1.Transcription level of mitochondria gene Cyt b in different stages of Pagumogonimus skr j abini

ABSTRACT:In this study ,we aimed to analyze the transcription level of mitochondria gene Cytb in metacercariae ,larva‐30d ,larva‐60d ,adult ,and egg of Pagumogonimus skrjabini .The mRNA of metacercariae ,larva‐30d ,larva‐60d ,adult and egg of P .skrjabini were extracted with genomic extraction kit ,and transcripted reversely into cDNA .With 18SrDNA of Par‐agonimus westermani as an internal standard primer ,Cytb genes were amplified by real‐time PCR for establishing standard curves to evaluate the copy number of genes .Those quantitative analyses were reliable because the R value of standard curves was 0 .994 (>0 .98) ,and the melting curve showed a single peak .There were increasing trend of transcription of Cytb gene at metacercariae stage ,larva‐30d stage ,and larva‐60d stage .There were less transcription of Cytb gene at adult stage and no transcription at egg stage .There were differences about the transcriptional level of mitochondria Cytb gene at different stages of Pagumogonimus skrjabini ,and peaked at larva‐60d stage ,suggesting that Cytb gene may play a role in the development and migrating of larva .
2.Effects of different frequencies of pricking-bloodletting at auricular points plus auricular point sticking therapy on the serum levels of related factors in patients with acne vulgaris

Objective: To observe the effects of different frequencies of pricking-bloodletting at auricular points plus auricular point sticking therapy on the levels of the serum immunoglobulin G (IgG), immunoglobulin M (IgM), testosterone (T), and insulin-like growth factor-1 (IGF-1) in patients with acne vulgaris (AV). Methods: Ninety patients with AV were randomly assigned to treatment group 1, treatment group 2, and treatment group 3 according to the random number remainder grouping method, with 30 cases in each group. All three groups received pricking-bloodletting at auricular points plus auricular point sticking therapy. Treatment group 1 was treated once a week, treatment group 2 was treated twice a week, and treatment group 3 was treated 3 times a week. Four-week treatment was taken as 1 treatment course, and 3 treatment courses were observed. On the day before the start of the study and on the day next to the end of each course, the global acne grading system (GAGS) score was recorded, and 3 mL of blood from the median cubital vein was collected to test the serum levels of IgG, IgM, T, and IGF-1. After 3 courses of treatment, the efficacy index was calculated according to the GAGS score, and the serum indicators in patients with effective treatment (efficacy index ≥20%) and complete sampling were analyzed. Results: A total of 60 patients were included in the final blood indicator analysis. After 3 courses of treatment, the intra-group comparisons showed that the serum IgG level increased significantly in patients in treatment group 1 (P<0.01); the serum T level decreased significantly in the female patients in treatment group 2 (P<0.05); the IGF-1 level significantly decreased in the patients in all three groups (P<0.05 or P<0.01). There were no significant differences in the serum levels of IgG, IgM, T, and IGF-1 among the three groups at each time point (P>0.05). Conclusion: Pricking-bloodletting at auricular points plus auricular point sticking therapy can affect the levels of serum IgG, T, and IGF-1 in AV patients. The level of the serum IGF-1 can be reduced by treatment once a week, twice a week, or 3 times a week. Treatment once a week can increase the patients' serum IgG level, and treatment twice a week can significantly decrease the serum T level in female patients. Reducing the serum IGF-1 level may be one of the action mechanisms of pricking-bloodletting at auricular points plus auricular point sticking therapy in treating AV.
3.A Clinical Analysis of Spontaneous Intracerebral Hemorrhage.

We have experienced 40 cases of spontaneous intracerebral hemorrhage admitted to the Chung Ang University Hospital. We analized the result of the treatment of spontaneous intracerebral hemorrhage and assessed prognostic factors affecting the result of treatment of intracerebral hemorrhage. 1) The age distribution was ranged from 26 to 69 and 67.5% of them were in fourth and fifth decade. The ratio of male to female was about 5:3 and the age of the patients did not significantly influence on the result of operative or non-operative cases(P:0.22 P>0.05). 2) The predilection sites of hemorrhage were basal ganglia(60%) and thalamus(15%). 3) There was a correlation between the conscious status on admission and that of two weeks after admission, not so strong, in the operated cases. I.e., the higher Glasgow Coma Score on admission, the better results two weeks after admission. But such a correlation was not found in the non-operated cases(P:0.5732 P<0.01, P:0.1581 P>0.05). The hemorrhage in the dominant hemisphere had little correlation with the outcome(P:0.5068 P<0.01). 4) The pre or post-operative conscious status appeared to have no correlation with the hematoma volume. The large hematoma not always lead to poor prognosis when it was removed in proper time unless the secondary change of the hematoma did not develop(P:0.4257 P<0.05, P:0.4644 P<0.05). 5) The blood pressure on admission, not his proper pressure, had no correlation with the result two weeks after admission in the operated or non-operated cases(P:4988 P<0.01, P:0.4120 P<0.05). 6) The midline shift on the carotid angiogram had a closed correlation with the conscious status of the operated or non-operated cases in spite of little influence of hematoma volume to the conscious state(2:0.7350 P<0.01, P:0.5377 P<0.01).

4.Intracranial Arachnoid Cyst: Case Report.

Intracranial arachnoid cysts are uncommon congenital lesions of considerable interest and importance, the origins and exact nature of which remain uncertain. They account for about 1% of all intracranial space-occupying lesions4). Its predilection sites are in the sylvian fissure, the interhemispheral fissure, the cerebral convexity, the base of the brain, and over the midline of the cerebellum in the posterior fossa6)11). We are reporting a case of arachnoid cyst, developed in the left sylvian fissure in a ten years old male patient.

5.A Case of a Recurrent Postoperative Discitis.

The etiology of the postoperative discitis is not fully known, but most likely, the cause seems to be of a low grade infection and chemical and mechanical injuries. The incidence of these complication is from 0.12% to 2.6% of all cases. The characteristics of this disease are recurrence of severe lower back and gluteal pain, spasm of lumbar paravertebral muscle and elevated E.S.R. following the asymptomatic post-operative period, a roentgenogram showed an early destructive change in the adjacent vertebra, the narrowing of the intervertebral disc space, sclerotic changes in the vertebral bone and occasional fusion of the spine, and relatively good prognosis. We present a case of recurrent postoperative discitis with E.S.R. change and roentgenographic change.

6.Primary Osteosarcoma of the Skull.

Osteosarcoma is the most frequently encountered primary malignant tumor of the bone. But sarcoma of the skull are rare tumor to be observed even in an extensive neuropathological practice. We presents a case of skull neoplasm identified as osteosarcoma. A twelve year old male patient was admitted to our department because of the large palpable mass on the posterior parietal and occipital region with headache, nausea and vomiting. On admission moderate papilledema was noted in the both eyes. Other neurologic finding were within normal limits. Chest film and long bone series showed no evidence of abnormality. Skull films revealed a large round destructive area on the posterior parietal and occipital region and the right carotid angiogram revealed patch densities in the mass lesion which was supplied by the right occipital artery and possibly the right middle meningeal artery. Serum Alkaline Phosphatase was 14 BL Units. The findings on the pathology side was compatible with osteosarcoma.

7.Effect of dexamethasone on peroxisome proliferator activated receptor-gamma mRNA expression in 3T3-L1 adipocytes with the human recombinant adiponectin.

BACKGROUNDThe fat derived protein adiponectin plays an important role in the regulation of glucose metabolism. The aim of this study was to provide the experimental basis for further investigating on adiponectin (ADPN) function. Its eukaryotic recombinant was constructed and expressed in precursor cells of 3T3-L1 adipocytes. The effects of dexamethasone on peroxisome proliferator activated receptor-gamma (PPAR-gamma) mRNA expression in 3T3-L1 cells with human recombinant adiponectin were assessed.
METHODSThe recombinant plasmid pMD18-T-hADPN and eukaryotic expression vector pcDNA3.1(+) were digested by two restrictive endonucleases and adiponectin and linear pcDNA3.1(+) were obtained. Then, they were ligated and translated into JM109. The recombinant pcDNA3.1(+)-hADPN so obtained was identified by digestion by restrictive endonuclease and nucleotide sequencing. The 3T3-L1 precursor cells were transfected using SuperFect Transfection Reagent (Qiagen). Furthermore, 3T3-L1 cells with human recombinant adiponectin incubated with dexamethasone (0.5 mmol/L) for 24 hours, cells were collected and total RNA was extracted. The PPAR-gamma mRNA expression was quantified by semiquantitative reverse transcription-polymerase chain reaction (RT-PCR).
RESULTSAfter eukaryotic recombinant was digested by Hind III and EcoR I, fragments of 800 bp and 5.4 kb were identified by nucleotide sequence scanning and consistent with theoretical values. Electrophoretogram of RT-PCR in 3T3-L1 precursors showed only one band in front of 250 bp, which was consistent with theoretical value 234 bp. In the 3T3-L1 cells, 3T3-L1 cells with plasmid and 3T3-L1 cells human recombinant adiponectin, treatment with dexamethasone (0.5 mmol/L) decreased PPAR-gamma mRNA expression compared to untreated controls (P < 0.01). Effect of dexamethasone on PPAR-gamma mRNA expression in 3T3-L1 cells was reversed by stably transfected human recombinant adiponectin.
CONCLUSIONThe 3T3-L1 cells stably transfected human recombinant adiponectin had increased PPAR-gamma mRNA expression. Dexamethasone suppressed PPAR-gamma mRNA expression in the 3T3-L1 cells. Effect of dexamethasone on PPAR-gamma mRNA expression in 3T3-L1 cells was reversed by stably transfected human recombinant adiponectin.

8.Comparison of dopamine levels in the lateral geniculate nucleus in guinea pigs with flickering light?induced and form deprivation myopia

Objective To observe the dopamine(DA)concentrations in the lateral geniculate nucleus in guinea pigs with flickering light-induced and form deprivation myopia,and to compare and analyze the pathogenetic mechanisms in the centers of vision of these two different myopia models. Methods Twenty-four two-week-old guinea pigs were randomly assigned to three groups(n=8):flickering light-induced myopia(FLM)group, form deprivation myopia(FDM)group and control group. All the groups were fed for 8 weeks. The refraction and axial length(AL)were measured before and af-ter modeling. After eight-week modeling,the contents of dopamine in the left lateral geniculate nucleus were detected by high performance liquid chromatography with electrochemical detection(HPLC-ECD). Results Before modeling,no sig-nificant difference was found in refraction and AL among the three groups. After eight-week modeling,in contrast with the control group,significant differences were found in changes of both refraction(P<0.001)and AL(P<0.05)in the right eyes of FLM and FDM groups,indicating that the two myopic models were successfully established. The result of HPLC-ECD showed that the contents of DA in the left lateral geniculate nucleus in FLM group were significantly higher than the control group(P=0.01),while in the FDM group it was lower than the control group(P=0.021). The average contents of DA were as follows:(37.04 ± 1.18)pg/μL in the control group;(24.27 ± 3.46)pg/μL in the FDM group; and (45.58 ± 1.98)pg/μL in the FLM group. Conclusions The content of DA in the lateral geniculate nucleus is increased in FLM,while decreased in FDM,indicating that the expression of DA in LGN and the mechanisms of formation of these two experimental myopia are different.
9.Changes in the vessel densities of macular and optic nerve head and their relationships with axial length in pathological myopia

Objective:To observe the changes of vessel densities (VD) in the macula and optic disc and its correlation with axial length (AL) in pathological myopia (PM).Methods:A retrospective clinical study. A total of 171 eyes from 171 patients admitted to Department of Ophthalmology of Jinshan Hospital of Fudan University from June 2019 to December 2019 were included in this study. Among them, there were 72 males and 99 females; age was 35.0±10.8 years old. The patients were divided into PM group, high myopia (HM) group and non-HM group, 51 cases with 51 eyes, 70 cases with 70 eyes, and 50 cases with 50 eyes, respectively. Optical coherence tomography angiography was used to scan the macular and optic disc areas of all the examined eyes in the range of 6 mm×6 mm. According to the early treatment of diabetic retinopathy study, the 6 mm macular and optic disc scan range was centered on the macular fovea and optic disc, respectively, then divided into two concentric circles with diameters of 1 mm of central area, an annulus between 1-3 mm circles of paracentral area. The paracentral area was divided into superior, inferior, nasal, temporal four quadrants by 2 radiation lines. The VD of superficial capillary plexus (SCP), deep capillary plexus (DCP), outer retina, and choriocapillaris layer were calculated in the central, superior, inferior, nasal, and temporal areas, respectively. The VD of PM, HM and non-HM groups were compared. The variance analysis was used to compare the VD among the three groups; Pearson’s correlation was used to assess the correlation between VD and AL.Results:The perifoveal VD of the SCP, outer retina and choriocapillaris layers were all lower in the PM than those of HM and non-HM group, and the differences were statistically significant ( P<0.05). The VD of DCP macular central was higher in the PM than in the HM group, and the difference was statistically significant ( P=0.020). In the optic disc, the VD were lower in the PM group than in the non-HM group except for the area of DCP superior, inferior, temporal, outer retinal center, and the differences were statistically significant ( P<0.05). The results of correlation analysis showed that the VD in the DCP macular central, ONH superior and the choriocapillaris ONH central were not correlated with AL ( P=0.647, 0.688, 0.146), and the other VDs were negatively correlated with AL ( P<0.05). Conclusion:Compared with HM and non-HM groups, the majority of VDs in macular and ONH are lower in participants with PM.
10.Correlation between anxiety among public hospital medical personnel and social support, perceived social support and coping styles

Objective This research aimed to explore the mental status of public hospital medical personnel and social support for them, social support they perceive, and their coping styles. This study was designed to investigate on the relationship between their anxiety and social support, perceived social support and their coping styles. Methods A questionnaire was adopted to collect data, including participants’ background, measurement of anxiety, as well as social support, perceived social support and their coping styles. Results(1)The anxiety level of medical personnel was significantly higher than that of domestic norm 37.23±12.59(t=14.370, P<0.001);(2)Anxiety levels appeared to be significantly different in occupation, educational background and working years;(3)Results shown in the three scales were different among participants with different levels of anxiety in perspectives of every dimension in the scales; (4) With the help of Spearman correlation analysis, it showed that anxiety levels of medical personnel was negatively correlated with objective support (r=-0.098, P=0.051), subjective support (r=-0.189, P<0.001) ,utilization of social support (r=-0.164, P=0.001), friend support (r=-0.356, P<0.001),family support (r=-0.330, P<0.001),other support (r=-0.238, P<0.001) and positive response (r=-0.282, P<0.001), and positively correlated with negative response (r=0.385, P<0.001). Conclusions Mental health of medical personnel should be noticed and enhanced, and social support for public hospital medical personnel should be strengthened. Thus, the performance of the medical team can be developed in order to support further public medical service construction.
