1.Effects of ERC61-Extract from Rhizoma Curcumae on Hematopoietic Functions in Mice Treated by Chemotherapy

OBJECTIVE:To explore the effects of ERC61,an extract from Rhizoma Curcumae on hematopoietic functions of mice pretreated by cyclophosphamide(CTX) and the proper regime of the administration of ERC61.METHODS:The experimental mice were treated with CTX(100mg? kg-1? d-1) by ip for 3 consecutive days to establish leucopenia model.ERC61 was ip administered to the prevention group,the treatment group,and the combined group(prevention+ treatment).The numbers of blood leucocytes and bone marrow cell were counted by conventional methods;spleen index was measured as well.RESULTS:ERC61 enhanced the count of leucocytes and nucleated cells in bone marrow and improved the level of spleen index.CONCLUSION:ERC61 is able to improve the disorders of hematopoietic functions induced by CTX,and the combination therapy of prevention and treatment is the optimum one in efficacy.
2.Determination of Curdione in Nano-micelles Loaded with Curdione by UV-Vis Spectrophotometry

OBJECTIVE:To establish a UV-Vis spectrophotometric method for content determination of Curdione in Nano-micelles loaded with Curdione.METHODS:Curdione was colored by sulfuric acid-vanillin reagent.The absorbance was measured by visible-spectrophotometry at a wavelength of 500nm.RESULTS:The content of Curdione in the range of 0.050~ 0.400mg? mL-1 was in accordance with the Lambert-Beer law(r=0.999 3).The average recovery was 99.14%,and the RSD was 1.23%(n=9).CONCLUSION:This method is simple,accurate and suitable for the content determination of curdione in Nano-micelles loaded with Curdione.
4.Changes of brain - derived neurotrophic factor expression in gene modified bone marrow mesenchymal stem cells

AIM: To study the changes of brain - derived neurotrophic factor ( BDNF ) expression in gene modified bone marrow mesenchymal stem cells ( BMSC) .
●METHODS:BMSC were divided into blank control group ( without transfected BMSC ) , negative control group ( empty vector without BDNF gene transfected BMSC) and experimental group ( BDNF gene transfected BMSC) . The expression of BDNF mRNA in BMSC was measured by Realtime PCR, and the expression of BDNF in BMSC was measured by ELlSA.
●RESULTS:The BDNF mRNA expressions of 3, 4, 5, 6, 7 and 8-generation BMSC cells in the experimental group were higher than those in the blank control group and negative control group. The differences were statistically significant (P3: F=491. 788, P<0. 05; P4: F=380. 112, P<0. 05;P5:F=1854. 929, P<0. 05; P6: F=224. 540, P<0. 05;P7:F=619. 155, P<0. 05; P8: F=10. 092, P<0. 05). As the BMSC cells in the experimental group passaging, the BDNF mRNA expressions in the experimental group decreased. The difference of BDNF mRNA expression among different passage cells was statistically significant (F=298. 603, P<0. 05). The BDNF secretion of 3, 4, 5, 6, 7 and 8-generation BMSC cells in the experimental group were higher than those in the blank control group and negative control group. The differences were statistically significant (P3: F=520. 609, P<0. 05; P4: F=734. 520, P<0. 05;P5:F=152. 847, P<0. 05;P6:F=80. 372, P<0. 05; P7:F=96. 083, P<0. 05;P8:F=38. 532, P<0. 05). As the BMSC cells in the experimental group passaging, the BDNF secretion decreased. The difference of BDNF secretion among different passage cells was statistically significant (F=230. 084, P<0. 05).
●CONCLUSION:Long-term expression of BDNF in BMSC can be enhanced by genetic engineering.
5.Effects of vitamin A deficiency and its supplementation on tibial growth in rats

Objective To study the effects of vitamin A deficiency(VAD)and its supplementation on the skeletal growth in rats.Methods Eighteen female Wistar rats were randomly divided into VAD(Vit A 400 IU/kg diet,n=3),supplement(n=9)and control groups(normal diet Vit A 6 500 IU/kg,n=6).The supplement groups were randomly divided into 3 sub-groups,and their diet was changed to Vit A 6 500 IU/kg diet from 14 d after pregnancy,0 d and 4 weeks after birth respectively until they were 7 weeks age old.When they were 0 day,4 and 7 weeks old,their body weight,the length of the tibia,serum Vit A level were measured,and the histology of growth plate was also observed.Results At every measuring times,the serum Vit A level was significantly lower,and the tibial length was shorter in VAD group than in the control(P

Objective To explore the methods of thoracoscopic intrapleural perfusion hyperthermic chemotherapy(TIPHC)on diagnosing and treating malignant pleural effusion caused by lung cancer,as well as its effect.Methods From February 1999 to March 2006,seventy patients with malignant pleural effusion caused by lung cancer were randomly divided into therapeutic group(35 cases)and control group(35 cases).Pleural biopsy and TIPHC under general anesthesia with unilateral ventilation were performed in the therapeutic group,and intrapleural injection of cisplatin was administered in the control group after drainage of pleural effusion.The effect on malignant pleural effusion,the change for the concentration of carcino-embryonic antigen(CEA),cytokeratin-19 fragments(CYFRA21-1),neuron-specific enolase(NSE)and the side effect were compared before and after the treatment.Results The therapeutic group achieved total response rate of 100%,but only 54.3% in the control group,with significant difference(P

Objective To study the adjustment principle and optimizing methods of the image processing parameters of Computed Radiography(CR), including the image density, contrast degree, tissue equalization, spatial frequency, LUT curve,etc. Methods Konica REGIUS MODEL170 CR was applied to adjust various image processing parameters on its CS-1 console. Results Images of all parts of body could be optimized and improved. Conclusion High-quality medical images are available only if the adjustment methods of image processing parameters are completely comprehended and reasonably applied.
8.Genetic polymorphism study of the Y chromosome in pedigree with the surname Kong and their forensic implications

Objective Acquiring genetic information of Y-SNPs and Y-STRs genetic makers from samples with the surname of Kong, which is useful for exploring the correlation between surname and Y chromosome in forensic applications studies.Methods Two multiplex genotyping assays and SNaPshot assay were used to analyze 255 unrelated male blood samples who share the same surname Kong and 330 unrelated male blood samples obtained randomly. 17 Y-STRs were typed for the surname Kong population samples. The software Arlequin 3.5.1.2 and the program Network 4.6.1.1 were used for data statistical analysis.Results 13 haplogroups were observed. The highest haplogroup frequency in the two populations were O3a2c1a-M117 (21.57%, 14.85%). 196 haplotypes in Kong population deifned by 17 Y-STRs locus were obtained and the haplotype diversity was 0.9939. 14-12-25-28-19-15-12-19-12-11-12-22-12-11-14-10-19 is the typical haplotype. Median Joining algorithm and Mismatch Distribution were adopted to analyze the Y-STR haplotye under haplogroup O3-M122, and the result shows that there are two “central star” distribution. Conclusion Combined with Y-SNP and Y-STR analysis showed that the Kong population had experienced complicated exchanges and expansion or continued growth, which has more than one surname origin. Hence, its population genetic structure and historical differences have potential applications in forensic science.
9.Delayed acute kidney injury recognition is not associated with poor short-term outcomes: a propensity score matched study

Objective Acute kidney injury (AKI) is common but usually under-diagnosed in hospitalized patients,of the impact of which on patients is still unclear.The paper was aimed to investigate the impact of delayed recognition of AKI on short-time prognosis of patients through a propensity score matched study.Methods From Oct 2013 to Sep 2014,1401 adult hospitalized patients with AKI in the First Affiliated Hospital of Nanjing Medical University were divided into delayed recognition group and timely-diagnosed group according to propensity score matching (1∶ 1) without replacement method.Primary endpoint was 30-day all-cause mortality,and secondary endpoints included recovery of kidney at discharge,length of hospitalization,length of intensive care unit stay and hospital costs.Results There were significant differences in age,department distribution,complications,stage of AKI,Charlson index,APACHE Ⅱ score,SOFA score between the two groups before matching.After matching,there were no significant difference in demographic data,department distribution,complications,stage of AKI,Charlson index,APACHE Ⅱ score,SOFA score between the two groups except in blood urea nitrogen (P=0.039) and use of diuretics (P=0.018).Delayed recognition of acute kidney injury was not associated with 30-day all-cause mortality in univariate (P=0.711) and multivariate Logistic regression analyses.The secondary endpoints did not differ in two groups.Conclusion Delayed acute kidney injury recognition did not associate with poor short-term outcomes in adult hospitalized patients.
10.Percutaneous vertebroplasty for treatment of Kümmel's disease with vacuum signs

Objective To study the clinical outcome of percutaneous vertebroplasty (PVP) for the s disease who had been suffering severe back pain even after conservative therapy for months were treated with PVP. Their preoperative CT images indicated nonunion of factures and "vacuum signs". Dynamic X-ray films demonstrated formation of pseudoarthrosis in the involved vertebral bodies in some eases. Their back pain was evaluated with visual analogue scale (VAS). Their preoperative and postoperative VAS scores and radiological indexes were compared. Results The mean VAS scores were 7.0±1.2 preoperatively, but 3.1±1.5 at the follow-up (P < 0.05) . The height of anterior margin of involved vertebral body was (2.1±0.3) cm pre-operatively, but (2.3±0.2) cm at the follow-up (P < 0.05). The ratio of anterior margin height to posterior margin height of the involved vertebral body was 0. 67±0. 10, but 0.84±0.08 at the follow-up (P<0.05), The focal kyphosis angle was 27.3°± 6.4° preoperatively but 20.7°±5.0° at the follow-up (P < 0.05). No pulmonary embolisms or neurological injuries happened. Conclusion PVP is an effective method for anterior margin of the involved vertebral body partially, and decrease the focal kyphosis.
