Objective To study the expression of GPC3 mRNA and GPC3 protein in ovarian carcinoma, and to investigate the clinical significance of their expression. Methods 35 specimens of ovarian carcinomas and 23 specimens of normal ovarian tissues were obtained from the patients undergoing operation during Jan. 2004 to Oct. 2005. The expressions of GPC3 mRNA in ovarian carcinomas and normal ovarian tissues were detected by real-time fluorescence quantitative PCR (RT-PCR). The clinical and pathological characteristics of ovarian carcinomas were analyzed. The expression of GPC3 protein was detected in 4 specimens of ovarian carcinomas and 3 specimens of normal ovarian tissues by Western bloting. Results The expression level of GPC3 mRNA in ovarian carcinomatous was down-regulated compared with that in normal ovarian tissues (P0.05). The expression of GPC3 protein was consistent with the expression of GPC3 mRNA. GPC3 protein was doubly or trebly expressed in normal ovarian tissues compared with ovarian carcinomas. Conclusion The present study suggests that GPC3 may be related to the occurrence and development of the ovarian carcinoma. The simultaneous determination of GPC3 and CA125 may increase the sensitivity of diagnosis for ovarian carcinoma.

Background Flicker light can induce myopia, but its mechanism remains unclear. As one of immediate early genes, early growth response-1 (Egr-1) gene can generate rapid response to visual stimulation, however, its effect on the formation and development of myopia is below understood. Objective This study was to investigate the dynamic expression of Egr-1 gene in retinas of flicker light-induced eyes (FL) and compare the results with form deprived eyes (FD).Methods One hundred and fifty 28-day-old C57BL/6J mice were randomly assigned to the normal control group,FD group and FL group. The right eyes of mice were occluded with a semitransparent hemispherical thin plastic shell for 2 weeks in the FD group, and the right eyes of mice were stimulated by 2 Hz flicker light for 2 weeks in the FL group, and then the mice were fed in the normal light environment for 1 week. The refractive state and axial length of the model eyes were measured by murine-specific eccentric infrared photorefraction and A-scan ultrasonography before modeling and 1 hour,I day,1 week,2 weeks after modeling as well as 1 week after termination, respectively. The mice were sacrificed in above-mentioned time points to isolate the retinas. The expressions and location of Egr-1 protein and mRNA in the retinas were detected by Western blot, and reverse transcription PCR (RT-PCR) and immunochemistry. The expressions of Egr-1 markers, neuron and protein kinase C (PKC)-α,in the retinas were assayed by using immunofluorescence. The care and use of the animals followed the administration regulations for experimental animals of Jiangsu Province. Results Two weeks after modeling, the refraction of the FL group was (0.32±0.14) D,which was significantly lower than (-0.66±0.43)D in the FD group (t=6.78,P=0.00).One hour after modeling,The expression levels of Egr-1 mRNA in mouse retinas were 0.626±0.044 and 0.695±0.058 in the FD group and FL group, which were significantly declined in comparison with 1.009±0.089 of the normal group (t=14.81,P=0.01;t=9.15,P=0.03).In 2 weeks after modeling, the expression levels of Egr-1 mRNA were still lower in the FD group and F:L group compared with the normal group (all at P<0.05).However,the expression levels were significantly elevated in the FD group and FL group compared with the normal group (t=4.13,P=0.01;t=4.26,P=0.01) at 1 week after termination.Western blot showed a dynamic decrease in the expressions of Egr-1 protein with lapse of time in the FD group and FL group with the lowest expressing level in the second week after modeling. In I week after termination of modeling, the expressing level was raised in the FD group or the FL group, but it was still lower than that ir the normal group (t =6.32,P=0.00;t =5.45,P=0.01).Egr-1 protein was mainly expressed in the retinal ganglion cell (RGC) layer, inner nuclear layer and photoreceptor layer in the normal mice, and the expression intensity was obviously weaker in the FD mice and FL mice 2 weeks after modeling.Htowever, the expression was enhanced in 1 week after termination of modeling.Neuron and PKC-α were strongly expressed in the RGCs and bipolar cells in the normal mice. Conclusions The eyes show a myopic trend after induce of flicker light in B6 mice. The expression level of Egr-1 gene in the retina down-regulates with the reduce of refraction in FL eyes, and its dynamic expressing change is consistent between the FD eyes and FL eyes.

Objective:To analyze bone strength index, osteoporosis and fracture in ankylosing spondylitis (AS) patients with radiologic hip involvement and explore the characteristics of bone strength in these patients. Methods:According to bath ankylosing spondylitis radiology hip index (BASRI-hip) score, 339 AS patients were divided into two groups. The differences of bone strength in each group were analyzed by t-test, Mann-Whitney U test and χ2 test. Logistic regression was used to analyze the risk factors of bone strength index. The correlation between quantitative ultrasound and dualenergy X-ray absorptiometry (DXA) (total hip, g/cm 2) was analyzed. Pearson correlation analysis was used. Results:①Bone strength index of quantitative ultrasound was positively correlated with bone mineral density of DXA (total hip, g/cm 2), r=0.553, P<0.01. ② The age of 27(23, 37) years old in radiographic hip involvement was lower than 37(28, 48) years old in non-radiographic hip involvement, and the difference was statistically significant ( Z=-5.986, P<0.01). There were no differences in gender and course of disease between the two groups ( P>0.05).③ The radiographic hip involvement in AS patients whose ages were younger than 50, when compared with non-radiographic hip involvement patients, the bone strength index was lower (78±18 vs 84±16), while bone strength was lower than patients at the same age (41.0% vs 18.4%), however, the incidences of osteoporosis (42.7% vs 28.8%) and fragility fracture (3.7% vs 0%) were significantly higher ( t=3.028, P<0.01; χ2=16.758, P<0.01; χ2=5.886, P=0.019; χ2=4.67, P=0.038). For AS patients whose ages were ≥50, there were no significant differences between the two groups ( P>0.05). ④ Multivariate analysis showed that radiographic hip involvement [ OR (95% CI)=1.912(1.05, 3.48)], age [ OR (95% CI)=0.94(0.911, 0.97)] and body mass index (BMI) (kg/m 2) [ OR (95% CI) =0.875(0.807, 0.948)] were associated with lower bone strength. Conclusion:There is positive correlation between the bone strength index of quantitative ultrasound and bone mineral density of DXA. AS patients with radiographic hip involvement are characterized by decreased bone strength and are more likely to develop osteoporosis and fragile fractures. The risk factors of low bone strength are radiographic hip involvement, age and BMI.

Journal of Huazhong University of Science and Technology (Medical Sciences) 2012;32(5):746-9
This study examined the effect of electro-acupuncture (EA) combined with transcranial magnetic stimulation (TMS) therapy at different time windows on learning and memory ability of rats with cerebral infarction and the underlying mechanism. Two hundred SD rats were randomly divided into four groups: normal group, sham-operated group, model group and EA+TMS group, and each group was then divided into five sub-groups in terms of the different time to start treatment post operation: 6, 12, 24, 48 and 72 h. Cerebral infarction models were established in the model and the EA+TMS groups by left middle cerebral artery occlusion/reperfusion (MCAO/R). After treatment for 14 d, the Morris water maze test was applied to examine the spatial learning and memory abilities of rats. In infarcted area, the expression of caspase-3 was immunohistochemically detected, and real-time fluorescent quantitative PCR was used to measure the expression of Bcl-2 mRNA. The results showed that in EA+TMS group compared with model group at the same treatment time windows, the escape latency was substantially shortened, the expression of caspase-3 was considerably decreased and the expression level of Bcl-2 mRNA significantly increased (P<0.05). In the EA+TMS sub-groups, the escape latency was shortest, the expression level of caspase-3 lowest, and the expression level of Bcl-2 mRNA highest at the treatment time window of 24 h. It was concluded that EA combined with TMS can promote neurological function of rats with cerebral infarction by increasing the expression level of Bcl-2 mRNA and decreasing the expression of caspase-3. The best time window is 24 h after perfusion treatment to ischemia.
