2. Regularity of absorption and distribution of nitrogen, phosphorus, potassium, calcium, and magnesium in Artemisia annua

Objective: To study the regularity of dry matter accumulation and its absorption and distribution of nitrogen (N), phosphorus (P), potassium (K), calcium (Ca), and magnesium (Mg) in Artemisia annua, and to provide a scientific fertilizer application for the standardized cultivation. Methods: The sampling was carried out in the different growing periods of A. annua under field condition, and the amount of dry matter and the contents of N, P, K, Ca, and Mg in all parts of the plant were determined, respectively. Results: The dry matter mainly distributed in leaves within 40 d after transplanting, and this period could be defined as seedling period. The period within 40-100 d could be defined as vegetative growth period and the dry matter mainly distributed in leaves and branches. The period within 100-180 d could be defined as reproductive growth period. And the period of 100-140 d could be defined as earlier reproductive growth period, and the dry matter mainly distributed in branches and flower buds during this time. The period of 140-180 d could be defined as later reproductive growth period, the dry matter mainly distributed in branches and seeds during this time. The accumulation of nutrition was the lowest in seedling period; but the uptake of nutrition increased in vegetative growth period, and mainly distributed in leaves and branches. Nutrition mainly distributed in leaves earlier reproductive growth period, but transferred to flower buds gradually. Nutrition mainly distributed in seeds in the later reproductive growth period. The dry matter accumulation lineally increased with the assimilation and accumulation of N, K, Mg, Ca, and P. The uptake proportion of them was 4.21:4.17:0.74:0.63:0.24 in the whole growth period. Conclusion: Low nutrition should be applied during seedling stage to controlling the loss of nutrition. During the period of 40-140 d after transplanting, A. annua grows quickly, and mineral nutrition must be supplied especially N and K. The maximum uptake period of P, Ca, and Mg is in 100-120 d, so Ca, Mg, and P fertilizers should be applied earlier or by foliage spray to promote the formation of flower buds and seeds.
4.A rabbit model of cervical spondylosis established by stimulation of wind, cold and dampness

OBJECTIVE: To make an animal model of cervical spondylosis (arthralgia syndrome type) with stimulation of wind, cold, and dampness. METHODS: Twenty-four 8 months old male New Zealand white rabbits were randomly allocated into four groups: normal control group, light stimulation group, moderate stimulation group and severe stimulation group. The wind speed was 10.8-13.8 m/s, the temperature was (5+/-0.5)degrees centigrade, and the humidity was 100%. The rabbits of light, moderate, and severe stimulation groups were kept in the above-mentioned environments for 4 hours everyday, and for a total of 32, 64, and 128 hours, respectively. The intervertebral discs were stained with HE method, and observed with a light microscope. Prostaglandin E(2) (PGE(2)), 6-ketone-prostaglandin F1alpha (6-K-PGF(1alpha)) and thromboxane B(2) (TXB(2)) contents were measured by ELISA. Fas and Bcl-2 expressions were examined by immunohistochemical avidin-biotin peroxidose complex technique. Interleukin-1beta (IL-1beta), tumor necrosis factor alpha (TNF-alpha), and transforming growth factor beta (TGF-beta) mRNA expressions were examined by reverse transcription-polymerase chain reaction. RESULTS: The nucleus pulposus of rabbits in the light and moderate stimulation groups shrunken, and in the severe stimulation group, the anulus fibrosus loosed or ruptured, and the cartilage end-plate became proliferated. Compared with rabbits in the normal control group, the PGE(2) content rose in the light stimulation group, the contents of PGE(2), 6-K-PGF(1alpha), and TXB(2) increased, the expressions of IL-1beta and TNF-alpha mRNAs and Fas were up-regulated, and the expressions of TGF-beta mRNA and Bcl-2 were down-regulated in the moderate and severe stimulation groups. The expression of Fas was up-regulated mostly and Bcl-2 was down-regulated mostly in the severe group. CONCLUSION: Moderate and severe stimulations of wind, cold and dampness can lead to degeneration of cervical intervertebral discs of rabbits. The model corresponds to the theory of traditional Chinese medicine about arthralgia syndrome caused by wind, cold and dampness.

Objective:Immune cells and inflammatory mediators play important roles in the development of atherosclerotic vascular inflammation.This study focuses on the direct effect ofIL-27 on human coronary artery endothelial cells.Methods:In this study,the effects of IL-27 and TNF-α on inflammatory cytokines and chemokines were investigated.Results:IL-27 could significantly enhance the TNF-α-mediated upregulation of inflammatory cytokine IL-6 and chemokines CCL5 from human coronary artery endothelial cells.The release of IL-6 and CCL5 was significantly suppressed by specific signaling molecule inhibitors.Conclusion:The induction of these mediators from the human coronary artery endothelial cells could be differentially regulated by the c-Jun N-terminal kinase,p38 mitogen-activated protein kinase,and nuclear factor-κB pathways.These results provided new insights into the effect of IL-27 on the TNF-α mediated activation of human coronary artery endothelial cells in atherosclerotic vascular inflammation.
7.Effects of storage time on quality of Desmodium styracifolium seeds.

The dynamic changes of germination percentage, germination potential, thousand-seed weight, antioxidase activity in Desmodium styracifolium seeds with different storage time were tested, and electrical conductivity, contents of soluble sugar, soluble protein, starch in seed leach liquor were also determined in order to reveal the mechanism of seed deterioration. The results as the following. (1) The germination percentage, germination potential and thousand-seed weight of D. styracifolium seeds declined, while the seed coat color darkened with the extension of storage time. (2) The activities of superoxide dismutase (SOD) and peroxidase (POD) decreased with the prolongation of storage period. The SOD activity declined fastest in 1,095-1,185 d of storage, while the POD activity declined significantly in 365-395 d of storage. (3) The electrical conductivity and the contents of soluble sugar, starch in seed leach liquor increased, while the content of soluble protein declined with the extension of storage time. (4) Correlation analysis indicated that the germination percentage, germination potential and thousand-seed weight of D. styracifolium seeds have a significantly positive correlation with SOD and POD activity, while have a significantly negative correlation with the electrical conductivity, contents of soluble sugar and starch. It can be concluded that during the storage of D. styracifolium seeds, physiological and biochemical changes including decrease in antioxidase activity, rise in electrical conductivity, degradation effluent of soluble sugar and starch, degradation of soluble protein were the main factors leading to the seed deterioration.

8.Effect of electroacupuncture on hemodynamics at peri-intratracheal intubation period under general anesthesia.

OBJECTIVETo observe effect of electroacupuncture on hemodynamics at peri-intratracheal intubation period under general anesthesia.
METHODSSixty cases at peri-intratracheal intubation period under general anesthesia were randomly divided into electroacupuncture group [Before anesthesia induction, electroacupuncture stimulation was given at Neiguan (PC 6), Hegu (LI 4), Quchi (LI 11), Shenmen (HT 7)], and control group, 30 cases in each group. All the patients in the two groups received intratracheal intubation with light-guide laryngoscopy under general anesthesia with midazolam, fentanyl, isopropyl, succinylcholine, etc.; changes of various indexes of hemodynamics were detected at anesthesia induction and different time points of the intubation.
RESULTSAt the anesthesia induction, various indexes of hemodynamics in the electroacupuncture group were relatively stable, and decreased significantly in the control group (P < 0.01); during intratracheal intubation, the various indexes in the electroacupuncture group (P < 0.05) and in the control group (P < 0.01) increased significantly as compared with those before anesthesia induction; 5 min after intubation, the various indexes in both the two groups restored normal.
CONCLUSIONElectroacupuncture at Neiguan (PC 6) and other points can stabilize changes of hemodynamics at the intratracheal intubation and increase safety of operation under anesthesia.

9.Association between endothelial nitric oxide synthase gene 4a/b polymorphism and IgA nephropathy.

OBJECTIVETo investigate the relation between endothelial nitric oxide synthase (eNOS) gene polymorphism and IgA nephropathy (IgAN).
METHODSThe variable number of tandem repeats (VNTR) polymorphism of eNOS gene was analyzed with PCR in 296 Chinese Han patients with IgAN groups and 310 healthy Han subjects from Beijing, Tianjin and Hebei Province, and the gene frequency and allele frequency were compared between the two groups.
RESULTSIn these Chinese Han subjects involved in this study, VNTR of eNOS gene showed 3 genetypes, namely eNOS4a, eNOS4b and eNOS4a/4b. No significant difference was found in the gene frequency between IgAN group and the healthy subjects.
CONCLUSIONSIgAN is not associated with VNTR polymorphism of eNOS gene, and no sufficient evidence has been obtained to establish the relation of eNOS VNTR polymorphism with the genetic factors of IgAN. The role of eNOS gene polymorphism in IgAN can be heterogeneous in regard to different races or areas.

10.The expression of integrin-linked kinase(ILK) in the transdifferentiation of tubular epithelial cell and its correlation with senescence

Objective To investigate the expression and function of integrin-iinked kinase (ILK) in the transdifferentiation process of primary tubular epithelial cell( TEC) and its effect on the accumulation of extracellular matrix (ECM) in the aging process. Methods Primary TEC was cultured from kidneys of male Wistar rats aged 3 and 24 months. Then the primary TEC was stimulated by TGF-? at the concentrations of 0, 5, 10 and 20-40/?g/L for 24 h. The TEC expressions of ?-SMA,ILK and F-actin were detected by immunocytochemistry or indirect immunofluorescence. FN or ILK protein expression levels were tested by Western-blot. DIG-labelled cRNA probe of rat ILK in situ hybridization was obtained by transcription in vitro, with which ILK expression level and the location of TEC were detected. Results ?-SMA expression levels were very low in young or aging rat TEC on the base situation, no significant difference was found between the two groups. However, ILK and F-actin levels in 24 months group were higher than in 3 months group. Along with increasing concentration of TGF-?, ?-SMA, ILK and F-actin levels were increased.
