2.Effects of the prescription of reinforcing kidney, nourishing blood, improving eyesight on ARPE - 19 cells induced by acrolein

?AlM: To explore the effects of the prescription of reinforcing kidney, nourishing blood, improving eyesight on the oxidative stress model of ARPE-19 cells induced by acrolein.
?METHODS:SD rats serum containing the prescription of reinforcing kidney, nourishing blood, improving eyesight and the content of distilled water in serum were prepared. The effects of the prescription and distilled water in serum at different concentration ( 2. 5%, 5%, 10%, 20% and 40%) on cell vitality was observed by cell counting kit ( CCK-8 ) assay. the logarithmic phase of ARPE-19 cells were pretreated by different concentrations (1. 25%, 2. 5%and 5%) of the prescription serum and distilled water in serum for 24h. Then it was treated with 75μmol/L acrolein for 24h. Cell vitality was observed by CCK-8 assay. The change of cell nucleus was detected by DAPl staining .
?RESULTS: 2. 5% and 5% serum had no effect on cell viability (P>0. 05), while 10%, 20%, 40% serum could inhibit cell viability (P<0. 05). CCK-8 results showed that 2. 5% and 5% the prescription of reinforcing kidney, nourishing blood, improving eyesight serum was better than distilled water in serum (P<0. 05).
?CONCLUSlON: The prescription of reinforcing kidney, nourishing blood, improving eyesight has the protective effect on ARPE-19 cell damage induced by acrolein.

4.Transcriptome analysis of bioenergy plant Miscanthus sinensis Anderss by RNA-Seq.

Miscanthus sinensis Anderss is a perennial C4-grass. It is a promising bioenergy plant, which has been proposed as general feedstock for biomass and lignocellulosic biofuel production. In this study, the flower and leaf buds transcriptomes of Miscanthus sinensis Anderss were sequenced by the platform of Illumina HiSeq 2000. In total 98 326 Unigenes were generated by de novo assembly with an average length of 822 bp and N50 of 1 023 bp. Based on the NR, NT, Swiss-Prot, KEGG, GO and COG databases (Evalue < le-5), 74 134 (75.40%) Unigenes were annotated. A total of 45 507 Unigenes were mapped into different GO terms. In KEGG pathways identification, 36 710 sequences were assigned to 128 KEGG pathways. Sorghum bicolor (37 731, 60.86%), Zea mays (16 258, 26.22%), and Oryza sativa (3 065, 4.94%) showed high similarity to Miscanthus sinensis Anderss. And 24 photosynthesis-related enzyme genes were identified. The result provides a foundation for further characterizing the functional genes in Miscanthus sinensis Anderss.

Objective To explore the therapy measurement of iron deficiency anemia(IDA)associated helicobacter pylori(HP)infection among children.Methods Eight hundred and ninty nine cases entrusted children aged from 2 to 7 years old were measured hemoglobin(Hb),mean corpuscular volume(MCV)mean corpuscular hemoglobin(MCH),mean corpuscular hemoglobin concentration(MCHC)and serum ferri,serum fettin(SF),HP-IgG antibodies.Divided the HP-IgG positive cases into two groups for treatment,The treatment group adopted omeprazole,amoxicillin and metronidazole to cure them for two weeks,then oral ferrous sulfate sugar liquid.The contrast group purely take the ferrous sulfate sugar liquid.After eight weeks re-examined the Hb and SF.Results Totally 279 cases were diagnosed as Hp-positive,620 as HP-negative and 99 children diagnosed as IDA,among the 99 cases IDA,58 cases with HP infection(20.8%) and 41 cases with non-HP infection(6.7%).That means the number of IDA cases with HP infection was higher than those with non-HP infection(?2=39.46 P
6.A retrospective study of antiviral therapy in hospitalized children with infectious mononueleosis

Objective:To investigate the efficacy of antiviral therapy and its selection in hospitalized children with infectious mononucleosis (IM).Methods:The IM children admitted to Shenzhen Children’s Hospital from January 2017 to December 2021 were divided into acyclovir (ACV) treatment group and non-antiviral treatment group, and the clinical data were collected for retrospective analysis.Results:A total of 370 children were included, the median age was 3.6 years, each age group proportion: <3 years (25.9%), 3-<6 years (53.0%). 6-<10 years (17.6%) and ≥10 years (3.5%). Male∶Female=1.5∶1. While 42.4% (157/370) of the 370 children received ACV, 57.6% (213/370) did not receive antiviral therapy; 52 cases were treated in infectious disease department, and none of them received antiviral treatment, and 318 cases were treated in non-infectious disease department, of which 157 cases were in ACV group. There was significant difference between the two groups ( P <0.001); Thirty-eight cases of children in non-infectious disease department complicated with abnormal liver function, of which 23 cases (60.5%) were in the ACV group and 15 cases (39.5%) were in the non-antiviral treatment group. There was no significant difference between the two groups ( P=0.060). Compared with the non-antiviral treatment group, the ACV group has longer hospitalization days and higher hospitalization expenses ( P<0.01). There was no significant difference in duration of fever ( P>0.05). The number of atypical lymphocytes recovered faster in ACV group ( P=0.001). Conclusions:The hospitalized children with IM were mostly boys, and most of them were from 3 to 6 years old. ACV treatment could not shorten the duration of fever and hospitalization days of IM children, nor could it reduce the hospitalization expenses. Abnormal liver function did not affect the decision of antiviral treatment, and infectious disease specialists prefer not to use antiviral drugs. The authors advocate reducing the use of antiviral drugs for IM.
7.Comparison of surgical thrombectomy and interventional thrombectomy for acute deep vein thrombosis of the lower extremity.

OBJECTIVETo study the short- and long-term results for acute deep vein thrombosis (DVT) of the lower extremity treated by surgical thrombectomy and interventional thrombectomy.
METHODSOne hundred and thirty-six acute DVT cases treated by surgical thrombectomy or interventional thrombectomy from March 2000 to August 2008 were reviewed. There were 80 patients treated by surgical thrombectomy. Among them, 30 cases were male, 50 cases were female, aged from 26 to 81 years with a mean of (58 ± 14) years. The other 56 cases were treated by interventional thrombectomy. Among them, 25 cases were male, 31 cases were female, aged from 22 to 92 years with a mean of (57 ± 17) years. All the 136 patients received district anticoagulation with heparin and thrombolysis with urokinase after operation.
RESULTSAfter operation, the circumference difference between bilateral thigh in intervention group were less than that in surgical group [(0.8 ± 1.3) cm vs. (1.5 ± 1.7) cm, P = 0.002]. The circumference difference of bilateral calf had no significant difference [(0.7 ± 1.1) cm vs. (1.0 ± 1.1) cm, P = 0.152]. The average hospital stay in intervention group was shorter than that in surgical group [(7 ± 4) d vs. (15 ± 7) d, P = 0.000]. The morbidity of complications in intervention group was less than that in surgical group (8.9% vs. 32.5%, P = 0.000). One hundred and eight patients were followed up, who was 79.4% of the total patients, and were followed up for an average of (46 ± 29) months. The circumference difference of thigh and calf, the symptom grade, the pigmentation, varicose veins, intermittent claudication and ulceration between the two groups had no significant difference (P > 0.05).
CONCLUSIONCompared with surgical group, intervention group has a better short-term effect, shorter hospital stays, less complications and similar long-term result.

8.Optimation of the Fermentation Conditions for Human ApolipoproteinA-I Expression

Optimization of the fermentation condition for human apolipoproteinA-I expression in recombinant Escherichia coli was investigated. The recombinant plasmid pBV220-ApoA-I was transformed respectively into different E.coli hosts such as JM109, BL21(DE3),DH5?, BMH7118,and TG1. The best host E.coli was DH5? in which the recombinant ApoA-I expression percentage was 21.2% corresponding to that in BL21(DE3) in flask shaker cultivation,while the ApoA-I expressed percentage in E.coli TG1 was 11%.Fed-batch cultivation was performed in FMG-5L fermentor,the optimum fermentation cultivation conditions were as following :optimum pH value was 7.0 in growth phase and 7.4 in the expression phase. The initial glucose concentration in batch phase was 3 g?L -1.The optimum C/N ratio was 2∶1.The recombinant ApoA-I reached about 40% of the total protein, and concentration of ApoA-I was 2.86 g?L -1.

The temperature effect on the recombinant protein production formation was investigated in present study. The culture temperature of growth phase is 30℃, and the culture temperature of induction phase was arranged according to three modes. Hign cell-density and high expression culture of E.coli to product recombinant human apolipoprotein A-I Milano by two temperature-shifted induction . Two temperature-shifted induction was carried out high density and high expression recombinant human ApoA-1 Milano. The recombinant protein ApoA-I Milano reached 4.8 g?L -1 with the final cell density of OD 600 150. And the two temperature-shifted induction avoided the acetic acid successfully to the influence of the high density and high expression. Two temperature-shifted induction was viable in high density culture and high expression of heterogenous protein in recombination E.coli.The sduty provides a basic work for production of recombinant ApoA-I Milano in scale.

Objective:To observe therapeutic effect of small dose spironolactone combined valsartan and amlodipine on obese patients with hypertension and its influence on serum levels of visfatin and leptin.Methods:A total of 106 obese patients with hypertension treated in our hospital from Mar 2016 to Mar 2017 were selected.They were ran-domly and equally divided into routine treatment group(received valsartan,amlodipine and routine treatment)and combined treatment group(received small dose spironolactone based on routine treatment group),both groups were continuously treated for eight weeks.Systolic blood pressure(SBP),diastolic blood pressure(DBP),body mass in-dex(BMI),waist circumference(WC)and serum levels of aldosterone,visfatin and leptin before and after treat-ment were measured and compared between two groups.Results:Compared with before treatment,there were sig-nificant reductions in levels of SBP,DBP,serum aldosterone,visfatin and leptin in two groups after eight-week treatment,P=0.001 all;compared with routine treatment group after eight-week treatment,there were significant reductions in levels of SBP[(138.67 ± 9.34)mmHg vs.(127.52 ± 8.74)mmHg],DBP[(93.27 ± 8.26)mmHg vs. (86.43 ± 7.98)mmHg],serum aldosterone[(155.73 ± 16.43)ng/L vs.(121.49 ± 13.76)ng/L],visfatin[(28.49± 4.13)ng/ml vs.(22.31 ± 3.64)ng/ml]and leptin[(15.67 ± 4.82)ng/ml vs.(12.43 ± 3.74)ng/ml]in combined treatment group,P=0.001 all.Total effective rate of combined treatment group was significantly higher than that of routine treatment group(96.23% vs.83.02%),P=0.026. There was no significant difference in incidence rate of adverse reactions(11.32% vs.7.55%)between two groups,P=0.740. Conclusion:Small dose spironolactone combined valsartan and amlodipine possesses significant therapeutic effect on obese patients with hypertension,and it can reduce their serum levels of visfatin and leptin,which is worth extending.
