OBJECTIVE To prevent and control nosocomial infection(NI) among patients with hematological malignancies,and make clinical investigation.METHODS The random medical records of patients suffering from hematological malignancies in our hospital were analyzed retrospectively.RESULTS NI rate was 51.11%,in which the main infection site first was respiratory tract,next oral cavity and gastrointestinal tract.Gram-negative bacteria and fungi were the most common pathogens.CONCLUSIONS Patients with hematological malignancies are susceptible to NI,and they should be treated properly so as to prevent from infection effectively.
2.Detection of cdc6 gene in exfoliate cell in urine

Objective DNA replication increases during tumor cells limitlessly proliferate. To develop a method that can early diagnose and routine screen out bladder cancer, but it does not impair patients by detecting cdc6 in exfoliate cell in urine of patients with bladder cancer. Methods 200 ml fresh urine of patients with bladder cancer or non bladder cancer who voided urine after morning was collected and centrifugated for urine sediment, then total RNA was extracted with TRIZOL reagent from it and detected the expression of cdc6 gene by RT PCR. Results 28 samples were positive and 2 samples were negative in 30samples of bladder cancer, positive rate was 93.3%. Positive rate was 16.7% in 30 samples of non bladder cancer. To build the Dendrobium nobile -T2DM network, and elucidate the molecular mechanism of D. nobile to type 2 diabetes (T2DM). Collect the chemical composition of D. nobile and the targets on T2DM by retrieving database and documents, build the network of D. nobile to T2DM using the entity grammar systems inference rules. The molecular mechanism of D. nobile to T2DM includes: (1) regulating lipid metabolism by lowering triglyceride; (2) reducing insulin resistance; (3) protecting islet cells; (4) promoting the glucose-dependent insulin tropic peptide (GIP) secretion; (5) inhibiting calcium channel. Under the guidance of network pharmacology, through entity grammar systems inference rules we elucidate the molecular mechanism of D. nobile to T2DM, and provide the basis for the further development of health care products based on D. nobile.

4.Role of renal nerve in cardioprotection provided by renal ischemic preconditioning in anesthetized rabbits

The effects of renal ischemic preconditioning (RIP) on ischemia-reperfused myocardium were examined in the urethane-anesthetized rabbit to determine whether RIP may provide cardioprotection and to observe the role of the renal nerve in such condition. The results obtained are as follows: (1) During 45 min myocardial ischemia and subsequent 180 min reperfusion, blood pressure, heart rate and myocardial oxygen consumption decreased progressively. Epicardial electrographic ST-segment was elevated significantly in the period of ischemia and returned to the baseline gradually in the course of reperfusion. The myocardial infarct size occupied 55.80±1.25% of the area at risk. (2) RIP significantly reduced the myocardial infarct size to 36.51±2.80% (P<0.01), indicating the cardioprotective effect of such an intervention. (3) Renal nerve section (RNS) completely abolished the cardioprotection afforded by RIP, though RNS per se did not affect the myocardial infarct size produced by ischemia-reperfusion. (4) During 10 min renal ischemia, the averaged multi-unit discharge rate of the renal afferent was increased from 0.14±0.08 to 0.65±0.12 imp/s (P<0.01). (5) Pretreatment with an adenosine receptor antagonist 8-phenyltheophylline (10 mg/kg) markedly attenuated the discharge rate of the renal afferent induced by transient renal ischemia, implying that adenosine released in ischemic kidney activated the renal afferent. It is suggested that activation of renal afferents by transient renal ischemia-reperfusion plays an important role in the cardioprotection afforded by RIP.
5.Actin Scavenging System and Coagulation-fibrinolytic System in Peripheral Blood of Pre-eclampsia Patients

Objective To detect the levels of plasma gelsolin(GSN),vitamin D binding protein(VDBP)and coagulation?related parameters in peripheral blood between pre?eclampsia,then analyze their relationships with the onset or development of pre?eclampsia. Methods A total of 30 women with severe pre?eclampsia(SPE)and 30 women with mild pre?eclampsia(MPE)were enrolled for the study and randomly chosen as trial group. 30 normal pregnancies with matching gestational ages were selected as control group?1 and 30 normal full?term pregnancies were randomly chosen as control group?2. The levels of GSN,VDBP,platelet(PLT),prothrombin time(PT),thrombin time(TT),activated partial thromboplas?tin time(APTT),fibrinogen(FIB),and D?dimer were compared between those groups. Results The levels of GSN,VDBP and PLT,PT,APTT, FIB in SPE group were significantly lower than that of MPE group(P<0.05),and the level of D?dimer was significantly higher(P<0.05). In healthy pregnant women between control group?1 and control group?2,GSN fell during gestational weeks,with a concomitant rise in VDBP with no significance(P>0.05). There were no statistical difference among all the levels of coagulation?related parameters in healthy pregnant women(P>0.05). Conclusion GSN and VDBP levels in pre?eclampsia are significantly lower than that of normal pregnant women,along with pathological hy?percoagulative state,and which are related to severity of disease.

Objective To study the antitumor activity of steroid saponins,such as Rhizoma Paridis saponins,Rhizoma Paridis saponin Ⅰ,and diosgenin,extracted from Paris polyphylla var.yunnanensis against lung adenocarcinoma cells.Methods Taking mouse lung adenocarcinoma LA795 cell line and its T739-bearing mice as models,the antitumor activity of steroid saponins of P.polyphylla var.yunnanensis against lung adenocarcinoma cells was carried out in vitro and in vivo.The inhibitory activity of Rhizoma Paridis saponins,Rhizoma Paridis saponin Ⅰ,and diosgenin on the LA795 cell line was studied by means of MTT assay and their antitumor effects in vivo were carried on inbred strains of mice(T739) affected by LA795 metastatic lung cancer.Results The MTT assay of in vitro cytotoxicity of Rhizoma Paridis saponins,Rhizoma Paridis saponin Ⅰ,and diosgenin showed strong cytotoxicity against the growth of LA795 cells.IC50 Values were 24.33 ?g/mL,1.85,and 149.75 ?mol/L,respectively.The tumor weight in all treated groups was significantly lower than that in the control group and the inhibition of tumor growth were 41.82%,29.44%,and 33.94% in their high-dose groups.The metastasis of tumor cells was inhibited by different degrees in treated groups.Conclusion Steroid saponins of P.polyphylla var.yunnanensis have tumoricidal activity on lung adenocarcinoma cell line,both in vitro and in vivo.
8.Quantitative detection of minimal residual leukemia cells in peripheral blood and clinical application

Objective To explore the clinical application significance of quantitative detection of minimal residual leukemia cells in peripheral blood in acute leukemia patients.Methods Using PCR amplification of quantitative method of limited dilution,the FLT3 gene was detected in blast cells of peripheral blood from 25 newly diagnosed cases of acute leukemia,and the number of minimal residual leukemia cells in newly diagnosed cases and the cases after one course of chemotherapy was calculated respectively.10 healthy subjects were used as control group.Results ①The positive rate of FLT3 gene in 25 newly diagnosed cases of acute leukemia sample was 80%(20/25),the positive rate in acute myeloblastic leukemia(AML) patients was 88.2%(15/17)and in acute lymphocytic leukemia(ALL)was 62.5%(5/ 8).②The mean DNA content in peripheral blood in 20 cases with FLT3 positive expression in newly diagnosed group was(2.36?1.25)?108 ?g?L-1,being equal to(18.66?8.79)?106 leukemia cells in every microliter peripheral blood.After one course of chemotherapy ,there was 1 case without remission,the DNA content in peripheral blood was 1.69?107?g?L-1,being equal to 1.01?106 leukemia cells in every microliter peripheral blood;there were 3 cases with partial remission,the mean DNA content in peripheral blood was(0.57?0.24)?106 ?g?L-1,being equal to(1.82?0.19)?103 leukemia cells in every microliter peripheral blood.There were 9 cases with complete remission with FLT3 negtive expression,and 7 cases with complete remission with FLT3 positive expression,the mean DNA content in peripheral blood was(0.16?0.06)?106 ?g?L-1,being equal to(1.86?1.31)?102 leukemia cells in every microliter peripheral blood.Conclusion The quantitative and periodic detection of minimal residual leukemia cells would help to evaluate leukemia chemotherapy efficiency and to adjust treatment scheme in time.
9.Effects of aconitine on Ca2+ oscillation in cultured myocytes of neonatal rats.

In order to investigate the effects of aconitine on [Ca2+] oscillation patterns in cultured myocytes of neonatal rats, fluorescent Ca2+ indicator Fluo-4 NW and laser scanning confocal microscope (LSCM) were used to detect the real-time changes of [Ca2+] oscillation patterns in the cultured myocytes before and after aconitine (1.0 micromol/L) incubation or antiarrhythmic peptide (AAP) and aconitine co-incubation. The results showed under control conditions, [Ca2+] oscillations were irregular but relatively stable, occasionally accompanied by small calcium sparks. After incubation of the cultures with aconitine, high frequency [Ca2+] oscillations emerged in both nuclear and cytoplasmic regions, whereas typical calcium sparks disappeared and the average [Ca2+] in the cytoplasm of the cardiomyocyte did not change significantly. In AAP-treated cultures, intracellular [Ca2+] oscillation also changed, with periodic frequency, increased amplitudes and prolonged duration of calcium sparks. These patterns were not altered significantly by subsequent aconitine incubation. The basal value of [Ca2+] in nuclear region was higher than that in the cytoplasmic region. In the presence or absence of drugs, the [Ca2+] oscillated synchronously in both the nuclear and cytoplasmic regions of the same cardiomyocyte. It was concluded that although oscillating strenuously at high frequency, the average [Ca2+] in the cytoplasm of cardiomyocyte did not change significantly after aconitine incubation, compared to the controls. The observations indicate that aconitine induces the changes in [Ca2+] oscillation frequency other than the Ca2+ overload.

10.Point prevalence of healthcare-associated infection and antimicrobial use in Buyi autonomous prefecture hospitals in Guizhou Province

Objective To explore the status of healthcare-associated infection(HAI)in hospitals in Buyi autono-mous prefecture of Guizhou Povince,and provide basis for formulating HAI control measures.Methods A survey was conducted by combined methods of bed-side survey and medical record reviewing,prevalence rates of secondary and above hospitals in Buyi autonomous prefecture in Guizhou Province between September 10 and October 5,2014 were surveyed.Results 6 577 hospitalized patients should be investigated,6 541(99.45%)were actually investiga-ted.The prevalence rate and case prevalence rate of HAI were 1 .83% (n=120)and 1 .94%(n=127)respectively. The top three departments of HAI distribution were intensive care unit (26.32%),neurosurgery (6.10%),and neonatal intensive care unit(5.13%);the main infection site was lower respiratory tract(n=39,30.71 %),followed by skin-soft tissue (n=24,18.90%)and superficial incision (n=23,18.11 %).58 pathogenic isolates were detec-ted,gram-negative bacteria were the major pathogens (n=44),gram-positive bacteria and fungi were 10 and 3 iso-lates respectively.Antimicrobial usage rate at survey day was 42.12%,64.75% of which were for therapeutic, 26.83% for prophylactic,and 8.42% for therapeutic+prophylactic use;the percentage of mono-drug,two drugs combination,and three or more drugs combination use were 79.53%,19.89%,and 0.58% respectively;bacterial detection rate in patients receiving therapeutic as well as therapeutic+prophylactic antimicrobial use was 13.76%. Conclusion Survey on prevalence of HAI is helpful for understanding the current status of HAI,monitoring on HAI in key departments of hospital and key sites of patients should be strengthened to reduce the occurrence HAI effectively.
