1.Research advances in molecular prediction of recurrence and metastasis of primary liver cancer after liver transplan-tation

Liver transplantation is an essential treatment of primary liver cancer,but the postoperative high risk of recurrence and metastasis has seriously affected the effect of liver transplantation.With the development of molecular biology,researchers have found numerous molec-ular markers potentially related to the recurrence and metastasis of liver cancer after liver transplantation.These markers,including genes, proteins,tumor markers,and serum viruses,are summarized,and their predictive values for tumor recurrence and metastasis after liver transplantation are analyzed.A growing number of molecular markers have been found,but which is not enough for clinical guidance.There-fore,more studies are needed to identify appropriate molecular markers of tumor recurrence and metastasis after liver transplantation.
2.A survey of the status quo of pediatric dentists in China,2003

Objective:In 2003,a survey of the manpower of pediatric dentists was made in China in order to provide basis for database. Methods:Questionnaires were distributed to dentists presenced the 5th conference of pediatric dentistry in China,and 139 available answers were analysis. Results:In those 139 pediatric dentists, with a mean age of 38.2?8.7,females were significantly more than males, and 89.9% dentists had bachelor, master or doctor degree. Significant relations were found between self-estimate ability of dentists and their length of work, degree, character of clinic.Conclusion:To date, the team of pediatric dentists begins to take shape, the structure of age and education are rational, but the number of them is insufficient and it is necessary to improve the basic and continuing education.
3.Expression of chemokine CXCL16 in murine collagen-induced arthritis and the effects on the proliferation of lymphocytes

Objective To investigate the effect of CXCL16 on the development of murine collageninduced arthritis (CIA). Methods CXCL16 mRNA of the involved synovium and serum CXCL16 protein were determined respectively by reverse transcription-polymerase chain reaction (RT-PCR) and enzyme linked immunosorbent assay (ELISA) in murine collagen-induced arthritis. The proliferation of lymphocytes from murine spleen and the level of RANKL mRNA, stimulated by CXCL16 at different concentrations (0,100, 200, 400, 800 ng/ml), was detected respectively by CCK-8 and RT-PCR, then the level of IL-2 and IFN-γ in culture supernatant was detected by ELISA. Comparisons between groups were tested by t test and one-way ANOVA analysis. Results The serum CXCL16 [(127± 10) vs (72±8) pg/ml, P<0.05] and synovial CXCL16 mRNA (0.214±0.007 vs 0.375±0.009, P<0.01) in CIA were all significantly higher than those in normal controls. The proliferation of CXCL16 (200, 400, 800 ng/ml) in CIA mouse lymphocytes, was significantly higher than that of CXCL16 (0 ng/ml) (0.51±0.06, 0.56±0.05, 0.55±0.04, (0.41±0.04, P<0.05). And CXCL16 on the CIA stimulated lymphocyte proliferation was significantly higher than controls on normal lymphocytes (P<0.05). Compared with blank control group, the expression of IL-2, IFN-γ and RANKL mRNA of CIA CXCL16 (400, 800 ng/ml) groups was higher significantly (P<0.01). Conclusion CXCL16 plays an important role in the development of murine CIA by activating lymphocytes.
4.Effect of Jiawei Wendan decoction regulating CaMK/MAPK signal pathway in hippocampus of depression model rats

Objective To explore the effect of Jiawei Wendan decoction on whole regulating function of CaMK/MAPK signal pathway in hippocampus of depression model rats.Methods Isolated depression model rats as research object were living unpredictable chronic stress.Based on the examination of mRNA/protein expression which was the key factor of the signal pathway of CaMK/MAPK and the intersection CREB1 mRNA expression,the relationship between CaMK Ⅱ/RSK protein expression and the CREB1 mRNA expression were analyzed with relation analysis method.Results ① Bidirectional correlation analysis:the respective coefficient (r value) of CaMK Ⅱ protein expression and the CREB1mRNA expression in hippocampus of the model group,Chinese medicine treatment group and western medicine treatnent group were-0.502 (P < 0.01),-0.643 (P < 0.01),-0.408 (P< 0.05) ;the respective coefficient(r value) of RSK protein expression and the CREB1 mRNA expression in hippocampus of the model group,Chinese medicine treatment group and western medicine treatment group were 0.550 (P < 0.01),0.687 (P < 0.0 l),0.407 (P < 0.01).②Regression analysis:the respective regression coefficient of CaMK Ⅱ protein (positive direction),RSK protein (negative direction) and CREB1 mRNA expression in the model group,Chinese nedicine treatment group and western medicine treatment group were R 2 =0.472,F =12.983(P<0.01),R2=0.666,F=28.961(P<0.01),R2=0.356,F=8.004(P<0.01).Conclusion The anti-depressant effect mechanism of Jiawei Wendan decoction is to regulate the whole function of the CaMK and MAPK signal pathway and then further up-regulate CREB1 mRNA expression.

6.Detection of cancer stem cell like circulating tumor cells in peripheral blood of 45 patients with gastric cancer

Objective To explore the expression and significance of CD44 gastric cancer stem cell biomarker positive circulating tumor cells (CTC ) in peripheral blood of patients with gastric cancer . Methods From March 2010 to December 2012 ,45 patients with gastric cancer and 20 healthy individuals were enrolled .Peripheral blood samples of all the subjects were collected .The expression of CTC and CD44 postive CTC were detected by density gradient centrifugation method and double immunofluorescent staining .Chi‐square test and t‐test were performed for statistical analysis .Results CTC in peripheral blood of healthy controls were not detected .Among 45 patients with gastric cancer ,CTC were detected in 27 patients and 19 of them were CD44‐positive .The difference in positive rate of CTC expression was statistically significant between two groups (χ2 =10 .986 , P<0 .05) .Among 27 CTC‐positive patients , 19 were proximal gastric cancer and 16 of them were CD44‐positive;16 patients had lymph node metastasis and 14 of them were CD44‐positive;13 patients had distant metastasis and 12 of them were CD44‐positive;14 patients had recurrence and 13 of them were CD44‐positive .The CD44 positive CTC was correlated with tumor location ,lymph node metastasis ,distant metastasis and recurrence (χ2=5 .891 ,5 .527 ,5 .787 and 7 .052 ,respectively ,all P<0 .05) .Furthermore ,in 13 patients with recurrence and CD44 positive CTC ,the recurrence time was (10 .54 ± 5 .55) months which was shorter than that of one patients with CD44 negative CTC (19 months) ,and the difference was statistically significant (t= -3 .654 , P=0 .004) .Conclusions CTC which express cancer stem cell marker may be associated with recurrence and metastasis of gastric cancer .It has more important clinical significance to detect this subtype of CTC .
7.The Experimental Study on Dual Antiplatelet Drugs-Induced Intestinal Damage in Rats

Objective To investigate the effect of dual antiplatelet drugs on the intestinal damage in rats. Meth-ods Eighty SD rats were randomly allocated into four groups:control group (normal saline, n=20), aspirin group (10.41 mg/kg, n=20), clopidogrel group (7.81 mg/kg, n=20) and clopidogrel combined aspirin group (n=20). Each group was given intra-gastric administration of drugs once per day for 14 days. All rats received operation after the final intragastric administration. The intestinal injury was observed. The jejunal fluid was taken for bacterial culture. The intestinal permeability was detected by ethidium bromide (EB) method. The small intestinal mucosal injury was estimated by Chiu method. Results The differ-ent degrees of small intestinal mucosal injury were found in four groups. The scores of pathological lesions were significantly higher in aspirin group, clopidogrel group and clopidogrel combined aspirin group than those of control group (P<0.05). The dual antiplatelet group showed the highest score (3.450±1.356). The small intestinal permeability was significantly increased in experiment groups compared with that of control group (54.012±3.513μg/g, P<0.05). There was a significantly higher small intestinal permeability in dual antiplatelet group than that of aspirin group and clopidogrel group (μg/g:130.533 ± 29.631 vs 90.965±3.765 vs 66.800±4.853, P<0.001). The total jejunal bacteria was significantly increased in dual antiplate-let group than that of control group (CFU/mL:61924.805±1751.159 vs 18154.280±1153.376, P<0.001). The ratio of entero-bacterium and enterococcus was significantly decreased in dual antiplatelet group compared with that of control group (0.220±0.089 vs 1.007±0.148, P<0.001). Conclusion The routine dose of dual antiplatelet drug aggravates the small intes-tinal injury in rats compared with that of single drug. The manifestations of intestinal mucosal injury include increased intes-tinal bacteria, dysbacteriosis, and increased small intestinal permeability.
8.Detection and clinical significance of circulating tumor cells in pa-tients with gastric cancer

Objective: To detect circulating tumor cells (CTCs) in patients with gastric cancer and evaluate the relationship among CTCs, clinico-pathological characteristics, and prognosis of gastric cancer. Methods: Peripheral blood samples (10 mL in EDTA) were obtained from 45 patients with gastric cancer. CTCs were detected using density-gradient centrifugation and immunofluo-rescence staining. The clinical significance of the two methods were also compared and investigated. Results:CTC-positive case was defined by the presence of at least one CK19 (+)-CTC per 10 mL of the sample. CTCs were found in 27 of the 45 patients with gastric cancer. The presence of CTCs was significantly correlated with lymph node metastasis, distant metastasis, and recurrence (P=0.007, 0.035, 0.035, respectively). However, CTCs were not significantly correlated with sex, age, tumor location, TNM staging, and tumor differentiation (P>0.05). Conclusion:CTCs were associated with poor prognosis of gastric cancer.
9.Effects of hypoxia on circulating tumor cells in gastric adenocarcinoma in vivo

Objective To explore the correlation between the change of hypoxic in microenvironment and the genesis of circulating tumor cells (CTCs) in gastric adenocarcinoma.Methods After human gastric adenocarcinomn BGC-823 cells treated with different concentration of CoCl2 (0,100,200,300,400 μmol/L)for four hours,the expression levels of hypoxia inducible factor 1 alpha (HIF-1α)were detected by Western blot to select the optimal concentration.A total of 40 BALB/c mice were divided into two equal groups,named hypoxic group and normoxic group.Optimal concentration of CoCl2 treated BGC-823 gastric cancer cells and non hypoxic induced BGC 823 gastric cancer cells were injected through the tail vein.After three days blood samples were collected through the angular vein.CTCs were enriched by density gradient centrifugation,identified with cytokeratin19 (CK19) antibody marker by cell immunofluorescence and cell number was counted.Statistical significace test were performed by analysis of variance and Dunnett-T test.Results The expression of HIF-1α reached a maximun of 1.40±-0.16 when BGC-823 cells were treated with 200 μmol/L CoCl2 for four hours.The number of CTCs in hypoxic group was higher than that of normoxic group ((4.70± 1.72)/high power field (HPF) vs (1.15±0.56)/HPF,Z=-4.824,P<0.01).Conclusion Hypoxic microenvironment might promote the genesis of CTC in gastric adenocarcinoma.
10.Analysis of the clincal chracteristics of non-steroidal anti-inflammatory drugs caused gastrointestinal bleeding in the elderly patients

Objective To investigate the clinical features of non-steroidal anti-inflammatory drugs (NSAIDs) caused gastrointestinal bleeding in the elderly.Methods We performed a retrospective analysis ofNSAIDs caused gastrointestinal bleeding with 308 cases admitted to Department of Gastroenterology,The Second Hospital of Tianjin Medical University.Patients were divided into elderly group(≥65 years,n =208) and young group (< 60 years,n =100) to make a comparative analysis of two group differences in clinical characteristics.Results The NSAIDs in the elderly taking were mainly Aspirin (55.8 % (116/208)),which was significantly more frequent than that of young group (37.0% (37/100),x2 =9.517,P < 0.05).There was no significant difference on the blood routine and coagulation between the two groups (P > 0.05).The recent bleeding rate of the elderly group (21.2% (44/208)) was lower than the young group (63.0% (63/100))(x2 =52.161,P <0.05).The two groups had no significant difference on the history of uleer and Hp infection (P > 0.05).Conclusion The clinical characteristics of NSAIDs caused gastrointestinal bleeding on the elderly were different with young or middle-aged.This infers that elderly patients need individualized treatment.
