HLA (human leukocyte antigen) is critical molecule relevant to immunoresponse, which is always involved in antigen presentation and determining killing activity of immune cells against tumor. The abnormal expression of HLA in lung cancer is believed to be a common phenomena of tumor biology. It has been demonstrated that HLA expression varies in various types of lung cancer, even in differential stages of one type of lung cancer. HLA expression is highly associated with genre, progression and prognosis of lung cancer. The thorough investigations of the relationship between HLA and lung cancer and of mechanism of HLA abnormality will provide useful information for studying of development of lung cancer and tumor immunotherapy.

Objective To perform the performance verification of the Sysmex XT-2000i hematology analyzer.Methods Accord-ing to requirements of NCCLS EP5-A2,EP15-A2,EP6-A2,EP9-A2 documents,the accuracy,precision and linearity of the Symex XT-2000i hematology were evaluated and the detection results were compared with those detected by the Sysmex XE-2100i which participated in the external quality assessment(EQA)of Ministry of Public Health for long time.Results The accuracy,precision, linearity and contamination rate all are within the permissible range.Conclusion The performance of Sysmex XT-2000i is good,the determination results are reliable.As a perfect hematology analyzer,it can satisfy the hematological analysis of hospital clinical la-boratory.

ObjectiveTo investigate the success rate and energy requirements of ibutilide and direct current(DC)cardioversion,summarize the nursing experience.MethodsForty-six patients with persistent atrial fibrillation were enrolled and underwent DC cardioversion were selelct from May 2009 to January 2011.23 patients received pretreatment with ibutilide before DC (the ibutilide group),23 patients received non drugs before DC (the control group).The cardioversion energy started at 100 J and increased if patients could not acquire sinus rhythm.All the patients were given pertinent care before,during and posttreatment.The success rate and energy requirements of the two groups were compared.Results95.7％ of the patients were successfully cardioverted in the ibutilide group,which was significantly higher than 74.0％ of the control group.The energy required for cardioversion was ( 139.1±45.1 ) J in the ibutilide group,significantly lower than (197.8±53.3) J in the control group.Non patients terminated treatment or appeared complication because of improper nursing.ConclusionsPretreatment with ibutilide enhanced the efficacy of DC cardioversion for the patients with persistent AF at lower energy requirements.Proper nursing care can ensure the smooth process of treatment and decrease the complications.

Objective To explore the change of degrade of clonazepam in serum with single and repeated - dose administration in children with seizure, and find a reasonable method for using the clonazepam. Methods Children with seizures were divided into single - dose paradigam, repeated - dose paradigam, and decreased - dose paradigam. The concentration of CZP in serum was determined by high performance liquid chromatography( HPLC). Results The serum concentrations of clanazepain in single - dose paradigam were (101.9?12.1),(76.9 ? 5.8),(50.7?2.9),(30.9?5.4),(21.5?6.8)?g/L,the time point that the blood samples collected were 15,30,60,120 and 480 min. The serum concentrations in repeated - dose paradigam were (97. 2 ? 6. 1),(130.4? 13. 4), (99. 4 ? 9.8),(79.6?2.4)?g/L,in decreased-dose paradigam were( 101.1 ?13.1),(123.1 ?6. 6), (99.4 ?9. 8), (79. 3 ? 2. 2)?g/L,in these two groups,the time point were 15,45,60 and 120 min. Conclusion Repeated administration of CZP with decreased dose may increase its effectiveness in treatment without substantially increasing toxicity.

Background:MicroRNAs ( miRNAs ) is a class of small non-coding RNAs that regulate gene expression at posttranscriptional level. They are considered to play a critical role in the tumorigenesis and development of various tumors. Aims:To identify the recurrence-specific miRNAs in peripheral blood of patients with recurrent colorectal adenoma by miRNA array for providing sensitive and specific molecular markers for recurrence prediction. Methods:Fifty colorectal adenoma patients who had undergone polypectomy by colonoscopy were re-examined from Aug. 2012 to Sep. 2013. Four recurrent and four non-recurrent patients were selected and miRNA array was used to detect the expression profiles of miRNAs in peripheral blood samples. Differentially expressed miRNAs were identified between recurrent and non-recurrent patients. Real-time PCR was applied to verify the differentially expressed miRNAs identified by microarray. Results:Seven up-regulatedmiRNAs(hp hsa-miR-548ai,hsa-miR-4446-3p,hsa-miR-139-3p,hsa-miR-27a-5p,hsa-miR-10a,hsa-miR-23a and hsa-miR-486-3p)and 4 down-regulated miRNAs( hsa-miR-124,hsa-miR-3613-5p,hsa-miR-495 and hsa-miR-485-5p)were identified by microarray between recurrent and non-recurrent patients. Among them,seven miRNAs were validated by real-time PCR as consistent with microarray. Objective To study the clinical effect of Qiliqiangxin capsule (QC) in patients with chronic congestive heart failure (CHF). Methods According to random number table,64 CHF patients were randomly assigned into therapy group (n=33) and control group (n=31).During the treatment,sweat secretion,heart function improved rate,6 min walking distance,the side-effect and compliance were observed.The levels of serum Na+ and K+ of patients were measured on the 2nd,8th and 15th hospital day.Results Compared with control group,the remission rate of sweat secretion in therapy group was higher after treatment (P0.05);compared with control group,the improved rates of heart function and 6 min walking distance in therapy group were higher after two weeks (P0.05);on the 15th day,the level of serum Na+ in therapy group was higher than that in control group (P0.05).During treatment,33 patients had not any uncomfortable complain about this drug and nobody stopped using this capsule;while there was no complain from the patients of control group.Conclusion The effect of QC about releasing hyperidrosis in a short time is conspicuous.QC can improve heart function,but it plays this role gradually.QC can keep the level of serum Na+ to some extent.There is no influence on the level of serum K+.The tolerance of this capsule is well.

ObjectiveTo investigate the positive rates of serum antibodies to TORCH in women who want to have a baby or in the early period of pregnancy.MethodsSerum TORCH-IgM or TORCH-IgG in 500 women were measured with IgM capture ELISA.ResultsThe positive rates of serum TORCH-IgM were 0 in TOX samples,0.2% in RV samples,0.2% in CMV samples,1.4% in COXB samples and 0 in HSV samples;while the positive rates of serum TORCH-IgG were 0.2% in TOX samples,51.6% in RV samples.ConclusionSerum TORCH-IgM and TORCH-IgG screening in women who want to be a mother or in the early period of pregnancy should be strengthened.

Objective To explore the effect of sodium arsenite (iAs3+) and sodium hydrogen arseuate (iAs5+) exposure on rat multidrug resistance protein 2 (MRP2) expression,and the correlation of liver arsenic and its methylation products,and to provide a basis for further elucidating the mechanism of arsenic toxicity.Methods Seventy wistar rats were randomly divided into seven groups,10 rats in each group.Control group was administrated with deionized water.Sodiun arsenite high-dose group,middle-dose group,low-dose group were administrated with different concentrations of sodium arsenite:20.0,6.7 and 2.2 mg/kg BW every day,respectively.Animals were sacrificed 90 days later by cervical dislocation to collect liver,the expression of MRP2 in the membrane of hepatocyte was determined by Western blotting.HPLC-HGAFS was employed to determine the content or level of arsenic and its methylation products.Correlation between expression of MRP2 and arsenic methylation products was analyzed using Pearson's linear correlation method.Results The MRP2 protein levels of control group,iAs3+ and iAs5+ high-,middle-,low-dose groups were 1.21 ± 0.13,1.85 + 0.09,1.65 + 0.19,1.61 + 0.18,1.69 + 0.04,1.42 + 0.19,1.27 ± 0.10.Compared to control group,MRP2 protein levels of iAs3+ high-,middle-,low-dose group and iAs5+ high-dose group were increased (P ＜ 0.05); Compared with the low-dose group,MRP2 protein levels of iAs3+ and iAs5+ high dose groups were increased(P ＜ 0.05); Comparing the matched doses group of iAs5+ and iAs3+,MRP2 protein levels of iAs3+ high-,middle-,low-dose group were higher than iAs5+(P ＜ 0.05).Meanwhile,there was a significant positive relationship between the expression of MRP2 and the content of iAs3+,MMA and DMA in iAs3+ exposed group(r =0.575,0.678,0.395,all P ＜ 0.05).There was also a significant correlation between the expression of MRP2 and MMA and IMA in iAs5+ exposed group(r =0.593,0.643,all P ＜ 0.05).There was no significant relationship between the expression of MRP2 and the content of iAs5+in iAs5+ exposed group.Conclusions The expression of MRP2 is increased with increasing dose of arsenic exposure,thus resulting in compensatory changes on MRP2 expression in the liver cell membrane.MRP2 efflux may play an important role in the metabolic pathways of iAs3+.The level or load of arsenic methylation in liver is closely related with MRP2 expression.
