1.Preoperative neoadjuvant therapy for locally advanced rectal cancer

Preoperative radiotherapy combined with chemotherapy and (or) targeted therapy not only reduces the local recurrence of locally advanced rectal cancer and increases the rate of anal preservation and pathologic complete response,but also improves patient compliance.Preoperative neoadjuvant therapy will gradually become the standard treatment for locally advanced rectal cancer.

3.Study on preventive and therapeutic effects of astragali radix on denervated tibial muscle atrophy in rats.

OBJECTIVETo study the effect of Astragali Radix on the denervated tibial muscle atrophy in rats, and discuss its mechanism.
METHODTotally 60 SPF-grade Sprague-Dawley rats were selected in the common peroneal nerve crush model, and then randomly divided into 6 groups: Astragali Radix high-dose, medium-dose, low-dose groups, the Mecobalamin group, the model group, and the sham operation group. They were administered with drugs after the operation. At 18 d, the pathological section staining and morphological analysis were performed. The wet-weight ratio and section area of tibial muscles were also measured. The real-time fluorescence quantification was adopted to detect the differential expression between Angptl4 and PI3K genes.
RESULT(1) Wet-weight ratio: The wet-weight ratio in Astragali Radix high-dose, medium-dose groups was much higher than that in the model group (P < 0.05 or P < 0.01). (2) Section area: The sham operation group was higher, with regular morphology; Whereas the model group showed significant decrease, with chaotic structure and obvious connective tissue proliferation; Astragali Radix groups and the mecobalamin group showed relatively small section areas, with chaotic structure and unobvious connective tissue proliferation. Compared with the model group, Astragali Radix groups showed significant increase (P < 0.01). (3) Motor end plate: The sham operation group was in uniform brownish black color and oval or round shape; Astragali Radix medium-dose and high-dose group and the mecobalamin group showed rough line edges; Astragali Radix medium-dose and low-dose groups and the model group showed decline in the number, with irregular morphology, rough line edges and a light color. (4) Angptl4 and PI3K: Compared with the model group, the Astragali Radix high-dose group showed significant increase (P < 0.05).
CONCLUSIONAstragali Radix has a significant effect in preventing and treating denervated tibial muscle atrophy. It may delay the muscle atrophy by increasing Angptl4 and PI3K gene expressions.

6.Expression of delayed rectifier potassium channels in AGS cells and the effects of down-regulation of Kv1.5 on AGS cell proliferation

Objective To study the expression of 9 delayed rectifier potassium channel subunits in human gastric cancer cell line AGS, as well as the effect of down-regulation of the expression of Kv1.5 on AGS cells proliferation. Methods RT-PCR technique was employed to detect the mRNA expression of Kv1.1, Kv1.2, Kv1.3, Kv1.5, Kv1.6, Kv2.1, Kv2.2, Kv3.1 and Kv3.2 in AGS cells. Once the expression of Kv1.5 protein was confirmed by immunofluorescence, small interference RNA (siRNA) was applied to down-regulate the Kv1.5 protein expression, then MTT assay and flow cytometry were used to observe the cell proliferation and the cell cycle distribution. Results Totally 7 delayed rectifier potassium channel subunits were detected in AGS cells, among them the mRNA levels of Kv1.3, Kv1.5 and Kv2.1 were much higher than that of the others. It was confirmed by immunofluorescence that the Kv1.5 protein was expressed mainly in AGS cytomembrane. After the endogenous Kv1.5 expression in AGS cells was down regulated by siRNA, the cell proliferation obviously slowed down and the cell growth stopped in G1 period. Conclusion Multiform expression of delayed rectifier potassium channel subunits existed in AGS cells, and Kv1.5 may be involved in the regulation of gastric carcinoma cell proliferation by modulating the cell cycle.

8.Effect of millimeter wave radiation on apoptosis of human hepatoma cell

Objective To study the effect of millimeter wave radiation on human hepatoma cell. Methods BEL7404 hepatoma cells were cultured in vitro and randomly divided into 4 groups: the control group, the group radiated by millimeter wave for 30 min, the group treated with Fluorouracil(5 FU), and the group radiated by millimeter wave and treated with 5 FU simultaneously at same time. The ability of 35.8 GHz millimeter wave to induce the apoptosis of hepatoma cell was evaluated by analyses of fluorescence microscopy, DNA fragmentation assay and flow cytometry assay. Results BEL7404 cells radiated by the millimeter wave had the typical characteristics of apoptosis. Comparaed with the control group [(3.21? 1.06)%], the apoptosis rates were higher in 30 min radiating or/and 5 FU groups[ (14.33? 2.66)%, (18.58? 2.57)%, (27.91? 3.66)%]. Poly adp ribose polymerase(PARP) was found to be cleavaged in all the cells in millmeter wave radiation or/and 5 FU groups. Conclusion Radiation of 35.8 GHz could induce apoptosis of BEL7404 cell in vitro, and could act synergistcally with 5 FU treatment.
9.Expression of proliferating cell nuclear antigen, CDK4 and P16 in rat hepatocellular carcinoma by milli-meter wave radiation

Objective To study the expression of proliferating cell nuclear antigen (PCNA), CDK4 and P16 on rat hepatocellular carcinoma by millimeter wave radiation. Methods Fourty male Wistar rats were randomly divided into four groups. Group one to three were feeded by diethylnitrosamine (DEN). Group one was a tumor control group. In group two and three the liver was directly radiated by 35.8 GHz, 100 mW/cm 2 millimeter wave for 20 min, twice a week for 10 or 5 weeks. Group four was a normal control radiated group. Fourteen weeks later all rats were sacrificed to undergo serological test and immunohistochemical stain of liver. Results The serum levels of ? glutamyltransferase in group two and three were lower than that in group one. Adenocarcinoma was only existed in group one by histological examination of liver tissue. Other groups of DEN exposure only had basophilic and eosinophilic nodules. Liver tissue expression of PCNA and CDK4 in group two and three were significantly lower than in group one, but the expression of P16 in group two and three was higher than that in group one. Conclusions Radiation with millimeter wave can partially inhibit cell proliferation and suppress the DEN induced hepatocellular carcinoma in rats.
10. Inhibitory effects of L-type Ca2+-channel blocker nifedipine and magnesium sulfate on uterine contractility of full term pregnant women

Objective: To investigate the inhibitory effects of L-type Ca2+-channel blocker nifedipine and magnesium sulfate on uterine contractility of full term pregnant women and its relation with labour. Methods: Myometrial strips, prepared from biopsies taken at caesarean section from labouring and non-labouring women at full term pregnancy,were mounted in organ chambers for isometric tension recording. Oxytocin(10-9mol/L) was employed to induce stable contraction. The cumulative effects of nifedipine(10-8, 10-7, 10-6 and 10-5mol/L) and magnesium sulfate (1, 2, 4, 8 and 16 mmol/L) were studied by calculating the frequency, amplitude and AUC of contractions before and after drug applications, and the inhibitory rates were also calculated. Results: Nifedipine exerted a concentration-dependent relaxant effect on human term pregnant myometrial contractions induced by oxytocin. The relaxant effect was greater in the myometrium of non-labouring than that in the labouring group(P<0.05). Magnesium sulfate also inhibited the contractility in both groups, with no significant difference between the 2 groups(P>0.05). Conclusion: Decreased inhibition of human uterine contractility by L-type Ca2+-channel blocker nifedipine in term labouring myometrium may reflect sensitivity changes of Ca2+-channel to nifedipine after the onset of labour. The inhibitory effects of Mg2+ are not affected by labour, suggesting that Mg2+ may also exert its effect on contractility through other pathways besides Ca2+-channel.
