Objective To investigate the efficient methods used for tracing endothelial progenitor cells(EPCs)after transplantation in injured lung tissue.Methods Thirty clean grade healthy syngeneic SD female rats were divided into three experimental groups(n =10):(1)sham group,rats treated with intravenous phosphate-buffered saline(PBS)instead of lipopolysaccharide(LPS)followed by EPCs graft;(2)PBS-treated group,rats treated with intravenous PBS after intravenous injection of LPS to produce acute lung injury;(3)EPC-treated group,rats treated with EPCs after intravenous injection of LPS to produce acute lung injury.The transplanted EPCs were got from the same genetic species of the SD male rats.Rats of each group were sacrificed 7 days after EPCs transplantation.Their whole lung tissues were harvested to detect the expression of Y-chromosome by using hybridization in situ and RT-PCR assay.Statistic package of SPSS16.0 was used for the data analysis and significant differences between means were evaluated by ANOVA analysis.Results Compared with the other two groups,positive signals of sex-determining region y were found in lung endothelium from the EPC-treated group.Conclusions Y chromosme specific probe can be one of efficient methods for tracing stem cells after transplantation.

Objective:To study the effects of dexamethasone,cyclophosphamide,cyclosporin A and mycophenolate mofetil on the expression of Dectin-1 and TLR2 receptor in RAW264.7 cell line.Methods:In vitro culturing RAW264.7 macrophages ,the cells were given different doses of dexamethasone and cyclophosphamide ,cyclosporin A and mycophenolate mofetilfor 24 h.The influence of different immunosuppresants on cell proliferation was detected by CCK-8 assay.Dectin-1,TLR2 mRNA and protein levels of change were detected with RT-PCR technique and flow cytometry .Results:CCK-8 cell proliferation toxicity experiment results showed that with the increase of the dose of dexamethasone ,cyclophosphamide ,Cyclosporin A and mycophenolate mofetil ,the different degree of inhibi-tion of RAW264.7 cell proliferation in mice can be saw after the intervention of 24 h (P<0.05).After 24 h of dexamethasone stimula-tion,Dectin-1 mRNA and protein level decreased,TLR2 mRNA and protein levels elevated (P<0.05).However cyclophosphamide had no obvious influence on Dectin-1 and TLR2 ( P>0.05 ) .Mycophenolate mofetil and cyclosporin A could both inhibit the expression of Dectin-1 and TLR2 (P<0.05).Conclusion:There are significant differences among the regulatory functions of various immunosupres -sants on the expression of pattern recognition receptors .Different immunosuppressive agents exert their inhibitory effects on immune rec-ognition process of fungal pathogens not only by selectively regulating their target PRRs expression ,but also by inhibiting the growth and proliferation of macrophages .

Objective To observe the possible relationship of cardiac troponin T (cTnT) with cardiac disease and death events in hemodialysis patients Methods The cTnT was measured with dry chemical method in 57 hemodialysis patients The patients were followed up for one year Results The concentration of cTnT was more than 0 10 ?g/L in 12 of the 57 unselected hemodialysis patients 8 in the 12 patients were diagnosed as diabetes mellitus 7 of 12 patients died and the death reason was acute left ventricular failure(33 3%) in 4 patients In other 45 patients with normal cTnT, 3 died and only 1 died of heart failure(2 22%) The cTnT concentrations of pre-and post-hemodialysis were measured in 24 patients The concentration was more than 0 1 ?g/L in 6 patients before dialysis Post-dialysis concentration was increased in 5 patients', the value was decreased in the other one Conclusions In hemodialysis patients without acute myocardial infarction, mortality and cardiovascular events are closely correlated with the increase of cTnT concentration. Patients with diabetes mellitus have higher incidence in the abnormality of cTnT concentration.The process of hemodialysis does not affect the concentration of cTnT.

Objective:Our aim is to investigate the relationship of VEGF-C,VEGF-D and lymph node metastasis in pancreatic cancer,and to elucidate the role and significance of the cancerous peripheral lymphatic tissue.Methods: The expression of VEGF-C and VEGF-D was assayed by means of immunohistochemistry in 30 pancreatic carcinomas.Results:The positive rates of VEGF-C、VEGF-D were 73% (22/30)、57% (17/30) respectively in pancreatic cancer.The expression of VEGF-C、 VEGF-D in cancerous invasive edge was significantly higher than that in the center of cancerous tissues.There was no correlation between the expression of VEGF-C、VEGF-Dand the site,differentiation,histology types.Ⅲ~Ⅳ stages of pancreatic cancer showed strong expressions of VEGF-C、VEGF-D than that Ⅰ~Ⅱ stages of pancreatic cancer.The positive lymph node in positive VEGF-C and VEGF-D group were higher than that in the negative group.Conclusion:VEGF-C and VEGF-D induced lymphangiogenisis in pancreatic cancer,promoted the tumor cell lymph metastasis.

ObjectiveTo construct recombinant adenovirus vector containing human pancreatic and duodenal homeobox factor 1 (PDX1) and detect its expression in human umblical cord mesenchymal stem cells (HUCMSCs). MethodsPDX1 obtained by BgⅢ/XhoI enzyme digestion from pUC57-PDX1 was ligated into the recombinant shuttle vector pShuttle-GFP-CMV to obtain the recombinant shuttle plasmid pShuttle-GFP-CMVPDX1. pShuttle-GFP-CMV- PDX1 was shifted to pAdxsi vector to obtain pAdxsi-GFP-PDX1 virus plasmid. The recombinant plasmid was packaged and amplified in 293 cells. The expression of PDX1 gene and protein in HUCMSCs was detected by fluorescence microscopy, RT-PCB, immunofluorescence, immunohistochemistry, and Western Blot. ResultsPDX1 gene was inserted correctly into shuttle plasmid and the recombinant adenovirus vector was successfully constructed according to the results of sequence and enzyme digestion identification. The adenovirus was effectively transfected into HUCMSCs. RT-PCR verified that PDX1 mRNA was positively expressed in HUCMSCs. Expression of PDX1 protein in the nuclear of HUCMSCs was found by immunofluorescence assay, immunohistochemistry and Western Blot. ConclusionThe adenovirus vector containing PDX1 gene is successfully constructed and effectively expressed in HUCMSCs.

Objective To observe the change of the serum ABCG2 level of patients with non‐small cell lung cancer(NSCLC)be‐fore and after chemotherapy ,and explore its clinical significance .Methods Venous blood specimens of 15 healthy adults and 50 pa‐tients with NSCLC were collected before and chemotherapy ,the serum ABCG2 level of these specimens were detected by ELISA . the relation between the ABCG2 level and the chemosensitivity was investigated .Results The serum ABCG2 level of patients with NSCLC before chemotherapy was significantly higher than that in healthy adults(P<0 .05);the serum ABCG2 level was not related with TNM stage in NSCLC(P<0 .05);The serum ABCG2 level of patients with NSCLC after chemotherapy was significantly high‐er than that of before(P<0 .05);in chemosensitive patients ,the different of serum ABCG2 level was not significantly before and af‐ter chemotherapy(P>0 .05);among chemoresistant patients ,the serum ABCG2 level of patients with NSCLC after chemotherapy was significantly higher than that of before(P<0 .05);the serum ABCG2 level was higher in patients with pleural effusion than the patients without pleural effusion ,but the different was not significantly(P>0 .05) .Conclusion The serum ABCG2 level of patients with NSCLC is higher than that of healthy adults ;serum ABCG2 level may become a useful indicator in predicting the effect of NSCLC chemotherapy .

Objective To investigate the features and neural mechanisms of sustained attention and executive function in patients with acute mild traumatic brain injury (mTBI) by comparing and analyzing behavioral and event-related potentials of patients and healthy controls.Methods Seventeen patients with acute mTBI and seventeen healthy controls participated in a cued continuous performance test.Behavioral data and event-related potentials were collected and analyzed.Results 1.There were significant differences between the mTBI group and the control group in hitting number ((66.76±3.27), (69.12± 1.41)) ,reaction time((533.66±144.20) ms, (413.03±94.57) ms) and the number of errors of omission ((3.24±3.27), (0.88± 1.41)) (P＜0.05), but no significant differences in the number of false errors ((0.35±1.00), (0.53±0.87)) (P＞0.05).2.The amplitude of Go-N2 and Nogo-N2 were significantly smaller in mTBI group than that in control group (P＜0.05).The main effect of group was significant of N2 amplitude (P＜0.05), but main effect of condition and the interaction effect were not significant(P＞0.05).Group and condition had no significant main effect and interaction effect on the latency of N2 (P＞0.05).The amplitude of Go-P3 was significantly smaller in mTBI group than that in control group (P＜0.05),while not on the amplitude of Nogo-P3(P＞0.05).The main effect of group and condition were significant on P3 amplitude (P＜0.05),but the interaction effect was not significant(P＞0.05).Group and condition had no significant main effect and interaction effect on the amplitude of P3 (P＞0.05).Conclusion Patients with mTBI show impairments in sustained attention and conflict monitoring, but not in response inhibition.

Objective To introduce the technique and experience in reconstruction of posterior crueiate ligament-posterolateral comer(PCL-PLC)injuries with only one allograft of Achilles tendon. Methods The instable knees in 12 cases with PCL injury combined with three degree chronic PLC injury were treated with PCL reconstruction under arthroscope and PLC reconstruction through posterolateral arc incision.Single bundle grafts of PCL reconstructions in tibial and femoral tunnels were fixed by resorption screws.Fibular collateral ligament(FCL)and popliteofibular ligament(PFL)were reconstructed with reforming Larson(?)method.All reconstruction grafts only needed one Achilles tendon as donator.Total op- eration time was 130 minutes including 90 minutes of PCL reconstruction and 40 minutes of PLC recon- struction.Gradual weight loading was allowed after six weeks of bracing.Results Follow-up for mean 12 months(5-24 months)indicated that tibial“step off”reduction was 83%(10/12)and posterior drawer test of 0-1~+ 75%(9/12).Dial sign evaluated that normal external rotation angle was 75%(8/12).Nor- real varus stress test at 30?knee flexion accounted for 83%(10/12).Scores of Lysholm,Tegner and HSS were 90.5,5.1 and 84.5,respectively(P＜0.01=.Conclusion Synchronized reconstruction of PCL and PLC injuries with only one Achilles tendon can obtain satisfactory clinical result,with less expense and shorter operation time.

The present study examined the role of Wnt/β-catenin signaling pathway in the degeneration of nucleus pulposus cells and the protective effect of DKK1 on nucleus pulposus cells. The model of nucleus pulposus cell degeneration was induced by intra-disc injection of TNF-α, and the expression of β-catenin protein was detected by Western blotting. The cultured rabbit nucleus pulposus cells were divided into 4 groups. In group A, the cells were cultured with normal medium and served as control group. In group B, the cells were cultured with TNF-α and acted as degeneration group. In group C, the cells were cultured with TNF-α and transfected with Adv-eGFP and was used as fluorescence control group. In group D, the cells were cultured with TNF-α and transfected with Adv-hDKK1-eGFP, serving as intervention group. The expression of type II collagen, proteoglycan, β-catenin, and MMP-13 in each group was detected by immunocytochemistry and RT-PCR. The result showed that TNF-α increased the expression of β-catenin and MMP-13, and significantly inhibited the synthesis of type II collagen and proteoglycan, which resulted in the degeneration of nucleus pulposus cells. This effect could be obviously reversed by DKK1. We are led to concluded that TNF-α could activate the Wnt/β-catenin signaling pathway, and increase the expression of MMP-13, thereby resulting in disc degeneration. Specifically blocking Wnt/β-catenin signaling pathway by DKK-1 could protect the normal metabolism of intervertebral disc tissue. The Wnt pathway plays an important role in the progression of the intervertebral disc degeneration.

Breast cancer is the leading cause of malignancy-related mortality in women worldwide. The more accurate prediction of lymph node metastasis and evaluation of personalized prognosis of breast cancer patients could provide evidence and reference for individualized comprehensive treatment and clinical decision-making. Nomogram is statistical calculation model developed to generate individualized prediction of a certain clinical event through the factors associated with it. Currently breast cancer related nomogram models is most commonly used in the prediction of non-sentinel lymph node status in patients with sentinel lymph node-positive breast cancer, sentinel lymph node metastasis in clinical node-negative breast cancer and prognosis evaluation of breast cancer. This article reviewed the recent advances in breast cancer related nomograms according to the above mentioned three aspects, and evaluated respectively the predictive factors, accuracy, characteristics and clinical application potential.
