1.Effects of naoyikang on capability of learning and memory in Alzheimer’s disease model mice

AIM : To observe the effects of naoyikang on the capability of learning and memory in Alzheimer’s disease model mice. METHODS : The mice model was established by intraperitoneal injection of D BZ Gal and NaNO 2, the capability of learning and memory was tested on mice by electrical maze and water maze, the levels of superoxide dismutase (SOD) and malondialdehyde(MDA) of brain tissues were assayed by biochemical methods,and the changes in ultrastructure were observed by Transmission Electron Microscope. RESULTS : The capability of learning and memory of model mice decreased and the level of SOD of model mice decreased and MDA increased. 2.Total hip arthroplasty with no femoral shortening osteotomy for unilateral Crowe Ⅳ developmental dysplasia of the hip

Objective To evaluate the clinical efficacy of total hip arthroplasty (THA) with no femoral shortening oste?otomy for unilateral CroweⅣ developmental dysplasia of hip. Methods From October 2007 to January 2010, 32 patients with CroweⅣdevelopmental dysplasia of hip in one side underwent THA, including 20 females and 12 males, with an average age of 49.4 ± 9.7 years (range, 23-60 years). There were 15 cases as normal and 17 as mild developmental dysplasia of hip in the other side. The THA were performed with requisite soft tissue release and direct leverage using an elevator but with no femoral shorten?ing osteotomy. The patients' satisfaction, Harris hip score, bilateral leg?length discrepancy and pelvic obliquity was used to assess the clinical results. Results All of patient were followed up for 1-6 years, average 4.0 ± 1.5 years. No loosening or failure of component occurred by the end of follow?up. The Harris hip score was improved from preoperative 36.5±10.3 (20-63) to 89.8± 4.9 (80-97), and the excellent and good rate was 100% (excellent 16 cases, good 16 cases). The satisfactory rate was 93.8%(30/32). The leg?length discrepancy of the bilateral sides and the pelvic obliquity was corrected gradually and the gait returns to normal. Nine cases have valgus knee after THA and 4 cases of them felt uncomfortable after long?distance walk. Femoral nerve injury occurred in 2 cases. All of cases recovered after 1 and 3 months respectively. No infection and dislocation oc?curred. Conclusion THA with no femoral shortening osteotomy can achieve good clinical results in patients with unilateral CroweⅣdevelopmental dysplasia of hip. The discrepancy of leg?length will be diminished with the correction for pelvic obliquity.
3.Optimal dose of Ligustrazine nanoparticles in a rat model of experimental peritoneal adhesion

BACKGROUND:Ligustrazine can effectively prevent and treat peritoneal adhesion and adhesive ileus after surgical treatment. OBJECTIVE:To prepare Ligustrazine nanoparticle spray which is prepared to achieve sustained release and improve clinical efficacy and to explore its optimal dose for preventing experimental peritoneal adhesions in rats. METHODS:Eighty rats were randomly divided into five groups:model group, sodium hyaluronate group, high-, medium-and low-dose Ligustrazine nanoparticle groups, with 16 rats in each group. After peritoneal adhesion model was established with rasp method, model group was immediately subject to abdomen-closing, while sodium hyaluronate group and Ligustrazine nanoparticle groups received sodium hyaluronate smearing and Ligustrazine nanoparticle spray (2.5, 5, 10 mg/kg), respectively. Al rats were kil ed at 1 and 2 weeks after modeling, to observe adhesions. The adhesion score was recorded. The adhesive tissue sections were stained with hematoxylin-eosin for pathological changes. The level of transformation growth factor-β1 in peritoneal fluid of rats was detected with ELISA assay. RESULTS AND CONCLUSION:Ligustrazine Nanoparticle spray had a sustained release effect, and prolonged the duration of action of drugs, thus achieving a better anti-adhesion effect post-surgery. The medium-and high-dose Ligustrazine nanoparticle sprays exhibited better anti-adhesion effects, and improved the rise of transformation growth factor-β1 level in the peritoneal fluid. As the drug concentration is low, the intraperitoneal administration in a spray manner is preferred. Because the total dose is limited, we define the optimal effective dose as 5 mg/kg.
4.Effect Analysis of the Implementation of National Essential Medicine System on the Drug Utilization of Outpa-tients in a Shanghai Community Health Service Center

OBJECTIVE:To provide reference for promoting the further implementation of national essential medicine system and rational use of drugs in community health service center. METHODS:Outpatients’information in Changqiao Street Communi-ty Health Service Center from Sept. 2,2009 to Nov. 25,2014 was collected to statistically analyze the drug variety,the number of prescriptions,the consumption sum and the DDDs before and after the implementation of essential medicine system and after the supplement of non essential medicines. RESULTS:Essential medicines occupied the main position of drug utilization in each stage, with substantial growth in respects of consumption sum and number of prescriptions;the sum of Chinese patent drugs and essential cardiovascular drugs occupied more than 50% of the total consumption sum and number of prescriptions,with higher growth rate;essential medicines occupied the top 10 in respects of consumption sum and DDDs,among which most were the drug varieties in“Shanghai supplement essential drugs in community health service center”(2010 Edition),with some overlap the varieties in“Na-tional Essential Medicine System”(2012 Edition);cardio-cerebrovascular drugs occupied the dominant position;the growth rates of cefotiam and atorvastatin took up the top 2 ranking in the list of consumption sum,while the growth rates of calcium carbonate D3 and irbesartan and hydrochlorothiazide took up the top 2 ranking in the list of DDDs. CONCLUSIONS:Essential medicine cata-log covers the common drugs in the community health service center,in which the rational use of Chinese patent drugs and antibac-terial deserves needs to be noticed. The clinical rational use of the essential medicines should be strengthened,the essential medi-cine catalog should be adjusted and non essential medicines should be supplied when necessary in order to truly meet the needs of patients and optimize the allocation of medical resources.
5.Protective effect of naoyikang-containing serum on cultured hippocampus neuron injury induced by D-galactose

AIM: To observe the protective effect of naoyikang-containing serum on cultured hippocampus neuron injury induced by D-galactose(D-gal).METHODS: Naoyikang-containing serum was prepared in rats administered with aqueous extract of naoyikang(6.84 g?kg-1?d-1).MTT,MAO-B and ATPase assay were used to measure the viability of hippocampus neurons.RESULTS: D-gal at concentration of 100 ?mol/L caused significant decrease in the viability of hippocampus neurons 24 h after the treatment.Naoyikang-containing serum increased the viability,ATPase activity and the expression of bcl-xl mRNA,decreased the MAO-B activity and the expression of bax mRNA in D-gal injured hippocampus neurons.CONCLUSION: Naoyikang-containing serum prevents hippocampus neurons from D-gal induced cytotoxicity.
6.Role of heterogeneity of cancer-associated fibroblasts in targeted therapy of pancreatic cancer

The incidence of pancreatic cancer is increasing year by year, but the clinical diagnosis and treatment progress is limited and the prognosis is poor. Tumor microenvironment (TME) is closely related to the invasion, metastasis and chemotherapy resistance of pancreatic cancer. Cancer-associated fibroblasts (CAFs) are fibroblasts in a state of continuous activation, which are the most prominent components in TME. CAFs can promote the malignant biological behavior of pancreatic cancer through a variety of molecule-mediated mechanisms. Moreover, several attempts targeting CAFs for the treatment of pancreatic cancer have been largely unsuccessful. It may be related to the heterogeneity of CAFs in pancreatic cancer. Therefore, in-depth study of its heterogeneity and accurate targeting of some specific phenotypes and functional CAFs subtypes in the matrix on this basis may be more promising in the clinical treatment of pancreatic cancer.
7.The clinicopathology analysis of three cases of carcinoma with multi-directional differentiation derived from junction of bladder and prostate

To explore the clinical ,pathological and immunohistochemical features of carcinoma with multi-directional differentiation derived from junction of bladder and prostate .We collected clinical data and tissue samples of three typical cases of carcinoma from our hospital .Routine preparation of slides and immunohistochem-ical methods combined with clinical data and pathological changes analysis were adopted .We found that all 3 ca-ses occurred in the elderly lesions involving the bladder and prostate .Dysuria was the main common symptom .All the cases had a history of chronic inflammation in urethral and prostate gland .The following up data showed that survival time in two of them were no more than 15 months.The third patient without chemotherapy who took drugs of bicalutamide,enantone and pamidronate still was alive after 28 months.The pathological changes of these cases had the common features with diversity .They all showed the structure with nests ,papillary,solid,adenoid,single or syncytial cells .The nuclear of cancer cell was enlarged with hyperchromatic feature .The mitotic figures were easily found.The metaplasia and atypical hyperplasia of prostate were all found .The immunohistochemistry results showed positive results for HCK,LCK,p53,p63,PSA,P504S and negative for vimentin and S -100.The average proliferation index of Ki-67 was 77%.

In current study, the expressions of protein kinase C (PKC)-alpha, betaI and betaII as well as their correlation to the expression of transforming growth factor-betaI (TGF-betaI) and vascular endothelial growth factor (VEGF) were investigated in glomeruli of normal renal tissues taken from human kidney tumors and kidney tissues from patients with diabetic nephropathy (DN). The accumulation of glomerular extracelluar matrix (ECM) was determined by PAS staining, the expressions of PKC-a, PKC-betaI, PKC-betaII, TGF-betaI and VEGF were measured by semi-quantitative immunohistochemistry. Our results showed that in glomeruli of normal renal tissues, PKC-alpha and betaII had a strong expression whereas the expression of PKC-betaI was weak; in glomeruli of DN patients, the expressions of PKC-alpha, PKC-betaI, VEGF and TGF-betaI and the accumulation of ECM increased significantly, but the expression of PKC-betaII decreased markedly. Meanwhile, the expressions of PKC-alpha and betaI had a positive correlation to the expressions of VEGF and TGF-betaI respectively, whereas PKC-betaII showed no correlation to VEGF and TGF-betaI. It is concluded that the expressions of PKC-alpha, betaI and betaII in glomeruli of normal subjects and DN patients are different. PKC-alpha seems to play a critical role in human DN by up-regulating VEGF expression, whereas PKC-betaI is relatively important for the up-regulation of TGF-betaI and the accumulation of ECM under diabetic conditions.

The severity of disease can be used to evaluate the current situation of patients as well as to predict the diseases outcome.In the meantime, the severity among different diseases has been more and more applied in the evaluation of the patients at hospital level.This study collected and summarized different types of international disea-ses grouping tools'characteristics and applications based on the severity of disease, and their suitability and practical values in hospital performance evaluation were compared and analyzed.
