3.Transplantation of human umbilical cord mesenchymal stem cells at different gestational age for myocardium revascularization after myocardial infarction

BACKGROUND:Transplantation of stem cells has a beneficial effect on myocardium revascularization and improving cardiac function after myocardial infarction, and HLA-G protein contributes to the formation and maintenance of the immune tolerance.
OBJECTIVE:To investigate the transplantation effects of human umbilical cord mesenchymal stem cells at different gestational age with different HLA-G expression levels on myocardium revascularization after myocardial infarction in rabbits.
METHODS:Thirty healthy New Zealand rabbits were selected and were randomly divided into human smal gestational age umbilical cord-derived mesenchymal stem cells transplantation group, human ful-term umbilical cord-derived mesenchymal stem cells transplantation group and control group. After the rabbits models of acute myocardial infarction had been established, the former two groups were infused different umbilical cord-derived human umbilical cord mesenchymal stem cells labeled with 5-bromo-2-deoxyuridine into the edge and center of myocardial infarct region by multipoint injection. Rabbits in the control group were subjected to an equal volume of serum-free culture medium.
RESULTS AND CONCLUSION:Four weeks after celltransplantation, 5-bromo-2-deoxyuridine-positive cells were found surrounding the infarct site in both transplantation groups. Myocardial fibrosis and myocardial infarct size were significantly lower in both transplantation groups than those of the control group (P<0.05), and there was a significant difference between the two transplantation groups (P<0.01). The positive staining of factor VII indicated that capil ary density was increased significantly in the smal gestational age umbilical cord-derived mesenchymal stem cells transplantation group as compared with the ful-term umbilical cord-derived mesenchymal stem cells transplantation group (P<0.01), and a sstatistical difference was found between two transplantation groups and the control group (P<0.01). Transplantation of human umbilical cord mesenchymal stem cells with high HLA-G expression increases new capil ary vessels and improves myocardium revascularization. Al indicate that human smal gestational age umbilical cord mesenchymal stem cells have the potential to become the better source of cardiomyocytes transplantation.
4.Relationship between the intracellular calcium concentration changes and left ventricular hypertrophy and function in the spontaneously hypertensive rats

AIM: To elucidate the relationship between the intracellular calcium concentration changes and left ventricular hypertrophy and function in the spontaneously hypertensive rats (SHR). METHODS: Intracellular free calcium concentrations were measured by Fura 2 methodology and left ventricular function quantitated by cardiac catheterization in 20 SHR aged 10, 22, and 34 weeks and 20 age-matched Wistar-kyoto (WKY) rats. RESULTS: (1) The systolic blood pressure(SBP), intracellular calcium concentrations and left ventricular mass / body weight index ( LVM/BW) were significantly higher in all three age groups of SHR than the corresponding groups of WKY; (2) Compared with age-matched WKY groups, the peak left ventricular pressure descending rate(-d p /d t max ) decreased while left ventricular relaxation time constant (?)increased significantly in SHR aged 22 and 34 weeks. 5.Expressions of gelatinases in diffuse proliferative lupus nephritis and its clinical significance

Objective To investigate the roles and significances of MMP-2 and MMP-9 in diffuse proliferative lupus nephritis by repeated renal biopsy.Methods Seventeen patients diagnosed by renal biopsy as WHO typeⅣlupus nephritis were analyzed by immunohistochemistry staining for MMP-2 and MMP-9. Double staining for MMP-2 and MT1-MMP,MMP-9 and CD68 were also performed.Patients had repeated renal biopsy after followed up for 2.5 years.The relationship between expressions of gelatinases and pathological activity index and clinical data were studied.Results MMP-2 immunoreactivity was detected in normal controls and was increased in diffuse proliferative lupus nephritis.MMP-9 staining,which was almost negative in normal giomeruli,was increased much more significantly in diffuse proliferative lupus nephritis. The immunoreactivity of MMP-2 and MMP-9 was positive in MT1-MMP staining and CD68-positive macrophages, respectively.The expression of MMP-2 and MMP-9 was reduced by 70% and 62% in 10 patients whose clinical condition was partially alleviated,while the expressions in 7 patients whose clinical condition was not alleviated,were only reduced by 27% and 32%.The staining for MMP-2 and MMP-9 were correlated with activity index of lupus nephritis and proteinuria.Conclusion Up-regulation of gelatinases expression in diffuse proliferate lupus nephritis is correlated to activity index of the disease.
6.Relationship between job burnout and quality of life of factory staff.

OBJECTIVETo investigate the status of job burnout and the quality of life of factory staff and analyze the their relationship.
METHODSQuestionnaires were administered to investigate the status of job burnout and quality of life of all the staff in three randomly selected state factories, and then the methods of canonical correlation and hierarchical linear regression analysis were used to analyze the relationship between job burnout and quality of life.
RESULTSJob burnout and quality of life were somewhat different among staff in factories of different demography character. Emotional exhaustion was negatively associated with mental component summary (MCS). The predictive variables of physical component summary (PCS) were age, educational attainment, emotional exhaustion and professional efficacy and the standard partial regression coefficients of the four variables were -0.101, 0.055, -0.294, -0.058 respectively (P < 0.05). Educational level positively predicted the physical component summary, whereas age, emotional exhaustion and professional efficacy predicted physical component summary negatively. The predictive variables of mental component summary were emotional exhaustion, cynicism and professional efficacy and the standard partial regression coefficients of the three scale of job burnout were -0.546, -0.075, -0.184 (P < 0.05). All the three scales of job burnout predicted the mental component summary negatively. The emotional exhaustion was the most predictive variable for both the physical component summary and the mental component summary.
CONCLUSIONJob burnout is the important risk factor of quality of life of factory staff, especially influencing the staff's metal health significantly.

8.Inflammatory reaction changes with aging in kidneys of human TIMP-1 transgenic mice

Objective To explore the role of tissue inhibitor of metalloproteinase-1(TIMP-1) during renal senescence by using human TIMP-1 transgenic mice.Methods Renal histological changes of wild type mice and transgenic mice at the age of 3,12,24 months were observed by periodic acid-schiff(PAS)staining of paraffin sections.The numbers of F4/80 positive cells were detected by immunofluoreseence.The protein expressions of TIMP-1,TIMP-2,matrix metalloproteinase(MMP)-9,MMP-2,intercellular adhesion molecule-1(ICAM-1),transforming growth factor?1(TGF-?1),collagenⅢand collagenⅣwere detected by Western blot.The activities of gelatinases and TIMP-1 were examined by gelatin zymography and reverse zymography respectively.Results Focal renal fibrosis was found in two genotypes with aging.At the age of 24 months,compared with wild type,in kidneys of transgenic type,the expressions and activities of gelatinases were dowregulated (MMP-2:2.08?0.20 vs.3.39?0.43;MMP-9:4.02?0.82 vs.6.72?1.40,all P<0.05);the expressions of collagenⅢ,collagenⅣ,ICAM-1,and TGF-?1 were upragulated(0.72+0.11 vs.0.57?0.09;0.84?0.13 vs.0.6?0.11,0.72?0.12 vs.0.53?0.07; 0.69?0.12 vs.0.45?0.09,all P<0.05),and the numbers of F4/80 positive cells were increased (18.8?4.4 vs.12.7?3.6,P<0.05)with the upregulated expression and activity of TIMP-1(1.10?0.18 vs.0.62?0.09;50.75?7.25 vs.20.64?3.50,P<0.05).Conclusions TIMP-1 could promote age-related renal fibrosis through enhancing inflammation reaction by ICAM-1 upregulation.
9.Application of microimmunofluorescence test and PCR in the diagnosis of acute Chlamydia pneumoniae infections

Objective To explore the role of acute infection of Chlamydia pneumoniae (Cpn) in respiratory diseases. Methods Microimmunofluorescence test was used to detect IgG antibodies for Cpn in serum obtained from 93 inpatients and PCR was used to test Cpn in detection of Cpn DNA in throat specimens from 55 of the 99 patients. Results Acute Cpn infection was diagnosed in 35.5% of the respiratory diseases. Antibodies for Cpn (titer of ≥512) were present in 47.6% of the pneumonia group, which may suggest that during 1998 to 1999, Cpn caused an epidemic in Beijing. They were also present in 50% of asthma group, 50.0% of pulmonary heart disease group and 26.3% of lung cancer group. Only five patients (9.1%) were positive by PCR. There exists discrepancy between serological and PCR results. Conclusion Detection of IgG antibodies for Cpn conduces to diagnosis of acute Cpn infection and give advice for appropriate therapy.
