Objective To evaluate the effects of compound glycyrrhizin on the percentage of Th17 cells and expression of interleukin(IL)-17A in peripheral blood of patients with psoriasis vulgaris, and to explore their relationship with therapeutic effects. Methods A total of 30 patients with mild to moderate progressive psoriasis vulgaris were randomly and equally divided into two groups:group 1 treated with compound glycyrrhizin injection, antihistamines and topical drugs, group 2 treated with antihistamines and topical drugs. Twelve healthy human subjects served as the control group. Blood samples were collected from the patients 1 day before start of treatment and after 3 weeks of treatment, and from the control group. Flow cytometry and enzyme-linked immunosorbent assay (ELISA)were performed to determine the percentage of Th17 cells and expression level of IL-17A respectively in the peripheral blood samples. Results Before the treatment, the percentage of Th17 cells and expression level of IL-17A were both significantly higher in the two patient groups than in the control group (all P<0.05). After the treatment, group 1 showed significant decreases in the percentage of Th17 cells and expression level of IL-17A compared with those before the treatment (both P<0.05), while no significant differences were observed in group 2 between pre-and posttreatment IL-17A expression level or Th17 cell percentage (both P>0.05). Furthermore, both Th17 cell percentage and IL-17A expression were significantly different between the two patient groups after the treatment (both P< 0.05). Conclusion Compound glycyrrhizin may treat psoriasis vulgaris by regulating Th17 cell percentage and IL-17A expression in peripheral blood.

Journal of Zhejiang University. Medical sciences 2006;35(1):29-33
OBJECTIVETo determine the possible difference in vasodialtation effect of quercetin and rutin.
METHODSThe isolated rat thoracic aorta was treated with phenylephrine (PE), and the effects of quercetin and rutin on the preconstricted aorta rings with or without endothelium were determined by organ bath technique. Nitric oxide synthase inhibitor L-N(G)-nitroarginine methyl-ester (L-NAME), guanylyl cyclase inhibitor methylene blue, cyclooxygenase inhibitor indomethacin were used to explore the mechanism.
RESULTSQuercetin (10-160 micromol/L) caused vasorelaxation of aorta rings preconstricted with PE in endothelium-intact and denuded aorta rings in a dose-dependent manner. Rutin(10-160 micromol/L) caused dose-dependent vasorelaxation in endothelium-intact rings preconstricted with phenylephrine, but not in denuded aorta rings. The maximal response (Rmax) values calculated from vasorelaxation curves of quercetin and rutin were (77.20+/-6.11)% and (44.28+/-7.48)%, respectively. There was no difference between median effective concentration (EC(50)) values of quercetin and rutin. Pretreatment with L-NAME (0.1 mmol/L) abolished the vasorelaxation by rutin, but did not influence the vasodilating effect of quercetin in endothelium-intact rings. Pretreatment with methylene blue (10 mmol/L) canceled the vasorelaxation both by quercetin and rutin. Pretreatment with indomethacin (10 micromol/L) attenuated the vasodilatation of quercetin, but did not affect the vascular effect of rutin.
CONCLUSIONThe vasodilatation effect of quercetin is more potent than rutin. The vasodilatation effect of quercetin might be mediated by guanylyl cyclase and cyclooxygenase-dependent pathway, while the vasodilatation by rutin might be via nitric oxide-guanylyl cyclase pathway.

AIMTo investigate the relationship among sexual differences of motion sickness (MS), AVP levels of plasma and pituitary and the expression of pituitary V1b receptors for further understanding of the MS mechanisms.
METHODSThe conditioned taste aversion (CTA) of 0.15% saccharin sodium solution (SSS) was served as MS model. 98 (49 male and 49 female) rats were used in this experiment, 50 for the detection of the AVP level in plasma and pituitary with radioimmunoassay (RIA), 12 for the observation of the number of V1b receptor-positive neurons in the pituitary with the fluorescence immunohistochemistry method, the rest for the evaluation of the expression of V1b receptor in the pituitary by Western blot.
RESULTSWith regard to male rats, decrease of the drinking volume of 0.15% SSS was greater in female rats after rotatory stimulation. The plasma AVP concentration of female rats was significantly higher than that of males under normal conditions, but reduced significantly after rotatory stimulation. However, no significant change was found in male rats. In addition, the pituitary AVP level of the female rats was significantly higher than that of the male rats under normal conditions, but decreased at 8 h and significantly at 24 h after rotation. Similarly, the pituitary AVP level of male rats also decreased significantly at 8 h after rotation, but this decrease was not comparable to that of the females. At 24 h after rotation the pituitary AVP level almost recovered in male rats. In the pituitary, which was related to the stress response, the V1b receptor-positive neurons and the expression level of V1b receptor in female rats were significantly higher than those of the male rats, but they decreased significantly after rotation, while no apparent change was detected in the male rats.
CONCLUSIONThe changes of plasma and pituitary AVP and V1b receptor level of the pituitary after rotatory stimulation are different between male and female rats and the AVP secretion of the pituitary may be involved in the sexual difference in susceptibility to motion sickness.
