as the treatment group who hospitalized in our hospital from March 2002 to Feb 2003,and 54 cases of infant hepatitis syndrome as control group who hospitalized during March 2001 to Feb 2002. The treatment group were administrated with hepatocyte growth - promoting factor for 2 weeks. We obser-ved the recovery of patient's liver function (TBIL.ALT, AST) and the side effect of hepatocyte growth- promoting factor after two weeks of the treatment. Aleutian mink disease parvovirus (AMDV) causes a persistent infection associated with immune complex disease, hypergammaglobulinemia, and high levels of antiviral antibodies. Despite the presence of an antibody, the virus is not cleared in vivo. Pre-existing antibodies may enhance viral infections, by Fc-receptor-mediated antibody-dependent enhancement (ADE), but the mechanism that underlies ADE has not been fully defined. Three models have been proposed, including: (1) interactions between antibody and FcR, complement C3 fragment and CR, or between C1q and C1qR, which promotes viral attachment to cells; (2) suppression of IFN-gamma-mediated host-cell antiviral gene expression by the upregulation of negative regulators of pathogen pattern recognition; and (3) the promotion of early IL-10 secretion. In addition, the role of cytokine IL-6 in ADE mediated disease development is discussed, to facilitate a better understanding of the pathogenesis of AMDV infection, as well as give insights into rational vaccine design approaches.

9.Effect of procyanidins on gene expression of par-4 and bcl-2 in PC12 cells induced by A?_(25-35)

Objective To study the effect of procyanidins (PC) on mRNA and protein expression of par-4 and bcl-2 genes in PC12 cells induced by A?_ 25-35 . Methods Cell survival rate was evaluated by MTT assay and apoptosis was analyzed by Hoechst 33258-PI fluorescence staining. The expressions of mRNA and protein for par-4 and bcl-2 were tested by RT-PCR and Western blotting. Results Pretreatment with different concentrations of PC (5、10、20, and 30 mg/L) for 1 h increased the survival rate of PC12 cell in a dose-dependent manner. PC prevented the PC12 cells nuclei from shrinkage, condensation, and cleavage induced by A?_ 25-35 . PC decreased the expression of par-4 mRNA and protein, and increased the expression of bcl-2 mRNA and protein. Conclusion PC can protect PC12 cells from apoptosis induced by A?_ 25-35 in a dose dependent manner. The mechanism of protection is likely related to decreasing the par-4 gene expression and increasing the bcl-2 gene expression.
