Objective To investigate the relationship between STAT3 signal transduction pathway and proliferation as well as apotosis of vascular smooth muscle cells in rat.Methods VSMCs of rats were treated with AG490(a selective JAK2 inhibitor).MTT assay and flow cytometry were used to measure the proliferation and apoptosis.(RT-PCR) was performed to detect the levels of JAK2 mRNA expression from VSMCs.The expression of STAT3,(P-STAT3),JAK2,P-JAK2,cyclin D1 and BCL-2 proteins was measured by Western blot.Results High expression of JAK2 mRNA was detected in VSMCs and there was obvious decline of JAK2 mRNA after AG490 treating((P
3.Study on the Quality Standard of Anti-virus Chewable Tablet

OBJECTIVE:To establish the quality standard for Anti-virus chewable tablet. METHODS:TLC was used for the qualitative identification of Forsythia suspensa,Anemarrhena asphodeloides and Pogostemon cablin in Anti-virus chewable tablet, and HPLC was conducted to determine the content of forsythin in F. suspensa. The column was Diamonsil C18 with mobile phase of acetonitrile- water (20∶80,V/V) at the flow rate of 1.0 ml/min,the detection wavelength was 230 nm,column temperature was 25℃,and injection volume was 10μl. RESULTS:The test sample and the reference sample displayed same color spots on the cor-responding position in TLC diagram. The linear range of forsythin was 0.053 5-2.675 μg(r=0.999 9);RSDs of precision,stabili-ty and reproducibility tests were no more than 1.74%;average recovery was 98.94%(RSD=1.84%,n=6). CONCLUSIONS:The method is simple,rapid,accurate,reliable and reproducible,and can be used for quality control of Anti-virus chewable tablet.

OBJECTIVE: To evaluate the efficacy of reduced glutathione in the treatment of drug-induced liver disease. METHODS: Literatures were retrieved from PubMed, Medline, EMBASE, Cochrane Library, CBM, VIP, CNKI databases by computer and from relevant Chinese medical journals by hand. 10.Blocking PI3K/Akt signaling pathway strengthened the suppression of invasion and migration by CD40 in gastric cancer cells

Objective To investigate the effects of soluble CD40 ligand combined with LY294002,a specific inhibitor of phosphatidylinositol 3-kinase,on proliferation and invasion of gastric cancer cells (AGS ) and its potential mechanisms. Methods The effect of gradient concentration of sCD40L and its combination with LY294002 on cellular proliferation was determined using MTT assay. The ability of cellular invasion and migration was evaluated by wound healing and Transwell invasion assay. The protein expression of PI3K, p-Akt and vascular endothelia growth factor (VEGF) was tested by Western blotting. Results The inhibitory effect of cellular proliferation in sCD40L, LY294002 and sCD40L + LY294002 groups was 70. 1%, 65. 7% and 41. 3%, respectively, with significant difference (P<0. 05). And wound healing was also delayed in cells treated with sCD40L plus LY294002 when compared with those treated with sCD40L or LY294002. These results indicated that the inhibitory effect of cellular proliferation, migration and invasion was enhanced when cells were treated with sCD40L plus LY294002. The protein expression of PI3K,p-Akt and VEGF increased in cells treated with sCD40L, but reduced drastically in cells treated with sCD40L plus LY294002. Conclusion Inhibiting PI3K/Akt signaling pathway may enhance the inhibitory effect of proliferation, migration and invasion by sCD40L on AGS cells, which provide a new way for biochemical therapy of gastric cancer.
