1.Effect of terazosin on expressions of matrix metalloproteinase-9 and monocyte chemotactic protein 1 in a rat model of atherosclerosis

Objective To investigate the effects of terazosin on expressions of matrix metalloproteinase-9 (MMP-9) and monocyte chemotactic protein 1 (MCP-1) in a rat model of atherosclerosis and to explore the mecha-nism for reducing atherosclerosis by terazosin. Methods Normal rats were fed with high lipid feedstuff to set up a rat model of atherosclerosis. Blood lipid indexes were measured and the expressions of MMP-9 and MCP-1 in aorta were detected and further quantified by immunohistochemistry after 12-week high lipid feedstuff. Results The plasma concentration of TG, TC, LDL in the terazosin group were significantly lower than those in the high cholesterol-diet group (P<0.01). The expressions of MMP-9 and MCP-1 in the high cholesterol-diet group and in the terazosin group were significantly higher than those in the normal-diet group (P < 0.01), but expressions of MMP-9 and MCP-1 in the terazosin group were significantly lower than those in the high cholesterol-diet group (P<0.01). Conclusion Terazosin can lower the concentration of TC, LDL but increase the HDL concentration. The anti-atherosclerotic mechanism of terazosin is associated with the decrease of the expressions of MMP-9 and MCP-1 and plasma lipoprotein.
2.Protection of atorvastatin on cardiac function following ischemia-reperfusion injury in rabbits

AIM:To observe the effect of atorvastatin(ATV) on myocardial ischemia-reperfusion injury and the role of inducible nitric oxide synthase (iNOS). METHODS: The rabbits were randomly divided into control group, ATV group, ATV+S-Methylisothiourea sulfate (SMT) group, SMT group. All rabbits were subjected to 40 min ischemia and 240 min reperfusion. The hemodynamic variables, CK-MB, LDH-1 and nitric oxide synthase were detected after reperfusion. RESULTS: LVDP and +dp/dtmax decreased by 24.6% and 35.3% respectively following ischemia-reperfusion. Pre-treated by ATV (10 mg?kg-1?d-1) for three days, LVDP and +dp/dtmax decreased by 21.7% and 41.3%, CK-MB and LDH-1 by 31.4% and 19.1% and iNOS increased to 102.6%. The reduction of LVDP and +dp/dtmax, CK-MB, LDH-1 and iNOS in ATV+SMT group was no statistically significant with those in control group. CONCLUSION: ATV pretreatment protected myocardial ischemia-reperfusion injury by upregulating iNOS.
3.Association of rs501120 and rs17465637 gene polymorphisms with coronary heart disease in the Chinese Han population

Objective To investigate the rs501120 and rs17465637 gene polymorphisms,and their relationship with the risk of coronary heart disease(CHD)in Chinese Han population.Methods 775 CHD without treatment and 775 age and gender matched controls were selected for this study,the genotypes of two single nucleotide polymorphisms(SNP)rs501120 and rs17465637 were tested with TaqMan-MGB probes.Results There was no significant difference in the frequency of genotypes of the 2 SNPs between CHD group and control group(P >0.05).Stratified analysis showed that SNP rs501120 had significant protection with CHD in people younger than 60 years old(OR 0.4,95% CI 0.2-0.9,P < 0.05)or people with diabetes(OR0.3,95%CI0.1-0.7,P <0.05).Conclusions The results suggested that rs501120 was tightly associated with CHD in people younger than 60 years or had diabetes.
4.Testosterone suppresses rat vascular smooth muscle cell phenotypic transition and proliferation

Objective To investigate the inhibitory effect of testosterone on oxidized low-density lipoproteins ( ox-LDL)-stimulated phenotypic transition and proliferation of vascular smooth muscle cells ( VSMCs) in vitro, and to explore its possible mechanisms. Methods Rat VSMCs cultured in vitro were divided into control group, ox-LDL group(50μg/mlox-LDL),fetalbovineserum(FBS)group(10% FBS),andtestosteronegroups(5×10-8 or5×10-7 mol/L testosterone plus 50μg/ml ox-LDL) . The effect of testosterone on ox-LDL-induced proliferation of VSMCs was explored by WST-1 assay. The cell cycle distribution was determined using flow cytometry. Western blotting was used todetecttheexpressionsofmitofusin2(Mfn2),phosphorylatedextracellularsignal-regulatedkinases1/2(p-ERK1/2) , proliferating cell nuclear antigen ( PCNA) ,α-smooth muscle actin (α-SMA) ,and osteopontin ( OPN) . Results Compared with control group, the proliferation of VSMCs was promoted by ox-LDL, the number of VSMCs decreased in G0/G1 phase and increased in S phase significantly, the expression levels of Mfn2 and α-SMA were significantly reduced, and the expression levels of p-ERK1/2, PCNA, and OPN were significantly raised in ox-LDL group. Compared with ox-LDL group, the proliferation of VSMCs was inhibited, the number of VSMCs increased in G0/G1 phase and decreased in S phase in two testosterone groups, along with the increased expressions of Mfn2 andα-SMA, and the descended expressions of p-ERK1/2, PCNA, and OPN. Conclusions Testosterone inhibits phenotypic transition and proliferation of VSMCs induced by ox-LDL in vitro, which may be related to the up-regulated expression of Mfn 2 and the suppression of ERK1/2 pathway.

6. Follow-up study of 116 cases of transjugular intrahepatic portosystemic shunt in the treatment of cirrhotic portal hypertension

Objective:
To investigate the incidence rate of transjugular intrahepatic portosystemic shunt (TIPS) complications in the treatment of cirrhotic portal hypertension, and analyze the cause of complication to management methods.
Methods:
Data of 116 patients obtained from Zhongnan Hospital of Wuhan University were retrospectively analyzed. Portal venous pressure, routine blood test, coagulation test, liver and kidney function test, ammonia blood test, imaging and endoscopy reports were collected before and after procedure. The incidence rate of hepatic encephalopathy, gastrointestinal bleeding, ascites and shunt dysfunctions were observed. Data were expressed as mean ± Standard deviation and analyzed by t-test. A chi-squared test was used for comparison between categorical variables.
Results:
The success rate of TIPS operation was 97.41% (113/116). Two patients underwent prompt TIPS procedure due to active bleeding. Bleeding was successfully stopped. Portal venous pressure of 113 patients decreased from (42.73 ± 7.64) cmH2O to (24.92 ± 7.60) cmH2O, and the difference was statistically significant (
7.Clinical study on transesophageal electrocardiogram for measuring left atrioventricular interval in patients with coronary artery disease

Objective To explore the mechanism and clinical significance of left atrioventricular interval abnormality in the patients with coronary artery disease.Methods A total of 48 patients with coronary artery disease were selected and divided into the left interval atrioventricular shortening group(AV≤100 ms) and the left atrioventricular interval normal group(AR>100 ms) according to the left atrioventricular interval measured by the esophageal electrocardiogram.The clinical features,cardiac echocardiography and coronary vessel lesions were compared between the two groups.Results The age in the AR≤ 100 ms group was higher than that in the AR>10 ms group(P=0.018);the proportion of right coronary arterial lesion in the AR ≤ 100 ms group was higher in that in the AR>10ms group(P=0.038);the left atrial diameter in the AR ≤100ms group was enlarged compared with that in the AR> 100 ms group(P=0.041).Conclusion The cardiac electric transduction abnormality exists in the patients with coronary heart disease,the majority of the patients with left atrioventricular interval shortening have the right coronary artery lesion.
8.Preparation of cryoprecipitate coagulation factor from liquid-state plasma by supercooling method

【Objective】 To establish a novel preparation method of cryoprecipitate coagulation factor from overcooled liquid-state plasma. 【Methods】 The fresh liquid plasma was kept at -11℃ to -13℃ for a period of time. It can remain in the liquid state with some coagulation factors generated due to supercooling. Then cryoprecipitate can be obtained from the liquid plasma by siphon method. 【Results】 The average fibrinogen content yielded in cryoprecipitate, prepared from 50 samples of 16-hour-stored fresh liquid plasma, was (186.02±22.72) mg, with the average recovery rate of (37.51±7.42) %, and the average content of coagulation FⅧ was (104.66±22.88) IU, with the average recovery rate of (46.62±5.58) %. 【Conclusion】 The cryoprecipitate coagulation factors could be obtained not only from fresh frozen-thawed plasma, but also from overcooled liquid plasma which is simple and stable, also meets the requirements of relative standards.

The outbreak of coronavirus disease 2019 (COVID-19) caused by the severe acute respiratory syndrome coronavirus 2 has rapidly spread globally. Cancer patients are at a higher risk of being infected with the coronavirus and are more likely to develop severe complications, as compared to the general population. The increasing spread of COVID-19 presents challenges for the clinical care of patients with gynecological malignancies. Concerted efforts should be put into managing gynecological malignancies in an orderly manner by strictly implementing the measures that are specifically developed for controlling the spread of COVID-19. We have drafted Recommendations on Management of Gynecological Malignancies during the COVID-19 Pandemic based on our experience on controlling COVID-19 pandemic in China. We recommend that patients with gynecological malignancies should be managed in hierarchical and individualized manners in combination with local conditions related to COVID-19. Medical care decision should be balanced between controlling COVID-19 pandemic spread and timely diagnosis and treatment for gynecologic oncology patients.
