1.Experimental Study of Vascular Allograft of Autologous Venous Endothelial Cells in Seeded Rabbits

Objective To study the feasibility of transplanting autologous venous endothelial cells, as the liner, to the allogenic vein and to investigate the patency rate after such transplantation. Methods Autologous endothelial cells were gained after the administration of 0.2% collagenase and the centrifugalization of the enzyme liquid. The cells were not cultivated in a 60 ml plastic culture until the presence of the second generation. The cultivated cells were confirmed as endothelial cells by factor Ⅷ related antigen. The multiplied cells were lined in vitro onto the luminal surface of allogenic vein that was disposed by freeze-drying and radiation. The orthotopic transplantation of autologous venous endothelial cells was performed after the 9-day incubation. Results (9.47?0.35)?10~6 endothelial cells were obtained after the cultivation. Three hours after cell seeding, the luminal surface of allogenic vein was covered with vast endothelial cells but still had not formed an intact endomembrane. On day 9, the luminal surface was covered with a continuous endothelial monolayer and the arrangement and the shape of the cells all showed the perfect condition of endothelial cells. Eight weeks later, all the transplanted veins kept unobstructed. Conclusion The approach of lining allogenic vein with autologous endothelial cells in vitro may keep the vein unobstructed in the long term.
2.Titanium alloy steel artificial rib reconstruction of chest wall after tumor resection

Objective To evaluate the effects of chest wall reconstruction after tumor resection patients with chest wall defect. Methods From March 1999 to August 2008,28 patients with chest wall tumor underwent chest wall reconstruction.The number of rib resected was 3-6. Twenty-one huge chest wall defects underwent chest wall reconstruction using titanium alloy steel artificial rib. The materials for the soft tissue reconstruction include latissimus dorsi myocutaneous flap. Results No died of operation, postoperative respiratory function was good, no abnormal breathing exercises. Concurrent osseous tissue repair and rebuilt in 3 cases, the fluid recovered after conservative therapy. Followed-up 8-76 months,2 cases were lost, follow-up rate was 92.9% ,4 cases with benign tumor existed 7.8,7.2,5.4 and 3.6 years, 22 cases with malignant tumor after 1, 3, 5 years survival rate were 77.3% (17/22) ,63.2% (12/19),41.2% (7/17) respectively. Conclusion A favorable clinical outcome can be achieved by titanium alloy steel artificial rib reconstruction of chest wall.
3.Influence of Jiyin Capsule on apoptosis pathway of ovarian caspase de-pendent cells in rats with peri-menopausal syndrome

Objective To discuss the effective mechanism of Jiyin(Nourishing Yin)Capsule in interve-ning peri-menopausal syndrome.Methods Normal rats(n=10)were randomly chosen into blank group. The model of peri-menopausal syndrome was established with intraperitoneal injection of VCD once a day for 60 d.When estrous cycle was disorder,level of serum estradiol(E2)was lower than the lowest level of that in blank group and level of follicle-stimulating hormone(FSH)was higher than the highest level of that in blank group, the modeling was success.The modeled rats were randomly divided into 6 groups:model group,progynova group(PGV group),Kunbao(Female Treasure)Pill group(KBP group), and low-dose,mid-dose and high-dose Jiyin Capsule groups(each n=10).After grouping,blank group and model group were given distilled water,and other groups were given corresponding medicinals once a day for 30 d.The mRNA expressions of caspase-3,caspase-8,caspase-9 and Apaf-1 were detected by using re-al-time fluorescence quantitative polymerase chain reaction(RT-PCR).Results Compared with blank group,mRNA expressions of caspase-3,caspase-8,caspase-9 and Apaf-1 increased significantly in model group(P<0.01).Compared with model group, mRNA expressions of caspase-3, caspase-8, caspase-9 and Apaf-1 decreased significantly in Jiyin Capsule groups.Conclusion Jiyin Capsule can inhibit cell ap-optosis of ovarian through down-regulating conduction of apoptosis pathway of caspase dependent cells, which maybe one of its effective mechanisms of intervening peri-menopausal syndrome.
