1.Comparation of pharmacokinetics of midazolam tablet in Chinese Hui,Korean and Han healthy subjects

Aim To study the pharmacokinetics of midazolam tablet after a single oral dose in Hui,Koreanand Han healthy volunteers.Methods Ten Hui, ten Han and nine Korean healthy volunteers were involved in the study. Each subject received a single dose of 15 mg midazolam tablets. The plasma concentration was determined by HPLC. The pharmacokinetic parameters were calculated by DAS2.0 software and compared by one-way analysis of variance or KrusKal-Wallis rank test for PK parameters of different nationalities.Results There were statistically significant differences between Hui,Korean and Han nationality for PK parameters C_(max),MRT_(0~12 h) and T_(max)(P<0.05).The ranges of interindividual variation in the different ethnic groups and the same ethnic group were large.There were not satisfactory differences between young males and females for all pharmacokinetic parameters(P>0.05).Followimg oral administration, doublepeaks in midazolam blood concentration-time profiles were observed in more than half of subjects.Conclusion There are large interindividual variation and statistically significant difference in pharmacokinetics of midazolam tablet between Chinese Hui, Korean and Han.These observations suggest the dosage of midazolam should be adjusted in clinical practice.
2.The application of monoenergetic technology of dual-source CT in removing the artifact of metal fixation after the operation of lumbar spondylolysis

Objective To explore the clinical value of monoenergetic technology of dual-source CT in removing the artifact of me-tallic fixation after the operation of lumbar spondylolysis.Methods 24 cases of patients with metallic fixation after operation of lum-bar spondylolysis were scanned by dual-energy CT.To reconstruct the imaging using monoenergetic and 3D Inspace software,obtain 50,80,110,140,170 keV five groups of images and the multi-planar reconstruction*(MPR),volume vendering(VR),maximum intensity projection(MIP)imaging in different conditions.The five groups image and average weighted 120kev image are evaluate by two senior radiologist.The quality score of five groups of images and the average weighted images are analyzed statistically using Kruskal-Wallis test.The clinical value of images in the optimal condition are evaluated by two senior department of orthopedics phy-sicians.The results of two radiologists and department of orthopedics physician are examined by kappa test to evaluate consistency. Results In all patients after operation,the case of 110keV images above 3 points is 24,the case of 80keV images above 3 points is 14,the case of of 140keV images above 3 points is 13,the case of weighted average 120kev images above 3 points is 8.Images in 110keV have the best quality (F=98.523,P<0.01).MPR,VR,MIP images are reconstructed in 110keV.The ratio of image which are excellent and have high clinical value is 86.1%,especially the ratio of MPR image is 100%.Conclusion Monoenergetic technology of dual-source CT has a higher clinical value in removing the artifact of metallic fixation after the operation of lumbar spondylolysis.Under the condition of 110keV the quality of images are best,the reconstructed images using MPR Technology have the best clinical value.

4.Association between cytotoxic T lymphocyte-associated antigen 4 gene haplotype and ulcerative colitis in Chinese patients

Objective To investigate the association of gene polymorphism of cytotoxic T lymphocyte-associated antigen 4 (CTLA-4) with ulcerative colitis (UC) in Chinese patients. Methods The A+49G transition polymorphism at position 49 (exon 1) and C-318T transition polymorphism at position -318 in promoter of the CTLA-4 gene were determined by polymerase chain reaction with sequence specific primers (PCR-SSP) method in 82 Chinese patients with UC and 204 healthy controls of Han nationality. Results No significant differences in the distribution of genotype and allele frequencies were observed between CTLA-4 C-318T and A+49G gene polymorphisms in UC patients and normal controls. 5.DNA fingerprinting of crude and processed Atractylodes lancea

AIM: To study the DNA fingerprinting of crude and processed Atractylodes lancea. METHODS: DNA fingerprinting of samples of crude and processed Atractylodes lancea were examined by RAPD. RESULTS: All crude Atractylodes lancea showed similar fingerprinting characteristics,while the processed Atractylodes lancea products indicated considerable different results. CONCLUSION: DNA fingerprinting is a reliable method for di-(fferentiating) crude Atractylodes lancea.

7.Effect of parthenolide on neointimal hyperplasia after balloon injury

AIM:To detect the effect and underlying mechanism of parthenolide ( PN) on neointimal hyperpla-sia.METHODS:After 1 week of high-fat feeding, 30 male New Zealand white rabbits (2.0~2.3 kg) were randomly di-vided into 6 groups: sham +NS, rabbits received 0.9% normal saline after sham operation; sham +DMSO, rabbits re-ceived DMSO after sham operation;balloon injury(BI)+NS, rabbits received NS after balloon injury;BI+DMSO, rabbits received DMSO after balloon injury;BI+PN low, rabbits received PN at 1 mg/kg after balloon injury;BI+PN high, rab-bits received PN at 2 mg/kg after balloon injury .The drugs were intraperitoneal injected once a day after the operation until sacrifice.After fed with high-fat diet for 4 weeks, the intima-media thickness, the expression of caspase-1, IL-1β, the lev-els of IL-8, TC, TG, LDL and HDL in the serum were measured .RESULTS:Compared with sham +DMSO group, the thickness of intima, the amount of caspase-1, IL-1βand IL-8 in BI +DMSO group were significantly increased ( P <0.05).The levels of caspase-1, IL-1βand IL-8 were significantly decreased in BI +PN high group compared with BI +DMSO group (P<0.05).CONCLUSION:Neointimal hyperplasia is suppressed by PN after balloon injury , the potential mechanism may be associated with its anti-inflammatory role .
10.Activity of cathepsin D and alpha-1 antitrypsin in patients with hip or knee osteoarthritis

Objective: To assess the activity of cathepsin D (CAT-D) and alpha-1 antitrypsin (AAT) in blood in patients with hip or knee osteoarthritis, and to explore whether these two enzymes could be served as serum biomarkers for cartilage degeneration. Methods: hTe activity of CAT-D and AAT in blood serum of 44 women and 26 men with hip or knee osteoarthritis was determined by the method of ELISA before total joint replacement and on the 10th day atfer the surgery. One hundred healthy volunteers were chosen as the control. All datawere analyzed by using SPSS19.0 sotfware. Results: Compared with the controls, the activity of CAT-D in patients with osteoarthritis was decreased by 25% (P<0.05) and 50% (P<0.05) before and atfer the surgery, respectively. hTe activity of AAT in the osteoarthritis patients before the surgery was not signiifcantly changed compared with the control group (P>0.05), but it was increased by 80% after the surgery than that in the control group (P<0.05). hTere was no signiifcant difference in the activities of 2 enzymes between hip and knee osteoarthritis (P>0.05). hTe gender, hypertension, diabetes and age did not affect the activities of the 2 enzymes (P>0.05). Conclusion: AAT might be a possible inflammatory indicator in the osteoarthritis. CAT-D and AAT enzymes are not affected by gender, age, hypertension and diabetes, etc, and they might be served as potential biomarkers for cartilage degradation.
