1.Effects of plasminogen activator inhibitor-1 antisense RNA on aorta endothelial cells cultured

Objective To investigate the role of antisense RNA of plasminogen activator inhibitor 1 (PAI 1) in regulating the expression of PAI 1 and vascular endothelial growth factor (VEGF) in aorta endothelial cells (EC) cultured in vitro. Methods The second extron of PAI 1 was amplified by polymerase chain reaction(PCR) and the product was inserted into eukaryotic cell expression vector pcDNA3.1(-) to construct PAI 1 antisence RNA recombinant plasmid. The recombinant plasmid was transfected into EC and the PAI 1 expression was detected by immunohistochemistry, Westernblot and ELISA. The effects of PAI 1 variation on VEGF were examined by immunofluorescence method. Results PAI 1 antigen was the lowest (0 017 ng/ml) in cells and the immunofluorescence representing the expression of VEGF in the cytoplasm showed the weakest at the third day after transfection. At the fifth day, PAI 1 antigen increased to 0 093 ng/ml with VEGF expression increased correspondingly. At the seventh day, PAI 1 antigen(0 143 ng/ml) and VEGF increased closed to normal level. Conclusions PAI 1 antisense RNA blocked the translation of PAI 1 proteins effectively and inhibited the expression of VEGF in aorta endothelial cells.
2.Progress in the Study of Chemokine CXCL9/Mig

Chemokine CXCL9/Mig (monokine induced by IFN-?) belongs to the subfamily of chemotactic cytokines known as CXC-chemokines. In vivo CXCL9 is mainly induced by IFN-? in macrophages and primary glial cells. In vitro, CXCL9 can be secreted by cells such as macrophages, microvascular endothelial cells and neutrophils, in response to the synergy of IFN-? and TLR(toll-like receptor) ligands. CXCL9 is a chemoattractant for activated T lymphocytes, tumor-infiltrating T-lymphocytes, but not for neutrophils or monocytes. The receptor specific for CXCL9 is CXCR3, a G protein-coupled protein which has seven transmembrane domain. The structure and the chemical characterization of CXCL9, as well as its effects on autoimmune deseases, allograft rejection, cancer therapy were reviewed.

4.Correlation between Aquaporin-4 Antibody and Optic Neuropathy in Patients with Neuromyelitis Optica Spectrum Disorder

Objective To determine the correlation between aquaporin-4 antibody (AQP4-Ab) and optic neuropathy in patients with neuromyelitis optica spectrum disorder (NMOSD).Methods The clinical and biochemical data of 53 patients with NMOSD diagnosis based on AQP4-Ab level in Changhai Hospital between January 2010 and October 2015 were retrospectively analyzed.According to optic neuropathy occurrence,the NMOSD patients were divided into optic neuropathy and non-optic neuropathy groups.Clinical and biochemical characteristics were compared between the two groups.According to the serum AQP4-Ab levels,the NMOSD patients were divided into AQP4-Ab seropositive and seronegative groups.The incidence of optic neuropathy was compared between the two groups.The correlation between optic neuropathy and AQP4-Ab levels was analyzed.Results Between the optic neuropathy and non-optic neuropathy groups,no significant differences in sex,age at onset,disease course,serum alanine aminotransferase levels,protein levels in cerebral spinal fluid,IgG index,and oligoclonal band were observed (P > 0.05).However,statistically significant differences were found in frequency,superficial sensory impairment,serum creatinine level,and serum AQP4-Ab level (P < 0.05).Between the AQP4-Ab sempositive and semnegative groups,a statistically significant difference in the incidence of optic neuropathy was observed (F =4.93,P < 0.05).The incidence of optic neuropathy positively correlated with AQP4-Ab levels (r =0.297,P < 0.05).Conclusion NMOSD patients with AQP4-Ab seropositivity could be prone to optic neuropathy,and the correlation may be beneficial to early diagnosis,therapy,and monitoring of NMOSD.
5.Effect of comprehensive schistosomiasis control measures in Kaihua County from 1996 to 2015

Objective To evaluate the control effect of comprehensive measures of schistosomiasis after its transmission in?terruption in Kaihua County,Zhejiang Province,so as to provide the references for further consolidation of schistosomiasis con?trol. Methods The data of Oncomelania hupensis snail survey and control,as well as environmental reform in Kaihua County were collected and analyzed from 1996 to 2015. Results From 1996 to 2015,totally 2 635 snail habitats and 102.75 hm2 area with snails were found,and 125.4 thousand snails were dissected and no one was schistosome infected. The accumulated snail control area was 4 932.98 hm2,and the area with snails was effectively reduced by the comprehensive control measures. Conclu?sion The schistosomiasis control effect could be consolidated by the comprehensive control measures emphasizing environmen?tal reconstruction,and the snail surveillance work still should be strengthened.
6.Detection of microvesicle miRNA expression in ALL subtypes and analysis of their functional roles.

Microvesicles (MVs) are the heterogeneous mixtures of vesicles. MVs released by leukemia cells constitute an important part of the leukemia microenvironment. MVs might act as important reservoirs of microRNAs (miRNAs). It is worth evaluating whether MVs possess some unique miRNA contents that are valuable in understanding the pathogenesis. In this study, we investigated the miRNA expression patterns of Nalm-6-derived MVs, Jurkat-derived MVs and normal cell-derived MVs using miRNA microarrays. The potential target genes regulated by differentially expressed miRNAs were also predicted and analyzed. Results demonstrated that 182 miRNAs and 166 miRNAs were differentially expressed in Nalm-6-MVs and Jurkat-MVs, respectively. Many oncogenes, tumor suppressors and signal pathway genes were targeted by these aberrantly expressed miRNAs, which might contribute to the development of B-ALL or T-ALL. Our findings expanded the potential diagnostic markers of ALL and provided useful information for ALL pathogenesis.
8.Chemical constituents from herb of Epimedium brevicornum.

OBJECTIVETo investigate the chemical constituents of Epimedium brevicornum.
METHODThe chemical constituents were isolated by using silica gel column chromatography and preparative TLC. The structures were identified on the basis of physical-chemical constants and spectral data.
RESULTFive compounds were isolated and identified as hyperoside, icariin, epimedin B, epimedin C, inositol.
CONCLUSIONCompound I and III - V were isolated from the plant for the first time.

10.Influences of long-term and repetitive exposure to hyperbaric oxygen on oxidative stress

Objective To evaluate the levels of oxidative stress in patients undergoing long-term and repetitive exposure to hyperbaric oxygen (HBO) treatment. Methods 16 healthy volunteers and 58 patients with sub-acute sudden hearing loss (SHL) exposed to HBO were included in the study. Oxidative stress indices (malondialdehyde, MDA, advanced oxidation protein products, AOPP; superoxide dismutase, SOD) were measured in peripheral blood samples collected at the 5th,10th, 20th and 30th HBO treatments sessions (PO2 0.18 MPa, 1 session per day and 5 sessions per week) and under normal ambient pressure respectively. Results After 5th,10th, 20th and 30th sessions of HBOT, no relevant differences in these three indices were detected compared to pre-HBO exposure, between healthy volunteers (P > 0.05). Conclusions The long-term repetitive HBO treatment for 0.18 MPa of PO2 and 30 sessions could not affect in particular the response of the oxidative stress in healthy persons and patients with sub-acute SHL. The influence on three indices of patients with abnormal situation of oxidative stress undergoing lower pressure of HBO (0.18 MPa) is under investigation.
