1.Clones identification of Sequoia sempervirens (D. Don) Endl. in Chile by using PCR-RAPDs technique.

A protocol of polymerase chain reaction-random amplified polymorphic DNAs (PCR-RAPDs) was established to analyse the gene diversity and genotype identification for clones of Sequoia sempervirens (D. Don) Endl. in Chile. Ten (out of 34) clones from introduction trial located in Voipir-Villarrica, Chile, were studied. The PCR-RAPDs technique and a modified hexadecyltrimethylammonium bromide (CTAB) protocol were used for genomic DNA extraction. The PCR tests were carried out employing 10-mer random primers. The amplification products were detected by electrophoresis in agarose gels. Forty nine polymorphic bands were obtained with the selected primers (BG04, BF07, BF12, BF13, and BF14) and were ordered according to their molecular size. The genetic similarity between samples was calculated by the Jaccard index and a dendrogram was constructed using a cluster analysis of unweighted pair group method using arithmetic averages (UPGMA). Of the primers tested, 5 (out of 60) RAPD primers were selected for their reproducibility and high polymorphism. A total of 49 polymorphic RAPD bands were detected out of 252 bands. The genetic similarity analysis demonstrates an extensive genetic variability between the tested clones and the dendrogram depicts the genetic relationships among the clones, suggesting a geographic relationship. The results indicate that the RAPD markers permitted the identification of the assayed clones, although they are derived from the same geographic origin.

2.Percutaneous Endovascular Aortic Aneurysm Repair with INCRAFT Endograft Guided by CO2 Digital Subtraction Angiography in Patients with Renal Insufficiency

Iodinated contrast is the most common contrast agent used during endovascular abdominal aneurysm repair (EVAR). However, its use may worsen kidney function in patients with renal insufficiency. Previous studies have demonstrated the safety and effectiveness of carbon dioxide (CO2)-EVAR. Here, we report cases of three male patients with mild renal insufficiency (mean age: 79 years) that successfully underwent CO2-EVAR using INCRAFT ultra-low profile endografts. CO2 angiography provided the necessary vascular roadmap for safe and effective percutaneous EVAR, eliminating the need for iodinated contrast media and preventing contrastinduced nephropathy.
3.IKKβ overexpression together with a lack of tumour suppressor genes causes ameloblastic odontomas in mice.

Odontogenic tumours are a heterogeneous group of lesions that develop in the oral cavity region and are characterized by the formation of tumoural structures that differentiate as teeth. Due to the diversity of their histopathological characteristics and clinical behaviour, the classification of these tumours is still under debate. Alterations in morphogenesis pathways such as the Hedgehog, MAPK and WNT/β-catenin pathways are implicated in the formation of odontogenic lesions, but the molecular bases of many of these lesions are still unknown. In this study, we used genetically modified mice to study the role of IKKβ (a fundamental regulator of NF-κB activity and many other proteins) in oral epithelial cells and odontogenic tissues. Transgenic mice overexpressing IKKβ in oral epithelial cells show a significant increase in immune cells in both the oral epithelia and oral submucosa. They also show changes in the expression of several proteins and miRNAs that are important for cancer development. Interestingly, we found that overactivity of IKKβ in oral epithelia and odontogenic tissues, in conjunction with the loss of tumour suppressor proteins (p53, or p16 and p19), leads to the appearance of odontogenic tumours that can be classified as ameloblastic odontomas, sometimes accompanied by foci of secondary ameloblastic carcinomas. These tumours show NF-κB activation and increased β-catenin activity. These findings may help to elucidate the molecular determinants of odontogenic tumourigenesis and the role of IKKβ in the homoeostasis and tumoural transformation of oral and odontogenic epithelia.
4.A preliminary study of platelet hyperactivity in the chronic indeterminate phase of Chagas’ disease

The chronic indeterminate phase of Chagas’ disease is asymptomatic despite
positive test results for antibodies specific to Trypanosoma cruzi. CD62P-APC (P-selectin)
and PAC-1 FITC (GpIIb/IIIa) may improve diagnosis as biomarkers of platelet activity. Nine
asymptomatic seropositive subjects, previously untreated, were selected from a blood bank
within a year of Chagas’ disease detection, in addition to a control group of four. All subjects
were evaluated by flow cytometry for CD62P, PAC-1 and CD41, and in a complementary
study, by Tissue Doppler Echocardiography for isovolumic relaxation times (IVRT) and E/A
ratios. The subjects were classified as positive or negative for CD62P and PAC-1 by a cut off
obtained from their mean±2SD. For IVRT and E/A ratios, cut offs were obtained from the
American Society of Echocardiography and the European Association of Cardiovascular
Imaging recommendations. Fisher’s exact test was used for associated findings. Pre-test and
post-test probability, sensitivity, specificity, positive and negative predictive values and
likelihood ratios were calculated. Abnormalities were expressed as platelet hyperactivity
and ventricular dysfunction in CD62P, PAC-1, IVRT and E/A ratios. CD62P appears to have
greater sensitivity (0.75) and PAC-1, more accurate specificity (0.75), which may explain
thrombotic events in Chagas’ disease. We recommend the use of CD62P and PAC-1 as biomarkers
of platelet hyperactivity in patients in the chronic indeterminate phase of Chagas’ disease.
