1.The antibacterial activity of imipenem in combination with cefoperazone-sulbactam against carbapenem-resistant Acinetobacter baumannii

Objective To investigate the interaction of in vitro antibacterial activity between cefoperazone-sulbactam and imipenem against carbapenem-resistant Acinetobacterbaumannii.Methods The minimum inhibitory concentrations (MIC) of imipenem and cefoperazone-sulbactam against Acinetobacter baumannii were detected and the fractional inhibitory concentration (FIC) index of imipenem/cefoperazone-sulbactam combination was calculated by broth micro-dilution method.The interactions of imipenem and cefoperazone-sulbactam were assayed by K-B agar plate method.Results The MIC50 of cefoperazone-sulbactam and imipenem were both 16 mg/L,and MIC90 of the two drugs were both 64 mg/L.Among the 26 strains of Acinetobacter baumannii,16 strains had FIC indexes ≤0.5,and 10 had FIC indexes≥2.0.K-B agar plate assay showed that the cefoperazone-sulbactam/imipenem combination was synergistic for strains with FIC indexes≤0.5,and antagonistic for strains with FIC indexes ≥ 2.0.Conclusions Imipenem in combination with cefoperazone-sulbactam can be either antagonistic or synergistic against carbapenem-resistant Acinetobacter baumannii.The clinical use of cefoperazone-sulbactam/imipenem combination for the treatment of carbapenem resistance Acinetobacter baumannii infections should be cautious in case of antagonism.
3. Detection of in vitro anti-fungal activity of 20 tetralin compounds

Objective: To investigate the anti-fungal activities of 20 tetralin compounds in vitro. Methods: We adopted the M27-A project recommended by the National Committee for Clinical Laboratory Standard (NCCLS). Tetralin compounds were tested and selected with several candidal strains and non-candidal strains with different susceptibilities to fluconazole. After obtaining the susceptibility results, we plotted the time-growth curves of several typical tetralin compounds, including compound 22-1, 31-1 and their muriate 22, 31, as well as compound 34-1, 26-1 and their bromate 34, 26. The combination effects of compound 22, 26, 34, 31-1 with other anti-fungal agents (with different structures) were also determined. Results: The 20 tetralin compounds were proven to have different degrees of anti-fungal activities. Compound 31-1 had a stronger anti-fungal activity to FLC-susceptible strains than fluconazole did, and its effect lasted for over 54 h at the concentration of 6 μg/ml. Compounds 22-1 and 22 markedly suppressed the growth of Candida albicans, Candida parapsilosis, and Trichophyton rubrum, with all the MIC50 values less than 0.125 μg/ml. Time-growth curves indicated that the anti-fungal activity of 22-1 on fluconazole-resistant strain was more powerful than that of ketoconazole. Furthermore, tetralin compounds had a synergistic effect with terbinafine and berberine on fluconazole-resistant strains. Conclusion: Tetralin compounds have powerful anti-fungal activities and their structures are different from those of other anti-fungal agents currently used, which provide a basis for developing new anti-fungal agents.
4. Target sites of new anti-Candida albicands agents

With the wide use of azoles, drug tolerance and cross tolerance of Candida albicands has seriously hampered the clinical treatments of Candida albicands. New antifungal agents (potent and effective) have been developed over the past years based on the discovery of many new target sites of Candida albicands. For example, Echinocandins (caspofungin and micafungin), aiming at the cell wall of Candida albicands, showed strong antifungal activities to fluconazole resistant candidas and fungal biofilm. Moreover, because β-glucan synthase does not exist on the cell membrane of mammalian cells, Echinocandins has a low toxicity and a promising clinical future. Though the mechanism of Histatin (targeting fungal membrane) is not clear, it has strong activity not only on candida resistant of polyene and azole, but also to Candida parapsilosis, Candida krusei and Cryptococcus neoformans. Berberine and Ocimum gratissimum L. were also found to have prominent anti-fungi activities. Berberine extract can intensively inhibit 24-SMT in a dose-depended manner; besides, it has more potent inhibitory activity against the growth of mycelia than against that of yeast. Various new methods can be used to increase the susceptibility of Candida albicands to antifungal agents, such as altering fungi membrane composition, changing some genes, adopting the photodynamic inactivation method, employing hypersensitization agents and combining antifungal agents. This article reviews the newly-developed antifungal agents and newly-proposed target sites of Candida albicands.
5.Treatment of experimental femoral artery obliterans in rat by transplantation with endothelial progenitor cells from bone marrow

ObjectiveTo observe the treatment possibility and effects on rats models with experimental femoral artery obliterans(EFAO) by transplantation of endothelial progenitor cells(EPC) from bone marrow. MethodsEFAO rat models were successfully made. Bone marrow mesenchymal stem cells(MSCs) were isolated from allogeneic male Wistar rat,cultured in DMEM in vitro. EGF,bFGF and IGF-1 were added into culture medium on the 10th day and the active EPC were labled with BrdU. 5×106 of the above cells were transplanted into the right hindlimbs i.m. as Group A in the 16 rats,and the same volume of normal saline into the opposite as Group B as control. Laser Doppler perfusion imaging was taken on the 0 day、the 14th day and the 28th day after transplantation. On the 14 day and the 28 day 8 rats were sacrificed respectively and muscles of all hindlimbs were extracted for immunohistochemical examinations (by FⅧ and BrdU).Results1. In group A the skin blood perfusion in hindlimb were significantly increased as compared with group B(P<0.05). 2. Some positive stained EPC by BrdU were found in Group A but not in Group B. 3. The numbers of blood vessels with positive staining of FⅧ in hindlimb on the 28 day in Group A were more obviously than that in Group B (P<0.01). ConclusionTransplantation of EPC from bone marrow can significantly increase the skin blood perfusion and capillary density in ischemic hindlimbs in EFAOrats.
7.Effects of different titers of bacteriophage D29 on growth and function of airway epithelia cell 9HTE

Objective To research the effects of different titers of bacteriophage D29 on growth and function of airway epithelial cell 9HTE.Methods Cell viability rates was analyzed after applying high(109 PFU/mL)and low(107 PFU/mL)titers of bacteriophage D29 and phage buffer respectively by MTT colorimetry.Additionally,the secretion levels of IL-6,IL-8 in cell culture supernatant were detected by ELISA.RT-PCR was performed to detect the expression of ICAM-1 mRNA.Cell apoptosis rate was analyzed by flow cytometry.Results There was no difference in cell growth,secretion levels of IL-6,IL-8,ICAM-1 mRNA and cell apoptosis rate between cells treated with high and low titers of D29 and phage buffer(P>0.05).Conclusion Neither high nor low titer of bacteriophage D29 exerts effect on growth and function of airway epithelial cell 9HTE in vitro.
8.The Application of Satir Family Therapy in Medical Students with Learning Difficulties

9.An investigation on the levels of the specific antibody in the serum of convalescent SARS patients

Objective:To investigate the levels of the specific antibody in the serum of convalescent SARS patients.Methods:The specific antibody in the serum of convalescent SARS patients were detected by indirect ELISA and double antigen sandwich method.Results:The levels of the specific IgG antibody in the serum of convalescent SARS patients is up to the maximum in the fifth week, the levels of the specific IgM is up to the maximum in the third week. The levels of the specific IgG in the serum of convalescent SARS patients is 1-7 times than that of the specific IgM. The positive detection rate of the specific antibody during 5-7 weeks in the serum of convalescent SARS patients is up to 100%.Conclusion:The levels of the specific antibody in the serum of convalescent SARS patients is up to the maximum in the fifth week, and the positive detection rate of the specific antibody is 100%.
