ObjectiveTo investigate the pattern of the relationship of craniopharyngiomas with pituitary stalks and hypothalamus. Furthermore, based on different patterns affirmed during the operations, surgical strategies were designed for radical excision. MethodsA series of 66 patients with primary suprasellar craniopharyngiomas were treated with microsurgical techniques and the relationships of the craniopharyngiomas with pituitary stalk and hypothalamus were analyzed prospectively. Appropriate surgical approaches were selected preoperatively according to the lesion's sizes and expanding directions. The relationships of the lesions to the pituitary stalks and hypothalamus were identified in detail and dissected carefully with protection of perforating vessels. ResultsThe patterns that pituitary stalks were involved by craniopharyngiomas were identified in 36 cases and divided into four types: no relation (5 cases), adhesion (10 cases), partial invasion (8 cases) and total invasion (13 cases). The anterior floor of the third ventricle was involved in 42 cases, in which it disappeared totally in 10 cases. Total resection was achieved in 58 case, subtotal resection in 8 cases. Diabetes insipidus (DI) was identified in 44 cases postoperatively. No surgical mortality, no recurrence was found during follow-up of mean 35 months. ConclusionThe appropriate selection of surgical approaches preoperatively, careful identification and according treatment of the relationships of the tumors with pituitary stalks and hypothalamus are the keys to protect the pituitary stalk; hypothalamus in craniopharyngiomas resection in adults.

According to that Y-chromosomal sequence in characterised by Y-specific repeat DNA family (DYZ1 ), which contain 800-5000 copies, a pair of primers Y3, Y4 is designed to amplify their 446bp long of specific DNA segment by polymerase chain reaction (PCR), so as to detect male fetal cells in materal blood. In this paper male fetal cells in maternal blood can be detected by PCR amplification of unpurified DNA from maternal peripheral blood during various stages of gestation (early, middle, late). Compared with villi, ammotic fluid and deliverd neonate sex their coincident rate are 93%. 100%, 87. 5% respectively among three periods. It is revealed that noninvasive examining fetal cells from peripheral blood of pregnant women for diagnosis of sex-linked inherited diseases is significant valuable.

Objective To study the role of pro inflammatory cytokines tumor necrosis factor?(TNF ?),interleukin 1beta(IL 1?),interleukin 6(IL 6) and anti inflammatory cytokines interleukin 4(IL 4),interleukin 10(IL 10),interleukin 13(IL 13) in acute lung injury(ALI). Methods Wistar Rats (200?50g) were randomly divided into the oleic acid(OA) and Lipoplysaccharide(LPS) induced lung injury experimental group and the only OA induced lung injury control group. The LPS(2mg/kg) was injected in rat's tail vein at the 1, 4, 12 and 24 hours after administration of OA(0.2ml/kg, iv). Reverse transcription polymerase chain reaction(RT PCR) were used to detecting the expression of TNF ?, IL 1?, IL 6 and IL 4, IL 10, IL 13 mRNA in the lung tissue of rats in two groups. Results The results showed that the peak expression of TNF ? and IL 1? mRNA at one hour after first hit by oleic acid, and IL 6 mRNA at four hours were observed. Others, such as IL 4, IL 10 and IL 13 mRNA, the highest levels in lung tissue at four hours after lung injury induced by oleic acid. Conclusions The results suggested that TNF ? and IL 1? are presenting early phase of ALI, and that IL 6 played its important role in delayed development. The over expression of anti inflammatory cytokines IL 4, IL 10 and IL 13 may be as a promoters rather than protectors in inflammatory amplification.

?Objective: To study the methodology of the culture of Schwann cells derived from degenerated peripheral nerve. Method: Sciatic nerve of adult rats was surgically cut. 14 days after operation, the degenerated nerve tissue was obtained and treated with trypsin and collagenase typeⅡ to prepare single cell suspension, the cells were purified by different speed of attachment and digestion, and incubated on ZQ membrane in the presence of 10 -5 mol/L cytosine arabinoside. The growth of the cells of passage 2 was studied by MTT assay. Schwann cells were identified with anti S100 immumohistochemistry. Results: The cultured cells were spindly in shape and 95% of them were S100 positive. The population doubling time of passage 2 cells was 72 h.The cells attached and stretched on ZQ membrane as well as on the culture vessel surface. Conclusion: Schwann cells can be cultured and purified by predegeneration of the peripheral nerve, different speed of attachment and digestion and the presence of cytosine arabinoside. The cells can grow well on ZQ membrane.

According to ZFY gene sequence on Y-chromosome, which supposedly codes for the testis determining factor (TDF), a pair of primers F3.2 and F4.2 were designed to specifically amplify 650 bp DNA fragment. The amplified product was electrophoresized on 1.5% agarose gel stained with EB, and then directly visualized under UV. This protocol resulted in constantly producing 650 bp band of the specific product by amplification of 500 pg male genomic DNA and had the same result when mixed with 100 fold amount of female genomic DNA. When this method was used to detect Y-ZFY of chorionic villi and amniotic fluid in 20 samples respectively, its accuracy for determining fetal sex reached 100 percent.

Objective To study the growth effect of human cholangiocarcinoma cell line QBC939 treated by norcantharidin (NCTD) and preliminary illustrate the potential mechanism. Methods The human cholangiocarcinoma cell line QBC939 was detected by MTT assay, flow cytometry, immunocytochemistry after the treatment of NCTD in vitro. Results NCTD displayed inhibitory effect on growth of QBC939 from different doses of 0.125,0.75,2.5,10,120 μg/ml after 48 h (P <0.05).It was in a dose and time dependent manner.Dose-effect curve was drawn and IC50 value was (3.66±1.14) μg/ml.The flow cytometric profiles showed that the rate of cell apoptosis enhanced following increasing the concentration of NCTD[(8.6±0.4) %,(17.6±0.3) %,(22.9±0.4) %,(25.5±0.9) % and (31.1±1.5) %,respectively]and cells blocked in the G2/M phase after treatment with 2.5 μg/ml NCTD[(14.1±1.0) % and (5.7±0.3) %].The expression of the protein caspase-3 elevated after different concentrations of NCTD co-cultured with QBC939 compare with contrast group. Conclusion NCTD has an inhibitory effect on proliferation of QBC939 cell line, and the mechanism might be related to the induction of cell apoptosis and blockade of cell cycle.

Aim To investigate the effect of Dicranostiga Leptodu (Maxim) Fedde (DLF) on experimental liver injury induced by Calmette Guerin bacillus vaccine (BCG) and Lipopolysaccharide (LPS) in mice. Methods Experimental liver injury was induced by Calmette Guerin bacillus vaccine and Lipopolysaccharide in male mice by injecting via tail vein (50 ?g?kg -1 ), and administering LPS (10 ?g?mouse -1 ) 10 d later. Therapeutic groups were given respectively with DLF (0 5,1 0,2 0 g?kg -1 ) before administration of LPS. The level of ALT, AST and LDH and the concentration of ALB and GLB in serum were measured by automatic biochemistry analyzer, and MDA in liver plasma was measured by TBA method and the level of SOD was measured by pyrogallol auto oxidation method. Pathological morphological changes in liver tissue were observed. Results The three experimental groups were found with significant decrease in the elevation of serum GPT and LDH level and the content of MDA in the liver plasma. The liver tissue damages were also ameliorated. Conclusion DLF has protective effect against the experimental liver injury induced by BCG and LPS in mice.
