ABSTRACT:In this study ,we aimed to analyze the transcription level of mitochondria gene Cytb in metacercariae ,larva‐30d ,larva‐60d ,adult ,and egg of Pagumogonimus skrjabini .The mRNA of metacercariae ,larva‐30d ,larva‐60d ,adult and egg of P .skrjabini were extracted with genomic extraction kit ,and transcripted reversely into cDNA .With 18SrDNA of Par‐agonimus westermani as an internal standard primer ,Cytb genes were amplified by real‐time PCR for establishing standard curves to evaluate the copy number of genes .Those quantitative analyses were reliable because the R value of standard curves was 0 .994 (>0 .98) ,and the melting curve showed a single peak .There were increasing trend of transcription of Cytb gene at metacercariae stage ,larva‐30d stage ,and larva‐60d stage .There were less transcription of Cytb gene at adult stage and no transcription at egg stage .There were differences about the transcriptional level of mitochondria Cytb gene at different stages of Pagumogonimus skrjabini ,and peaked at larva‐60d stage ,suggesting that Cytb gene may play a role in the development and migrating of larva .
2.Impact of individualized diabetes education on disease awareness and blood glucose control in type 2 diabetic patients

This study was to evaluate the effect of individualized health education on blood glucose control in type 2 diabetic(T2D)Patients.Two hundred and thirty-eight T2D patients were randomly received individualized diabetes education(intervention group)or general education(control group).At 48 week,change of blood glucose control from baseline was observed.The blood glucose level and HbA1c in the intervention group were lower than those in the control group[(7.1±0.8)vs (9.3±6.5)mmol/L,(7.0 ±1.3)%vs(8.0%±1.0)%;P<0.05].Thus,individualized diabetes education might be helpful for blood glucose control in T2D patients.
3.Effect of electroacupuncture on angiogenesis in rat ischemic hippocampus after focal cerebral ischemia/reperfusion and its mechanism

Objective To explore the effect of electroacupuncture on CD 34 +VEGFR2 +endothelial progenitor cell (EPC)-derived vessels and stromal cell-derived factor-1α(SDF-1α)/CXCR4, and study its mechanism of promoting an-giogenesis in hippocampus after focal cerebral ischemia /reperfusion .Methods A total of 180 healthy male adult Sprague Dawley (SD) rats were randomly divided into sham operation (sham) group, model (I/R) group, electroacupuncture (I/RE) group, I/RE plus AMD3100 (A specific antagonist of CXCR4) group (I/REA) and AMD3100 (I/RA) group. The rats received filament occlusion of the right middle cerebral artery for 2 hours followed by reperfusion .Electroacupunc-ture was applied to “Baihui” (GV20)/“Siguan” (Hegu LI 4/Taichong LR 3) acupoints for 30 min, once a day.The mR-NA expression of SDF-1αand CXCR4 were detected by reverse transcription-polymerase chain reaction ( RT-PCR) .Double immunofluorescence was used to stain CD 34 +VEGFR2 +EPC-derived vessels.Results Compared with the sham group, the mRNA expressions of SDF-1αand CXCR4 were significantly upregulated in I/R and I/RE group ( P<0.05 ) , but that in I/RE group was more significantly increased than I/R group(P<0.05).In addition, the mRNA expression of SDF-1αand CXCR4 were highly increased on day 1 in the I/REA group than that of I/RE group, but decreased than that of I/RE group on day 7 after reperfusion (P<0.01).CD34 +VEGFR2 +EPCs-derived vessels were obviously increased on 3d and 7d in the I/RE group compared with that of the I/R group, and significantly decreased on 7d in the I/REA group compared with that of the I/RE group ( P<0.01) .Conclusions Electroacupuncture can effectively promote an-giogenesis through upregulating the expression of SDF-1αand CXCR4 in rat ischemic hippocampus after focal cerebral is-chemia/reperfusion.
4.Preparation of glycerol reference material

Objective To prepare a glycerol reference material.Methods The material was prepared and characterized according to the primary standard substance technological specification(JJG 1006- 1994).Glycerol was dissolved in water containing 0.5% sodium azide and dispersed to glass ampules.The homogeneity and stability of this material were tested with an HPLC method.Glycerol concentration was determined by a titration method as specified in the Pharmacopoeia of China.Results The three time measuring result of glycerol reference material was 1.297 5?0.014 3,1.302 0?0.008 9,1.313 7? 0.007 8,respectively.Statistical analysis showed that this material was homogeneous (F=1.462,P=0.166) .It was stable for at least 4 years at 4℃.The assigned reference value was 0.103 6 g/g and the expanded uncertainty was 0.000 4 g/g.Conclusions This material meet the technical requirements of national primary standard reference material.It is approved as the Certified Reference Material (GBW 09149) by General Administration of Quality Supervision,Inspection,Quarantine of the People's Republic of China in May,2006.
5.Analysis of appropriate ecological environment of Himalayan marmot based on remote sensing and geographic information system in Qinghai province

Objective To assess the quantitative relationship between the distribution of Himalayan marmot and its ecological environment,the terrain,the temperature and the precipitation,using remote sensing and geographic information system in Qinghai province.Methods The distribution of Himalayan marmot was located by Google Earth and ArcGIS software and by using field survey data provided by Chinese Center for Disease Control and Prevention.The corresponding ecological environment of marmot including terrain,temperature and precipitation were derived from the spatial information datasets.All results were processed according to the overlay and statistics analysis using ArcGIS software.Results Seventy-seven point twenty-seven percent(153/198) of Himalayan marmot were distributed in the area of elevation between 3000 and 4000 meters.The number of marmot reached the highest when the slope was between 0 and 17 degrees,and aspect range was between 91 and 270 degrees,180 degree was as south direction.During the period with the maximum temperature of the warmest month of 14.3-17.5 ℃,17.6-20.8 ℃ and 20.9-24.0 ℃,the distribution of marmot reached 95%(186/198) of the total area.Meanwhile,most of the marmot were presented in the area with average precipitation of 46-108 mm.Conclusions A quantitative analysis of appropriate ecological environment of Himalayan marmot in a large scope is carried uul successfully using remote sensing and geographic information system.The study indicates that spatial information technology has important applications in plague prevention and control.

OBJECTIVETo study the analgesic, antipyretic and anti-inflammatory effect of Bailian Caogen granule.
METHODThe antipyretic effects of Bailian Caogen granule was evaluated in rabbit fever model induced by peptone. The analgesic effect of the drug was studied with pain model of mice induced by acetic acid and hot plate, The severity of oedema in inflamed animal was observed to study the anti-inflammatory effects of Bailian Caogen granule.
RESULTBailian Caogen granule could obviously inhibit the fever of rabbits. The number of writhing induced by acetic acid was reduced and the pain threshold of mice was increased by Bailian Caogen granule. Bailian Caogen granule also had anti-inflammatory activity against xylene-induced mouse ear swelling and carrageenin-induced rat paw edema.
CONCLUSIONBailian Caogen granule has significant analgesic, antipyretic and anti-inflammatory activities.

8.Correlation among cognition impairment, P300 value and MR diffusion tensor imaging data in patients with depression

Objective To investigate the cognition impairment in depression patients with its event-related potential P300 and MR diffusion tensor imaging (DTI) and explore the mechanism of depression combined with cognition impairment Methods We conducted a study of 60 people: 30 patients with depression and 30 healthy controls group-matched by age, educational level andsocioeconomic status. The latency period and amplitude of P3, and the data of fractional anisotropy (FA) were measured by P300 examination and DTI, respectively, in the bilateral white matter of interested region. Correlation analysis of these 3 factors was performed in the 2 groups. At the same time, the scores of Wisconsin card sorting test (WCST) were detected. Results The WCST scores of each sub-item,and the P3 latency and amplitude between the control group and depression group had statistically significant differences (P<0.05). FA value in the white matter of the both frontal lobe, the anterior cingulate gyms, the supramarginal gyrus, splenium of the corpus callosum in the patients was significantly lower than that in the controls (P<0.05). P3 latency and percentage of persistent errors in depression patients were positively correlated (r= 0.677, P=0.009). P3 amplitude and both percentage of persistent errors and percentage of not being able to maintain a complete classification were negatively correlated, respectively (r=0.765,P=0.001; r=-0.654, P=0.012). FA values and both percentage of persistent errors and percentage of not being able to maintain a complete classification were negatively correlated in patients with depression in the bilateral frontal white matter, respectively (left: r=-0.544,P=0.003; r=0.489, P=0.023; right: r=0.665, P=0.002; r=0.448,P=0.027). Conclusions Neuropsychology and event-related potential P300 reflected the cognition impairment in patients with depression; the latency period and amplitude of P3 could be a reference index of evaluating the cognitive function. The outcome of DTI can reveal the possible abnormality of neurofibra in the white matter region, which may be one of its neuropathology in depression patients with cognition impairment.
9.Expression and clinical significance of HBsAg and HBcAg in hepatocytes in chronic hepatitis B.

OBJECTIVETo study the expression of HBsAg and HBcAg in hepatocytes in CHB patients, and analyze the correlation among the expression of HBsAg and HBcAg, the quantity of HBV DNA in serum, the pathology of liver tissue and the clinical manifestation.
METHODSQuantitative polymerase chain reaction was used to assay the quantity of HBV DNA in serum in 351 CHB patients. Furthermore pathological diagnosis was performed using liver biopsy to assay the expression of HBsAg and HBcAg in hepatocytes by an immunohistochemical staining technique.
RESULTSThe positive expression rate of HBsAg and HBcAg in hepatocytes was 92.3% and 76.9% respectively. Cytoplasm-membrane HBcAg expression type (75.6%) was observed in the CHB with more active inflammation, while Nucleus HBcAg expression type (24.4%) was observed in the CHB with more sedative one (P < 0.0001). The expression of HBsAg was correlated with the quantity of HBV DNA in serum (rp = 0.24, P = 0.0129), while inversely correlated with the inflammation and the fibrillation of liver tissue (rp = -0.22, P = 0.0279; rp = -0.23, P = 0.0186). The expression of HBcAg was correlated with the quantity of HBV DNA in serum (rp = 0.52, P < 0.0001), while was inversely correlated with the inflammation and the fibrosis of liver (rp = -0.33, P < 0.0001; rp = -0.34, P < 0.0001).
CONCLUSIONCytoplasm-membrane HBcAg expression type was observed in the CHB with more active inflammation, while Nucleus HBcAg expression type was observed in the CHB with mild change. In the immunopathogenesis of the liver damage in CHB, HBcAg might be a main target antigen. HBsAg might be a sensitive index to screen HBV infection; HBcAg might probably be a reliable index to evaluate the replication of HBV

10.Optimization of two-phase aqueous extraction for Astragali Radix by central composite design-response surface method

AIM To optimize the two-phase (ethanol-dipotassium phosphate) aqueous extraction for Astragali Radix by central composite design-response surface method.METHODS With ethanol concentration,dipotassium hydrogen phosphate solution concentration and Astragali Radix feed ratio as influencing factors,together with extraction rates of total flavonoids and total saponins as evaluation indices,the technology optimization was accomplished by central composite design-response surface method.RESULTS The optimal conditions were determined to be 27.97% for ethanol concentration,22.03% for dipotassium hydrogen phosphate solution concentration,and 1 ∶ 58.85 for Astragali Radix feed ratio.The extraction rate of total flavonoids reached 74.13% (concentrated in the upper layer),while that of total saponins reached 81.34% (concentrated in the lower layer).CONCLUSION With good predictability,this simple and accurate method can be used for the extraction of total flavonoids and total saponins in Astragali Radix.
