2.The experimental study on melatonin gastro intestinal motility and plasma levels of stress hormones in overtraining rat.

OBJECTIVETo study the effect of melatonin on the gastrointestinal motility and plasma levels of the stress hormone in overtraining rats.
METHODThirty adult SD rats were randomly divided into three groups (n = 10): control group, over-training group, melatonin intervention group. 30 min before each training, rats in the control and over-training groups were fed with normal saline (15 mg/kg) once a day and 5 times per week, while rats in the melatonin intervention group were administrated with melatonin, perfusion in the intervention group (15 mg/kg). Excessive training group and melatonin intervention group rats were subjected to excessive training at 5 times a week for 6 weeks. After 6 weeks, the gastric emptying rate, small intestinal propulsion ratio and levels of plasma motilin (MTL) and calcitonin gene-related peptide (CGRP), cortisol (CORT) and catecholamines (CA) were observed in all groups.
RESULTSCompared with the control group, the gastric emptying rate, small intestinal propulsion ratio and levels of plasma MTL, CORT and CA were increased significantly (P < 0.01) while the content of CGRP was reduced (P < 0.01) in over-training group. After treated with melatonin, this trend was reversed, that was, the gastric emptying rate, small intestinal propulsion ratio and levels of plasma MTL, CORT and CA were surpressed significantly (P < 0.01) while the content of CGRP was improved obviously (P < 0.01) in over-training group.
CONCLUSIONMelatonin plays an important role in protecting gastrointestinal tract from dysfunction, in which MTL, CGRP, CORT and CA are all involved.

3.Preliminary report of expression of hepcidin gene in Wistar rats ACD model is affected by EPO

Objective To investigate the expression changes of the liver hepciding gene in the formation process of Wistar male rats anemia in chronic disease(ACD) model. Methods ACD rat model was established by repeated injection of Freund' s complete adjuvant. The relationship between hepcidin mRNA expression levels of normal rats, ACD rats and ACD rats after intra -peritoneal injection of EPO. Results Anemia was induced in the rat model. In the ACD group, hepcidin mRNA expression levels increased; after intra-peritoneal injection of EPO, hepcidin mRNA expression levels decreased, anemia is improved. Conclusion Hepcidin plays an important role in pathogenesis of ACD. EPO could be decreased hepcidin mRNA expression levels by anti- inflammatory reaction and decrease cell factors. Decrease of hepcidin level as early as possible will benefit to block the development of anemia, that will provide a new strategy of therapy for ACD.

5.Influence of dose distribution selected from different beam orientation for a nasopharyngeal carcinoma IMRT plan

Purpose:To evaluate the different dose distribution created by three different plans on a nasopharyngeal carcinoma case with simultaneous integrated boost intensity modulated radiation therapy (SIB-IMRT). Methods:we designed three different SIB-IMRT plans on a nasopharyngeal carcinoma case, one was from anterior direction of the head, 5 non-equally spaced fields, another was from posterior direction of the head, 5 non-equally spaced fields also, third was 5 equally spaced fields around the head. Each plan had its dose distribution to the target and main critical structures.Results:No significant difference of dose distribution to the target when comparing the previous three different IMRT plans. However, dose to the parotids delivered from 5-equally spacd was obviously lower than the other plans, dose to the optic chiasm was much lower created by the plan designed from the posterior direction of the head with 5 non-equally spacea field.Conclusions:For a nasopharyngeal carcinoma case, there is no significant difference of dose distribution to the target compared with three different 5-beam orientation SIB-IMRT plans. To protect parotids, we recommend designing a plan with equal beam direction around the head. When the target is very close to the optic chiasm, plauning from posterior direction of the head should be better.
7.Expression of TfR2 in the anemia of chronic disease of rats

Objective Based on establishing a rat model of anemia of chronic disease (ACD), expression of transferrin receptor 2(TfR2) in the liver cultured with erthropoietin(EPO) and cultured without EPO to the action of TfR2 in the ACD were detected. Methods Anemia was induced by injecting freund adjuvant for third times to the traditional model rats of adjuvant arthritis to enhance their immune responses the levels of the expression level of TfR2 mRNA in the livers of rats among different treated groups were compared. Results In the ACD group the expression of TfR2 mRNA in the livers of rats was higher than in the normal control goup(P =0.001), is significant in anemia; while in the EPO therapy group the expression of TfR2 mRNA was less than in the ACD goup (P=0.001), is improved in anemia. Conclusion TfR2 may be revalant to ACD.
8. Research status of Lonicera Japonicae Flos and Lonicerae Flos and its key issues for quality control

More and more disputes have happened to confront continuously since the separation of Lonicera Japonicae Flos and Lonicerae Flos in the 2005 edition of Chinese Pharmacopoeia, however, there are no differences in the properties, flavors, and channel tropism, functions of curing, usage and dosage in the 2015 edition of Chinese Pharmacopoeia. By consulting literatures on the medical history, identification, chemical composition, pharmacological action, quality control, and other aspects of Lonicera Japonicae Flos and Lonicerae Flos at home and abroad in recent years, we explore the similarities and differences between Lonicera Japonicae Flos and Lonicerae Flos, and the key issues of quality control in order to solve the problem of the combination and separation between them, which can lay a foundation for promoting the development of traditional Chinese medicinal materials industry.
9.The study of genome DNA methylation in rheumatoid arthritis

Objective To study the genome DNA methylation in rheumatoid arthirits(RA)and the re- lated factors of DNA methylation.Methods Twenty-first cases with RA and 20 controls were recruited to par- ticipate the study.Plasma Hcy,SAM,SAH,the MTHFR gene C677T polymorphism and the expression of LFA-1 in CD4~+T cells was measured in all patients and controls.Results①The SAM levels were lower sig- nificantly in RA groups than in controls.The SAH levels were higher significantly in RA groups than in con- trols.②There was significant inverse correlation between plasma Hcy level and SAM level(r=-0.932,P<0.01). There was significant positive correlation between plasma Hcy level and SAH level(r=0.924,P<0.01).③The expression of LFA-1 in CD4~+T cells was higher significantly in RA groups than in controls.There was a signif- icant positive correlation between LFA-1 expression level and Hcy level(r=0.557,P<0.01),a significant in- verse correlation between LFA-1 expression level and SAM level(r=-0.651,P<0.01).④The MTHFR gene mu- tation lead to dramatically increase of Hcy,SAH level and the expression of LFA-1 level in CD4~+T cells and genome DNA hypomethylation.Conclusion①Hypomethylation of genome DNA is found in most RA pa- tients.②The factors associated with genome DNA hypomethylation include MTHFR gene mutation and hyper- homocysteinemia.③The expression of LFA-1 in CD4~+ T cells is higer in RA groups than in controls,which re- lates to the DNA methylation level and the MTHFR gene C677T polymorphism.
