1.Antioxidant properties and inhibitory effects of Satureja khozestanica essential oil on LDL oxidation induced-CuSO(4) in vitro.

OBJECTIVETo assess various antioxidative activities of Satureja khozestanica essential oil (SKE) and its effect on oxidation of low density lipoprotein (LDL) induced by CuSO4 in vitro by monitoring the formation of conjugated dienes and malondialdehyde (MDA).
METHODSThe formation of conjugated dienes, lag time and MDA were measured. Inhibition of this Cu-induced oxidation was studied in the presence of several concentrations of SKE. Also total antioxidant activity and free radical scavenging of SKE were determinated.
RESULTSIt was demonstrated that SKE was able to inhibit LDL oxidation and decrease the resistance of LDL against oxidation. The inhibitory effects of SKE on LDL oxidation were dose-dependent at concentrations ranging from 50 to 200 µg/mL. Total antioxidant capacity of SKE was (3.20±0.40) nmol of ascorbic acid equivalents/g SKE. The SKE showed remarkable scavenging activity on 2, 2-diphenyl-picrylhydrazyl, IC50 (5.30±0.11) ng/mL.
CONCLUSIONSThis study shows that SKE is a source of potent antioxidants and prevents the oxidation of LDL in vitro and it may be suitable for use in food and pharmaceutical applications.

2.2-Arylbenzofuran derivatives from Morus wittiorum.

The investigation on the stem bark of Morus wittiorum was carried out to find its chemical constituents possessing anti-oxidative activity. The isolation and purification were performed by various chromatographies such as silica gel, Sephadex LH-20, RP-C18 column chromatography and so on. Based on the spectral analysis such as NMR, MS, etc., seven 2-arylbenzofuran derivatives were identified as wittifuran D (1), wittifuran E (2), moracin C (3), moracin M (4), moracin P (5), 2-(3,5-dihydroxyphenyl)-5,6-dihydroxybenzofuran (6) and mulberroside C (7). Compounds 1-7 were isolated from this plant for the first time. Among them, 1 and 2 were new compounds. Compounds 3-7 were used to assay antioxidant activity, the inhibitory ratios of compounds 3, 4, 6, at a concentration of 1 x 10(-5) mol x L(-1), to malondialdehyde (MDA) produced during microsomal lipid peroxidation induced by ferrous-cysteine were 73%, 69% and 89% respectively.

3.Effect of noise on antioxidant capacity of brain tissue in guinea pigs.

OBJECTIVETo investigate the effect of noise on the antioxidant capacity in different regions of brain tissue in guinea pigs.
METHODSThirty male white red-eye guinea pigs were equally and randomly divided into five groups: 1-, 3-, 7-, and 14-day groups after noise exposure and control group. The guinea pigs of the experimental groups were exposed to steady white noise with a sound pressure level at 100 dB for 8 h per day and for 2 consecutive days. The auditory brainstem response (ABR) of guinea pigs, as well as the glutathione (GSH) level, methane dicarboxylic aldehyde (MDA) level, and superoxide dismutase (SOD) activity in the cerebrum, cerebellum, and hippocampus, was determined prior to and 1, 3, 7, and 14 days after noise exposure.
RESULTSAfter noise exposure, the shifts in ABR threshold of the experimental groups were significantly higher than that of the control group (P<0.05). Compared with those in the control group, the SOD activity and GSH level both significantly decreased in the cerebrum tissue of each experimental group after noise exposure (P<0.05) and MDA content significantly increased in the 1-day group (P<0.05). As for cerebellum tissue, the SOD activity and GSH level in the 7-day group were significantly lower than those in the control group (P<0.05), but there was no difference in MDA level between each experimental group and the control group (P>0.05). In comparison with those in the control group, the GSH and MDA levels in the 1-day group after noise exposure were significantly higher, and the GSH and MDA levels in the 3-day group and the MDA level in the 7-day group after noise exposure were significantly lower (all P<0.05).
CONCLUSIONNoise exposure can lead to hearing loss and affect the antioxidant capacity of brain tissue, which indicates that the improvement in antioxidant levels may alleviate noise-induced damage.

4.Effect of in ovo ghrelin administration on serum malondialdehyde level in newly-hatched chickens.

OBJECTIVETo investigate effects of in ovo ghrelin administration on serum malondialdehyde (MDA) level in newly-hatched chickens.
METHODSFertilized eggs were divided into 7 groups: group T1 as control (without injection), group T2 (in ovo injected with 50 ng/egg ghrelin on day 5), group T3 (in ovo injected with 100 ng/egg ghrelin on day 5), group T4 (in ovo injected with 50 ng/egg ghrelin on day 10), group T5 (in ovo injected with 100 ng/egg ghrelin on day 10), group T6 (in ovo injected with solvent: 1% acetic acid, without ghrelin on day 5) and group T7 (in ovo injected with solvent without ghrelin on day 10). After hatching, serum MDA concentrations were determined.
RESULTSGhrelin administrated groups (T2, T3, T4 and T5) had lower serum MDA level in comparison with control group (T1) or solvent injected groups (T6 and T7). T2 and T3 (ghrelin injection on day 5) had significantly lower MDA concentrations (4.10 and 4.60 nmol/mL, respectively) in comparison with other groups. In T4 and T5, MDA levels were lower than T1, T6 and T7 (non-ghrelin administrated groups) (9.53 and 9.50 in comparison with 10.73, 10.03 and 10.13 nmol/mL) and were higher than T2 and T3.
CONCLUSIONSIt can be concluded that in ovo administration of ghrelin can have anti-oxidative protection and reduce serum MDA level. Ghrelin administration on day 5 of incubation is more efficient.

5.Level and effect factors of superoxide dismutase and malondialchehyche of the old people aged 90 and over in longevity regions, China.

OBJECTIVETo explore the level and effect factors of superoxide dismutase (SOD) and malondialchelyche (MDA) of the people aged 90 years old and above in longevity regions in China.
METHODSFrom Xiayi county (Henan province), Zhongxiang county (Hubei province), Mayang county (Hunan province), Foshan county Sanshui district (Guangdong province) and Yongfu county (Guangxi province), the people of hundred-year were selected and numbered. The old people aged 90 - 99 according to the age and sex were appointed by the centenarians' code and 496 people were selected in total. Questionnaire investigation was conducted on nationality, education, nutrition, smoking, alcohol drinking, tea drinking and physical activities.436 blood sampling (4 ml Per sampling) were conducted and the level of triglyceride (TG), SOD, MDA, Ca, Mg, and some microelements were determined. Non-conditional logistic regression analysis was conducted to explore the effect factors of SOD and MDA in plasma.
RESULTSAnalysis showed that the activity of plasma SOD was (31.19 +/- 5.51) U/ml, and the content of MDA was (4.68 +/- 1.98) micromol/L. The SOD level of minority people of 90 years old and over was (27.55 +/- 4.97) U/ml, lower than that of Han nationality ((31.73 +/- 5.39) U/ml; t = 5.85, P < 0.01); the MDA level of minority people of 90 years old and over was (5.58 +/- 1.95) micromol/L, higher than that of Han nationality ((4.54 +/- 1.95) micromol/L; t = -3.94, P < 0.01). logistic regression showed, comparing with the Han nationality, the minority had lower level of SOD (OR = 0.493), and higher level of MDA (OR = 5.412, P < 0.05). Beside nationality, the factors significantly affecting the activity of SOD included the intake frequency of meat, intake frequency of eggs and plasma concentration of Se (OR values were 0.388, 2.260 and 1.906, P < 0.05). The factors significantly affecting the level of MDA included the intake frequency of milk, physical activity and the level of TG (OR values were 0.435, 1.850 and 1.647, P < 0.05).
CONCLUSIONThe level of SOD of the oldest-old people aged 90 years old and over is relatively higher than the younger ones, and the level of MDA is relatively lower in longevity region, China. The effect factors of SOD are nationality, meat intake frequency, eggs intake frequency and the level of Se in plasma. The effect factors of MDA are nationality, milk intake frequency, physical activity and the level of TG in plasma.

6.Effects of fungi fraction on growth and anti-oxidative activity of Eleutherococcus senticosus.

The present study was conducted to explore the effect of endophytic fungi fraction on growth and anti-oxidative activity of Eleutherococcus senticosus. The growth,yield,contents of MDA,and antioxidant activities were assessed in E. senticosus under five fungi fractions,namely BZ,MH,DT,JS,and XFZ. The results showed that fungi fractions and component significantly affected the growth,low concentration of DT fungi fraction significantly increased the biomass of E. senticosus,reduced the MDA content in cells,and the antioxidant activities of the aqueous extracts were superior to the others. The results indicated that low concentration of DT fungi fraction was the optimum fraction to achieve high yield and quality of E. senticosus.

7.Effect of a simulated He-O2 saturation dive to 65 msw on oxidative stress in humans.

OBJECTIVETo investigate whether a simulated He-O2 saturation dive to 65 msw would affect oxidative balance in humans.
METHODSSeven divers participated in a simulated saturation dive to 0.75 MPa (65 msw). 24-h urine samples were collected twice before, twice during, and twice after the dive, then were analyzed for contents of superoxide dismutase (SOD), malondialdehyde (MDA), total amino acid (T-AA) and total anti-oxidant capacity (T-AOC). Meanwhile, total urine volume and body weight were measured.
RESULTSThe content of T-AA was higher. (P < 0.05) than the base value in final decompression, but reverse to normal at one week after decompression. There were no changes in contents of SOD, MDA and T-AOC during and after the dive compared with their basic value. Total urine volume was lower (P < 0.05, vs basic value) at first day in chamber, then returned to normal. Body weight gradually increased after compression till the end of decompression (higher than basic value, P < 0.05).
CONCLUSIONThese data indicate that simulated saturation dive to 65 msw may not induce obvious oxidative damage, but it is necessary to monitor 24-h urine volume and oxidative sress by time in order to prevent from tissue injury.

9.Effects of moxa smoke with different concentrations on expression of SOD and MDA in lung and serum of rats.

OBJECTIVETo observe the effects of intervention of moxa smoke with different concentrations on superoxide dismutase (SOD) and malondialdehyde (MDA) in serum and lung of male rats, so as to explore the safety concentration of moxa smoke.
METHODSA total of 32 Wistar male rats were randomly divided into a control group, a low-concentration group, a moderate-concentration group and a high-concentration group, 8 rats in each one. All the rats were exposed in the full-automatic toxicant exposure cabinet, and the overshadow of moxa smoke was set at 0%, 10%, 40% and 70%, respectively. Each rat was exposed for 20 min per day. After 26 weeks, the activities of SOD and content of MDA in serum, lung organ and bronchoalveolar lavage fluid were tested.
RESULTSCompared with the control group, the activities of serum SOD in the high-concentration group were reduced (P< 0. 05), but those in the low-concentration group and moderate-concentration group were not significantly different (both P>0. 05). Compared with the control group, the content of serum MDA in the low-concentration group, moderate-concentration group and high-concentration group was increased insignificantly (all P>0. 05). There were no significant differences regarding activities of SOD and content of MDA in lung organ and bronchoalveolar lavage fluid among each moxa smoke group (all P>0. 05).
CONCLUSIONThere is no obvious toxic reaction in the low-concentration group and moderate-concentration group; in the high-concentration group the antioxidant ability is damaged due to long-term exposure.

10.Structural modification of cyclovirobuxine D and their inhibition activity on lipid peroxidation.

AIMTo search for compounds through structural modification of cyclovirobuxine D, using 20 or 21-aminosteroids as lead compound for the treatment of cerebrovascular diseases with better lipid peroxidation inhibitory activity.
METHODSAccording to rational drug design principle, a series of cyclovirobuxine D analogues were prepared, and their bioactivities were tested.
RESULTSFour new compounds were obtained and confirmed by spectra.
CONCLUSIONLipid peroxidation inhibitory effects of cyclovirobuxine D analogues were tested by using TBA method. Some compounds showed better activity than that of cyclovirobuxine D.
