1.Regulation of organic anion transporting polypeptides expression and activity.

Organic anion transporting polypeptides (OATP), a member of solute carrier (SLC) superfamily, is considered as an important transmembrane uptake transporters. OATP is involved in the transport of a variety of endo- and xenobiotics (bile acids, bilirubin, prostaglandin, thyroid hormones, steroid hormone conjugates), drugs and toxins in a Na+ and ATP independent manner. Multiple factors (eg. hormones, proinflammatory cytokines, drugs) can affect the distribution, expression and activity of OATPs, leading to an altered accumulation of OATP substrates and related food-drug and drug-drug interactions. Changes in the distribution and expression of OATPs in malignant tissues may be related to the pathological process of cancer, while the modulation epigenetic mechanism also contributes to its distribution patterns. This review describes the factors that can affect the expression or function of OATPs, which may provide a valuable reference for drug development and the clarification of pathogenesis.

2.Ginkgo biloba extract attenuates advanced glycation end products-induced TGF-?_1,CTGF mRNA expression and oxidative stress in NRK-49F cells

Objective To elucidate the effects of Ginkgo biloba extract on advanced glycation end products-induced TGF-?_1,CTGF mRNA expression and oxidative stress in cultured NRK-49F cells.Methods From Jan.to Jul.2005,NRK-49F cells were incubated with serum-free medium with or without 25 mmol/L glucose as control.Cells were incubated with medium containing AGEs-BSA with or without 25 mmol/L glucose for 24 h.Cells were pretreated with EGb(50,100mg?mL -1)for 24 h to evaluate effects of EGb.And then intracellular ROS generation was measured by flowcytometry,expression of TGF-?_1 and CTGF mRNA were performed by a semi-quantitative RT-PCR technique.Results Compared with control,AGEs significantly increased the expression of TGF-?_1 and CTGF mRNA and intercellular ROS generation in NRK-49F cells.Incubation with both AGEs and high glucose(25 mmol/L)can upregulate TGF-?_1 and CTGF mRNA expression and intercellular ROS generation,compared with AGEs and high glucose(25 mmol/L)respectively.Preincubation with EGb attenuated AGEs-induced TGF-?_1 and CTGF mRNA overexpression and intercellular ROS generation.Conclusion Attenuation of AGEs-induced TGF-?_1 and CTGF mRNA overexpression in renal fibroblasts through scavenging of free radical and inhibition of oxidative stress may be one of mechanisms underlying EGb in prevention and therapy of DN.

To build the Dendrobium nobile -T2DM network, and elucidate the molecular mechanism of D. nobile to type 2 diabetes (T2DM). Collect the chemical composition of D. nobile and the targets on T2DM by retrieving database and documents, build the network of D. nobile to T2DM using the entity grammar systems inference rules. The molecular mechanism of D. nobile to T2DM includes: (1) regulating lipid metabolism by lowering triglyceride; (2) reducing insulin resistance; (3) protecting islet cells; (4) promoting the glucose-dependent insulin tropic peptide (GIP) secretion; (5) inhibiting calcium channel. Under the guidance of network pharmacology, through entity grammar systems inference rules we elucidate the molecular mechanism of D. nobile to T2DM, and provide the basis for the further development of health care products based on D. nobile.

5.The analysis of the temperament types and influence factors of school-age twins

Objective To explore the distribution of temperament types and its related influencing factors of school-age twins.Method Childhood temperament were evaluated by standardized Middle Childhood Temperament Questionnaire (MCTQ) in a total of 125 pairs of 8 to 12 years old twins,and temperament related factors were measured by FACES Ⅱ-CV.Results The majority of temperament types of school-age twins were easy and intermediate-low.Easy,intermediate-low,difficult,intermediate-high and start-to-warm up took a percent of 41.6%,38.0%,11.2%,6.4% and 2.8%,respectively.The heritability of temperament types was 0.454.The distribution of temperament types were influenced by the family cohesion,parenting rearing style,zygotic,age,father's occupation,mother's educational level and the method of delivery.Conclusion The temperament types of school-age twins were both influenced by genetic and environmental factors.
6.Comparative Study of Clinical Efficacy of Ultrafine Extracted Granule Preparation and Traditional Herbal Decoction of Li Zhong Tang

Objective To compare the clinical efficacy of different doses of ultrafine extracted granule preparation (EGP) and traditional herbal decoction (THD) of Li Zhong Tang in treatment of epigastric pain. Methods Sixty cases of epigastric pain patients in accordance with the diagnostic criteria of deficiency and cold pattern of spleen and stomach in TCM were randomly divided into THD group, 1/3 dose group and 1/5 dose group, and were given THD, 1/3 dose of ultrafine EGP and 1/5 dose of ultrafine EGP, respectively. The clinical efficacy of the three groups after one course of medication was comparatively analyzed. Results There were no significant differences in age, course of disease, symptom score before treatment, epigastric pain efficacy and syndrome curative effect among the three groups, the differences had no statistical significance (P>0.05). The severity, frequency and duration of epigastric pain were all reduced in the three groups, with significant differences between before and after treatment (P<0.05). Conclusion There are no significant differences in the clinical efficacy on epigastric pain among THD, 1/3 dose of ultrafine EGP and 1/5 dose of ultrafine EGP. In addition, the effect of 1/3 dose group is very close to the THD group.
7.Effect of 5-Aza-CdR on pancreatic carcinoma Capan-2 cell proliferation and PCDH8 gene expression

Objective To discuss the effect of 5-Aza-CdR on pancreatic carcinoma Capan-2 cell proliferation and PCDH8 gene expression.Methods Pancreatic carcinoma Capan-2 cells were treated with different doses of 5-Aza-CdR with or without gemcitabine,negative control group without drug,0.08μmol/L group with 0.08μmol/L 5-Aza-CdR,0.40 μmol/L group with 0.40 μmol/L 5-Aza-CdR,2.00 μmol/L group with 2.00 μmol/L 5-Aza-CdR,10 μmol/L group with 10 μmol/L 5-Aza-CdR,50 μmol/L group with 50 μmol/L 5-Aza-CdR. The inhibition ratio of Capan-2 cell proliferation were observed by MTT assay and PCDH8 gene and protein expression were detected by RT-PCR and Western blot. Results The inhibition ratio was increased with 5-Aza-CdR dose increasing(P<0.01),but decreased apparently with times extending(P<0.01). Inhibition ratio in 5-Aza-CdR and gemcitabine group was higher than those with only 5-Aza-CdR or gemcitabine groups(P<0.05 or P<0.01).The levels of PCDH8 gene and protein expression were increased significantly in 5-Aza-CdR treatment groups,with dose dependent (P <0.01 ).Conclusion 5-Aza-CdR can inhibit the proliferation of pancreatic carcinoma Capan-2 cell proliferation,and increase PCDH8 gene expression. The inhibition effect is strong when combined with gemcitabine.
8.Ultrasonic manifestations of heptic mesenchymal hamartoma in children

Objective To describe the sonographic appearance of mesenchymal hamartoma of the liver(MHL)and to analyze the diagnostic value of ultrasound.Methods Eleven surgically and pathologically confirmed cases of MHL from January 2005 to May 2011 in the Beijing Children′s Hospital were retrospectively reviewed.Results Ultrasound examinations showed 9 cystic hamartomas,including 7 multiseptate cystic and 2 monocystic lesions.Of the 7 multiseptate cystic hamartomas,4 had a honeycomb appearance and 3 had irregularly-distributed multiple cysts with varied septations.Of the 2 monocystic hamartomas,1 had a large cystic portion while the other was mainly solid with approximately 4% cystic portion of the tumor.Two cases in this group were solid,presenting with a well-defined homogenous mass.Conclusions Ultrasonography is an effective imaging modality for the diagnosis for MHL.A mixed or a mainly-cystic liver mass found in a child less than 2 years old should be suspicious for MHL.
9.Quantitative detection of minimal residual leukemia cells in peripheral blood and clinical application

Objective To explore the clinical application significance of quantitative detection of minimal residual leukemia cells in peripheral blood in acute leukemia patients.Methods Using PCR amplification of quantitative method of limited dilution,the FLT3 gene was detected in blast cells of peripheral blood from 25 newly diagnosed cases of acute leukemia,and the number of minimal residual leukemia cells in newly diagnosed cases and the cases after one course of chemotherapy was calculated respectively.10 healthy subjects were used as control group.Results ①The positive rate of FLT3 gene in 25 newly diagnosed cases of acute leukemia sample was 80%(20/25),the positive rate in acute myeloblastic leukemia(AML) patients was 88.2%(15/17)and in acute lymphocytic leukemia(ALL)was 62.5%(5/ 8).②The mean DNA content in peripheral blood in 20 cases with FLT3 positive expression in newly diagnosed group was(2.36?1.25)?108 ?g?L-1,being equal to(18.66?8.79)?106 leukemia cells in every microliter peripheral blood.After one course of chemotherapy ,there was 1 case without remission,the DNA content in peripheral blood was 1.69?107?g?L-1,being equal to 1.01?106 leukemia cells in every microliter peripheral blood;there were 3 cases with partial remission,the mean DNA content in peripheral blood was(0.57?0.24)?106 ?g?L-1,being equal to(1.82?0.19)?103 leukemia cells in every microliter peripheral blood.There were 9 cases with complete remission with FLT3 negtive expression,and 7 cases with complete remission with FLT3 positive expression,the mean DNA content in peripheral blood was(0.16?0.06)?106 ?g?L-1,being equal to(1.86?1.31)?102 leukemia cells in every microliter peripheral blood.Conclusion The quantitative and periodic detection of minimal residual leukemia cells would help to evaluate leukemia chemotherapy efficiency and to adjust treatment scheme in time.
10.Role of Interleukin-10 in the Maintenance of Hemodialysis in Patients With Immune Deficiency

Objective: To determine the role of interleukin-10 (IL-10) secreted by peripheral blood mononuclear cells (PBMC) in the maintenance of hemodialysis in patients with immune deficiency and to evaluate thot if IL-10 can bea biocompatibility index of dialyzer. Methods: Twenty patients with end-stage renal disease were divided into maintenance hemodialysis group (MHD group) and non-hemodialysis group (NHD group). Patients in MHD group were dialyzed with cellulose (CA group) and polysulfone (PS group) dialyzers respectively. Ten healthy persons were as controls (CON group). The levels of IL-10 in supernatants of PBMC cultured with or without lipopolysaccharide (LPS) were measured by using ELISA method in all groups. Results: There was no spontanous IL-10 secretion in all groups. The level of IL-10 in all supernatants secreted by PBMC cultured with LPS increased significantly, and the increased levels of IL-10 in MHD and NHD patients were lower than that in controls. The PBMC stimulated by LPS secreted more IL-10 in patients dialyzed by PS with better biocompatibility than those of CA dialyzers with poor biocompatibility. Conclusion: Patients immune deficiency could be improved by using better biocompatible dialyzers more than poor ones, particularly in humoral immunity induced by urimia. The IL-10 could be a parameter in evaluating the biocompatibility of dialyzer.
