1.The variation of IGF-1,IGFBP-3 in the development of rats’ non-alcoholic fatty liver disease

Objective To observe the expression of insulin-like growth factor 1 (IGF-1),insulin-like growth factor binding protein 3 ( IGFBP-3) in rats’ serum with non-alcoholic fatty liver disease ( NAFLD) and the impact of Polyene phosphatidyl choline .Methods NAFLD model was induced by feeding the SD rats with a high-fat diet, with Polyene phosphatidyl choline to intervene .Observe the pathological changes of the rats ’ liver tissue dynamically after HE staining . Detect the contents of IGF-1,IGFBP-3 in serum dynamically through the immuno radio metric assay ( IRMA).Results There were no obvious exception of the liver tissue pathology in the normal group at each time .With the extension of a high-fat diet,the liver tissue of the model group increased on fatty change ,the degree of inflammation ,the balloon sample change , the NAFLD activity score at 4,8,12weeks.IGF-1,IGFBP-3 in serum were decreased significantly ,and the model set was significantly lower than the normal group at the same phase .After the application of polyene phosphatidyl choline ,the degree of rat liver tissue inflammation and the NAFLD activity score were reduced significantly when compared with model group , while the level of IGF-1,IGFBP-3were significantly higher .Conclusion The levels of IGF-1,IGFBP-3 in the rats’ serum reduce in the development of non-alcoholic fatty liver disease .
2.Effect of high fat diet on the serum biochemical parameters and histopathology of main organs in Mongolian gerbils

Objective To explore the effect of high fat diet on serum biochemical parameters and histopathology of main organs in Mongolian gerbils.Methods Forty-eight healthy adult male Mongolian gerbils were randomly and equally divided into model and normal groups.The gerbils in the model group were fed with high fat diet while the normal group with standard diet.Eight gerbils in each group were killed at the end of 4th,8th and 16th week,respectively,and the body weight, serum levels of Glu, TG, CHOL, HDL-C, LDL-C, UA, CREA, BUN, TBil, TP, ALB, ALT, AST and AMS were determined.The histopathological changes of main organs were observed.Results Compared with the normal group,the blood lipid of the model gerbils was significantly increased, the liver function was impaired, the blood uric acid level was higher, and the blood glucose was decreased at the end of 16th week.The AMS was increased at the end of 16th week,but the renal function showed no significant changes.The liver tissue of the model group gradually showed steatosis, inflammation, fibrosis and cirrhosis, accompanied by splenomegalia. The lung tissue and myocardium showed fatty degeneration and obvious damages in the later period,the pancreatic islets were enlarged and the amount of endocrine cells was increased,and the small intestine and kidney didn’ t show any distinct changes.Conclusions A gerbil models of hyperlipidemia and nonalcoholic steatohepatitis and cirrhosis can be well established by high fat diet feeding,and may serve as good models for research of hyperlipidemia-related hyperuricemia, and lung and myocardial damages.
3.Analysis of hemorrhagic transformation following thrombolytic therapy in acute cerebral infarct patients

Objective To investigate the risk factors of hemorrhagic transformation (HT) following thrombolytic therapy in acute cerebral infarct patients. Methods The clinical data of 246 cases with acute cerebral infarct treated with rt-PA within 4.5 h from the onset were reviewed. According to the results of brain CT imaging after intravenous thrombolysis for 24-36 h, the patients were divided into HT group and non HT group. The factors including age, gender, NIHSS scores, drinking, smoking, hypertension, diabetes, atrial fibrillation. The univariate analysis and Losgistic regression analysis were further assessed. Results The results of univariate analysis indicated that there were no significant differences between the two groups in age, gender, smoking , drinking, time from onset to thrombolysis, diabetes (P > 0.05), but there were significant differences between two groups in NIHSS scores [(14.53 ± 6.06) scores vs.(9.98 ± 6.26) scores, P = 0.000], hypertension [86.1%(31/36) vs.70.0%(147/210), P = 0.046] and atrial fibrillation [63.9%(23/36) vs. 24.3%(51/210), P = 0.000]. Logistic regression analysis showed that NIHSS scores (OR = 1.079, 95% CI = 1.014- 1.147, P = 0.016) and atrial fibrillation (OR=3.298, 95%CI=1.481-7.345, P=0.003) were the risk factors associated with hemorrhagic transformation after intravenous thrombolysis for acute cerebral infarction. Conclusions NIHSS scores and atrial fibrillation are the risk factors associated with HT after thrombolytic therapy in acute cerebral infarct patients.
4.Effect of rt-PA intravenous thrombolytic therapy on serum levels of neuro-specific endolase, C-reactive protein and fatty acid binding protein in patients with early cerebral infarction

Objective To analyse effect of rt-PA intravenous thrombolytic therapy on serum levels of neuro-specific endolase, C-reactive protein and fatty acid binding protein in patients with early cerebral infarction.Methods 54 patients who were diagnosed with early acute cerebral infarction in Dalian Municipal Central Hospital were collected.All patients were randomly divided into experimental group and control group,27 cases in each group.Two groups were given conventional treatment,such as reduction of intracranial pressure,improve circulation and nutrition brain cells.Control group was given low molecular weight heparin calcium injection 5000U, one times per 12 h for 7 consecutive days, and experimental group was given rt-PA intravenous thrombolytic therapy on the basis of the control group for 7 consecutive days, post-treatment,the serum levels of NSE, CRP and FABP were detected in all patients.Results Compared with control group post-treatment, the serum CRP,NSE and FABP level was lower in experimental group ( P<0.05).Conclusions The rt-PA intravenous thrombolytic therapy can significantly reduce the serum CRP, NSE and FABP levels in the patients with early cerebral infarction, improve the prognosis of patients.
5.Present Situation and Development and Utilization Suggestions for TCM Resources in Changshou District of Chongqing

Objective To investigate TCM resources in Changshou District of Chongqing based on the fourth national census of TCM resources; To provide theoretical references for the protection and exploitation of TCM resources. Methods Sample-plot survey, line survey and interview survey were used in this study. Results Totally 142 families, 482 genera and 639 species (including subspecies and varieties) were collected from the general survey of TCM resources in this region. There were a total of 575 species of medicinal plants, including 456 kinds of ordinary medicinal plants and 119 species of national key medicinal plant which were recorded in the Chinese Pharmacopoeia (2015 Edition). But there was no department leader of the Chinese medicine industry and TCM cultivation was almost blank. Nowadays there was no TCM processing enterprise, and the quality could not be controlled. The continual increase of production cost restricted the development of TCM industry. Conclusion TCM resources in Changshou District of Chongqing are abundant, while the TCM industry is almost blank. The support and guidance from government are needed to develop TCM industry in this area.

Objective To construct the recombinant eukaryotic expression plasmids of human adenylyl cyclase-associated protein 1 (CAP1)and to explore its intracellular location and functions.Methods By using Hela cDNA as the template,the cDNAs encoding CAP1 was amplified by PCR and inserted into pCMV-Myc vector to construct the recombinant plasmid.The recombinant plasmid was transfected into 293 cells using lipofectamine 2000.The protein expression and the intracellular location of the inserted gene were confirmed by Western blotting and immunofluorescence,respectively.Scratch-repair experiment was used to detect the cancer cells’ migration ability.Results The recombinant eukaryotic expression plasmid of human CAP1 was successfully constructed and transfected into eukaryote cells.The recombinant plasmid was successfully expressed in eukaryote cells.CAP1 was located in the cytoplasm.The results of scratch-repair experiment showed that the overexpression of CAP1 could significantly inhibit the cells’ migration.Conclusion CAP1 recombinant plasmid was successfully expressed in eukaryotic cells.CAP1 protein was located in the cytoplasm.The overexpression of CAP1 inhibited cell migration. The present study provides important experimental evidence for further study on CAP1.
7.Clearance of insoluble depleted uranium particles in lungs by citric acid and ambroxol

Objective To investigate the effect of citric acid and ambroxol on clearing insoluble particles of depleted uranium in rat lungs by establishing a tracheal perfusion model.Methods One hundred and fifty male Wistar rats were randomly divided into model exposure group, normal control group(NC group), depleted uranium exposure group(DU), citric acid treatment group( CA) , ambroxol treatment group( AM) and citric acid+ambroxol treatment group( CA+AM) . The rats were sacrificed on 7, 15 and 30 days.Uranium content in the lungs was detected by microwave digestion method, pathological changes in the lungs were observed, and inflammatory factors of lung homogenates were detected.Results Compared to DU control group, the intrapulmonary uranium deposit amount in experimental groups was significantly reduced on 7 and 15 days (P<0.05).HE stained lung tissue showed that the pathological changes in treatment groups were less significant than in DU control group.The level of IL-1α,IL-1β,and IL-2 was significantly lower than in DU control, but the level of MCP-1 and MIP-1 was observably higher.Conclusion Citric acid and ambroxol can evidently improve the clear-ance of lung uranium and reduce damnification of lung tissues.Drug treatment can reduce the level of pulmonary inflamma-tory cytokines alleviate the chronic inflammation in the lungs, and enhance the capacity of macrophage to recruitment.
8.Role of Hes1 in malignant transformation of human bronchial epithelial cells induced by tobacco

AIM: To investigate the role of transcription factor hairy and enhancer of split 1 (Hes1) in the malignant transformation of human bronchial epithelial cell line BEP2D induced by tobacco.METHODS: The BEP2D cells were chronically exposed to cigarette smoke condensate (CSC) at 1 cigarette per L until the 70th generation.The phenotype of malignant transformation of the cells induced by CSC was detected by soft agar clony formation assay.RT-PCR and Western blot were used to determined the expression of Hes1 at mRNA and protein levels in each generation of the cells.The proliferation and apoptosis of the BEP2D cells exposed to CSC were analyzed with the methods of MTT assay, flow cytometry and cell colony formation assay after treatment with Notch pathway bloker DAPT or liposome transfection with Hes1-siRNA.The expression of Hes1 in the peripheral small airway tissues of the smoking rats was evaluated by immunohistochemical staining.The expression of Hes1 in non-small-cell lung cancer and normal airway tissues was also detected by the methods of immunohistochemistry and RT-PCR.RESULTS: The BEP2D cells in the 70th generation had a malignant transformation phenotype.The expression of Hes1 in the BEP2D cells exposed to CSC for different time showed an increa-sing trend.DAPT and liposome transfection with Hes1-siRNA down-regulated the expression of Hes1, inhibited the cell proliferation and induced cell apoptosis.The expression of Hes1 in the airway mucosa of the rats exposed to cigarette smoke for 1 month and 6 months was significantly higher than that in control group.Cigarette smoking induced the expression of Hes1 in lung cancer and normal airway tissues.CONCLUSION: Hes1 may be involved in smoking-induced lung cancer by promoting the imbalance between apoptosis and proliferation.
9.Trend on Mortality of Gastric Cancer and Cause-eliminated Life Expectancy from 1987 to 2019 in Harbin

Objective To analyze the mortality characteristics and trends and the cause-eliminated life expectancy of gastric cancer in Harbin City from 1987 to 2019. Methods Mortality data of residents with gastric cancer from 1987 to 2019 in Harbin was analyzed to describe the mortality characteristics and trends of gastric cancer. Abridged life table and cause-eliminated life table were applied to calculate life expectancy and cause-eliminated life expectancy. Average annual percentage change (AAPC) was calculated with Joinpoint 4.2 software to evaluate the trends of mortality and cause-eliminated life expectancy of gastric cancer. Results From 1987 to 2019, the crude mortality, ASMRC and ASMRW and the truncated rate (35-64) were 14.3/105, 10.9/105, 10.9/105, and 13.5/105, respectively. 10.Curative effect analysis of thrombolytic therapy in acute cerebral infarct patients with atrial fibrillation

Objective To investigate the risk of hemorrhagic transformation and the curative effect in patients with atrial fibrillation(AF)treated by intravenous thrombolysis with alteplase after acute cerebral infarct. Methods The clinical data of 246 patients with acute cerebral infarct treated with intravenous alteplase within 4.5 h from the onset were analyzed.According to the presence or absence of AF, the patients were divided into AF group (74 cases) and non AF group (172 cases). The outcomes were the incidence of hemorrhagic transformation within 36 h and the curative effect after 2 weeks. Results The incidence of hemorrhagic transformation in AF group was 31.1%(23/74), in non AF group was 7.6%(13/172),and there was significant difference(χ2=22.917,P=0.000).The effective rate in AF group was 54.1%(40/74), in non AF group was 76.7%(132/172), and there was significant difference (χ2=12.665,P=0.000).Conclusions Patients with acute cerebral infarction combined with AF have a high risk of hemorrhagic transformation and poor prognosis after intravenous thrombolysis.
