3.Levels of serum interleukin-6 and interlenkin-18 in newly diagnosed type 2 diabetes mellitus

Objective To explore the levels of serum interleukin-6 and interleukin-18 in newly diagnosed type 2 diabetes mellitus (T2DM) and investigate their roles. Methods The serum interleukin-6 and interleukin-18 levels were detected in 50 newly, diagnosed T2DM patients (T2DM group) and 40 normal subjects (control group). Fasting blood glucose, fasting insulin, blood lipid, body mass index (BMI) and waist/hip ratio (WHR) were measured. HOMA-IR and HOMA-β were calculated. Results The serum interleukin-6 and interleukin-18 levels in T2DM group were significantly higher than those in control group [(2.56 ± 1.09) ng/L vs (1.81±0.80) ng/L, (5.38±0.91) ng/L vs (4.62±0.59)ng/L] (P <0.01).Interleukin-6 level was positively correlated with HbA_1c,while interleukin-18 level was positively correlated with BMI and WHR. Interleukin-6 and interleukin-18 levels were positively correlated. Interleukin-6 and interhukin-18 could predicted the insulin resistance. Conclusion Interleukin-6 and interleukin-18 take part in the development of T2DM, and can be related to the insulin resistance.
4.An improved method for isolation of mouse islets and islet transplantation

Objectives To study of the method for isolation and transplantation of mouse islets. Methods The method for isolation of mouse islets described by Gotoh G was modified. The solution of digestion was injected not through the common bile duct but through gallbladder. Soybean trypsin inhibitor and BSA were added into the digestive and Ficoll separation solutions. Results The yield of islets was increased from 41.7±13.2 to 266.5±32.1(P<0.01). Islet viability was more than 95!%. Among the purified islets, there was no exocrine tissue but few ductal fragments. Conclusions By the improved method, digestive solution could be injected into pancreas without inverted microscope, which made manipulation easier and more successful. Having avoided the digestive effect of trypsin on islets, the yield of islets was increased and good repetitiousness was obtained.
5.Preparation of monoclonal antibody to human augmenter of liver regeneration:screening of hybridomas with unpurified antigen expressed by E.coli

Objective:To prepare monoclonal antibody(McAb) against human augmenter of liver regeneration(hALR) by hybridoma technique with unpurified recombinant protein expressed by E.coli as screening antigen.Methods:BALB/c mice were immunized with hALR thioredoxin(hALR Thi) fusion protein twice.The splenocytes were fused with myeloma cells by polyethylene glycol and the hybridomas were selected in HAT medium.The positive hybridomas secreting anti hALR McAb were screened by means of ELISA,with the lysates of E.coli containing the recombinant plasmid pQE30 hALR induced by IPTG as screening antigen and containing plasmid pQE30 as controlling antigen.The reactivity of McAb secreted by the positive hybridoma to hALR expressed in eukaryocyte and native hALR in human serum was confirmed by means of ELISA and Western blot methods.Results:A positive hybridoma was screened successfully;the specific antigen antibody reaction was confirmed between the McAb prepared and hALR expressed in eukaryocyte and native hALR in human serum.Conclusion:Without any purification procedure,recombinant protein expressed by E.coli can be used for screening bybridomas in preparing of McAb with the expressed product of empty plasmid as control;anti hALR McAb can be used to the further study of hALR.

7.Comparison of mechanisms and cellular uptake of cell-penetrating peptide on different cell lines.

Cell-penetrating peptide (CPP) can be used in pharmaceutics as a highly efficient drug delivery transporter. In this study, four tumor cell lines (MCF-7, MDA-MB-231, C6, and B16F10) were used to observe the uptake of fluorescein isothiocyanate (FITC) labeled CPP and the effects of time and concentration of CPP on cell penetration was studied. The CPP exocytosis on C6 cell line was observed, and its exocytosis kinetics was described by zero order equation. In addition, low-temperature condition (4 degrees C) and endocytosis inhibitors were utilized to investigate the mechanism of CPP uptake by cells. Low-temperature condition did not show significantly inhibition on CPP uptake. Heparin, a membrane glycoprotein receptor inhibitor, showed strong inhibition effect (only 3%-10% of the control) on CPP uptake. Chlorpromazine, chloroquine and 5-(N-ethyl-N-isopropyl)-amiloride (EIPA) showed little effect on CPP uptake. This study indicated that CPP penetration had little selectivity on cell type, but the amount and rate of CPP penetration into cells were related to the type of cell lines. The adsorption of CPP on cell membrane induced by sulfate proteoglycan plays an important role on CPP penetration.

9.Significance of clinical pathology in diagnosis and treatment of autoimmune liver diseases

Liver biopsy plays an important role in the diagnosis of autoimmune liver diseases,particularly in patients without specific clinical manifestations or negative for autoantibodies.The histological features of several autoimmune liver diseases are reviewed.The characteristic features of autoimmune hepatitis in histology include interface hepatitis with lymphoplasmacytic infiltration,hepatic rosette formation,and emperipolesis,while chronic nonsuppurative destructive cholangitis and epithelioid granuloma are frequently found in primary biliary cirrho-sis.Primary sclerosing cholangitis is characterized by onion skin-like periductal fibrosis,while storiform fibrosis with IgG4-positive plas-ma cell infiltration often indicates IgG4 -related sclerosing cholangitis.Enhanced communication between clinicians and pathologists can help improve the diagnosis and treatment of autoimmune liver diseoses.
10.Bronchial asthma and autophagy
