Acta Anatomica Sinica 2002;0(05):-
Objective The purpose of this study was to develop a new cultural method, so that the rat bone marrow stromal cells (MSCs) can well differentiate into osteoblasts in vitro and the cultured osteoblasts can be identified. Methods MSCs of rats were isolated and cultured, then the cultured cells were identified with inverted microscopy and undergone an HE stain for observing the morphology of living cells, and the cells were histochemically stained for examining the activity of cellular alkaline phosphatase (ALP), as well as were radio-immunologically analyzed for detecting the secretion of osteocalcin. Results The MSCs were typical fibroblast-like and possess a strang proliferative capability after cultured in vitro. The cellular ALP activity and osteocalcin secretion achieved the peak value from the fortieth day and maintained the status until the sixtieth day after cultured in vitro. Conclusion The data demonstrated that the new developed culture method has the advantages of short time, less risk of contamination and higher efficiency, and conducive to MSC's differentiating and proliferating into osteoblasts that do function well.

3.Hemo oxygenase-1 and its relationship with svere acute pancreatitis
International Journal of Surgery 2010;37(2):121-124
Recently, heme oxygenase-1 (HO-1) has been emphasized in the area of intensive care unit (ICU) and other domains. HO-1 can catalyze the decomposition of heine and the products of its enzymatic activity, including carbon monoxide, biliverdin, and bilirubin and ferritin can play a significant cytoprotec-tive role in antagonizing inflammation, protecting cells from oxidative injury and cellular stresses. In the re-search of severe acute pancreatitis (SAP), researchers have realized that it is of great importance to antago-nize the overflow of cytokines in order to improve the therapeutic effect of SAP. In this article, we give a re-view of the research progress of HO-1 and its relationship with SAP.

Methods 72 patients with gallstones were treated as research subjects and 30 cases with normal physical examination as the control group, the serum interleukin and T lymphocyte subsets levels of observation group were detected before and after surgery. Results The preoperative serum IL-6, IL-8 and TNF-α levels of observation group were(61.5 ±4. 8) ng/L, (246. 2 ±15.3)ng/L and(1.79 ±0.72)mg/L respectively which were significantly higher than postoperative and control group, the difference was statistically significant (P < 0. 05 ) , while there was no significant difference between postoperative and normal control group ( P > 0.05); The preoperative CD3+ , CD4+ and CD8+ T cells ratio of observation group were (0. 53 ± 0. 06) % , (0. 28 ±0.05)% and(0.21±0.04)% respectively which were significantly higher than postoperative and control group, the difference was statistically significant(P < 0.05) , while there was no significant difference between postoperative and normal control group(P>0.05). Conclusion Patients with gallstone had obvious abnormal serum IL-6, IL-8, TNF-α,CD3+ ,CD4+ and CD8+ T cells levels, surgery could not only eliminate inflammation, but also help the recovery of immune function in patients.
6.EFFECTS OF RADIX HEDYSARI POLYSACCHARIDE (RHPS) & ASTRAGALI POLYSACCHARIDE (APS) ON IMMUNE FUNCTIONS
Chinese Pharmacological Bulletin 1986;0(06):-
The effects of RHPS & APS on immune function in normal mice and immunosuppressed mice were studied. The results showed that RHPS and APS enhanced not only the phagocytosis of peritoneal mac-rophages in normal mice, but also antagonized completely the immun- Osuppresslve effect of prednisoione. RHPS and APS increased both the lymphocyte transformation rate induced by PHA and ANAE+ rate in normal mice, and antagonized also the suppressive effects of GY or PDS. No significant influence was found on the proliferation of rabbit lymphocyter treated with PHA in vitro. It seems that no obvious difference of effect was found on immune function between RHPS and APS.
7.Study on the Effect and Its Mechanism of Dexmedetomidine on the Inflammatory Response in Septic Mice
China Pharmacy 2017;28(19):2651-2654
OBJECTIVE:To study the effect and its mechanism of dexmedetomidine(Dex)on inflammatory response in septic mice. METHODS:Mice were randomly divided into normal control group, model group,miR-146a inhibitor (50 mg/kg)+Dex (50 μg/kg)group,Dex low-dose,medium-dose,high-dose groups(10,30,50 μg/kg),10 in each group. Except for normal con-trol group, other groups were intraperitoneally injected lipopolysaccharide to induce septic models, intraperitoneally injected rele-vant medicines after 0.5 h. After drug intervention for 6 h,miR-146a expression,IRAK1 and TRAF6 mRNA expressions in periph-eral blood mononuclear cells in each group were detected by real-time fluorescence quantitative polymerase chain reaction method. IRAK1,TRAF6 protein expressions in peripheral blood mononuclear cells in each group were detected by Western blot method. TNF-α,IL-6 levels in serum were detected by enzyme-linked immunosorbent method. RESULTS:Compared with normal control group,miR-146a expression,TNF-α and IL-6 levels,IRAK1,TRAF6 mRNA and protein expressions in peripheral blood mononu-clear cells in model group were increased(P<0.01). Compared with model group,miR-146a expression in peripheral blood mono-nuclear cells in Dex medium-dose,high-dose groups were increased;TNF-α and IL-6 levels,IRAK1,TRAF6 protein expressions were decreased (P<0.05 or P<0.01);while IRAK1,TRAF6 mRNA expressions changed less obviously (P>0.05). Compared with Dex high-dose group,miR-146a expression,in peripheral blood mononuclear cells in miR-146a inhibitor+Dex group was de-creased;TNF-α and IL-6 levels,IRAK1,TRAF6 protein expressions were increased (P<0.05 or P<0.01);while IRAK1, TRAF6 mRNA expressions changed less obviously(P>0.05). CONCLUSIONS:Dex can inhibit the inflammatory response in sep-tic mice. The mechanism may associate with inducing miR-146a expression and inhibiting the 2 important adaptor proteins IRAK1, TRAF6 expressions in Toll-like receptor 4/nuclear factor-κB pathway.
8.Effect of Head Extension Angle to Glottis Exposure : A Simulation Evaluation Based on CT Reconstruction Images of Upper Airway
Journal of Medical Research 2017;46(8):169-171
Objective To investigate the effect of head extension angle to glottis exposure by using three-dimensional CT reconstruction sagittal plane images of upper airway. Methods There were 80 patients, aged 7-78 years old,who had been scanned by helical CT in the head and neck.Reconstructed images were obtained by AW4.4 workstation and the following parameter was measured:angle α(A0 group),the angle created by the upper central incisor and root of epiglottis(angular vertex) and midpoint of glottis.Reconstructed images were printed according to the actual size of human body in a ratio of 1∶ 1,then the images were cut off from the upper central incisor to root of epiglottis by a section of arcs.Increasing the degree of angle α by clockwise to hypothetically increase the angle of head extension. The glottis was exposed by using a paper-made laryngoscope(Macintosh) model, and the success rate of glottis exposure was recorded when the degree of angle α was respectively increased 10°(A10 group),20°(A20 group),and 30°(A30 group). Results Compared with A0 group, the success rate of glottis exposure increased significantly in A10 group,A20 group and A30 group. The success rate of glottis exposure in A20 group and A30 group was significantly higher than A10 group.Compared with A20 group, the success rate of glottis exposure obviously increased in A30 group. Conclusion Based on CT reconstruction images of upper airway,we can analog study the effect of head extension angle to glottis exposure.
9.Intraoperative and postoperative impact of CO2 temperature and humidity to gynecologic laparoscopic surgery: a meta-analysis
China Journal of Endoscopy 2017;23(4):49-56
Objective To evaluate the impact of warmed and humidified CO2 on the gynecological laparoscopic operation. Methods We searched keywords related to gynecological laparoscopic and CO2 pneumoperitoneum temperature and humidity on Pubmed and Wanfang database, then analyze the postoperative pain, analgesic consumption, core temperature, operation time, length of hospital stay. Results 17 publications included into the meta analysis after screening. Compared with the standard laparoscopy, warmed and humidified CO2 can reduce postoperative pain and prevent intraoperative hypothermia. No significant difference in postoperative analgesic consumption, postoperative core temperature, duration of recovery and hospital stay, endoscope lens fog and operation time. Conclusions Only few beneficial effects on postoperative pain and core temperature have been identified in this meta-analysis, though more studies are probably needed to study the real impact of warmed and humidified CO2 for laparoscopic procedures.
